Molecular Authentication of Pinelliae Tuber from its adulterants by the analysis of DNA barcodes, matK and rbcL genes (matK와 rbcL DNA 바코드 분석을 통한 반하(半夏) 및 반하(半夏) 유사 한약재 유전자 감별)

  • Objectives : Pinelliae Tuber has been used as a typical unauthentic herbal medicines. Due to the morphological similarity between Pinelliae Tuber and adulterants, the correct authentication is very difficult. Therefore, we introduced DNA barcode to establish a powerful tool for the authentication of Pinelliae Tuner from adulterants. Methods : To obtain DNA barcode regions, genomic DNA was extracted from nineteen specimens of Pinellia ternata, Pinellia pedatisecta, Pinellia tripartita, and Typhonium flagelliforme, and matK and rbcL genes were amplified. For identification of species specific sequences and analysis phylogenetic relationship, a comparative analysis were performed by the ClastalW and UPGMA based on entire sequences of matK and rbcL genes, respectively. Results : In comparison of two DNA barcode sequences, we elucidated the phylogenetic relationship showing distinct four groups depending on species and identified 40 and 20 species specific nucleotides enough to distinguish each species from matK and rbcL gene, respectively. The sequence differences at the corresponding positions were avaliable genetic marker nulceotides to discriminate the correct species among analyzed four species. These results indicated that phylogentic and comparative analysis of matK and rbcL genes are useful genetic markers to authenticate Pinelliae Tubers. Conclusions : The marker nucleotides enough to distinguish P. ternata, P. tripatrita, P. peditisecta, and T. flagelliform, were observed at 40 positions in matK gene and 20 positions in rbcL gene sequence, respectively. These differences can be used to authenticate Pinelliae Tuber from adulterants as well as discriminate each four species.

  • Graphene-based materials have been gaining the unprecedented academic and industrial applications, due to the unique charge transport as a new kind of 2-dimensional materials. The applications incorporate electronic devices, nonvolatile memories, batteries, chemical sensors, etc. based on the electrical, mechanical, structural, optical, and chemical features newly reported. The current work employs thermal chemical vapor deposition involving H2 and CH4, in order to synthesize the 2-dimensional graphene materials. The qualitative/quantitative characterizations of the synthesized graphene materials are evaluated using Raman spectroscopy and Hall Measurements, In particular, the effect of processing variables is systematically investigated on the formation of graphene materials through statistical design of experiments. The optimized graphene materials will be attempted towards the potential applications to flat-panel displays.

  • For a class of nonlinear systems which satisfy a certain condition so called output feedback exponential passivity (OFEP), it is well known that one can easily design a high-gain output feedback control system. The designed high-gain controller has simple structure and high robustness. However, from the viewpoint of practical application, it is important to consider a robust control scheme for controlled systems for which some of the assumptions of output feedback stabilization are not valid. In this paper. we deal with a design problem of the robust high-gain adaptive output feedback control for the above-mentioned class of nonlinear systems with uncertain nonlinearities and/or disturbances.

  • Alumina electrostatic chucks for silicon wafer process with wide range of electrical resistivity were fabricated by controlling the amount of $TiO_2$ addition(0, 1.3, 2.0, 2.8 wt%). The dependence of electrostatic force on applied voltage, temperature and humidity was investigated. In addition, response characteristics on applied voltage and relationship between electrical resistivity and electrostatic force characteristics such as Coulomb force and Johnsen-Rahbeck force were discussed.

Determination of Model Parameters of Surface Cover Materials in Evaluation of Sediment Reduction and Its Effects at Watershed Scale using SWAT (토양유실 저감을 위한 지표피복 저감효과 변수 결정 및 SWAT 모형 유역단위 효과 분석)

  • The purpose of this study was to determine parameters of surface cover materials and evaluation the effects on runoff and sediment reductions with rice straw mat with PAM at watershed scale using the SWAT model. In this study, 1) regression equation of CN for rice straw mat + PAM using SCS curve number method was developed, 2) the USLE P factor, being able to reflect simulation of rice straw mat + PAM in the agricultural field, was estimated for various slope scenarios with VFSMOD-w. Then regression equation for CN and USLE P factor were used as input data in the SWAT model. Assuming rice straw mat + PAM is applied to radish and potato fields, occupying 24% of agricultural fields at the study watershed. Result of direct runoff without rice straw mat + PAM was $65,964,368\;m^3,$ with rice straw mat + PAM, direct runoff was $65,637,336\;m^3$, $327,031.8\;m^3$ reductions compared without it. Also, result of sediment without rice straw mat + PAM was 163,531 ton, with rice straw mat + PAM, sediment was 84,779 ton, 78,752 ton reduction compared without it. This analysis showed that about 48% sediment reductions would be expected with rice straw mat + PAM. As shown in this study, rice straw mat + PAM would be used as an efficient site-specific BMPs to reduce runoff and sediment discharge from field.

  • The 3D numerical analysis is carried out to investigate the settlement behavior of flexible mat foundations subjected to vertical loads. Special attention is given to the improved analytical method (YS-MAT) that reflects the mat flexibility and soil spring coupling effect. The soil model captures the stiffness of the soil springs as well as the shear interaction between the soil springs. The proposed method has been validated by comparing the results with other numerical methods and a field measurement on mat foundation. Through comparative studies, the settlement of the proposed method was in relatively good agreement with those of a field measurement and other numerical methods. The results permit us to estimate the response of the mat foundation subjected to vertical loads that should be taken into account in the combination of mat flexibility and soil continuity characteristics.

  • Recently, because of environment, cost, supply and demand factors, though applying sea-sand as horizontal drains is getting difficult that usage of Gravel has been growing in large size of construction sites, Study on engineering properties and behavior characteristics of Gravel stratum is not thoroughgoing enough. We have applied Gravel Mat as the horizontal drains in O O construction site. We also conducted several field tests such as Material property test, Geosynthetics damage test with Repeated load, Discharge capacity test performed by inflow of upper soil and In-situ PBD Penetration test to review the application of Gravel Mat. Test results show that Gravel Mat is not only advantageous in Trafficability and Water drainage by Consolidation due to its great Internal friction angle and Permeability, but also easy to penetrate with Mandrel and has great discharge capacity and guarantee of the stability against geosynthetics damage at the same time. With these benefits Gravel Mat shows great application in fields.
