The researchers studied the composition of latent buds by surveying the embryo of harvested seed, and the developmental pathway of latent buds by analyzing the characteristics of local ginseng. One-year-old ginseng seedlings were transplanted into the field and harvested two years later. The developmental pathway of the main bud, which would be the shoot of ginseng in the fourth year, was also investigated. The main bud of the seedling was formed from the region between root and shoot of the germinating seed. Primary and axillary latent buds protruded in a dome-shape from the cortex and separated from the main bud. Ninety percent of the single main bud was derived from the primary latent bud. Twin main buds were derived from a primary latent bud and one axillary bud, and triple main buds were derived from primary latent bud and two axillary latent buds. In the field, the researchers could not find 2-stem plants in 2-3 years old plants. However, the researchers found a 2-stem plant in a 4-year-old plant because its two main buds developed from a 3-year-old plant. We can conclude that a 2-stem plant was observed in the plant that was at least 4 years old. The main buds of the 4-year-old plant were formed at the primary and axillary latent bud of seed and cortex, the latent bud of rhizome in a 2-3 year old plant. In older plants, twin and triple main buds were derived mostly from the cortex latent bud than the primary latent bud.

This study investigated the effective harvest of 'Kyoho' grapevines by examining their characteristics including bud development, necrosis types and flower primordium formation. The size of the axillary bud did not show any difference in the 5th node or more, but it was smaller because it was closer to the base in the 4th node or less. In the 1st node, the rates of main bud necrosis (MBN), accessory bud necrosis (ABN), and whole bud necrosis (WBN) were high, and the rate of flower primordium formation was low, but there was no significant difference in the other nodes. Therefore, it was expected that using other nodes than the 1st node would be advantageous to secure production. Because the growth progresses after sprouting, the main bud necrosis rate increases, showing a very low flower primordium formation rate in March of the following year. Therefore, a method is needed to increase the storage nutrients in the winter and the rate of flower primordium formation after March. This study found that the thickness of the shoots should be less than 8.5 mm between the 3rd and 4th nodes, and the length should be less than 60 cm for nodes up to the 10th node.
In this study, we investigated the cumulative effect of low temperature on bud dormancy release and bud break characteristics in 'Campbell Early' grapevine (Vitis labruscana B.) cuttings grown in water culture. Additionally, we observed the development of buds while exposed to low temperatures in an attempt to improve our understanding of dormancy and bud break. The shoots were collected 120 days after full bloom (DAFB; leaf abscission period), and the accumulated chill unit (CU) value was calculated by reducing the temperature to $7.2^{\circ}C$ at 125 DAFB. The rate of bud break was 100% in shoots collected at 150 DAFB, The period until the first bud break was two times longer than in the shoots collected 165 DAFB, and bud break speed was significantly reduced. These results indicate that buds are released from endodormancy after 165 DAFB, because at this point the bud break was complete (bud break rate 100%) and it occurred in a very short time period. During this period, when the low-temperature accumulated value was 321h and 442CU according to the CH and Utah models, respectively. Furthermore, the survival rate of main buds decreased rapidly after 165 DAFB, and survival rate of accessory buds was maintained at more than 90% without seasonal differences. The rate of flower bud formation of main buds was much higher than in accessory buds (1:0.23) before the release from endodormancy at 150 DAFB. The final ratio of accessory buds to main buds was high, 1:1.54, at 255 DAFB. Correlation analysis of each investigated factor revealed that bud survival rate and bud formation rate were related only for the main buds, and there was a close relationship between the survival rate of main bud and time. In addition, the survival rate of main buds was positively correlated to the rate of flower bud formation.
This study aims to identify the cause for low appearance of flowers in 'Heukgoosul' grapevines, and determine basic characteristics of the axillary buds and types of necrosis in order to find out their influence on necrosis. As for $1^{st}$ to $4^{th}$ node, the closer buds were located to cane, the smaller the size of buds became. However, there was no difference in the size of an axillary bud in the upper part of the shoot above the 4th node. The bud necrosis occurrence in the $1^{st}$ node was the highest with 32% while buds in $4^{th}$ to $10^{th}$ node were normal with 84~96%. The size of buds in July was the largest with 6.40 mm, while buds showed no difference from August to October. The bud necrosis and main bud necrosis occurred most frequently in October, however accessory bud necrosis took place from July demonstrating no difference since then. The analysis on the relationship between shoot vigor and necrosis of axillary buds showed that the shoot diameter and internodes' length have no co-relationship with axillary bud necrosis, but there were negative relationship between the size of buds and necrosis occurrence, which was the most related to accessory bud necrosis. Therefore, despite the low occurrence of bud necrosis and healthy buds in 'Heukgoosul' grapevines, there was little appearance of flowers in the grapevines, which was attributable to the necrosis in the first bud. In conclusion, the research suggests spur pruning with three nodes kept intact yield.

The germination of the buds in 'Hongisul' grapes is poor and especially the high ratio of shoots where the flowers failed to emerge it was difficult to secure a sufficient amount of harvest. Thus, in this study, the necrosis and the periodical change in the buds were observed morphologically and also the cause of necrosis of the bud was investigated in order to understand what was causing the low germination and flowering. There was no change in the size of the bud after June and based upon external observation, there wasn't any retrogression or withering. However, based on a microscopic examination, the ratio of whole bud and main bud necrosis of the 'Hongisul' grapes continued to increase after August, and specially the ratio of the main bud necrosis continued to increase up to October. As for the size of the buds on the shoots, the buds located on the $1^{st}-3^{rd}$ nodes from the basal part were small whereas the buds located on the $4-10^{th}$ nodes were comparatively larger in its size. The ratio of necrosis of the bud was the highest at the $1^{st}$ and $2^{nd}$ bud, meaning that the buds located at the basal part of the shoot were defective compared to those located at the end of the shoot. It was also found that when the growth of the shoot is active and the shoot diameter is thick, it hindered the development of the buds. Therefore, it was judged that long pruning of 'Hongisul' grapes would help the emergence of the flowers as you could use the buds that have comparatively developed better.
This study was carried out to study the effect of emergence type of multiple stem and main bud on the quality of fresh and red ginseng in Korean ginseng. To achieve the aim of this study, characteristics of roots and quality factors were investigated. Single stem plants were 62.9% of the total samples, and the remainder(37.1%) were multiple stem plants. The number of stems affected considerably on root weight. Root weight of triple stem type with triple main bud was the largest among the types. As the number of stem per plant increased, the root quality became worse. In both single and multiple stem, the more the number of main buds was, the lower the quality grade was. Yield of the red ginseng was about 30%, showing little difference between single stem and multiple stem plant. As the number of stem increased, yield of Bonsam decreased. The quality grade of red ginseng of single stem was better than that of multiple stem. Multiple stem plant produced relatively more Yangsam and Japsam. As the number of main bud increased, the quality grade of ginseng decreased.
Background: After the Fukushima Daiichi Nuclear Power Plant (FDNPP) accident, biological alterations in the natural biota, including morphological changes of fir trees in forests surrounding the power plant, have been reported. Focusing on the terminal buds involved in the morphological formation of fir trees, this study developed a method for estimating the absorbed radiation dose rate using radionuclide distribution measurements from tree organs. Materials and Methods: A phantom composed of three-dimensional (3D) tree organs was constructed for the three upper whorls of the fir tree. A terminal bud was evaluated using Monte Carlo simulations for the absorbed dose rate of radionuclides in the tree organs of the whorls. Evaluation of the absorbed dose targeted 131I, 134Cs, and 137Cs, the main radionuclides subsequent to the FDNPP accident. The dose contribution from each tree organ was calculated separately using dose coefficients (DC), which express the ratio between the average activity concentration of a radionuclide in each tree organ and the dose rate at the terminal bud. Results and Discussion: The dose estimation indicated that the radionuclides in the terminal bud and bud scale contributed to the absorbed dose rate mainly by beta rays, whereas those in 1-year-old trunk/branches and leaves were contributed by gamma rays. However, the dose contribution from radionuclides in the lower trunk/branches and leaves was negligible. Conclusion: The fir tree model provides organ-specific DC values, which are satisfactory for the practical calculation of the absorbed dose rate of radiation from inside the tree. These calculations are based on the measurement of radionuclide concentrations in tree organs on the 1-year-old leader shoots of fir trees. With the addition of direct gamma ray measurements of the absorbed dose rate from the tree environment, the total absorbed dose rate was estimated in the terminal bud of fir trees in contaminated forests.
Growth hadit and demograph in a mature population of polygonatum and polygonatum humile in kanghwa island were studied for two growing seasons. Shoots of two species emerged early spring from the growing apices of the underdground rhizomes which had persisted for up to 1~5 year.after flowering, the ramets produced two rhizome buds at the shoot base. Bacause of the apical dominance in the rhizome system, a new rhizome was developed from only one bud, an actual bud,and the other latent buds were suppressed sothat remained dormant. The latent dud produced a new rhizome only when the actual dud was severed by the herbivores or by the physical obstacles. Therefore, the ramet number is not increased by the new rhizome from the latent bud. however, new ramets dould sometimes grow from latent buds which had been produced more than a year ago. Production of these ramets was main means increasing the ramet numbew and widening the potential zone of exploitation. Changes in size class of each ramet were noyiceable after a tear in small size-classes. Small ramets replaced themselves with larger-sized ramet, while large ramets with similar-sized or smallar-sized ramets. ramet numbers were average 0.82and 1.14 times of those fromthe previous year inp. involucratum and p. humile, though there was between-site variation. Almost all the ramets in the quadrats were alive during the growing season. when the entire rhizome systems were excavater next spring, there were many rhizomesegments without shoots, especially in p. involucratum. therefore, the drcrease of ramet number in p. involucratum in probably due to the climatic factors of winter.

This study was carried out to examine the base temperature to flowering and the average days to flowering by accumulated hours of low temperature ($5.0^{\circ}C$) or changing temperature after bud breaking. Over-all, the prediction of flowering time in the commercial apple cultivars ('Fuji' and 'Tsugaru') and apple cultivars ('Chukwang', 'Gamhong', 'Hongan', 'Honggeum', 'Hongro', 'Hongso', 'Hwahong', 'Summer dream', 'Sunhong') bred in Korea at the Gunwi region for 4 years (from 2009 to 2012) was investigated. Also, this study estimated the flowering time when the air temperature of Gunwi region rises at $5.0^{\circ}C$ was investigated using the same data. The range of accumulated hours of low temperature (chilling requirement) was from 0 hour to 1,671 hours, and the range of high temperature (heat requirements) to flowering after low temperature treatment was from $5.0^{\circ}C$ to $29.0^{\circ}C$. The treatments of changing temperature after bud breaking were classified as constant temperature treatment (control) and $5.0{\sim}10.0^{\circ}C$ elevation or descent treatments. The results show that the average days to flowering was longer with shorter accumulated hours of low temperature, and the average days from bud breaking to flowering of 0 hour treatment was longer about 2~4 weeks than that of 1,335~1,503 hours treatments. In comparing to apple cultivars, the all cultivars were not flowered under $10.0^{\circ}C$ after bud breaking, and the cultivars with low chilling requirements needed low heat requirements for flowering. The average days to flowering of treatments that the air temperature after bud breaking was controlled about $15.0^{\circ}C$ was shorter about 1~3 weeks than that of treatments was controlled about $10.0^{\circ}C$. In the treatment of changing temperature after bud breaking, the average days from bud breaking to flowering of temperature elevation treatment was shorter than that of constant temperature treatment. By use of these results, the base temperature to flowering of main apple cultivars in Korea was seemed to $10.0^{\circ}C$, and if the air temperature of Gunwi region rises about $5.0^{\circ}C$ than that of current, the flowering time was estimated to be delayed by 1 week.

Plant biotechnology involving genetic modification has been rather controversial. However, the major issues related to safety are being addressed by continued improvements in technology. Some of the related facts will be highlighted to set the tone for a scientific discussion on the possibilities of using the technology for crop improvement. Our main research interest is to understand the molecular regulation of shoot bud regeneration in plant tissue culture, which is essential for crop improvement by biotechnology. We have isolated and characterized some genes that are associated with adventitious shoot regeneration. These include a MADS-box cDNA (PkMADS1) from paulownia kawakamii, which regulates vegetative shoot development and in vitro shoot regeneration from leaf explants. Another gene we have characterized from petunia codesfor a cytokinin binding protein (PETCBP). Preliminary functional analysis of this gene indicated that this also affects adventitious shoot bud initiation. Also, the antisense suppression of this gene in petunia causedexcessive branching. Results from our work and selected other publications will be used to highlight the possibilities of manipulation of such genes to improve crop species.
