열처리 방법에 따른 대두의 이화학적 특성 변화 (Effect of heat treatment on physicochemical properties of soybean)

  • 대두는 된장, 간장, 스낵 등 전통식품의 주요 원료이며, 열처리 공정은 대두의 가공과정에서 대부분 수반되는 단위 조작이다. 본 연구에서는 열처리가 대두의 이화학적 특성에 미치는 영향을 조사하기 위하여 상업적인 조건에서 대두를 증자, 가열팽화, 튀김 처리한 다음 단백질의 분자량 분포와 용해도, 수분과 유지 흡착력 및 열 특성과 산패도의 변화를 조사하였다. 대두는 가열처리에 의하여 단백질이 10-40 kDa 범위의 작은 분자량으로 분해되는 경향을 나타내었다. 대두의 용융 엔탈피는 199.62 J/g이었으며 열처리에 의하여 123.07-135.90 J/g 범위로 엔탈피가 감소하였고 지질 산화를 보여주는 fluorescence intensity도 열처리로 증가하였으며 열처리 효과는 튀김, 증자, 가열팽화의 순으로 높았다. 또한 대두의 수분 흡착력은 열처리 한 경우가 비열처리 대두보다 상대적으로 높았으며, 단백질의 용해도는 산성 영역(pH 3-6)에서 같은 경향을 보였다. 결과적으로 대두는 가열처리에 의하여 용융 엔탈피가 감소하고 단백질이 분해되며 지질 산화와 용해도 및 수분흡착력이 증가하는데 대두에 대한 열처리의 효과는 튀김 공정에서 뚜렷하였다.

Effects of Insulin and IGFs on Phosphate Uptake in Primary Cultured Rabbit Renal Proximal Tubule Cells

  • The aim of present study was to characterize phosphate uptake and to investigate the mechanism for the insulin and insulin-like growth factor(IGF) stimulation of phosphate uptake in primary cultured rabbit renal proximal tubule cells. Results were as follows : 1. The primary cultured proximal tubule cells had accumulated $6.68{\pm}0.70$ nmole phosphate/mg protein in the presence of 140 mM NaCl and $2.07{\pm}0.17$ nmole phosphate/mg protein in the presence of 140 mM KCl during a 60 minute uptake period. Raising the concentration of extracellular phosphate to 100 mM$(48.33{\pm}1.76\;pmole/mg\;protein/min)$ induced decrease in phosphate uptake compared with that in control cells maintained in 1 mM phosphate$(190.66{\pm}13.01\;pmole/mg\;protein/min)$. Optimal phosphate uptake was observed at pH 6.5 in the presence of 140 mM NaCl. Phosphate uptake at pH 7.2 and pH 7.9 decreased to $83.06{\pm}5.75%\;and\;74.61{\pm}3.29%$ of that of pH 6.5, respectively. 2. Phosphate uptake was inhibited by iodoacetic acid(IAA) or valinomycin treatment $(62.41{\pm}4.40%\;and\;12.80{\pm}1.64%\;of\;that\;of\;control,\;respectively)$. When IAA and valinomycin were added together, phosphate uptake was inhibited to $8.04{\pm}0.61%$ of that of control. Phosphate uptake by the primary proximal tubule cells was significantly reduced by ouabain treatment$(80.27{\pm}6.96%\;of\;that\;of\;control)$. Inhibition of protein and/or RNA synthesis by either cycloheximide or actinomycin D markedly attenuated phosphate uptake. 3. Extracellular CAMP and phorbol 12-myristate 13 acetate(PMA) decreased phosphate uptake in a dose-dependent manner in all experimental conditions. Treatment of cells with pertussis toxin or cholera toxin inhibited phosphate uptake. cAMP concentration between $10^{-6}\;M\;and\;10^{-4}\;M$ significantly inhibited phosphate uptake. Phosphate uptake was blocked to about 25% of that of control at 100 ng/ml PMA. 3-Isobutyl-1-methyl-xanthine(IBMX) inhibited phosphate uptake. However, in the presence of IBMX, the inhibitory effect of exogenous cAMP was not significantly potentiated. Forskolin decreased phosphate transport. Acetylsalicylic acid did not inhibit phosphate uptake. The 1,2-dioctanoyl-sn-glycorol(DAG) and 1-oleoyl-2-acetyl-sn- glycerol(OAG) showed a inhibitory effect. However, staurosporine had no effect on phosphate uptake. When PMA and staurosporine were treated together, inhibition of phosphate uptake was not observed. In conclusion, phosphate uptake is stimulated by high sodium and low phosphate and pH 6.5 in the culture medium. Membrane potential and intracellular energy levels are also an important factor fer phosphate transport. Insulin and IGF-I stimulate phosphate uptake through a mechanisms that involve do novo protein and/or RNA synthesis and decrease of intracellular cAMP level. Also protein kinase C(PKC) is may play a regulatory role in transducing the insulin and IGF-I signal for phosphate transport in primary cultured proximal tubule cells.

Leuconostoc fallax LH3이 생산하는 효소에 의한 Ginsenoside Rd의 Ginsenoside F2로의 전환 (Transformation of Ginsenoside Rd to Ginsenoside F2 by Enzymes of Leuconostoc fallax LH3)

  • Ginsenosides have been regarded as the principal components, responsible for the pharmacological and biological activities of ginseng. Absorption of major ginsenosides at the gastrointestinal tract was extremely low, when ginseng taken orally. In order to improve the absorption and bioavailability, transformation of major ginsenosides into more active and valuable minor ginsenoside is much required. In this present study, We isolated a lactic acid bacteria Leuconostoc fallax LH3 from the Korean fermented food Kimchi, which have higher ${\beta}$-glucosidase activity. Using the ethanol precipitated curd enzyme of Leuconostoc fallax LH3, we investigated the biotransformation of ginsenoside Rd at different experimental condition to increase transformation. The maximum convertion was supported at 30 $^{\circ}C$ and decreased when temperatures increased. In order to optimize the effect of pH, the curd enzyme was mixed 20 mM sodium phosphate buffer (pH 3.5 to pH 8.0). Ginsenoside Rd was almost hydrolyzed between pH 7.0 and pH 9.0, but not hydrolyzed above pH 10.0. Ginsenoside Rd was hydrolyzed after 24 hrs incubation, but whereas the ginsenoside F2 was appeared from 36 hrs, and all ginsenoside Rd was transformed to F2 after the 60 hrs incubation. Based on this study, the curd enzyme of Leuconostoc fallax LH3 transformed the ginsenoside Rd at the 30$^{\circ}C$ and the pH optimum of 7.0 to 9.0 after the 60 hrs incubation time.

제주시 도심지역 미세먼지의 화학적 조성 특성 (Chemical Composition of Fine Particulate Matter in the Downtown Area of Jeju City)

  • This study observed particulate matter ($PM_{2.5}$ and $PM_{10}$) in the downtown area of Jeju City, South Korea, to understand the chemical composition of particulates based on an analysis of the water-soluble ionic species contained in the particles. The mass fraction of the ionic species in the sampled $PM_{10}$ and $PM_{2.5}$ was 44.3% and 42.2%, respectively. In contrast, in Daegu City and Suwon City, the mass fraction of the ionic species in $PM_{2.5}$ was higher than that in $PM_{10}$. The chloride depletion percentage of $PM_{10}$ and $PM_{2.5}$ in Jeju City was higher than 61% and 66%, respectively. The contribution of sea-salt to the mass of $PM_{10}$ (5.9%) and $PM_{2.5}$ (2.6%) in Jeju City was similar to that in several coastal regions of South Korea. The mass ratio of $Cl^-$ to $Na^+$ in the downtown area of Jeju City was comparable to that in some coastal regions, such as the Gosan Area of Jeju Island, Deokjeok Island, and Taean City. The mass fraction of sea-salt in $PM_{10}$ and $PM_{2.5}$ was very low, and the concentration of sodium and chloride ions in $PM_{10}$ was not correlated with those in $PM_{2.5}$ ($R^2$ < 0.2), suggesting that the effects of sea-salt on the formation of particulate matter in Jeju City might be insignificant. The relationship between $NH_4{^+}$ and several anions such as $SO_4{^{2-}}$, $NO_3{^-}$, and $Cl^-$, as well as the relationship between the measurement and calculation of ammonium ion concentration, suggested that sea-salts may not react with $H_2SO_4$, and $(NH_4)_2SO_4$ may be a major secondary inorganic aerosol component of $PM_{2.5}$ and $PM_{10}$ in Jeju City.

고등학생 흡연자와 비흡연자의 1일 영양소 및 간식 섭취 (The Daily Intakes of Nutrients and Snacks of High School Smokers and Non-Smokers)

  • This study investigates the relationship of smoking on daily intake of nutrients and snacks in the Chungnam and Daejeon high school students. Up to date scientific nutrition education and counseling programs in the regular school system is needed for a professional nutrition education teacher. The primary objective of this study was to provide useful information to nutrition education teachers. A survey was conducted with 400 high school students in the Chungnam and Daejeon areas. 381 out of 400 questionnaires were analyzed with SPSS 12.0K. The subjects were composed of 49.8% male, 50.1% female and 40.9% regular high school students, 59.1% business high school students and smokers 43.1%, non-smokers 56.9%. 43.4% of smokers had been smoking since middle school. On analysis of daily nutrient intakes, 16 out of 19 nutrients except animal calcium, Vitamin A and Vitamin C were much more consumed by the smoking group than the non-smoking group non-significantly. Especially vegetable fat and Vitamin E were higher in the smoking group than the non-smoking group (p<0.05). The intake ratio of carbohydrates: protein: fat was similar in the two groups (smoking group 55:15:29, nonsmoking group 56:15:28). Intakes of Vitamin B1 and potassium in comparison with the Korean dietary reference intakes(KDRI) were under 50% in both groups. However, sodium was taken over 200% compared to KDRI in both groups. Intakes of Vitamin C in the smoking group were as low as 76.5% in comparison to KDRI. Smokers need to increase the intakes of Vitamin C considering that smokers need to intake Vitamin C two times than non-smokers. Nutrient intakes from snacks in the smoking group were higher than the non-smoking group. Nutrients that originated from snacks which took over 20% among daily nutrient intakes were 12 nutrients(energy, fat, carbohydrate, calcium, P, Fe, K, Vitamin $B_1$, Vitamin $B_2$, Vitamin C, Vitamin E, dietary fiber) in the smoking group compared to 7 nutrients(energy, vegetable protein, fat, carbohydrate, calcium, Vitamin $B_2$, Vitamin C) in the non-smoking group. The smoking group was significantly paying more money for snacks each month than the non-smoking group was(p<0.01). Periods of consumption were irregular in the smoking group(p<0.05) and the smoking group was used to taking snacks in the morning compared to the non-smoking group. The smoking group preferred sweets and high calorie food over other snacks in comparison of the non-smoking group. The nonsmoking group had better eating habits than the smoking group.

  • 에이니그마타이트($Na_4(Fe^{2+},Ti,Fe^{3+}){_{12}}(Fe^{3+},Si){_{12}}O_{40}$)는 Na가 풍부한 알칼리 화성암에 흔히 산출되는 광물이며, 분지 개방형 단쇄형 규산염광물로 분류된다. 3개의 산출이 다른 자연산 에이니그마타이트를 대상으로 뫼스바우어 분광분석을 실시하였으며 상세한 결정화학적 연구가 수행되었다. 뫼스바우어 분광분석 결과 9개의 흡수선이 분리되었는데, 3쌍의 흡수선은 팔면체 자리를 점유하는 $Fe^{2+}$의 피크이며, 저속도 구간의 독립적인 2개의 흡수선과 대응하는 한 개의 중첩된 흡수선은 각각 사면체와 팔면체 자리의 $Fe^{3+}$의 피크로 밝혀졌다. $Fe^{2+}$$Fe^{3+}$의 정확한 함량비에 의해 화학분석치가 재계산되었으며, Fe 양이온의 사면체 자리와 팔면체 자리에 대한 정확한 자리점유율로 상세한 결정화학적 정보를 제공하였다. 상당한 함량의 $Fe^{3+}/tet$는 사면체 자리점유에 있어 $Fe^{3+}$$Al^{3+}$보다 우위를 보여준다. 결정화학적로 규명된 에이니그마타이트(AEN1)의 상세한 화학조성은 $(Na_{3.97}Ca_{0.03})(Ca_{0.11}Mn_{0.59}Fe^{2+}{_{8.07}}Ti_{2.07}Mg_{0.70}Fe^{3+}{_{0.43}}Al_{0.04})(Fe^{3+}{_{0.56}}Al_{0.18}Si_{11.26})O_{40}$으로 밝혀졌다.

비타민 D 결핍성 구루병에 동반된 근위 신 세뇨관성 산증 1례 (A Case of Proximal Renal Tubular Acidosis Accompanied by Vitamin D Deficient Rickets)

  • 최근 들어 모유 단독 수유의 장기화 및 잘못된 식이 제한 등으로 인한 비타민 D 결핍성 구루병의 발병이 점차 증가하고 있는 시점에서, 비타민 D 결핍성 구루병 및 이와 동반된 저칼슘혈증성 경련에 대한 예는 다소 보고된 바 있으나, 비타민 D 결핍으로 인해 골 병변 뿐 아니라 저칼슘 혈증, 부갑상선 기능 항진증, 나아가 신세뇨관성 산증까지 초래한 예는 보고된 바가 없었다. 저자들은 8개월간 모유 단독 수유만 유지하던 중 저칼슘혈증성 경련을 주소로 내원하여 골 병변을 동반한 비타민 D 결핍성 구루병과 부갑상선 항진 및 근위 세뇨관성 산증을 진단 받은 후, 적절한 칼슘, 비타민 D, 중탄산염 투여 후 완전히 회복된 환자의 1례를 경험하였기에 보고하는 바이다.

유통 축·수산물 중 잔류벤질페니실린의 검출 및 모니터링 (Detection and Monitoring of Benzylpenicillin Residues in Livestock and Marine Products)

  • 국내 유통 중인 돼지고기, 소고기, 닭고기, 넙치 232건의 시료에 대해 벤질페니실린의 잔류량을 조사하기 위해 현행 식품공전상 축 수산물 중 벤질페니실린 분석법에 대한 검증을 실시하고 모니터링을 수행하였다. 그 결과 벤질페니실린에 대한 분석법은 CODEX에서 요구하는 기준에 적합한 정밀성과 정확성을 보였다. 벤질페니실린의 잔류량 모니터링 결과 축산물 193건 중 11건, 수산물 39건 중 2건 총 13건이 검출되었다. 검출된 수준은 축 수산물 12건에서 0.001-0.004 mg/kg으로 잔류허용기준 이하로 검출되었으나, 축산물 중 국내산 돼지고기 1건에서 잔류허용기준을 초과한 0.134 mg/kg 수준으로 검출되어 해당 유관기관에 부적합 통보 및 사후관리를 요청하였다. 따라서 벤질페니실린 등 항생제는 다른 동물용 의약품에 비해 사용빈도가 많은 만큼 지속적으로 잔류모니터링과 관리가 필요할 것으로 판단된다.

분석측정범위의 실증적 평가 (An Empirical Study of the Analytical Measurement Range in Clinical Chemistry)

  • The analytical measurement range (AMR) is the range of analyte values that a method can directly measure on a specimen without any dilution, concentration, or other pretreatment not part of the usual assay process. The linearity of the AMR is its ability to obtain test results which are directly proportional to the concentration of analyte in the sample from the upper and lower limit of the AMR. The AMR validation is the process of confirming that the assay system will correctly recover the concentration or activity of the analyte over the AMR. The test specimen must have analyte values which, at a minimum, are near the low, midpoint, and high values of the AMR. The AMR must be revalidated at least every six months, at changes in major system components, and when a complete change in reagents for a procesure is introduced; unless the laboratory can demonstrate that changing the reagent lot number does not affect the range used to report patient test results. The AMR linearity was total protein (0-16.6), albumin (0-8.1), total bilirubin (0-18.1), alkaline phosphatase (0-1244.3), aspartate aminotransferase (0-1527.9), alanine aminotransferase (0-1107.9), gamma glutamyl transpeptidase (0-1527.7), creatine kinase (0-1666.6), lactate dehydrogenase (0-1342), high density lipoprotein cholesterol (0.3-154.3), sodium (35.4-309), creatinine (0-19.2), blood urea nitrogen (0.5-206.2), uric acid (0-23.9), total cholesterol (-0.3-510), triglycerides (0.7-539.6), glucose (0-672.7), amylase (0-1595.3), calcium (0-23.9), inorganic phosphorus (0.03-17.0), potassium (0.1-116.5), chloride (3.3-278.7). We are sure that materials for the AMR affect the evaluation of the upper limit of the AMR in the process system.

넙치(Paralichthys olivaceus)의 생존과 생리상태에 미치는 오존의 급성 독성 (Acute Toxicity of Ozone on Survival and Physiological Conditions of Olive Flounder, Paralichthys Olivaceus)

  • 오존처리 해수중에 함유하는 잔류 옥시던트(TRO)가 양식 넙치의 생존, 혈액성상과 삼투질 농도, 산소소비 및 아가미 조직에 미치는 독성 영향을 조사하였다. TRO에 노출된 넙치의 48-hr 및 96-hr $LC_{50}$은 각각 26.4 와 22.3ppb이었고, 47ppb에서는 3시간 이내에 모두 치사하였으나 13ppb에서는 96시간 동안 폐사발생이 없었다. TRO 24~39ppb 농도범위에 노출된 넙치의 hematocrit, 헤모글로빈, 적혈구 수, 혈중 sodium과 chloride 이온 및 삼투압 농도는 TRO 농도가 높을수록 유의하게 증가하였고, 아가미 조직은 아가미 상피층이 모세혈관으로부터 박리되고, 괴사와 상피새포의 증생이 관찰되었다. 산소소비율은 TRO 농도가 높고, 노출이 계속될수록 감소하였다. 13ppb에서는 아가미 조직과 생리상태에 미치는 변화는 관찰되지 않았으나 혈당은 유의하게 증가하였다. 그러나 활성탄으로 TRO를 제거한 대조구에서는 오존의 독성 영향은 관찰되지 않았다. 잔류오존에 노출된 넙치는 삼투질 농도의 생리적 불균형 및 아가미 손상과 상피세포의 증생에 의한 산소섭취 부족이 치사를 유발하는 것으로 나타났다. 본 연구의 결과 해산어 양식장에서 오존을 사용할 경우에는 오존처리수중의 TRO는 제거하여야 하며, 비록 저농도라도 장기간 노출되면 양식어류의 생존과 생리상태에 미치는 영향은 현저할 것으로 평가되었다.
