Bioactivity of Metabolites from Actinomycetes Isolates from Red Sea, Egypt

  • Actinomycetes isolated from marine habitats represent a promising source of bioactive substances. Here, we report on the isolation, identification, productivity enhancement and application of the bioactive compounds of Streptomyces qinglanensis H4. Eighteen marine actinomycetes were isolated and tested for resistance to seven bacterial diseases. Using 16S rRNA sequencing analysis (GenBank accession number MW563772), the most powerful isolate was identified as S. qinglanensis. Although the strain produced active compound(s) against a number of Gram-negative and Gram-positive bacteria, it failed to inhibit pathogenic fungi. The obtained inhibition zones were 22.0 ± 1.5, 20.0 ± 1, 16.0 ± 1, 12.0 ± 1, 22.0 ± 1 and 24.0 ± 1 mm against Bacillus subtilis ATCC 6633, Escherichia coli ATCC 19404, Enterococcus faecalis ATCC 29212, Pseudomonas aeruginosa ATCC 9027, Candida albicans ATCC 10231 and Staphylococcus aureus ATCC6538, respectively. To maximize bioactive compound synthesis, the Plackett-Burman design was used. The productivity increased up to 0.93-fold, when S. qinglanensis was grown in optimized medium composed of: (g/l) starch 30; KNO3 0.5; K2HPO4 0.25; MgSO4 0.25; FeSO4·7H2O, 0.01; sea water concentration (%) 100; pH 8.0, and an incubation period of 9 days. Moreover, the anticancer activity of S. qinglanensis was tested against two different cell lines: HepG2 and CACO. The inhibition activities were 42.96 and 57.14%, respectively. Our findings suggest that the marine S. qinglanensis strain, which grows well on tailored medium, might be a source of bioactive substances for healthcare companies.

  • A new ${\alpha}-pyrone$ derivative, violapyrone J (1), and along with the two known violapyrones B (2) and C (3) were isolated from the fermentation broth of a marine actinomycete Streptomyces sp. SC0718. The structure of violapyrone J (1) was elucidated from 1D and 2D NMR spectroscopic analyses.

Biosurfactant Production by Marine Actinomycetes Isolates Streptomyces althioticus RG3 and Streptomyces californicus RG8 as Promising Sources of Antimicrobial and Antifouling Effects

  • Two marine actinobacterial isolates, RG3 and RG8, were identified using 16Sr DNA as Streptomyces althioticus RG3 and Streptomyces californicus RG8 and submitted to the database of genetic information with accession numbers MW661230 and MW661234, respectively; they were found to have emulsification indexes of 60 ± 2.5% and 53 ± 2.2%, respectively. The biosurfactants obtained were stable at a temperature of 35℃ for both strains; they were stable at 10% NaCl, in the case of S. althioticus RG3 and at 10-15% NaCl in the case of Str.californicus RG8; both strains produced the most biosurfactant when exposed to alkaline conditions. We characterized the biosurfactants, including features such as their chemical composition, using Fourier transform infrared spectroscopy analysis. The antimicrobial activity of the biosurfactant extracts was evaluated using the well diffusion method against Vibrio alginolyticus MK170250, Escherichia coli ATCC 8739, Pseudomonas aeruginosa ATCC 4027, and Staphylococcus aureus ATCC 25923. S. althioticus RG3 biosurfactants were found to have better antimicrobial activity than those of Str. californicus RG8, indicating that they may be used in pharmaceutical industries and in the manufacture of antifouling products.

해양방선균으로부터 Haloperoxidase의 검색과 특성 (Screening and Partial Purification of Haloperoxidase from Marine Actinomycetes)

  • Haloperoxidase를 생산하는 미생물을 분리하기 위하여 국내 연근해와 남북극 등의 해양시료에서 분리된 방선균 균주를 대상으로 탐색을 수행하여 남해 백도 해조류 추출물로부터 분리된 한 종류의 방선균(#1460)에서 높은 haloperoxidase 활성이 확인되었다. 본 균주의 생리.생화학적 특성은 Streptomyces 속과 유사하며 생산되는 haloperoxidase는 세포 조 추출물로부터 ammonium sulfate precipitation, High-Q column chromatography, gel permeation chromatography, Hydroxyapetite chromatography 그리고 hydrophobic interaction chromatography를 통하여 42%의 수율과 purification fold 70으로 정제하였다. 본 효소의 최적 반응 pH는 7이고 pH 8에서 더 높은 안정성을 보여 $60^{\circ}C$에서 1시간 반응에 효소활성의 50%가 생존한다. 또 cyanide와 azide 이온에 의해 강한 저해현상을 보인다.

해양방선균 Streptomyces sp. ACT-1의 동정 및 항산화 활성 (Identification and Antioxidant Activity of Marine Actinomycetes Streptomyces sp. ACT-1)

  • 이 연구는 해양 유래의 천연 항산화물질에 관한 연구로서 항산화물질 생산 해양 방선균은 제주연안 해수에서 분리되었다. 균주는 16S rRNA 염기서열, 전자현미경에 의한 형태학적, 생리, 생화학적 및 세포 지방산 분석을 기초로 동정되었다. 분리균주 ACT-1은 그람양성, 호기성, 비운동성, 기질균사체색이 red, 기중균사체 색이 gray계열 이었다. 세포지방산 분석결과 주요 지방산으로 $C_{15:0}$ anteiso (39.33%), $C_{16:1}$ cis 9 (11.96%), $C_{16:0}$ (13.08%)와 $C_{17:0}$ anteiso (10.99%)로 분석되었으며, 최종적으로 Streptomyces sp. ACT-1으로 동정되었다. Streptomyces sp. ACT-1 메탄올 추출물의 항산화활성은 DPPH (1,1-diphenyl-2-picrylhydrazyl), hydroxyl, alkyl radical 소거활성을 ESR (electron spin resonance) spectrometer를 이용하여 측정되었다. SBME-1 (Streptomyces broth methanol extract)의 DPPH 라디컬 소거활성은 $1,000{\mu}g$/ml 농도에서 67%, Hydroxyl radical 소거활성은 $500{\mu}g$/ml에서 84%, Alkyl radical 소거활성은 $1,000{\mu}g$/ml 농도에서 71%를 보였다.

  • This study investigated the production of antioxidant from Actinomyces culture supernatant. For the research of the natural marine antioxidant, several bacteria were isolated from the coast of Je-ju in Korea. An actinomycetes strains, S-1 was identified to a genus level 16S ribosomal DNA sequence and fatty acid analysis. From these results and other characteristics described in the Bergey's Manual, this strain was identificated as a Nocardiopsis dassonvillei. Strain S-1 showed high activity of 1,1-diphenyl-2-prcrylhydrazyl radical scavenging. The hydroxyl radical scavenging ability of Nocardiopsis sp. S-1 supernatant was 53%. Nutritional and cultural conditions for the production of antioxidant by this organism under shake-flask conditions have optimized. Similary initial medium pH 7.6, incubation temperature of $25^{\cicr}C$, sodium chloride concentration 2.5 and incubation time of 8 day were found to be optimal.

  • 본 연구는 제주 연안 해수로부터 분리된 항산화물질을 생산하는 해양 방선균의 천연해양항산화물질에 관한 연구이다. 균주 ACT-18은 16S rDNA 염기서열, 전자현미경을 이용한 형태학적 분석, 생리학적, 생화학적 특성 및 세포 지방산분석을 통하여 동정되었다. 분리균주 ACT-18은 Gram 양성, 호기성, 비운동성 포자, 배지상에서 substrate mycelia의 색이 어두운 초록색, aerial mycelia는 노란색의 회색을 나타내였다. 세포사이즈는 약 $0.5{\sim}1.0\;{\mu}m$였다. 16S rDNA 염기서열 분석결과 Gram 양성박테리아인 Streptomyces 종들과 그룹을 형성하였다. 지방산분석결과 주요 지방산은 $C_{15:0}$ anteiso(39.33%), $C_{16:1}$ cis 9(11.96%), $C_{16:0}$ (13.08%)과 $C_{17:0}$ anteiso(10.99%)로 분석되었다. Streptomyce sp. ACT-18의 배양액 메탄올 추출물의 항산화 활성은 electron spin resonance (ESR) spectrometer에 의한 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl, alkyl 라디컬 소거활성을 측정하여 나타내었다. 그 결과 배양액 메탄올 추출물의 DPPH 라디컬 소거활성은 0.1 mg/mL의 농도에서 46%의 라디컬 소거활성을 보였으며, Hydroxyl 라디컬 소거활성은 0.1 mg/mL에서 63%, alkyl 라디컬 소거활성은 0.1 mg/mL에서 39%의 소거 활성을 보였다.

해양 방선균 유래 물질의 정제 및 항균 활성 (Purification and Antibacterial Activity of Compound Derived from Marine Actinomycetes)

  • Antibiotics are substances produced by microorganisms that kill or inhibit and are essential for infectious diseases management. This study aimed to provide basic data for overcoming antibiotic resistance in the marine bacterium LJ-18. The API 20NE and API 50CH kits were used to identify this microorganism. Morphological, physiological, and biochemical properties were investigated using MacFaddin's manuals. Subsequently, isolated LJ-18 was found to belong to a genus of Streptomyces that forms mycelia. LJ-18 also grew well at 28-32℃ on modified Bennett's agar. To isolate and purify the antibacterial compound, LJ-18 culture was divided into ethyl acetate and distilled water fractions. Considerable antimicrobial activity against various pathogenic microorganisms, including methicillin-resistant Staphylococcus aureus (MRSA), was confirmed in the C18 ODS open column fractions. Peak 2 compound was obtained using reversed-phase HPLC. As a result, this compound had a significant antimicrobial activity against various pathogenic microorganisms. In particular, it showed strong activity against MRSA, Mycobacterium smegmatis, Bacillus subtilis, Bacillus cereus, and Staphylococcus aureus.

제주 연안에서 분리된 해양방선균의 이화학적 특성 및 다양성 (Physico-chemical Characteristics and Diversity of Marine Actinomycetes Isolated from the Coast of Jeju Island)

  • 제주도 연안해역 4개 지역(한림, 애월, 신촌, 함덕) 해수의 온도, 염분농도, 용존산소량(DO), 화학적 산소요구량(COD), 부유물질(SS), 암모니아성 질소($NH_3-N$), 질산성 질소($NO_3-N$) 및 아질산성 질소($NO_2-N$)와 같이 다양한 이화학적 특성을 확인하였다. 해수의 평균 온도는 $26.23{\sim}28.6^{\circ}C$, 염분농도는 31.4~32.88‰, pH는 8.15~8.35, COD는 0.48~0.91 mg $L^{-1}$, DO는 6.78~6.87 mg $L^{-1}$로 나타났다. 해수에서 분리된 해양방선균은 총 52종으로 제주시 동부지역(A, B)에서는 24균주, 서부지역(C, D)은 28균주가 분리되었다. 분리된 해양방선균은 16S rRNA 염기서열 분석을 이용하여 최종적으로 동정되었으며, 염기서열 정보를 기초로하여 유사도(similarity)를 조사하였다. 분리된 방선균의 16S rRNA 염기서열 분석을 통하여 계통 분류학적으로 어떤 division에 속하는지를 확인하여 본 결과 제주도 제주시 동부지역(Site A, B) 해양에서 분리된 방선균 24균주는 Gram positive bacteria (division)/Actinobacteria (class)/Actinomycetales (order)/Streptomy-cineae (suborder)/Streptomycataceae (family)/Streptomyces(genus)에 22 균주, Actinomycetales (order)/Streptosporangineae (suborder)/Nocardiopsaceae (family)/Nocardiopsis (genus)에 2 균주가 분리되었다. 제주시 서부지역(Site C, D) 해양에서 분리된 방선균 28균주는 Gram positive bacteria (division)/Actinobacteria (class)/Actinomycetales (order)/Streptomycineae (suborder)/Streptomycataceae (family)/Streptomyces (genus)에 27균주, Actinomycetales (order)/Streptosporangineae (suborder)/Nocardiopsaceae (family)/Nocardiopsis (genus)에 1균주가 분리되었다.

Purification and Structure Determination of Antifungal Phospholipids from a Marine Streptomyces

  • A series of antifungal compounds were obtained from the methanol extract of the mycelium from marine actinomycetes M428 which was identified as a Stereptomyces species by fatty acid composition and biochemical characteristics. These compounds were purified by combined chromatographic techniques and the structures were characterized with spectroscopic methods including 1D and 2D NMR, and mass spectrometry as sn-l lysophosphatidyl inositols. The side chains were established by chemical degradation followed by GC analysis to be 14-methyl pentadecanoic acid (iso-palmitic acid, i-C16:0, compound A) and 13-methyl tetradecanoic acid (iso-pentadecanoic acid, i-C15:0, compound B). These compounds displayed highly selective antifungal activity against C. albicans with MIC values of $5{\;}\mu\textrm{g}/ml$ (compound A) and $2.5{\;}\mu\textrm{g}/ml$ (compound B), while it had almost negligible antibiotic activity against E. coli and P aerogenosa with MIC value higher than $50{\;}\mu\textrm{g}/ml$ and no cytotoxic activities against human myeloma leukemia K562 ($IC_{50}>100{\;}\mu\textrm{g}/ml$).
