Growth responses of kelp species Ecklonia cava to different temperatures and nitrogen sources (온도와 질소원 종류에 따른 대형갈조류 감태(Ecklonia cava)의 생장)

  • We investigated the seasonal variations in growth and physiological responses of the kelp species Ecklonia cava to different nitrogen sources to establish indoor culture conditions for mass production. Ecklonia cava was cultivated for 10 days in 16 combinations of seawater temperatures (15, 17, 21, and 25℃) and different nitrogen sources (control; NH-NH4+ 100 μM; NO-NO3- 100 μM; and NHNO-NH4+ 50 μM+NO3- 50 μM). The growth and growth rate of the blade were affected by temperature. The mean fresh weight and area-based daily growth rate were the highest (5.8±0.5 and 6.6±0.5% day-1, respectively) at 15℃ and the lowest (2.2±0.2 and 3.0±0.3% day-1, respectively) at 25℃. The daily growth rate was the highest in the NH and NO treatments and lowest in the control. The nitrate reductase activity of E. cava varied with water temperature (season). The highest activity was in the control (1.32±0.10 μmol NO2- g-1 dry weight h-1) and the lowest was in the NH treatment (0.25±0.02 μmol NO2- g-1 dry weight h-1). The photosynthetic pigment concentrations reached a maximum value in the NHNO treatment and a minimum value in the control. These results showed that water temperature played an important role in the cultivation of E. cava and that a single supply of NH4+ or NO3- may induce the accelerated growth of E. cava. The growth and physiological responses of E. cava to different nitrogen sources during each season provide valuable information for determining the optimal nitrogen source in E. cava cultivation under indoor conditions.

  • Sweet sorghum [Sorghum bicolor (L)] is one of the major crops for biofuels such as sugarcane and sugar beet which raw materials rich in saccharide. Sweet sorghum juice was extracted from the stem. It's composed of fermentable sugars such as glucose, fructose and sucrose. Ethanol from the extracted sweet sorghum juice can be easily produced by yeast fermentation process. Sweet sorghum juice is consisted of not only sugars but also various nutrients like nitrogen and phosphate. For commercial production of bioethanol, seed culture is one of the important parts of fermentation, so that optimal culture medium should be selected for the reduction of processing costs. In this study, sweet sorghum juice was estimated as a culture medium for seed culture of cellulosic bioethanol. For the comparison of cultures with various substrates, it used YPD including each 5 g/L yeast extract and peptone, sweet sorghum juice and hydrolyzed Miscanthus was taken part in the culture with 2%, 5% and 10% sugar conditions. Based on media of YPD and sweet sorghum juice, cell-mass concentration was obtained maximum more than $2.5{\times}10^8CFU/mL$ after 24 h of cultivation. Consequently sweet sorghum juice is suitable for the cell culture with more than $1.0{\times}10^8CFU/mL$ after 12 h of cultivation. This can be used as a culture medium for the cellulosic bioethanol industry.

A Study on Interpreting People's Enjoyment under Cherry Blossom in Modern Times (벚꽃을 통해 본 근대 행락문화의 해석)

  • In landscape architecture, plants play an important role in realizing the intention of the architect and user- behavior as well as an ecology and appearance of the space for them. However, it is true that many researches have focused on ecological characteristics of plants, their cultivation environment and symbolic meanings in traditional terms, while relatively few for the analysis of the aspects of each period through plants. For this, cherry trees that we often see around are selected and their introduction, propagation, development and symbolism from the view of chronicle are studied and the results are followings; Firstly, three-year seedlings of 1,500 pieces of cherry tree from Osaka and Tokyo were planted for the first time in Oieseongdae, Namsan Park, Seoul. Since then, they had been widely planted at traditional sites, modern parks, newly-constructed roads for street trees, and for this, the Japanese Government-General of Chosun had actively supported by its direct cultivation and selling of cherry trees. The spread of cherry trees planted raised the question of whether or not Prunus yedoensis is originated from Jeju Island. Secondly, such massive and artificial planting of them had become attractions over the time and mass media at that time also had actively promoted it. And such trend made the day and night picnic under the cherry blossoms one of the most representative cultures of enjoying spring in Seoul. Thirdly, although general people enjoyed cherry blossoms, but they had dual view and attitude for cherry trees, which were well expressed in their use of them: for example, cherry blossoms, aeng and sakura were used altogether for same meaning, but night aeng or night picnic under cherry blossoms were especially used instead of yojakura when mentioning just pleasure, which meant some saw night enjoying cherry blossoms a low culture. Fourth, symbolic space of Chosun had been transformed into the space for enjoyment and consumption. Anyone who paid entrance fee could enjoy performance of revugirl, cinema and entertainment along with enjoying cherry blossoms. The still-existing strict differentiation of enjoyment culture by social status, class and ethnicity was dismantled from that trend and brought about a kind of disorder. From this, we could find that cherry blossoms had made a great contribution to the change of traditional enjoyment culture over the Japanese colonial period and become a popular spring enjoyment.

Effect of droplet vitrification on mitochondrial membrane potential and developmental competence in two-cell mouse embryos

  • The accelerated cooling rate associated with vitrification reduces injuries attributed to cryopreservation and improves the post-freezing developmental competence of vitrified embryos. In this study, embryos were vitrified and warmed and morphologically evaluated for their development to blastocysts. Survival rates between the fresh ($96.7%{\pm}3.8%$) and vitrified embryos ($90.7%{\pm}5.1%$) did not differ significantly (P>0.05). The mitochondrial membrane potential of fresh control cells measured by 5,5',6,6'-tetrachloro-1,1',3,3'-tetraethylbenzimidazolyl carbocyanide iodide staining was similar to that of cryoprotected and vitrified embryos. Mitochondrial staining with rhodamine 123 did not differ among the fresh, cryoprotected, and vitrified embryos. Moreover, the distribution of $H_2O_2$, assessed by 2',7'-dichlorodihydrofluorescein diacetate staining, did not differ among the groups. The results showed that the developmental rate did not differ significantly among the fresh ($87.8%{\pm}11.3%$), cryoprotected ($83.2%{\pm}7.6%$), and vitrified 2-cell embryos ($75.8%{\pm}14.2%$). The mean number of the inner cell mass (ICM), trophectoderm (TE), and apoptotic cells was counted and statistically compared, and although the number of ICM and TE was decreased in the cryoprotected and vitrified embryos, there were no significant differences among the groups (P>0.05). During the cultivation period, randomly selected blastocysts from each group were stained using either 4',6-diamidino-2-phenylindole and bisbenzimide or the terminal deoxynucleotidyl transferase mediated dUTP nick-end labeling technique. The incidence of apoptosis appeared to be almost identical in all the groups. Droplet vitrification could subsequently lead to high survival and developmental rates of cryopreserved mouse embryos.

Study on Eco-friendly Control Effect of Natural Plant Extract Mixtures on Mulberry Popcorn Disease and Mulberry Sucker (천연 식물추출물 복합제를 이용한 오디균핵병 및 뽕나무이에 대한 친환경 방제기술 개발)

  • BACKGROUND: Two Field tests were conducted at Buan and Yangpyung in 2012 and 2013 to confirm controlling effect of mulberry popcorn disease and mulberry Sucker pests which are becoming serious in mass cultivation area of eco-friendly farming mulberry. METHODS AND RESULTS: As the treatments, 4 Natural products and 3 microbials were applied and 4 mulching materials were used. On the prevent effect of mulberry popcorn disease(Sclerotinia shiraiana) by using mulching materials, nonwoven fabric mulching showed worse effect than non-mulched treatment plot. Moreover, rice straw mulching showed significantly worse effect compare to nonwoven fabric and herb mulching treatments. Natural plant extracts and microbials showed 40~65% control value in 2013, which is little bit worse than 2012 results. On the control effect of mulberry Sucker(Amomoneura mori), organic products which combined with neem, sophora and derris showed excellent result as similar control level of Thiophanate-methyl. It means the chemical products can be replaced with organic product. CONCLUSION(S): According to the 2 years studied results, integrate eco-friendly farming measures are recommended for control of mulberry popcorn disease, because any single method is seemed not sufficient enough. However, natural plant extracts mixture is recommended as a product to control of mulberry Sucker.

Studies on the Life History of Nysius plebejus Distant (Hemiptera: Lygaeidae) an Insect Pest of Chrysanthemum (국화 해충의 1종인 애긴노린재의 생활사에 관한 연구)

  • Life histoy of Nysius plebejrrs, the most injurious pest in chysanthemun cultivation in Korea, was studies both in field and inseday in Chinju during 1991 and 1992 seasons, and resulis are summarized as below 7hls insect has 3 generation per year, adult peaks on the mid April, mid June and late August, and the highest number of adults were attracted to the light tmp from the late August through mid September. Overwntered generation and needed 6-7weeks from egg rnatulity m spring, and 5-6 weeks, for both the first and second generation m summer, Incubation egg periods were 142, 9.3 and 7.2 days, nymphal penods were 45.2, 32.1 and 31.2 days, and total growth periods, were 59.4. 41 4 and 38.4 days, respect~vely for the overwintered, flst and second generation. Ovipositional penods were 25.6, 23.4 and 21.2 days. and number of eggs Iald by each female were 41.5. 37.2 and 35.4 days, respedively of each generation Longevities of female were 326, 27.5 and 25.1 days. and of male were 15 2, 12.3 and 12.1 days. respectively of each generation. Sex ratlo was about 1:1 in all generation, and eggs were laid in mass on the surface of leaves or stems of the host plants. This insed over-wmters as adult (90%) or 5th instar nymph (10%), however, over-wintering adults seem to be sexually premature, and mating take place actively in the following spring.

A High Viscosity of Curdlan at Alkaline pH Increases Segregational Resistance of Concrete (염기성 pH에서의 고점도 커들란에 의한 콘크리트의 재료분리 억제 효과 증진)

  • In order to use a polysaccharide, curdlan, as a concrete admixture, we first developed a pilot-scale fermentation process for the mass production of curdlan. We also examined the rheological properties of curdlan, and tested how well the curdlan obtained in this work increased the segregational resistance of the cement slurry. Fermentation was performed in a 300-liter fermenter equipped with 3 disk-turbine impellers. Since curdlan production is stimulated under nitrogen-limiting conditions, the culture pH was shifted from the optimal pH for cell growth (pH 7.0) to the optimal pH for curdlan production (pH 5.5) at the onset of ammonium exhaustion. We obtained a curdlan production of 65 g/L in 120 hr batch cultivation of Agrobacterium species. The insoluble curdlan at the final stage of fermentation was readily harvested by centrifugation together with the cells. The freeze-dried sample contained 78% (w/w) of curdlan. The solubility and viscosity of the curdlan increased with the increase of the solution pH, which enhances the viscosity of concrete since the pH of concrete is extremely high (pH 13.0). Test results of the curdlan as a concrete admixture with cement slurry demonstrated that it prohibits the leakage of water. In conclusion, this work certifies and enlarges curdlan's industrial potential as a concrete admixture.

Growth Characteristics of Microalgae Scenedesmus obliquus by LED Light Source (LED 광원에 따른 미세조류 Scenedesmus obliquus의 성장 특성)

  • Microalgae are independent organisms that perform photosynthesis and can alter the culture environment to increase accumulation of useful substances derived from microalgae. In this study, cell growth was measured by incubation for 39 days using MBBM, Neo medium, and seven light sources, which is the main factor affecting cell growth of microalgae S. obliquus. In the case of S. oliquus, which grew in MBBM and Neo medium, cell growth was highest under fluorescent light sources and Red2 LED (R660) light sources, and cell growth was lowest under Infra Red LED (R741) light sources. The average cell growth rate was 17.7% for MBBM and 15.4% for Neo. Comparing the effects of dry cell weight of Neo medium containing nutrients on the production of aquatic plants, MBBM and dry cell weight of Neo resulted in higher cell growth than Neo medium under all LED light sources except for Blue LED (B450). This proves that MBBM is more suitable for increasing the cell growth of microalgae than Neo medium and confirms that light source selection is important in the production of useful materials through mass cultivation of microalgae in the future.

Optimal Conditions for the Laccase Production from Fomitopsis pinicola Mycelia (Fomitopsis pinicola 균사체로부터 Laccase의 최적생산조건)

  • The culture conditions to maximize the production of laccase (EC 1.10.3.2) from Fomitopsis pinicola mycelia were investigated. Among the tested media for the enzyme production, mushroom complete medium (MCM ; 2% dextrose, 0.2% peptone, 0.2% yeast extract, 0.05% $KH_2PO_4$, and 0.05% $MgSO_4{\cdot}7H_2O$) showed the highest activity of the enzyme. To optimize the culture condition for the laccase activity, influence of various carbon and nitrogen sources was investigated in MCM. Among various carbon and nitrogen sources, 2% glucose and 0.4% peptone showed the highest production of the enzyme, respectively. For the phosphorus and inorganic source, 0.05% $NaH_2PO_4$ and 0.05% $CaCl_2$ were best for the enzyme activity. The enzyme production was reached to highest level after the cultivation for 8 days at $25^{\circ}C$. Native polyacrylamide gel electrophoresis (PAGE) followed by the laccase activity staining using 2, 2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) as the substrate was performed to identify the laccase under culture conditions studied. Zymogram analysis of the culture supernatant showed a laccase band with molecular mass of 52 kDa. The optimum pH and temperature for the enzyme activity were $80^{\circ}C$ and pH 3.0.

Application of Lignocellulosic and Macro-algae Hydrolysates for Culture of Chlorella saccharophila (Chlorella saccharophila 배양을 위한 목질계 및 해조류 바이오매스 가수분해물의 이용)

  • In this study, we investigated the possibility of using hydrolysates of lignocellulosics (rapeseed straw, barley straw, rice straw) and marine macro-algae (Undaria pinnatifida, Laminaria japonica, Enteromorpha intestinalis, and Gracilaria verrucosa) to cultivate Chlorella saccharophila. The growth of C. saccharophila was inhibited by 7 hydrolysates without active carbon treatment. In contrast, hydrolysates treated with active carbon increased the cell growth and product (oil and chlorophyll) formation by C. saccharophila. The oil contents of C. saccharophila treated with each hydrolysate were $41.26{\pm}0.69%$ (glucose), $22.06{\pm}1.21%$ (rapeseed straw), $28.65{\pm}1.08%$ (barley straw), $31.15{\pm}0.76%$ (rice straw), $31.50{\pm}2.12%$ (U. pinnatifida), $31.49{\pm}4.53%$ (L. japonica), $29.63{\pm}3.93%$ (E. intestinalis), and $26.15{\pm}1.99%$ (G. verrucosa), respectively. Lignocellulosics and marine macro-algae may be useful resources for improving the mass cultivation of C. saccharophila.
