Cloning, Expression, and Characterization of a Cold-Active and Organic Solvent-Tolerant Lipase from Aeromicrobium sp. SCSIO 25071

  • The gene encoding lipase (Lip98) from Aeromicrobium sp. SCSIO 25071 was cloned and functionally expressed in Escherichia coli. Lip98 amino acid sequence shares the highest (49%) identity to Rhodococcus jostii RHA1 lipase and contains a novel motif (GHSEG), which is different from other clusters in the lipase superfamily. The recombinant lipase was purified to homogeneity with Ni-NTA affinity chromatography. Lip98 showed an apparent molecular mass of 30 kDa on SDS gel. The optimal temperature and pH value for enzymatic activity were recorded at 30℃ and 7.5, respectively. Lip98 exhibited high activity at low temperatures with 35% maximum activity at 0℃ and good stability at temperatures below 35℃. Its calculated activation energy was 4.12 kcal/mol at the low temperature range of 15-30℃. Its activity was slightly affected by some metal ions such as K+, Ca2+, and Na+. The activity of Lip98 was increased by various organic solvents such as DMSO, ethanol, acetone, and hexane with the concentration of 30% (v/v) and retained more than 30% residual activity in neat organic solvent. The unique characteristics of Lip98 imply that it is a promising candidate for industrial application as a nonaqueous biocatalyst and food additive.

  • In this study, we estimated the performance of PEMFC's unit cell as changing operating temperature in different inlet humidity condition at cathode side but anode dry, and tried to match experimental results with 1-dimensional simulation. We used $Nafion^{\circledR}112$ membrane and a self-manufactured PEMFC with active area of $25cm^{2}$ was used in this study. The range of operating temperature was $40{\sim}70^{\circ}C$ and oxygen through bubbled humidity chamber was supplied $0{\sim}80$% humidity condition as changing water temperature in humidity chamber. For figuring out governing equations, represent water contents in electrolyte membrane, the linear forward difference method was applied about time progress and quadratic central difference method was used about space progress. It was assumed that pressure terms were linearly changed due to thin electrolyte membrane. In low operating temperature condition, $40{\sim}60^{\circ}C$, increasing temperature rarely effected cell performance but we can see performance drop at $70^{\circ}C$. By modifying Henrry's constant and/or diffusion coefficient, the modified one-dimensional model was accomplished for calculation.

Characteristic of Growth and Active Ingredient in Angelica gigas Nakai according to Forest Environment by Climate Zone (기후대별 산림환경에 따른 참당귀의 생육 및 지표성분 특성)

  • Background: Angelica gigas Nakai, that belong to the Umbelliferae family, is one of the traditional medicinal plants in Korea. Its roots have been used to treat gynecological diseases. In this study, growth characteristics and index components were compared with the forest microclimate at several forest sites. Methods and Results: A. gigas was planted in three climatic zones according to the temperature (southern temperature zone - Hamyang, central temperature zone - Bonghwa, and northern temperature zone - Jeongseon) and growth characteristics were investigated in comparison with the forest microclimate. Our results indicated that the root diameter and length, and fresh and dry weight were the highest in Jeongseon. The total content of decursin was the highest in Jeongseon (9.52%), followed by those in Hamyang (8.07%) and Bonghwa (7.48%), respectively. Additionally, the yield of decursin (1.39 g) was the highest in Jeongseon. Conclusions: The yield and index components were influenced by the microclimate in the forests, and it was assumed that high altitude and low temperature affected the increase in growth and index components. These results will be useful as basic data to study the correlation among environmental conditions, growth, and index components.

  • Flexible electronics, a future technology of electronics, require a low cost integrated circuit that can be built on various types of the flexible substrates. As a potential candidate for this application, a single walled carbon nanotube network is studied as an active device with a scheme of thin film transistor. Transistors are formed on a plastic foil by the Roll-to-Roll (R2R) and the Roll-to-Device (R2D) printing method. For both printing methods, electrical transports for the transistors are presented with the temperature dependence of threshold voltage (V_Th) and mobility from the measured transfer curves at temperatures ranging from 10 K to 300 K. It is observed that ${\mu}=0.044cm^2/V{\cdot}sec$ and V_Th=7.28V for R2R and ${\mu}=0.025cm^2/V{\cdot}sec$ and V_Th=3.10V for R2D, both for the temperature at 300K. Temperature dependence of mobility and V_Th is observed. However for R2R, the temperature dependence of V_Th is constant. It is the difference between, R2R and R2D.

A Self-Biased Current Reference in $0.25{\mu}m$ CMOS Technology

  • A self-biased CMOS current reference is described which provides supply and temperature independent bias current. The supply independency is obtained by subtracting two bias currents which have the same supply dependency. Unlike the conventional self-bias CMOS current reference, excellent supply independency can be obtained even with the minimum channel length devices and thus smaller area implementation becomes possible. The supply independent bias current is then applied to a temperature compensating circuit and as a result supply and temperature independent bias current is obtained. The current reference has been implemented in a $0.25{\mu}m$ standard CMOS technology. The active silicon area is only $45{\mu}m{\times}45{\mu}m$. The simulated temperature coefficient is 64ppm/$^{\circ}C$ in temperature range between $0^{\circ}C$ and $120^{\circ}C$. Supply voltage can be as low as 1.3V and the supply dependency of the current reference is measured to be smaller than 4500ppm/V. While providing $10.25{\mu}A$ output current, the current reference consumes $160{\mu}W$.

Isolation and Purification of Novel Anti-Fungal Peptides from Hemolymph of Immunized Larvae of Housefly, Musca domestica (집파리유충 hemolymph 중신형의 anti-fungal peptides의 분리정제)

  • To isolate and purify anti-fungal active substances from immunized housefly (Musca domestica), low dose of Candida albicans was injected into the larvae of the housefly to induce the appearance of potent anti-fungal active substances in the hemolymph. This purification work was performed by the routine isolation and purification processes of protein, namely, solid phase extraction (SPE), SDS-PACE electrophoresis, HPLC purification. Three 4-16 kDa peptides which exhibited antifungal activity against Candida albican and other fungi were isolated from induced hemolymph. Consequently, further anti-fungal activity study showed that these three peptides were different either in molecular weight or in anti-fungal activity. All isolated substances were proved to be active and resistant to high-temperature. It was deduced that these peptides isolated from induced housefly were novel members of the insect defensin family and they were inducible.

  • In general, electrolytic capacitors are used to reduce power pulsations on PV-panels. However, this can reduce the reliability of the PV AC-module system, because electrolytic capacitors have a shorter lifetime than PV-panels. In addition, PV-panels generate irregular power and inject it into the grid because the output power of a PV-panel depends on the surrounding conditions such as irradiation and temperature. To solve these problems, a grid-connected photovoltaic (PV) AC-module with active power decoupling and energy storage is proposed. A parallel bi-directional converter is connected to the AC module to reduce the output power pulsations of PV-panels. Thus, the electrolytic capacitor can be replaced with a film capacitor. In addition, the irregular output power due to the surrounding conditions can be regulated by using a parallel energy storage circuit. To maintain the discontinuous conduction mode at low irradiation, the frequency control method is adopted. The design method of the proposed converter and the operation principles are introduced. An experimental prototype rated at 125W was built to verify the performance of the proposed converter.

  • Perimeter zone has been reinforced by active systems, such as fan-coil units, because it causes an increase in heating and cooling loads, dew condensation in winter, or discomfort with cold-draft to residents in buildings, through poor insulation by light-weighed skin due to progressing multi-storied buildings and skyscrapers. However, because these active systems raise Its capacity so that fossil fuel is used as much as they are added, and ultimately, greenhouse effect is urged, we proposed BIPV system functioned as solar collector which can substitute active system. As an early stage, heat balance equation in steady-state by Fortran was used not only for pre-heating effect and electric power capacity during the day in winter, but also for electric power capacity during day in slimmer and sky radiation effect during night in summer. Especially, we should have considered shading on PV, since even a little bit of it makes the efficiency too low for the PV to work. Still, when the flux of pre-heated air was increased to make air-barrier, its temperature was not enough to make it because the speed of heat exchange was too fast to warm up the air, thus the capacity to meet the condition was evaluated, and electric power from PV was made used for it.

However, domestic vehicle emissions test temperature of $20{\sim}30^{\circ}C$ is it does not reflect this situation the actual test temperature to accurately measure the exhaust gas volume of the vehicle is difficult. In this study, domestic automobile exhaust gas test conditions of a test temperature $20{\sim}30^{\circ}C$ various temperatures, including (35, 25, 0, -7, -15, $-25^{\circ}C$) under the two vehicles (2.0L MPI, 2.4 L GDI) as was discussed with respect to the exhaust gas characteristics of the vehicle according to the ambient temperature gas. As a result, domestic emissions test temperature of $25^{\circ}C$ than average conditions were temperature decreases greenhouse gas emissions and increase overall increased by up to 15 times higher. Effects of Simultaneous Bending and Heating on Characteristics of Flexible Organic Thin Film Transistors

  • Recently, active materials such as amorphous silicon (a-Si), poly crystalline silicon (poly-Si), transition metal oxide semiconductors (TMO), and organic semiconductors have been demonstrated for flexible electronics. In order to apply flexible devices on the polymer substrates, all layers should require the characteristic of flexibility as well as the low temperature process. Especially, pentacene thin film transistors (TFTs) have been investigated for probable use in low-cost, large-area, flexible electronic applications such as radio frequency identification (RFID) tags, smart cards, display backplane driver circuits, and sensors. Since pentacene TFTs were studied, their electrical characteristics with varying single variable such as strain, humidity, and temperature have been reported by various groups, which must preferentially be performed in the flexible electronics. For example, the channel mobility of pentacene organic TFTs mainly led to change in device performance under mechanical deformation. While some electrical characteristics like carrier mobility and concentration of organic TFTs were significantly changed at the different temperature. However, there is no study concerning multivariable. Devices actually worked in many different kinds of the environment such as thermal, light, mechanical bending, humidity and various gases. For commercialization, not fewer than two variables of mechanism analysis have to be investigated. Analyzing the phenomenon of shifted characteristics under the change of multivariable may be able to be the importance with developing improved dielectric and encapsulation layer materials. In this study, we have fabricated flexible pentacene TFTs on polymer substrates and observed electrical characteristics of pentacene TFTs exposed to tensile and compressive strains at the different values of temperature like room temperature (RT), 40, 50, $60^{\circ}C$. Effects of bending and heating on the device performance of pentacene TFT will be discussed in detail.
