계난백(鷄卵白) Lysozyme의 N-말단(末端)과 C-말단(末端) 항원결정기(抗原決定基)에 대한 연구(硏究) (Further Studies on the Specificity of the N- and C-terminal Antigenic Determinant of Hen Egg-white Lysozyme)

  • 계난백(鷄卵白) Lysozyme N-와 C 말단(末端) 항원결정기(抗原決定基)($P_{17}$: sequence $Lys^1-{cys-}^6-Asn^{27},\;{Trp^{12}}_2-Cys^{127}-Leu^{129}$)의 특이성(特異性)에 대(對)하여 연구(硏究)했다. $^{14}C-acetyl$ Lysozyme 과 정제(精製)한 guinea pig 항(抗)-$P_{17}$ 항체(抗體)와의 결합(結合)을 Scatchard plot 상(上)에서 나타낸 결과(結果) 그 실험치(實驗値)가 거의 r=1였다. 이것은 Lysozyme에 대(對)하여 각각(各各) 다른 친화성(親和性)을 가진 2개(個)의 항체군(抗體群)의 존재(存在)나 그렇잖으면 제(第)1의 항체결합부위(抗體結合部位)에 최초(最初)의 Lysozyme 분자(分子)가 결합(結合)함으로 인(因)하여 항체분자(抗體分子)의 제(第)2의 결합부위(結合部位)에 다른 Lysozyme 분자(分子)의 결합(結合)을 방해하는 즉 steric hindrance에 의한 가능성(可能性)을 시사(示唆)한다. 여러가지 peptides의 항원활성(抗原活性)을 $^{14}C-acetyl-P_{17}$과 항(抗)-$P_{17}$ 항체(抗體)와의 결합저해(結合沮害)시험에서 측정(測定)했다. 그 결과(結果) 단지 $P_{17}$$P_{17}t$(sequence $Lys^1-cys^5-Homoser^{12},\;Trp^{123}-cys^{127}-Leu^{128})$)만이 억제되었고 그 $K_1$치(値)가 각각(各各) $2.0{\times}10^4$$8.1{\times}10^3$이였다. 이들 결과(結果)를 종합(綜合)하면 $P_{17}$의 항(抗)-$P_{17}$ 항체(抗體)와의 직접적 결합부위(結合部位)는 $P_{17}$의 말단부위(末端部位)에 국재(局在)해 있는 것을 암시해 준다. 한편 $P_{17}$의 나머지 부분(部分)은 이 항원결정기(抗原決定基) 구조(構造)를 유지(維持)하는데 중요(重要)한 역할(役割)을 할 것으로 생각되며 또한 이 결정기내(決定基內)의 한 개의 disulphide 결합(結合)은 면역학적(免疫學的) 활성(活性)을 나타내는데 필수적(必須的)인 것으로 믿어진다.

A Long-day-stimulus Induced the Expression of c-Fos-like Molecules in the Hypothalamus of Japanese quail

  • In birds, the photoperiodic seasonal breeding involves encephalic photoreception at the initial step of triggering the well-known endocrinal cascade. Especially in Japanese quail (Coturnix coturnixjaponica), the reproductive neuroendocrine function responds to a single long day, and hypothalamic regions are known to be important for the reproductive response. However, little is known about where and how the light and time signals are integrated to detect daylength information and transduced to the endocrinal responses. To gain insights into this issue, we are interested in the c-Fos expression in the hypothalamus of the Japanese quail. Meddle and Follett (1997) previously identified two hypothalamic regions where c-Fos-like immunoreactivities were induced in response to a long day by using an antibody to carboxyl terminal region of human c-Fos (Lys$^{347}$ -Leu$^{367}$ ). In the present study, we used a different anti-c-Fos antibody recognizing a region from Lys$^{128}$ to Ala$^{152}$ of human c-Fos, and found in long-day- stimulated quails many c-Fos-like immunoreactive nuclei localizing within two regions, nucleus anterior medialis hypothalami and nucleus periventricularis hypothalami, which are distinct from those identified in the previous study. Then, we focused on the difference in the cross-reactivities of the antibodies used, and determined the whole coding sequence of quail c-Fos to compare the antigenic sequences of the two antibodies with the amino acid sequence of quail c-Fos. We found that the antibody we used would recognize quail c-Fos more specifically than that used in the previous study.

  • Alkaline .alpha.-amylase expressed in the transformant, Baciollus subtills MB809, containing alkaline amylase gene cloned from alkalophilic Bacillus sp. AL-8, was purified through for step separation processes. The purified alkaline .alpha.-amylase had molecular weight of app[roximately 59, 000 daltons on SDS-PAGE and Sephaex G-100 gel filtration. Amino acid sequence of terminal portion of the enzyme was analyzed with pure amylase eluted form the SDS-PAGE gel. N-terminal amino acid sequence of .alpha.-amylase was determined by the Edman degradation method and resulted in $NH_{2}$-ser-thr-ala-pro-ser-(ile)-lys-ala-gly-thr-(ile)-leu. For C-terminal amino acid sequencing, purified .alpha.-amylase was digested with carboxypuptidase A and B, and reverse-phase HPLC gradient elution system resulted in -thr-trp-pro-lys-COOH.

Studies on the active site of the Arabidopsis thaliana S-Adenosylmethionine Decarboxylase: $Lys^{81}$ residue involvement in catalytic activity

  • The Arabidopsis thaliana S-Adenosylmethionine decarboxylase (AdoMetDC) cDNA ($GenBank^{TM}$ U63633) was cloned, then the AdoMetDC protein was expressed and purified. The purified AdoMetDC was inactivated by salicylaldehyde in a pseudo first- order kinetics. The secondorder rate constant for inactivation was 126 $M^{-1}min^{-1}$ with the slope of n=0.73, suggesting that inactivation is the result of the reaction of one lysine residue in the active site of AdoMetDC. Site-specific mutagenesis was performed on the AdoMetDC to introduce mutations in conserved $lysine^{81}$ residues. These were chosen by examination of the conserved sequence and proved to be involved in enzymatic activity by chemical modification. Changing $Lys^{81}$ to alanine showed an altered optimal pH. The substrate also provided protection against inactivation by salicylaldehyde. Considering these results, we suggest that the $lysine^{81}$ residue may be involved in catalytic activity.

구연산 생성 Candida lipolytica의 원형질체 융합 (Intraspecific Protoplast Fusion of Citric Acid Producer, Candida lipolytica)

  • 내산성이 우수하고 citric acid와 SCP를 많이 생산할 수 있는 효모균주를 육종하기 위하여 구연산 생성효모인 Candide lipolytica의 종내 원형 질체융합 조건을 검토하였다. 배양기간과 효소처리조건을 최적화함에 의해 98%의 protoplast가 형성되었다. 3 % agar와 30mM $CaCl_2$를 함유한 재생용최소배지에 동일배지의 중층에 의해 약 20-30%의 protoplast가 재생되었다. 2개의 영양요구성이 상보적인 영양요구성원형질체, L-14($lys^-$)와 T-24($try^-$)에 100mM $CaCl_2$를 함유하는 30% PEG 6000을 $30^{\circ}C$ 에서 20분간 처리하여 4-5${\pm}$$10^{-4}$의 융합빈도가 얻어졌다.

Isolation, Cloning and Co-Expression of Lipase and Foldase Genes of Burkholderia territorii GP3 from Mount Papandayan Soil

  • Lipases are industrial enzymes that catalyze both triglyceride hydrolysis and ester synthesis. The overexpression of lipase genes is considered one of the best approaches to increase the enzymatic production for industrial applications. Subfamily I.2. lipases require a chaperone or foldase in order to become a fully-activated enzyme. The goal of this research was to isolate, clone, and co-express genes that encode lipase and foldase from Burkholderia territorii GP3, a lipolytic bacterial isolate obtained from Mount Papandayan soil via growth on Soil Extract Rhodamine Agar. Genes that encode for lipase (lipBT) and foldase (lifBT) were successfully cloned from this isolate and co-expressed in the E. coli BL21 background. The highest expression was shown in E. coli BL21 (DE3) pLysS, using pET15b expression vector. LipBT was particulary unique as it showed highest activity with optimum temperature of $80^{\circ}C$ at pH 11.0. The optimum substrate for enzyme activity was $C_{10}$, which is highly stable in methanol solvent. The enzyme was strongly activated by $Ca^{2+}$, $Mg^{2+}$, and strongly inhibited by $Fe^{2+}$ and $Zn^{2+}$. In addition, the enzyme was stable and compatible in non-ionic surfactant, and was strongly incompatible in ionic surfactant.

Allelic variation of melanocortin-1 receptor locus in Saudi indigenous sheep exhibiting different color coats

  • Objective: This study was designed to characterize the DNA polymorphisms of the melanocortin-1 receptor (MC1R) gene in indigenous Saudi Arabian sheep breeds exhibiting different color coats, along with individuals of the Sawaknee breed, an exotic sheep imported from Sudan. Methods: The complete coding region of MC1R gene including parts of 3' and 5' untranslated regions was amplified and sequenced from three the indigenous Saudi sheep; Najdi (generally black, n = 41), Naeimi (generally white with brown faces, n = 36) and Herri (generally white, n = 18), in addition to 13 Sawaknee sheep. Results: Five single nucleotide polymorphisms (SNPs) were detected in the MC1R gene: two led to nonsynonymous mutations (c.218 T>A, p.73 Met>Lys and c.361 G>A, p.121 Asp>Asn) and three led to synonymous mutations (c.429 C>T, p.143 Tyr>Tyr; c.600 T>G, p.200 Leu>Leu, and c.735 C>T, p.245 Ile>Ile). Based on these five SNPs, eight haplotypes representing MC1R $E^d$ and $E^+$ alleles were identified among the studied sheep breeds. The most common haplotype (H3) of the dominant $E^d$ allele was associated with either black or brown coat color in Najdi and Sawaknee sheep, respectively. Two other haplotypes (H6 and H7) of $E^d$ allele, with only the nonsynonymous mutation A218T, were detected for the first time in Saudi indigenous sheep. Conclusion: In addition to investigating the MC1R allelic variation in Saudi indigenous sheep populations, the present study supports the assumption that the two independent nonsynonymous Met73Lys and Asp121Asn mutations in MC1R gene are associated with black or red coat colors in sheep breeds.

Growth Performance, Carcass Characteristics, Nutrient Digestibility and Serum Biochemical Parameters of Broilers Fed Low-protein Diets Supplemented with Various Ratios of Threonine to Lysine

  • This study was conducted to evaluate performance, breast (thigh) muscle yield, nutrient digestibility and serum biochemical parameters of broilers fed low-protein diets supplemented with various threonine to lysine ratios (Thr/Lys). Three hundred and twenty four day-old male Arbor Acres broilers were randomly allotted to six dietary treatments with six replicates per treatment and nine broilers per replicate. Six treatments included one control diet [formulated according to NRC (1994)], and five experimental diets (treatments 1-5). In treatments 1-5, the crude protein level was 2% lower than that of the NRC control diet. The Thr/Lys in treatments 1-5 was 0.65, 0.70, 0.75, 0.80 and 0.85 during the starter and grower phases and 0.70, 0.75, 0.80, 0.85 and 0.90 during the finisher phase with other nutrient levels kept consistent. The results showed that: (1) With increasing Thr/Lys, ADG of treatments 1, 3 and 4 were lower (p<0.05) than those of the NRC control diet during the starter phase; ADG of treatments 1-5 were lower (p<0.05) than those of the NRC control during the grower phase, and ADG of treatment 1 was lower (p<0.05) than that of the NRC control during the finisher phase. Average daily feed intake in treatments 1 and 4 were lower (p<0.05) than that of broilers on the NRC control diet during the grower phase. Feed/gain (F/G) of the NRC control diet was lowest (p<0.05) during the starter phase. F/G in treatments 2 and 5 were higher (p<0.05) than that of NRC during the grower phase. (2) Breast muscle proportion on d 21 increased linearly (p<0.05) in response to graded levels of Thr/Lys. (3) No differences were detected for dry matter, energy and crude protein digestibility among all seven treatments. (4) On d 21, serum triglyceride of broilers on dietary treatments 3 and 5 was higher (p<0.05) than that of broilers in control. The results indicate that the low-protein diets supplemented with an appropriate Thr/Lys could not support the same performance that was achieved by the broilers fed the NRC control diet.
