산가수분해법과 반응표면분석법을 이용한 해조류 청각으로부터 레불린산의 생산 (Production of Levulinic Acid from Marine Algae Codium fragile Using Acid-Hydrolysis and Response Surface Methodology)

  • This work is focused on the possibility of marine biomass Codium fragile as renewable resources for production of levulinic acid. In an effort to optimize the reaction conditions of levulinic acid production from Codium fragile, response surface methodology was applied. A total of 18 individual experiments were designed to investigate the effect of reaction temperature, catalyst amount, and reaction time. As a result, 4.26 g/L levulinic acid from Codium fragile was produced in the condition of $160.7^{\circ}C$ of reaction temperature, 3.9% of sulfuric acid, and 39.1 min of reaction time. This result will provide the useful information for chemical production from marine resource.

  • Foods and related substances from diverse sources known to have a potential for disease risk reduction are called functional foods, while nutraceuticals are bioactive compounds isolated from food and sold in dosage form. Nutraceutical and functional food industries are rapidly growing in recent years and most of the cases development of these functional materials involves certain biotransformation processes. A number of bioactive compounds has been identified up to date and isolated from seafood related products through enzyme-mediated hydrolysis. The enzymatic bioconversion process require suitable biocatalysts and appropriate bioreactor systems to incubate byproducts with digestive enzymes. Membrane bioreactor technology is recently emerging for the development of bioactive compounds from seafood processing byproducts.

해양 호염성 세균 Vibrio alginolyticus가 생산하는 Extracellular Amylase의 특성 (Properties of an Extracellular Amylase Produced by the Marine Halophilic Bacterium Vibrio alginolyticus)

  • V. alginolyticus 138-2, a marine halophilic bacterium, produced an extracellular amylase with a molecular weight of ca. 56,000. The analysis of the digestion products of soluble starch by thin layer chromatography(TLC) revealed that the extracellular amylase of V. alginolyticus 138-2 is a saccharifying-type alpha-amylase. The alpha-amylase activity of the culture supernatant of soluble starch was optimal at pH 6.0 and 45$^{\circ}C$. Ca2+ slightly increased the alpha-amylase activity, whereas Hg2+, An2+, Cu2+, Ni2+, Fe2+, and Mn2+inhibited the enzymatic activity. Alkylating thiol group agent, iodoacetic acid did not affect the alpha-amylase activity, but reduced thiol reagents such as dithiothreitol, cysteine, and beta-mercaptoethanol stimulated theenzymatic activity. On the other hand, even if V. alginolyticus 138-2 is a marine halophilic bacterium, its alpha-amylase activity was significantly inhibited by NaCl.

Enhancing Photon Utilization Efficiency for Astaxanthin Production from Haematococcus lacustris Using a Split-Column Photobioreactor

  • A split-column photobioreactor (SC-PBR), consisting of two bubble columns with different sizes, was developed to enhance the photon utilization efficiency in an astaxanthin production process from Haematococcus lacustris. Among the two columns, only the smaller column of SC-PBR was illuminated. Astaxanthin productivities and photon efficiencies of the SC-PBRs were compared with a standard bubble-column PBR (BC-PBR). Astaxanthin productivity of SC-PBR was improved by 28%, and the photon utilization efficiencies were 28-366% higher than the original BC-PBR. The results clearly show that the effective light regime of SC-PBR could enhance the production of astaxanthin.

  • 1-Acetyl-$\beta$-carboline was isolated as an anti-MRSA agent from the fermentation broth of a marine actinomycete isolated from marine sediment. The producing strain was identified to be Streptomyces sp. by phylogenetic analysis of the 16S rRNA gene sequence. The anti-MRSA agent was isolated by bioactivity-guided fractionation of the culture extract by solvent partitioning, ODS open flash chromatography, and purification with a reversed-phase HPLC. Its structure was elucidated by extensive 2D NMR and mass spectral analyses. Combination of 1-acetyl-$\beta$-carboline with ampicillin exhibited synergistic antibacterial activity against MRSA.

Scalarane-type Sesterterpenes from the Philippines Sponge Hyrtios sp.

  • The marine sponge Hyrtios sp. collected in the Philippines was extracted and partitioned. The resulting organic layer was purified by C18 reversed-phase column chromatography and HPLC to achieve the separation of nine scalarane-type sesterterpenes, including one new compound with eight known scalarane analogs. The chemical structures of the isolated compounds 1-9 were elucidated by 1D and 2D NMR and MS data analysis. All nine compounds were evaluated for their antibacterial activities against three Gram-positive and three Gram-negative bacteria. The compound 3 exhibited potent antibacterial activities against Bacillus subtilis and Micrococcus luteus. The compounds 7 and 9 displayed considerable activities against Bacillus subtilis and the others had moderate results.

Influence of Organic Carbon Sources on Growth and Lipid Content of Marine Green Alga Dunaliella tertiolecta

  • This study investigated the potential use of various organic carbon sources (glucose, glycerol and acetate) and different concentrations of $CO_2$ for culturing marine microalga Dunaliella tertiolecta. Cell growth and lipid production were monitored under heterotrophic, mixotrophic and photoautotrophic modes of cultivation. D. tertiolecta showed the ability to grow under mixotrophic (acetate and glucose), heterotrophic (glucose) and photoautotrophic condition under high $CO_2$ concentration (15%). With all the organic carbon sources (glucose, glycerol and acetate) tested in this study, 1~5% acetate enhanced cell growth rate and lipid content, while higher concentrations of acetate (10% and 15%) were inhibitory and resulted in cell death.

Seasonal Assessment of Biomass and Fatty Acid Productivity by Tetraselmis sp. in the Ocean Using Semi-Permeable Membrane Photobioreactors

  • A green microalga, Tetraselmis sp., was cultivated in the coastal seawater of Young-Heung Island using semi-permeable membrane photobioreactors (SPM-PBRs) in different seasons. The microalgae in the SPM-PBRs were able to grow on nutrients diffused into the PBRs from the surrounding seawater through SPMs. The biomass productivity varied depending on the ion permeabilities of the SPMs and environmental conditions, whereas the quality and quantity of fatty acids were constant. The temperature of seawater had a greater influence than solar radiation did on productivity of Tetraselmis sp. in SPM-PBRs. SPM-PBRs could provide technologies for concurrent algal biomass and fatty acids production, and eutrophication reduction in the ocean.

A new sesterterpenoid showing anti-inflammatory effect from the Marine Sponge Haliclona species

  • Four spiroketal sestertepenoids (1 ~ 4) were isolated from the marine sponge Haliclona species. Their planar structures were completely determined from a combination of extensive 1D and 2D NMR experiments, and also the relative stereochemistry on the chiral centers were established by the ROESY experiment. Compounds 1 ~ 3 were determined as the same planar structures with different stereochemistry on the chiral centers C-11 and C-13. Of these, 1 was identified as a new stereoisomer. Four compounds showed the inhibition effect of nitric oxide (NO) production in LPS-stimulated RAW 264.7 cells (0.7~2.0 g/ml).

R&D Trends of Brown Algae as Potential Candidates in Biomedical Application

  • Seaweeds contain various bioactive compounds. Especially, brown algae (Phaeophyceae), the second abundant group of seaweeds, contain numerous nutraceutical and pharmaceutical substances. In this review, we investigated on the brown algae-related patents and literature. Consequently, the research and development (R&D) trends of patent related to brown algae showed that the large majority was applied as the composition of stem cell culture medium and mostly used as active substances. In conclusion, we suggested that many researchers try to investigate and develop applications of brown algae as the sophisticated-level biomedical materials because brown algae are actively developing as simple-level biomedical materials.
