Umbilical Cord Arterial Concentrations of Isoprostane(8-iso-PGF2α) in Newborn Infants (신생아에서 제대 동맥혈 Isoprostane(8-iso-PGF2α) 농도에 관한 연구)

  • Purpose : We measured the umbilical cord arterial concentrations of isoprostane($8-iso-PGF_{2{\alpha}}$) and intended to decide whether the umbilical cord arterial concentrations of isoprostane could be used as a useful parameter for lipid peroxidation in newborn infants. Methods : The isoprostane and malondialdehyde(MDA) concentrations of the umbilical cord were measured by enzyme immunoassay and TBARS(thiobarbituric acid reactive substance) assay in 33 preterm and 28 term infants, respectively. The concentrations of isoprostane and MDA were compared between preterm infants and term infants, and were analysed for association with perinatal risk factors and neonatal complications. Results : Umbilical cord arterial concentrations of isoprostane were $704.7{\pm}635.6pg/mL$ and $421.9{\pm}306.5pg/mL$ in preterm and term infants, respectively. Umbilical cord arterial concentrations of MDA were $44.0{\pm}22.9{\mu}M/L$ and $26.2{\pm}10.7{\mu}M/L$ in preterm and term infants, respectively. Umbilical cord arterial concentrations of isoprostane and MDA in preterm infants were significantly higher than those in term infants(P<0.05). The umbilical cord arterial concentrations of isoprostane were significantly associated with perinatal risk factors such as fetal distress, oligohydramnios, and breech delivery in preterm infants and pregnancy-induced hypertension in term infants(P<0.05). Conclusion : Umbilical cord arterial concentrations of isoprostane in preterm infants were higher than those in term infants, and those are significantly associated with some perinatal risk factors.

Differences of Garlic Powder Ingestion and Exercise Training on Blood Lipids, MDA and SOD in Rats (운동훈련과 마늘가루 섭취에 의한 흰쥐의 혈중 지질 및 MDA, SOD 활성 차이)

  • This study investigated the effect of exercise training and garlic powder ingestion on blood lipids and antioxidants activity in rats. Twenty-eight male Sprague Dawley rats were fed a high-fat diet with or without garlic powder (500 mg/kg) for four weeks as grouped in control (CON), exercise (EXE), garlic (GAR), and garlic + exercise training (GAREXE), respectively. EXE and GAREXE were trained on the treadmill for the same periods. Weight of fats (mesentery, perirenal, and epididymal) were weighed and blood glucose, triglycerides (TG), total cholesterol (TC), and high density lipoprotein- cholesterol (HDL-C) were analyzed and low density lipoprotein-cholesterol (LDL-C) was calculated. Malondialdehyde (MDA) and superoxide dismutase (SOD) for lipid peroxidation were analyzed in liver tissue. Body weight in GAREXE was significantly lower in the statistics than that in other groups (p<0.05), and the volume of fat in GAR and GAREXE was also much lower (p<0.05). Blood glucose was significantly lower in EXE and GAR (p<0.05), however, there was no effect of exercise training. Blood TG was lower in GAR and GAREXE (p<0.05), however, there was no effect of exercise training. HDL-C was significantly improved in EXE and GAR compared to CON (p<0.05), and GAREXE was higher than EXE (p<0.05). MDA content was considerably lower in GAREXE compared to EXE (p<0.05), and SOD activity was much higher in other groups compared to CON (p<0.05). In addition, GAREXE was significantly higher than EXE and GAR, thus there was significant increase when a garlic diet was carried out together with exercise (p<0.05). These results suggested that garlic powder ingestion during the training periods had a beneficial effect of lowering glucose and enhancing blood lipids profiles. Moreover, it also has antioxidant effects, which means that it could possibly suppress aging. It is necessary to inspect various effects of garlic with a variety of research methods regarding sampling process, production process, intake method, etc.

The Effect of protein and lipioperoxide on White Ginseng(WG) and Fermenta Ginseng(FG) Extracts on the liver in Mice that was irradiated by radiation (방사선이 조사된 생쥐 간에서 백삼과 발효인삼추출물이 단백질 및 지질과산화에 미치는 효과)

  • The effects of ginseng extracts on liver damage induced by high energy x-ray were studied. To one group of ICR male mice were given white(50 mg/kg/day for 7 days, orally) and fermenta ginseng extracts(500 mg/kg/day for 7 days, orally)before irrdiation. To another group were irradiated by 5 Gy dose of high energy x-ray. Contrast group were given with saline(0.1 ml). This study also investigated the effect between MDA, protein content and ginseng extracts on hepatic damage. This study measured the level of MDA(malondialdehyde), protein content in liver tissue. Administrating orally white (50 mg/kg/day for 7 days, orally)and fermenta ginseng extracts(500 mg/kg/day), the level of MDA were generally decreased and the inhibition was increased. And the protein contents were identical with control group. After irradiation, the protein contents were increased and MDA(malondialdehyde) was increased. Therefore, ginseng extracts increased antioxidative enzyme activity. And We know that the antioxidatant effect of extracts from white and fermenta ginseng protect radiation damage by direct antioxidant effect involving SOD, CAT, GPX. It was included that ginsengs can protect against the lipid peroxidation in radiation damage through its antioxidatant properties.

Application of Edible Red Algae Paper Coated with Green Tea Extract for Shelf Life Extension of Kimbab

  • Edible red algae paper coated with green tea ext Tact was prepared and determined the microbial growth and quality change of kimbab wrapped with the paper during storage. The paper coated with green tea extract had the antimicrobial activity against Listeria monocytogenes and Escherichia coli. After 12 hr of storage of kimbab, packaging with the paper coated with green tea extract decreased populations of total aerobic bacteria by 1 log cycle. Kimbab wrapped with the red algae paper coated with green tea extract had 5.5 mg malondialdehyde (MDA)/kg after 12 hr, in contrast to the control of 7.4 mg MDA/kg. Kimbab with red algae paper with green tea extract was better than the control in terms of sensory qualities. These results clearly indicate that kimbab can be packaged with red algae paper coated with green tea extract, resulting in extending its shelf life.

Antioxidative and Probiotic Properties of Lactobacillus gasseri NLRI-312 Isolated from Korean Infant Feces

  • We selected a Lactobacillus spp. from Korean healthy infant feces based upon their antioxidant activity. This strain was identified as Lactobacillus gasseri by 16S rDNA sequencing, and named Lactobacillus gasseri NLRI-312. In the present study, we investigate the protective effect of this strain on the $H_2O_2$ induced damage to cellular membrane lipid and DNA in Jurkat cells. To estimate the extent of cellular lipid peroxidation inhibition, MDA (malondialdehyde) was measured, and DNA damage was tested by the comet assay. We also examined probiotic properties including tolerance to acid and bile, antibiotic resistance. From the results obtained, the supplementation of Jurkat cells with NLRI-312 decreased in DNA damage, while no effect was shown on MDA decrease. In probiotic properties, this strain was resistance to both acid and bile, showed considerably higher survival when incubated in pH 2 or 1% bile salts (w/v). We concluded that the NLRI-312 could be used as potential probiotic bacteria, with the effect of reducing DNA damage induced by $H_2O_2$.

Effects of Chilbokyeum(七福飮) on the Biochemical Changes in Brain Tissue of Senile Rats (칠복음(七福飮)이 노화(老化) 백서(白鼠) 뇌조직(腦組織)의 생화학적(生化學的) 변화(變化)에 미치는 영향(影響))

  • The present experiment was designed to examine catecholamines, 5-hydroxytryptamine, amino acids. malondialdehyde(MDA) and free radical scavenging activity, by administering Chilbokyeum extracts of a variety of concentration to senile brain rats. The results were summarized as followings ; 1. Chilbokyeum significantly increased noradrenalin in the hippocampus and hypothalamus of the brain tissue of senile rats, ad even though Chilbokyeum increased noradrenalin also in other brain tissue, there was no significance. 2. Chilbokyeum had no effects on dopamine changes in all brain tissue of senile rats.3. Chilbokyeum significantly increased 5-hydroxy-tryptamine in cerebellum, but decreased in other brain tissue.4. Chilbokyeum increased amino in the brain tissue of senile rats. 5. Chilbokyeum significantly decreased MDA and free radical in the brain tissue of senile rats. According to the above results, Chilbokyeum is assumed to improve brain function by reaction by reacting on biochemical of the senile brain, and that Chilbokyeum can be used to treat regressive brain disease carrying symptoms of psychoactive disorders.

Effectso fvitamin E Supplementation on the Lipid Perosides and Activities of Antioxidative Enzymes in the Pancreas of diabetic KK Mice (비타민 E 보강식이가 당뇨 KK 마우스의 췌장에서 지잘과 산화물의 항산화 효소 활성에 미치는 영향)

  • The purpose of this study was to investigate the effect of vitamin E supplementation on the lipid peroxidation and activities of antioxidative enzymes in the pancreas of diabetic KK mice. KK mice were fed high ft diet containing 20% corn oil(wt/wt), and sacrificed at 2 months of diabetes. A hish vitamin E diet consisted of the high fat diet supplemented with an excessive amount of 이-$\alpha$-tocopheryl acetate (2080IU/kg diet). The incidence of diabetes mellitus was 61% when mice were fed the high fat diet, but was 44% when mice were fed the high vitamin E diet, Vitamin E supplementation fhus seems to have the effect of decreasing of decreasing the onset of diaetes. In the diabetic group, we found increases of MDA (malondialdehyde) and antioxidative enzyme activities. Treatment with vitamin E did not modify the level of fasting blood glucose. However, MDA and antiosicative enzyme activities in diabetic mice were decreased by the high vitamin E diet. Increased levels of lipid peroxidation products suggests the occurrence of oxidative damage in the pancreas of diabetic mice. The increased level of antiosicative enzyme activities could be due to an adaptive response to conditions of increased peroxidative stress. Significant normalization on catalase activity was noted in vitamin E supplemented animals.

Effect of Galhwahyejung-tang (GHT) on Alcohol-induced Oxidative Stress in Rats

  • Objectives: This study was aimed to investigate the effects of Galhwahyejung-tang (GHT) on alcohol-induced oxidative stress in rat model. Methods: Twenty SD rats were orally administrated with 40% ethanol (mL/kg) combined with GHT (50, 100, 200mg/kg) or distilled water for 2 weeks. Biochemistry in blood, malondialdehyde (MDA), total reactive oxygen species (ROS), and total antioxidant capacity (TAC) in serum, liver, brain, and kidney were determined. Results: GHT treatment significantly ameliorated the alcohol-induced alteration of hepatic enzyme; especially AST and ALT. GHT treatment also ameliorated the increase of MDA in liver, ROS level in serum and brain. GHT treatment reduced the depletion of antioxidant capacity in serum and brain. Conclusion: These results that GHT has antioxidant properties explaining the relevance of clinical application and its partial mechanisms of GHT.

The Extract of Limonium tetragonum Protected Liver against Acute Alcohol Toxicity by Enhancing Ethanol Metabolism and Antioxidant Enzyme Activities

  • The protective effect of EtOAc fraction of Limonium tetragonum extract (EALT) against alcohol-induced hepatotoxicity was assessed following acute ethanol intoxication in Spraque-Dawley rats. EALT (200 mg/kg p.o.) was administrated once before alcohol intake (8 g/kg, p.o.). Blood ethanol concentration, and the activities of alcohol metabolic enzymes, alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) in the liver were measured. Also, the formation of malondialdehyde (MDA) and the activities of antioxidant enzymes, superoxide dismutase (SOD), glutathione peroxidase (GSH-px), catalase were determined after acute alcohol exposure. Pretreatment of rats received ethanol with EALT significantly decreased blood ethanol concentration and elevated the activities of ADH and ALDH in liver. The increased MDA level was decreased, and the reduced activities of SOD, GSH-px and catalase were markedly preserved by the treatment with EALT. This study suggests that EALT prevent hepatic injury induced by acute alcohol which is likely related to its modulation on the alcohol metabolism and antioxidant enzymes activities.

Effects of Houttuynia cordata Thunb on Lipidperoxide and Cholesterol in 2,3,7,8-TCDD-damaged Rats (2,3,7,8-tetrachlorodibenzo-p-dioxin(TCDD)로 처치된 흰쥐에서 어성초가 과산화지질 및 콜레스테롤에 미치는 영향)

  • TCDD, one of the notorious toxic environmental pollutants, damages various organs including liver and is regarded as an endocrine disrupter. To investigate the effects of Houttuynia cordata Thunb (HCT) on the biochemical parameters of function, liver and serum of TCDD-treated rats were used. Seven days after the injection of TCDD (1 $\mu\textrm{g}$/kg), HCT (200 mg/kg) was administered to rats on every other day for four weeks. The lipidperoxide activity was examined by measuring the level of total cholesterol, HDL-cholesterol, LDL-cholesterol, total lipid and triglyceride (TG) in serum, and malondialdehyde (MDA) in liver tissue of rats. Result showed that lipidperoxidation was inhibited In the significant level when 2,3,7,8-TCDD-damaged rats were treated with HCT.
