Alleviation Technology of Cold Stress of Maize(Zea mays L.) by Low Temperatures Damage

  • Maize is one of the world's three largest crops and has a long cultivation history, and is an important crop used for various purposes such as food, feed, and industrial raw materials. Recently, the agricultural environment is changing, in which the limit of cultivation of crops is shifted to the north due to the rise in temperature due to climate change. This study was conducted in experimental field of Suwon in 2022 by setting a seeding period earlier than the sowing time to establish the North Korean agricultural climatic zone and meteorological conditions. The test cultivars were silage cultivars, Kwangpyeongok and Dacheongok. As a priming test method, it was used to directly plant seeds in the field through immersion using 4mM zinc (Zn) and 2.5mM manganese (Mn), which are trace elements for seeds. The planting season was early on March 15th, April 1st, and April 15th. The number of days from sowing to silk stage of the two cultivars sown on March 15, April 1, and April 15 was 107, 93, and 85 days for Kwangpyeongok and 109, 95, and 87 days for Dacheongok, respectively. The seed priming test did not show any difference from the control group in the growth survey up to the middle stage of growth. In another test, low-temperature recovery was confirmed through nitrogen (2-5%) foliar fertilization after 3 days, 5 days, and 7 days in refrigeration (0 degrees), a selective low temperature treatment for com in the third leaf stage. As a result of this study, it was confirmed that the low-temperature damaged com treated at 0℃ showed the same growth as that of the untreated com through nitrogen foliar fertilization. These results suggest that urea foliar fertilization for low-temperature damage reduction of corn for silage in high-latitude climates will be helpful. In addition, through the results of the study, additional studies are needed on the recovery mechanism and field application through urea foliar fertilization.

남부지역 시설하우스 벼 극조기재배의 안전작기 설정 (Optimum Transplanting Time for Extremely Early Rice Greenhouse Cultivation in the Southern Area)

  • Optimum transplanting time for extremely early rice cultivation as an after-crop of fruit and vegetables under greenhouse conditions in the southern area was determined. Rice was transplanted on March 10, March 20, March 30, April 10 and April 20 far three years from 1998 to 2000. Meteorological computations for rice production were high for heading between early May and early July, but they were too low for heading between late July and early August. Especially the expected yield predicted with 35,000 spikelets, the average spikelets per $m^2$ for extremely early transplanting. Computation for heading between late July and early August was low by 106 kg/10a compared with that yield at heading during the same period in the field. As the transplanting date in extremely early rice cultivation was earlier) rice growth at early stages was more retarded by low temperature. Rice growth at heading stage recovered with high temperature, showing less difference for the transplanting date. Abnormal tillers occurred by 15.5∼22.2%. The contribution of 1,000 grain weight${\times}$ripened grain ratio to yield of the extremely early rice cultivation in the greenhouse was 50.6%, indicating 16% hi일or than the degree of panicle per $m^2$ on yield. The estimated optimum transplanting time on the basis of yield for the extremely early greenhouse rice cultivation ranged from March 19 to April 28, and the estimated critical transplanting date on the basis of accumulated effective temperature was March 12. Rice reduced the amount of NO$_3$-N by 97.1% and EC by 90.5% in greenhouse soil with continuous fruit/vegetables fer more than a 10-year period, and completely removed the root-knot nematodes.

생육온도에 따른 느타리류의 갓색, 수량 및 형태적 특성 변화 (Changes of pileus color, yield and morphological traits due to cultivation temperature in Pleurotus spp.)

  • 생육온도가 느타리류 품종의 갓색과 수량, 형태관련 형질에 어떤 영향을 미치는지 알아보고자 하였다. 주요결과는 다음과 같다. 느타리류의 모든 품종에서 생육온도가 12℃에서 21℃로 높아짐에 따라 갓색의 명도(L)가 높아졌지만, 산타리, 황금산타리, 순정은 21℃의 갓색 명도가 18℃ 보다 낮았다. 갓색의 적색도와 황색도는 생육온도가 높아 질수록 낮아졌는데, 유채색계열의 변화폭이 컸다. 유채색의 황색도는 생육온도 전반에서 무채색보다 높았다. 전체 수량은 생육온도가 높아질수록 많아지는 경향이었으나, 저온성 품종인 원형1호는 저온일수록 전체수량이 높았고 21℃에서는 발이가 불가능하였다. 유효경수는 대체적으로 18℃에서 가장 많았고, 수량과의 상관관계는 낮았다. 자실체의 전체 길이와 대두께는 생육온도의 상승에 따라 길어지고 뚜꺼워지는 경향이 뚜렸했으며, 흑타리의 전체길이와 온도사이의 결정계수(R2)는 0.514, 산타리의 대두께와 온도사이의 결정계수는 0.963로 나타났다. 상관분석의 결과 한 형질에 대해 복수의 형질이 높은 상관관계를 보였다. 앞으로 RNA 발현 분석을 통하여 생육온도에 따라 변화하는 느타리류 갓색물질과 관련 있는 유전자의 변화 양상을 밝힐 계획이다.

표고 톱밥 재배에서 저온 및 변온 처리가 표고 품질에 미치는 영향 (Effects of Low and Alternated Temperature Treatments on Quality of Oak Mushroom in Sawdust Culture)

  • Recently the sawdust culture of Lentinus edodes(Berk.) has been gradually extended replacing the log cultivation in Korea. It is indeed able to reduce the use of log and cultivation period in controlled facilities, but is not yet able to produce the high-quality mushroom. The objectives of this study were to examine the effects of low and alternated temperature treatments during the fruiting period on the quality of oak mushroom. At low temperature treatments of 1$^{\circ}C$ and 10$^{\circ}C$, the crack, lightness, hardness, and other characteristics for the high-quality oak mushroom were not improved. However, the crack, brightness, and hardness of cap were increased at alternated temperatures of 5-10$^{\circ}C$ than the other temperature treatments. In conclusion, the alternated temperature treatments were more effective than the low temperature treatment for improving the indices of high-quality oak mushroom such as the crack, brightness and hardness of cap.

Morphological and molecular characterization of germinability related to direct-seeding in rice varieties

  • Direct-seeding cultivation of rice is increasing in Asia instead of transplanting system, because of its lower cost and operational simplicity. Low-temperature germinability (LTG) and anaerobic germinability (AG) are important characters for breeding of varieties for wide-spread adoption of direct-seeding cultivation in rice. This study was performed to characterize LTG and AG of seven rice varieties and identify varieties with strong germinability on both low-temperature and anaerobic conditions. The mean germination rate and germination vigor of seven varieties were 51.7% and 6.0 under low-temperature condition, respectively. Among these varieties, Cheongcheongjinmi and Hwanggeumnodeul had the highest germination rate of 80%, indicating that Cheongcheongjinmi and Hwanggeumnodeul have a good LTG. In anaerobic conditions, the germination rate and coleoptile length for all varieties were 47.6% and 3.2 cm, respectively. Of them, the highest germination rate and coleoptile length were observed in Subo and Hopum, respectively, suggesting that these two varieties are tolerant to anaerobic during germination stage. Molecular characterization by SDS-PAGE revealed that the protein patterns differed at 50 kDa, 40 kDa, and 22 kDa between low-temperature and anaerobic conditions. Varieties identified as good LTG or AG in this study may be used for developing new direct-seeding rice cultivars through pyramiding these traits in the breeding program.

등숙기 온도에 따른 쌀가루 가공용 벼의 등숙특성 변이 구명 (Effects of Temperature on Grain Filling Properties of Rice Flour Varieties during the Ripening Stage)

  • 쌀가루용 품종의 등숙기 온도에 따른 등숙특성 변이를 구명한 내용은 다음과 같다. 1. 고온(28℃)에서는 바로미2의 천립중이 21% 감소하면서 새누리(6% 감소)와 설갱(12% 감소)에 비해 크게 줄어들었으며, 전분함량의 감소 또한 바로미2에서 10% 이상으로 새누리와 설갱의 1~3% 감소와 대비되었다. 2. 저온(18℃)에서는 새누리나 설갱에 비해 바로미2의 등숙 속도가 매우 감소하였으며 특히 등숙 초반에 종실중의 증가가 매우 느렸다. 그러나 최종 천립중 및 전분함량은 22℃보다도 약간 증가하며 다른 품종과도 비슷한 것을 확인하였다. 3. 앞서 언급한 것처럼 쌀가루 가공용 품종 중 분질미(바로미2)는 등숙기 온도에 민감하기 때문에 추후 분질미 재배시 등숙기에 고온을 피할 수 있도록 이앙시기를 늦추는 편이 좋으며, 다만 과도하게 늦출 경우 저온조건에서 등숙기간을 제대로 확보하지 못하여 등숙이 충분히 되지 않을 수 있기 때문에 주의가 필요하다.

대장균에서 chaperons 분자와 저온배양에 의한 CGTase의 가용성 발현 증대 (Enhancement of Soluble Expression of CGTase in E. coli By Chaperone Molecules and Low Temperature Cultivation.)

  • E. coli에서 B. macerans 유래 cyclodextrin glucanotransferase (CGTase)의 활성형 생산에 GroEL/ES chaperone과의 공발현과 저온 배양의 공동작용 효과에 대해 조사하였다. 실험에 사용된 cgt와 groEL/ES 유전자를 발현하는 pTCGTl과 pGroll은 각각 T7 promoter와 Pzt-1 promoter에 의해 조절되고 이들을 E. coli에 도입시켰다. 대수증식기 초기(2 hr)와 대수증식기 중기(3 hr)에 tetrarycline 10 ng/ml 과 IPTG 1 mM을 첨가하여 각각의 유전자를 발현시켰다. CGTase활성과 specific artivity 측정 시 $37^{\circ}C$에서 pTCGTl 단독 발현 보다 $25^{\circ}C$에서 chaperone과 함께 발현시킨 경우 2배나 높은 활성이 측정됐으며, SDS-PACE 분석결과 $37^{\circ}C$에서 단독 발현 시킬경우 20% 정도 가용성 형태로 발현되던 것이 $25^{\circ}C$에서 chaperone과 공발현 시에는 거의 3.5배가 넘는 69%가 가용성으로 전환됨을 알 수 있었다. 이와 같이 분자 chaperone과 $25^{\circ}C$에서의 저온 배양은 E. coli에서 활성형질 가용성 CGTase의 생산 증가에 큰 영향을 미치는 것으로 나타났다.

Effect of Seed Coating with Polymers on Seed Vigour and Seedling Stand in Direct Seeded Rice

  • These experiments were conducted to investigate water uptake, electrical conductivity, germination percentage, seedling growth, and seedling establishment rate in direct seeding cultivation of rice. The rice seeds of six japonica type varieties were coated with 12 kinds of polymers in a standard concentration of 0.2% using seed coating machine. The water absorption of the polymer-coated seeds under saturation conditions was not different among varieties, and was the highest in kulcel, maltrin, and waterlock on the polymer-coated seeds. The electrical conductivity with waterlock (55.0 $\mu scm^{-1}g^{-1}$) was higher than the control plot (45.6 $\mu scm^{-1}g^{-1}$) and other treatments. The germination of the polymer-coated seeds was 95.9% at control plot, 92.7% at low temperature and 35.7% at high temperature. The total dry weight of seed decreased in the order of low temperature, control plot, and high temperature, and was effective in pvp (polyvinyl pyrrolidone), opadry, and sacrust. The seedling establishment rate in direct seeding cultivation ranged from 74.9 to 81.0% in flooded paddy surface, and ranged from 64.7 to 76.6% in dry paddy. In both cases, it decreased in the order of early, medium and medium-late varieties, but was enhanced in daran 8600, sepirect, and sacrust. According to this study the recommended polymers for direct seeding cultivation of rice are daran 8600, sepirect, and sacrust.

Growth Characteristics And Yield of Corn (Zea mays L.) for Grain by Early Sowing Date in the Central Region of South Korea

  • The limit of crop cultivation is moving northward due to the temperature rise by climate change. There is a problem with crop growth if early sowing is performed at a time when the temperature is low. It is difficult to secure crop productivity and cultivation stability due to the low temperature and short cultivation period. Therefore, this study was conducted to analyze the change in growth characteristics and yield of corn for grain when early sowing is performed in central region of South Korea. This experiment was conducted at experimental field of Suwon in 2021. Three varieties of corn for grain such as Kwangpyeongok, Sinhwangok, and Hwangdaok were sown at intervals of 5 days from 20 March to 15 April. The planting density at this time was sown with a row interval of 70 cm and a plant interval of 25 cm. Nitrogen, phosphoric acid, and potassium fertilizers were applied at 17.4 kg, 3.0 kg, and 6.9 kg per 10a, respectively. Phosphoric acid and potassium fertilizers were all applied before sowing and nitrogen fertilizer was applied 50% before sowing and 50% in the fifth leaf period. The corn growth characteristics and yield components were investigated. The seedling establishment rate by sowing date was in the range of 68.5~88.5%, and it showed a difference depending on the variety. The range of days from sowing to tassel and silk emergence by sowing date was 79.9~98.4 and 81.0~98.9 days, respectively. As the sowing date was delayed, the days from sowing to tassel and silk emergence decreased. The growth characteristics and yield of corn by sowing date are as follows. Plant height was the highest at 241.3 cm at the sowing on 25 March, and Stalk diameter was the thickest at 25.6 mm at the sowing on 31 March. The fresh weight per plant was the highest at 728 g at the sowing on 25 March, and the dry weight per plant was the highest at 185 g at the sowing on 31 March. Corn growth characteristics did not show a certain trend depending on the sowing date, and corn growth was more vigorous at the sowing on March 25 and 31 than the others. In the case of ear weight, it was the heaviest with 344 g at the sowing on 25 March, and filled ear length ratio showed a tendency to decrease as the sowing time was delayed. The weight of 100 grains and grain yield per 10a of maize were the highest at 36.0g and 878.7kg/10a, respectively at the sowing on 25 March. Although the growth and yield of corn for grain were good during early sowing in the central region of South Korea, it is necessary to investigate the limit temperature for early sowing of corn by examining the annual variation according to weather conditions.

표고재배(栽培)의 관리분석(管理分析)과 종균활착(種菌活着)을 위한 골목관리(管理)에 관(關)한 연구(硏究) (The Analysis of Management and the Method of Cultivation of Lentinus edodes I. for Full-Development of Mycelium in Bed Logs)

  • 본 연구는 표고재배기술의 단계별 관리방법을 제시하며, 활착율 증대를 통하여 양질의 버섯생산을 도모하여 생산성을 향상시키는데 목적이 있다. 현지 재배자를 대상으로 재배현황에 대한 실태조사를 실시하여 재배현황을 분석하였고, 이를 토대로 현지 재배환경을 위주로 표고 종균활착율 향상과 재배 관리에 관하여 실험한 결과는 다음과 같다. l. 실태조사결과 많은 재배자가 활착촉진을 위한 재배관리에 등한히 하는 경우가 많았는데, 이는 노동력의 부족과 자본의 부족 그리고 재배관리 요령에 익숙하지 못한 이유 등이었다. 2. 원목내 수분함량은 3월 12일 l차 접종분이 28.63%, 3월 25일 2차분은 25.20% 그리고 4월 10일의 3차분은 23.19%로 점차 감소하였다. 종균의 활착에 적절한 원목내의 수분함량을 확보하며 활착증진과 노동력의 분산을 위해서는 3월 초부터 접종하는 편이 유리하였다. 3. 종균별 활착율은 10월 20일의 활착율조사에서 고온성은 모리465가 100%, 모리3046이 98.98%, 저온성에서는 모리290이 98.97%였다. 상대활착율은 고온성에서 모리465가 97.70%, 모리3046이 82.45%. 저온성에서는 모리290이 88.87%로 나타났다. 본 종균은 높은 활착율로 재배자들이 선호하는 품종으로 상대활착율에서 차이를 보여, 재배자의 종균선택에 유의하여야 할 부분으로 판단된다. 4. 접종시기별 활착율은 고온성 모리465와 저온성 모리290을 접종하여 고온성의 경우, 활착율은 1차(3월 중순) 100.0%, 2차(3월 하순) 98.98% 그리고 3차(4월 상순)에 96.79%로 약간의 차이를 나타냈고, 상대활착율은 1차가 97.70%, 2차 82.45% 그리고 3차가 8l.42% 순이였다. 저온성의 경우, 활착율은 1차가 99.09%, 2차가 98.97% 그리고 3차가 97.89%로 별차이가 없었으며, 상대활착율은 1차가 93.27%, 2차가 89.67% 그리고 3차가 88.87%로 나타났다. 접종시기가 빠를수록 활착율에서는 비교적 차이가 없었으나 상대활착율의 경우는 차이를 보였는데, 우리재배자의 접종시기가 3차인 4월에 집중되어 있는 점을 감안 한다면 앞으로의 접종시기는 조기접종인 3월초에 실시하도록 하여야 할 것으로 판단된다. 5. 가눕혀두기시 위치별로 골목내의 수분함량을 측정한 결과, 상단부일수록 수분함량이 낮아 가눕혀두가시 쌓는 높이결정에 있어서는 가능한한 60cm를 넘지 않도록 하여야 할 것으로 판단된다.
