  • The characteristics of high heat input (342kJ/cm) EG (Electro Gas Arc) welded joint of EH36-TM steel has been investigated. The weld metal microstructure consisted of fine acicular ferrite (AF), a little volume of polygonal ferrite (PF) and grain boundary ferrite (GBF). Charpy impact test results of the weld metal and heat affected zone (HAZ) met the requirement of classification rule (Min. 34J at $-20^{\circ}C$). In order to evaluate the relationship between the impact toughness property and the grain size of HAZ, the austenite grain size of HAZ was measured. The prior austenite grain size in Fusion line (F.L+0.1 mm) was about $350{\mu}m$. The grain size in F.L+1.5 mm was measured to be less than $30{\mu}m$ and this region was identified as being included in FGHAZ(Fine Grain HAZ). It is seen that as the austenite grain size decreases, the size of GBF, FSP (Ferrite Side Plate) become smaller and the impact toughness of HAZ increases. Therefore, the CGHAZ was considered to be area up to 1.3mm away from the fusion line. Results of TEM replica analysis for a welded joint implied that very small size ($0.8\sim1.2{\mu}m$) oxygen inclusions played a role of forming fine acicular ferrite in the weld metal. A large amount of (Ti, Mn, Al)xOy oxygen inclusions dispersed, and oxides density was measured to be 4,600-5,300 (ea/mm2). During the welding thermal cycle, the area near a fusion line was reheated to temperature exceeding $1400^{\circ}C$. However, the nitrides and carbides were not completely dissolved near the fusion line because of rapid heating and cooling rate. Instead, they might grow during the cooling process. TiC precipitates of about 50 ~ 100nm size dispersed near the fusion line.

  • This study was conducted to investigate methane production, emission and diffusion under organic matter application in paddy soil (Jeonbug Series). The rates of application of rice straw were 5,000kg/ha in combination with 110kg N/ha as chemical fertilizer. In seasonal variations of the $CH_4$ emission rates two maxima were found during the ear formation stage and the heading stage of the rice plant. Entrapped methane increased dining the early growing season, declined thereafter and especially increased during the heading stage. Methane concentration in the soil solution was the higest at 5cm depth, but decreased with upper and lower depth. The gas diffusion rate of $CH_4$ was very slow in the liquid phase. Eh of soil solution varied from -150~-160mV and methane prodution rate was highly correlated with Eh.

  • A new L-isoascorbic acid derivative epi-leptosphaerin (1) and two known compounds leptosphaerin (2), and verongamine (3) were isolated from sponges of the orders Verongida and Thorectidae. Compounds 1 and 2 are most likely of sponge-associated fungal origin. In the present study, isolated compounds were investigated for their inhibition of soluble epoxide hydrolase (sEH), which is considered a promising target for the management of pain, inflammation, and comorbidities associated with diabetes. Compound 3, verongamine, displayed weak inhibitory activity against sEH with an $IC_{50}$ value $51.5{\pm}1.0{\mu}M$.

  • Acoustic microscopy and scanning electron microscopy were applied to non-destructively evaluate the hydrogen-induced cracking of API X-80 steels and to find the initiation time of the crack. The API X-80 steel had the average grain size of about $4-10{\mu}m$. The hardness was reduced from 240 to 202 [Hv] after exposing in HIC environment for 2-days. Friction coefficient and wear loss were 0.745 and 0.392 mm, respectively. Empirical equation of corrosion potential and corrosion rate of the steel with HIC time in $5%NaCl-0.5%CH_3COOH$ at $25^{\circ}C$ were $Eh\;(up)=0.06^*t[day]+0.2951$, $Eh(down)=0.376^*t[day]+0.5938$, respectively. HIC grew with micro-size after 1-day exposure. The HIC tended to propagate on the surface with Al, Si, Ti, and Mn.

  • The microsomal epoxide hydrolase gene (referred to as mMCEH) of Mugil cephalus was cloned by PCR, and then inserted to pColdI and pET-21b(+) vector, respectively. The recombinant E. coli possessing the recombinant plasmids exhibited the enantioperference toward (R)-styrene oxide. When enantioselective kinetic resolutions were conducted with 20 mM racemic styrene oxide, enantiopure (S)-styrene oxide was obtained with high enantiopurity more than 99% enantiomeric excess (ee) and 24.50% yield by using the recombinant E. coli harboring pET-21b(+)/mMCEH.

  • We reported previously that glucagon decreased alpha- and pi-class glutathione S-transferases (GSTs) and microsomal epoxide hydrolase (mEN) protein levels in primary cultured rat hepatocytes. The present study examines the effects of Protein kinase A (PKA) inhibitor, KT5720, on the glucagon-mediated decrease in expression of GSTs and mEN. To assess cell viability. lactate dehydrogenase release and MTT activity were examined in hepatocytes treated KT5720. Cell viability was significantly decreased in a concentration dependent manner after incubation with KT5720 at the concentrations of 1 $\mu$M or above for 24 h, which was inhibited by the cytochrome P450 inhibitor SKF-525A. In contrast, another PKA inhibitor H89 (up to 25 $\mu$M) was not toxic to hepatocytes. The glucagon-mediated decrease in expression of alpha- and pi-class GSTs and mEH was completely inhibited by 25 $\mu$M H89 and attenuated by 0.1 $\mu$M KT5720. This study demonstrates that KT5720 may cause cytotoxicity in rat hepatocytes through cytochrome P450-dependent bioactivation. The present study implicates PKA in mediating the inhibitory effect of glucagon on expression of alpha- and pi- class GSTs and mEH.

  • Field water qualities (temperature, pH, Eh, EC, DO) was monitored by 6 times March to September 2009 on background water (BW) and acid mine drainage (AMD0, AMD1, AMD2 and AMD3 points), and flow rate was measured on AMD0 point. Acid mine drainage flowed out from abandoned Ilgwang mine were high acid waters that lower than pH 3, and Eh component was ranged 400 to 600 mV. EC measured on acid mine drainage were higher over 10 times than background water, DO component was increased by reaction on the air during the water flow from AMD0 point to AMD4 point. Heavy metal concentrations in acid mine drainage were ordered Fe > Cu > Zn > Mn > As > Cd, and Fe concentration was highest for 81.870~474.30 mg/L. Monitoring periods measured maximum concentrations of heavy metals were May for As and Cd, June for Fe, July for Cu, Zn and Mn. The periods measured minimum concentrations were monitored April for Cd and Mn, September for Fe, Cu, Zn and As. Discharge mass of heavy metal components were calculated 53.44 kg for Fe, 6.25 kg for Cu, 5.26 kg for Zn, 2.13 kg for Mn, 0.14 kg for As and 0.04 kg for Cd, respectively. Total discharge mass of heavy metal components were calculated 67.26 kg for 1 day, and Fe component was taken 79% of total mass.

  • The purpose of this research was to investigate the characteristics and the cleaning efficiency on HN(sub)4OH solutions added with H(sub)2O(sub)2 and surfactants. NH(sub)4OH solutions added with surfactants did not show much changes in pH and redox potential (Eh) as a function of NH(sub)4OH concentration compared with NH(sub)4OH solution. However H(sub)2O(sub)2 added NH\ulcornerOH solutions showed the decrease of pH and the increase of Eh as the concentration of NH(sub)4OH increased. The decrease of surface tension from 72 dynes/cm to 38 dynes/cm was observed in solutions added with surfactant but not in H(sub)2O(sub)2. The etch rates of silicon in NH(sub)4OH solutions(NH(sub)4OH:H(sub)2O= 1 : 5) showed at least 50 times higher than those in H(sub)2O(sub)2 and surfactant added NH(sub)4OH solutions(NH(sub)4OH:H(sub)2O(sub)2= 1 : 1 : 5) solution removed the PSL particles (0.67$\mu\textrm{m}$ in diameter) on Si wafers effectively at all temperatures investigated. NH(sub)4OH solution added with a surfactant could not remove particles at room temperature, however it was possible to remove particles at higher temperatures, 5$0^{\circ}C$ and 8$0^{\circ}C$.
