Effects of Hydrocortisone Administrations on Expressions of Casein and Prolactin Receptor mRNAs in Mammary Glands of Mid-Lactation of Korean Goats (Hydrocortisone 투여가 비유중기 재래산양의 유단백질과 유선세포 Prolactin Receptor mRNA 발현에 미치는 영향)

  • Glucocorticoid is activating mammary gland cells for lactating animals, resulting in increasing abilities of the milk synthesis. Expression of the prolactin receptor(PRL-R) in mammary gland cells was closely associated with milk production. To increase lactation ability for the Korean Native Goats at mid-lactation period. 0.05, 0.1. and 0.2 g of hydrocortisone was administrated with 5 $m\ell$ of saline. and injected into vein. For the control, 5 $m\ell$ of saline was administrated in to vein. After 24 H, the mammary gland tissue was collected, and mRNA expression rates were investigated for the alpha-casein and PRL-R using competitive PCR(polymerase chain reaction). There was no significant differences between treatment and control groups for the mRNA expression rate of PRL-R in mammary gland cells after 24 h of administration of hydrocortisone. The rate of mRNA expression for the alpha-casein was increased 37%, 630%, and 380% at 0.05, 0.1, and 0.2 g of hydrocortisone administration groups, respectively, comparing with control group. The results suggested that PR L-R mRNA expression of mammary gland cell by administration of hydrocortison was not significant, but increase of the alpha-casein mRNA expression my be differences of expression of functional proteins in the cell and expression patterns of protein secretion time to out of the cell. This study showed increase of alpha-casein mRNA expression by administration of hydrocortisone at mid-lactation period of Korean native goat.

Effect of Vitamin C, Silicon and Iron on Collagen Synthesis and Break-Down Enzyme Expression in the Human Dermal Fibroblast Cell (HS27) (피부 섬유아세포에서 비타민 C, Silicon, 철분 처리가 콜라겐 합성 및 분해 관련 효소의 발현에 미치는 효과 비교)

  • Collagen is the major matrix protein in dermis and consists of proline and lysine, which are hydroxylated by prolyl hydroxylase (PH) and lysyl hydroxylase (LH) with cofactors such as vitamin C, oxygen, iron (Fe$^{2+}$), ketoglutarate and silicon. The collagen degradation is regulated by matrix metalloproteinase-1 (MMP-1), of which is the major collagen-degrading proteinase whereas tissue inhibitors of metalloproteinase-1 (TIMP-1) bind to MMP-1 thereby inhibiting MMP-1 activity. In this study, we investigated the effects of vitamin C, silicon and iron on mRNA, protein expressions of PH, LH, MMP-1 and TIMP-1. The physiological concentrations of vitamin C (0-100 $\mu$M), silicon (0-50 $\mu$M) and iron (Fe$^{2+}$:0-50 $\mu$M) were treated to human dermal fibroblast cells (HS27 cells) for 3 or 5days. The expression level of mRNA and protein was increased in not only PH but also LH when cells were incubated with vitamin C. A similar increase in LH mRNA or protein expression occurred when cells were incubated with silicon. Our results suggest that treatment of vitamin C and silicon increased mRNA and protein expression of PH and LH in human dermal fibroblast.

Identification of Steroidogenic Acute Regulatory Protein mRNA in the Rat Ovary and Adrenal G land (흰쥐 난소 및 부신에서 Steroidogenic Acute Regulatory Protein mRNA의 발현에 관한 연구)

  • The synthesis of steroid hormone starts from cholesterol. Steroidogenic acute regulatory protein(StAR) transfers cholesterol acutely from the outer mitochondrial membranes to the inner in the early step of steroidogenesis. Many kinds of steroid hormones are mainly synthesized in adrenal grand, ovary and testis. The purpose of this study is to determine the distribution of StAR mRNA in the rat ovary and adrenal gland and to confirm the functions of StAR in these organs. In the ovary, StAR mRNAs were strongly expressed in the corpus luteum, where progesterone is synthesized, and these were weakly expressed in the theca layer of follicles, where androgen is synthesized. However, StAR mRNAs were not detected in the estrogen producing granulosa cells of growing follicles. In the corpus luteum, StAR mRNAs were strongly loclized in the zona fasciculata and zona reticularis, where glucocorticoid is mainly synthesized. StAR mRNAs were weakly expressed in the zona gromerulosa, where mineralcorticoid is synthesized. StAR mRNAs were not detected in the adrenal medulla. In our results, StAR mRNAs were expressed differentially in the steroidogenic cells of ovary and adrenal gland according to the types of steroid hormones, and the statges of corpus luteum development. We conclude that StAR is involved in the steroidogenesis at the very early step of steroid synthesis cascade.

  • 유전자의 발현이 다양한 형태로 억제되는 것을 silencing이라고 한다. 유전자 발현억제 방법 중 antisense RNA는 자연 상태의 mRNA에 역상보적인 RNA 분자로서 형질전환된 세포에서 그 mRNA의 전이을 억제하는 데 이용된다. RNA분자에 대하여 상보적 염기배열을 갖는 RNA는 분자간 결합을 연결하여 RNA의 기능 발현에 억제적으로 작용한다고 생각된다. 미생물에 나타나는 유전자발현 제어기작으로서 어느 특정한 mRNA에 대하여 상보적인 RNA가 유전자 발현의 억제인자로서 작용하고 있는 예가 몇 가지 알려져 있다. 이러한 경우 antisense RNA는 mRNA 상의 전이개시영역과 상보적 배열을 하고 있고, 전이과정을 방해한다고 추정되고 있지만 작용기작의 상세한 내용은 아직 명확하지가 않다. 한편 안티센스RNA는 임의의 표적유전자에 대하여 인위적으로 제작할 수가 있기 때문에 인위적인 유전자발현제어의 한 방법으로 이용되고 있다. 특정한 유전자에 대한 antisense RNA를, 발현하는 유전자를 인위적으로 제작하여 세포내에 도입하면 표적유전자의 발현을 특이적으로 억제 제어할 수 있는 것이 기대되어, 다양한 생물체를 대상으로 하여 많은 시도가 이루어지고 있으며 몇 가지 성공적인 보고가 있다. 그 중 식물이차대사과정에 관련 유전자를 대상으로 antisense RNA 기법으로 유전자의 발현억제와 이차대사산물 생산조절에 관한 연구를 이 논문에서 조사하고 정리하였다.

Interleukin-8 gene expression in the human colon epithelial cell line, HT-29, exposed to Entamoeba histolytica (이질아메바에 의한 인체 대장상피세포주 HT-29에서의 interleukin-8 유전자의 발현)

  • The protozoan parasite, Entcmoeba histoIWticc, is one of major causative agents of intestinal disease all over the world. In acute experimental infection, the early host response to 5. histoIHtica is characterized by an infiltration of neutrophils. However, the chemotactic signal for this response is not well known. Based on the (jading that human epithelial cells produce the potent neutrophil chemoattractant and activator, interleukin-8 (IL-8), IL-8 gene expression was examined thoroughly in human colon epithelial cells exposed to 5. histolvtica trophozoites. Cellular RNAs were extracted from HT-29 or Caco-2 human colon epithelial cells exposed to 5. histoLvtica trophozoites for 30 minutes, 1 and 3 hours. IL-8 mRNA transcripts were measured by reverse transcriptional polprnerase chain reaction (RT-PCR) using synthetic standard RNA. The number of IL-8 mRNA molecules increased from 30 minutes to 3 hours of exposure period, reaching 3.1 H 107 molecules/ug of total RNA. Expression pattern of IL-8 mRNA transcripts was parallel to the amounts of IL-8 protein measured by enzyme-linked immunosorbent assay (ELISA) . Lysates of 5. histoIVtica also induced expression of mRNA for IL-8 in colon epithelial cells. These results sugf:esc that acute inflammatory reaction by 5. histoIVticc may be initially triggered by proinflammatory cytokines such as IL-8 secreted from epithelial cells of the colon.

THE EFFECT OF XYLITOL ON THE EXPRESSION OF GTF GENE (gtf 유전자 발현에 대한 xylitol의 영향)

  • Xylitol, a five-carbon natural sugar alcohol, is widely used non-cariogenic sugar substitute. In present study, the effects of xylitol on the expression of mRNA for glucosyltransferase which synthesizes glucan from sucrose were detected by Fluorescent in situ hybridization (FISH) and flow cytometry. FITC fluorescences for mRNA of gtfB, gtfC and gtfD were decreased further with increasing concentration of xylitol from 1% to 10% when detected by FISH. Flow cytometric analysis also showed that the expression of gtfB, gtfC and gtfD was increased by the addition of sucrose and decreased by the addition of xylitol to BHI broth containing 1% sucrose. In conclusion, the expression of gtfB, gtfC and gtfD mRNA was decreased by the addition of xylitol.

  • The effect of radiofrequency radiation (RF) exposure on mouse associated with the exercise was investigated in the brain at the molecular level. The expression of tyrosine hydroxylase(TH), FoxO3a, AMPKα and mRNA was investigated by real-time RT-PCR in striatum and the hypothalamus. In the striatum, TH mRNA expression was decreased in the exercise and RF exposure group. FoxO3a mRNA expression was significantly increased in the spontaneous exercise group and a significant decrease was observed in the RF exposure and spontaneous exercise group. In the hypothalamus, TH mRNA expression was significantly decreased in the RF exposure and spontaneous exercise group. But, FoxO3a mRNA expression was significantly increased in the RF exposure and spontaneous exercise group. We will further investigate the expression of protein molecules in the hippocampus of the brain to reveal the effects of RF radiation on memory.

  • Insoluble glucan is the important component of oral biofilm, which is synthesized from sucrose through the action of glucosyltransferase (GTF) B and GTF C encoded by the gtfB and gtfC genes, respectively of Streptococcus mutans. In present study, the effects of various sugars on the mRNA expression of gtfB and gtfC of S. mutans Ingbritt were examined by fluorescent in situ hybridization (FISH). The mRNA of gtfB and gtfC was expressed normally in the BHI broth containing 1% and 5% sucrose. The mRNA expression was decreased by the addition of 10% of glucose, and 1%, 5% and 10% of fructose. Lactose had no great effect on the expression of gtfB and gtfC. 5% and 10% of xylitol greatly reduced the mRNA expression of gtfB and gtfC. Sorbitol slightly decreased the mRNA expression of gtfB and gtfC when compared to the control. In summary, mRNA expression of gtfB and gtfC was decreased by the addition of glucose, fructose, and xylitol.

Effects of compressive stress on the expression of M-CSF, IL-$1{\beta}$, RANKL and OPG mRNA in periodontal ligament cells (압박력이 치주인대 세포의 M-CSF, IL-$1{\beta}$, RANKL 및 OPG mRNA 발현에 미치는 영향)

  • Objective: The aim of this study was to determine if human PDL cells can produce osteoclastogenic mRNA and examine how compressive stress affects the expression of osteoclastogenic mRNA in human PDL cells. Methods: Human PDL cells were obtained from biscupids extracted for orthodontic treatment. The compressive force was adjusted by increasing the number of cover glasses. PDL cells were subjected to a compressive force of 0.5, 1.0, 2.0, 3.0 or $4.0\;g/cm^2$ for 0.5, 1.5, 6, 24 or 48 hours. Reverse transcription polymerase chain reaction (RT-PCR) analysis was performed to examine levels of M-CSF, IL-$1{\beta}$, RANKL, OPG mRNA expression. Results: Human PDL cells could produce M-CSF mRNA. Human PDL cells under compressive stress showed increased M-CSF, IL-$1{\beta}$ and RANKL mRNAs expression in a force (up to $2\;g/cm^2$) and time-dependent manner. However, OPG mRNA expression was constant regardless of the level and duration of stress. Conclusions: Continuous compressive stress induced the mRNA expression of osteoclastogenic cytokines including M-CSF, RANKL, IL-$1{\beta}$ in PDL cells. Together with an unchanged OPG mRNA level, these results suggest that compressive stress-induced osteoclastogenesis in vivo is partly controlled by M-CSF, RANKL and IL-$1{\beta}$ expression in PDL cells.

IFN-${\gamma}$mRNA Expression in Tuberculous Pleural Lymphocytes After in vitro Stimulation with M. tuberculosis Antigens (결핵균 항원 자극에 의한 결핵성 흉수 림프구의 IFN-${\gamma}$ mRNA 발현)

  • Background : IFN-${\gamma}$ is the main effector mediator of the host immune response against Mycobacterium tuberculosis. Evaluating the IFN-${\gamma}$ gene expression in response to M. tuberculosis antigens may help in elucidating the host defense mechanism against M. tuberculosis and in the development of a vaccine. Methods : The IFN-${\gamma}$ mRNA expression in the lymphocytes obtained from pleural effusions from tuberculous pleurisy patients (TB-PLC) after in vitro stimulation with whole cell M. tuberculosis(H37Rv), purified protein derivatives(PPD), man-lipoarabinamman (man-LAM), ara-LAM and Antigen 85B(Ag85B) were evaluated. The degree of IFN-${\gamma}$ mRNA expression was determined by a semiquantitative reverse transcriptase-polymerase chain reaction (RT-PCR) method. Results : M. tuberculosis induced the expression of IFN-${\gamma}$ mRNA in the TB-PLC in time and dose dependent manners. The PPD and Ag85B induced high levels of IFN-${\gamma}$ mRNA expression in the TB-PLC. However, man-LAM inhibited IFN-${\gamma}$ mRNA expression in the TB-PLC, while ara-LAM did not. Conclusion : IFN-${\gamma}$ mRNA expression in TB-PLC is stimulated by PPD and Ag85B, but inhibited by man-LAM.
