This study was to investigate the effect of neurotrophic factors on neural cell differentiation in vitro derived from human embryonic stem (hES, MB03) cells. For neural progenitor cell formation derived from hES cells, we produced embryoid bodies (EB: for 5 days, without mitogen) from hES cells and then neurospheres (for 7 - 10 days, 20 ng/㎖ of bFGF added N2 medium) from EB. And then finally for the differentiation into mature neuron cells, neural progenitor cells were cultured in ⅰ) N2 medium (without bFGF), ⅱ) N2 supplemented with brain derived neurotrophic factor (BDNF, 5ng/㎖) or ⅲ) N2 supplemented with platelet derived growth factor-bb (PDGF-bb, 20ng/㎖) for 2 weeks. In fast grown softwood, there are very large changes in material properties going outward from the pith to bark such as anatomical, physical and mechanical characteristics. Some of variations in anatomical properties with annual ring were then examined from Pinus koraiensis, Larix leptolepis, and Chamaecyparis obtusa, which are major softwoods of plantation in Korea. The large variations of annual ring width during young age of tree tended to stabilize after 25year through the transitional period in 17~23year. The ring density was 1.5~2.4 in 1~10year period, and 3.5~6.3 in 30~35year period, in which juvenile and mature wood were certainly assumed to be formed, respectively. Variations of tracheid length showed functional relationships with annual rings as logarithm. Demarcation between juvenile wood and mature wood could be 16~19year, which was determined from increase rate of tracheid length of 0.2%. Cell wall thickness increased with increase of annual ring even though large variations were observed as well. Variations of cell wall thickness within species were pronounced in latewood than earlywood. The increase of cell wall thickness from juvenile wood to mature wood was predominant in Larix leptolepis as 2.0times, and least in Chamaecyparis obtusa as 1.1 times. Cell diameters showed trends of increase during young age of 1~15year, and consistent afterward. The variations of cell diameter between radial and tangential direction were greater in latewood, and most pronounced in Chamaecyparis obtusa.
The present paper is a histological study of neurosecretory cells in the brain and the thoracic ganglion with the gonadal development in Palaemon serrifer. The reproductive cycle includes the successive stages of the growing period (February-March), the mature period(April-May), the ripe and spent periods(June-August) and the degenerative and resting periods(September-January). The neurosecretory cells are grouped into four types based on Matsumoto(1958) : A-,A'-, B- and E-cells. A- and A'-cells are $80-90{\mu}m,\;B-cell\;is\;30-40{\mu}m$ and E-cell is $10-15{\mu}m$. A- and B-cells are the positive to CHP and AF, while B-cell is the positive only to AF. The secretory grannules of a A-cell are transported to the axon, and at the same time they are discharged through the peripheral membrane. Of the four neurosecretory cells, A- and I-cells show the difference of secretory activity according to the gonad developmental process. In the female, A-cells show secretory activity for the ripe and spent periods, while I-cells show for the mature, ripe and spent periods. In the male, A-cells show secretory activity for the mature, ripe and spent periods, while I-cells show for the growing, mature, ripe and spent periods.
Cell and intercelullar space enlarged during maturation. The cell of soft persimmon was separated from each other. The degradation of middle lamella exhibited in mature persimmon and small vesicle appeared in cytoplasm of turning and mature persimmon. The middle lamella and cell wall were degraded during softening.
Alteration of molecular markers related to apoptosis of in vivo normal system and in vitro cancerous system by soy-isoflavonoid with estrogen was investigated. Down-regulation of Bcl-2 was accompanied by decreased expression of COX-2 (cyclooxygenase-2) in mature female rats treated with soy-isoflavonoid and estrogen. In ovariectomized rat system, Bax was regulated by higher concentration of soy treatment. Bax up-regulation by soy-isoflavonoid genistein treatment was observed in MCF-7 mammary cancer cell system. Estrogen without soy induced similar pattern of Bax expression as soy-isoflavonoid in vivo, but exhibited opposite trend in vitro. These findings suggest soy-isoflavonoid may have potential to induce apoptosis at higher concentrations through up-regulation of Bax or down-regulation of Bcl-2 expressions depending on normal or cancerous state, and physiological status of rats.
This study was carried out to determine the effect of cumulus cell attachment and various factors on in vitro maturation of pig foflicular oocytes. Oocytes with various configuration of cumulus cell mass were collected ftom ovaries of mature gilts by asperating with syringe equipped with needles of different gauges, follicle size and with or without cumulus cells. They were cultured in TCM-199 mediun containing FGS(fetal calf serum) for 30~48 hours in incubator with air containing 5% $CO_2$ at 38.5$^{\circ}C$. Mter orcein staining at in vitro maturation condition, GV, GVBD, anaphase, telophase and M II were observed. Results are surumarized as follows: 1. Recovery rates were 55.8, 55.5 and 34.4% when the cumulus-compacted oocytes were collected with 18, 21, 26 gauge needles of syringes, respectively. 2. 79% of oocytes with compacted cumulus cells were at GV stage and most of the oocytes with partially denuded and denuded cumulus cells were from GVBD to M- II stages. 3. Percentage of mature oocytes among those which are follicular diameter of 1~2, 3~6 and over 6 mm was 42.6, 53.2 and 60.8%, respectively. 4. Percentage of mature oocytes among those which are compacted, partially denuded and denuded was 60.5, 46.2 and 35.4% respectively. 5. Percentage of mature oocytes in co-cultured with monolayers of cumulus cells was higher (57.1%) than that found with oocytes cultured alone (53.4%).
Matured dendritic cells (DCs) begin migration with their release from the bone marrow (BM) into the blood and subsequent traffic into peripheral lymphoid and non-lymphoid tissues. Throughout this long movement, migrating DCs must apply specialized skills to reach their target destination. Non-invasive in vivo cell-tracking techniques are necessary to advance immune cell-based therapies. In this study, we used a DiD cell-tracking solution for in vivo dendritic cell tracking in naive mice. We tracked DiD (non-invasive fluorescence dye)-labeled mature dendritic cells using the Near Infrared (NIR) imaging system in normal mice. We examined the immunophenotype of DiD-labeled cells compared with non-labelled mature DCs, and obtained time-serial images of NIR-DC trafficking after mouse footpad injection. In conclusion, we confirmed that DiD-labeled DCs migrated into the popliteal lymph node 24 h after the footpad injection. Here, these data suggested that the cell tracking system with the stable fluorescence dye DiD was useful as a cell tracking tool to advance dendritic cell-based immunotherapy.
The RNA genome of poliovirus encodes a long polyprotein precursor and this polyprotein is cleaved proteolytically by viral protease to yield mature proteins. The mature proteins derived from the P1 polyprotein precursor are the component of capsids. To further delineate the process of capsid assembly and encapsidation, in a first attempt, a cell line which expresses the authentic P1 polyprotein was established. CV-1 cells were transfected with the pRCRSVS1P1 plasmid DNA which contains 5'ncr sequences, whole authentic capsid gene of poliovirus and neomycin resistance gene. These cells were treated with G418 for 3 months, and eventually G418 resistant cells were selected and formed colonies. Each colony was picked and grown in the media containing G418. DNA analysis indicated that 1 of 13 neomycin resistant cell lines (R2-18) contains whole poliovirus P1 capsid gene segment which was incorporated into the genome. Immuneprecipitation of cell lysates with sera from rabbit immunized with inactivateded Sabin type 1 particles demonstrated the constitutive expression of the poliovirus P1 capsid protein from R2-18.
IL-18. formerly known as IGIF(interferon -gamma inducing factor), is structurally IL-l related but functionally IL-12 related pro-inflammatory cytokine. The human IL -18(hIL-lS), like IL-$1{\beta}$, is synthesized as a biologically inactive precursor of 24kDa lacking a signal peptide, and then cleaved into an active mature form by cystein protease IL-$1{\beta}$ converting enzyme (ICE: caspase- 1), We tested if the mature hIL -18 can be expressed and secreted into culture medium by transforming the forming gene construct consisting of a mature hIL-18 gene fused to signal peptide of rice amylase lA. Secondly, we were tested if the pro- IL-18 could be processed into a biologically active form by caspase-l like protease in plant. Cell suspension culture was established from the leaf-derived calli of transgenic tobacco plant. Southern and Northern blot analysis indicated the expression of both pro-hIL-18 and mature hIL-18 plant cells. Western blot analysis introduced the protein products of pro- hIL -18 and mhIL -18 were observed in transigenic cell lines. In addition, the molecular size of recombinant pro-hILl-18 and mhIL-18 were estimated to be 24kDa and 18kDa, respectively. ELISA revealed that the amount of pro- hIL -18 was 1.3ug per gram of fresh weight calli. Moreover, the presence of mhIL-18 was detected in the culture medium and it appeared to be 25ug/L.
Ultrastructural studies of germ cell development during spermatogenesis and taxonomic values in mature sperm morphology of Argopecten irradians irradians were investigated by transmission electron microscopic observations. In the early stage of spermatid during spermiogenesis, a few granules and proacrosomal granules are formed by the Golgi complex. In the late stage of spermatid during spermiogenesis, a proacrosomal vesicle becomes an acrosomal vesicle in the acrosome through spermiogenesis. The sperm is approximately $ 45-48{\mu}m$ in length including a jar-shaped sperm nucleus (about $1.45{\mu}m$ long), an acrosome (about $0.34{\mu}m$ long) and tail flagellum. The axoneme of the sperm tail shows a 9+2 structure. As one of common characteristics of mature sperm morphologies in Pectinidae species in subclass Pteriomorphia, mature spermatozoon consists of the cone-shaped acrosomal vesicle and subacrosomal material on the invaginated jar-shaped nucleus. The acrosomal vesicle of this species is composed of electron high dense opaque part (material) from the base to the tip, as have seen in the species in the subclass Pteriomorphia. Exceptionally, five mitochondria are found in the sperm midpiece of this species, unlike four in most species of Pectinidae in subclass Pteriomorphia. However, the acrosomal vesicle of spermatozoa of A. irradians irradians resemble to those of other investigated Pectinidae species in subclass Pteriomorphia. Therefore, we can use sperm morphology as a tool in the resolution of taxonomic relationships within the Pectinidae species. These morphological charateristics of acrosomal vesicle belong to the family Pectinidae in the subclass Pteriomorphia.
