큰실말에서 분리된 식이섬유가 고지방식이로 유도된 흰쥐의 콜레스테롤에 미치는 영향 (Effects of Cladosiphon Okamuranus Dietary Fiber on Cholesterol in High Fat Diet-Fed Rats)

  • Cladosiphon okamuranus is edible brown algae cultured commercially and massively at Okinawa coast. Dietary fiber occupying most of C. okamuranus isn't digested and absorbed by digestive enzymes of human. But it is known to prevent and treat constipation as metabolic disease. This study was to investigate the effects of dietary fiber extracted from C. okamuranus on the damage of liver and the blood cholesterol level in the high fat diet-fed rats. The effects were measured by the levels of aspartate aminotransferase (ALT), alanine aminotransferase (AST), total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL) and high density lipoprotein (HDL) in sera. The levels of ALT, AST, TC, TG and LDL were significantly reduced in the C. okamuranus-treated group compared with the high fat diet group. But HDL level was markedly increased. The results showed that the dietary fiber extracts from C. okamuranus have the inhibitive effects of cholesterol biosynthesis in the high fat diet-fed rats.

창부도담탕이 비만 유도 흰쥐에 미치는 영향 (Effects of Changbudodam-tang on Obesity-induced Rats)

  • Objectives: This study was designed to evaluate the efficacy of Changbudodam-tang on obesity by using high-fat diet rats. Methods: Rats were divided into five groups. Normal group: Normal diet, Control group: High-fat diet, Positive control group: High-fat diet+Dietamin 4 mg/kg/day, Changbudodam-tang-Low group: High-fat diet+Changbudodam-tang 250 mg/kg/day, Changbudodam-tang-High group: High-fat diet+Changbudodam-tang 500 mg/kg/day. Weight, food intake were measured every week. After 7 weeks, total cholesterol, high density lipoprotein-cholesterol, low density lipoprotein-cholesterol, Triglyceride, free fatty acid, aspartate aminotransferase, alanine aminotransferase, complete blood count were measured and messenger ribonucleic acid expression of adiponectin, peroxisome proliferator-activated receptor-γ, leptin were observed using Reverse transcription polymerase chain reaction of liver cells. Results: There was no difference in food intake between groups. Body weight tended to decrease compared with the Control group, but it wasn't statistically meaningfull. The total cholesterol, low density lipoprotein-cholesterol, Triglyceride, free fatty acid tended to decrease compared with the Control group. High density lipoprotein-cholesterol tended to decrease compared with the Control group, but it wasn't statistically meaningfull. White blood cell, red blood cell, hemoglobin, platelet, aspartate aminotransferase, alanine aminotransferase were not affected by Changbudodam-tang. The messenger ribonucleic acid expression of Adiponectin, peroxisome proliferator-activated receptor-γ, leptin, which are involved in the differentiation of adipocytes, was decreased compared with the Control group. Conclusions: Based on the results above, it is suggested that Changbudodam-tang can be applied to improving serum lipid levels in obese patients caused by high fat diets.

천궁 열수추출물 첨가가 고콜레스테롤 식이를 급여한 흰쥐의 지방대사에 미치는 영향 (Effects of the Cnidium Officinale Makino Water Extract on the Lipid Metabolism of Rats Fed a Hypercholesterol Diet)

  • The study examines the effects of the Cnidium officinale Makino water extract on hyperlipidemia. For this, three levels (low, moderate, and high) of the Cnidium officinale Makino water extract were supplemented to six-week-old male sprague Dawley rats for five weeks. According to the results, serum total lipid, triglyceride, total cholesterol, and LDL cholesterol levels showed significant decreases (p<0.05) in the group with the Cnidium officinale Makino water extract. The serum total lipid level decreased by 8~22%; the triglyceride level, by 13~25%; the total cholesterol level, by 6~23%; and the LDL cholesterol level, by 14~25%. Total cholesterol and triglyceride levels in the liver were significantly lower in the group with the Cnidium officinale Makino water extract than in the control group (p<0.05), and fecal total cholesterol and triglyceride levels were significantly higher in the group with the Cnidium officinalle Makino water extract. These results suggest that the Cnidium officinale Makino water extract in the hypercholesterol diet may reduce hyperlipidemia, a major cause of cardiovascular disease, by reducing serum cholesterol and triglyceride level in the liver through the facilitation of their excretion by feces.

고지방식이와 병행 섭취한 뽕잎가루가 흰쥐의 혈중 지질관계 인자와 간 기능 개선에 미치는 영향 (Effect of Mulberry Extract on the Lipid Profile and Liver Function in Mice Fed a High Fat Diet)

  • In this study, we determined the effect of the mulberry leaf powder on blood lipid metabolism and liver function improvement of mice fed a high fat diet. The mulberry leaf showed a moisture content of $10.74{\pm}0.56%$, ash $9.67{\pm}0.56%$, crude protein $25.24{\pm}1.02%$, and crude fat $2.66{\pm}0.08%$. In the group with high fat diet supplemented with mulberry leaf powder, serum low-density lipoprotein (LDL-cholesterol) and total cholesterol levels were significantly lower (p<0.05); also, serum phospholipid and aspartate amino transferase (AST) levels were significantly lower (p<0.05). Serum insulin and leptin levels were high in mice fed a high fat diet; however, addition of mulberry leaf powder 10% in the diet had no significant effect on blood insulin and leptin control. The results of the study suggested that various plant sterols in mulberry leaf improve lipid metabolism and liver function in ICR-mice.

  • Objectives : This study is aimed to compare the changes in Antioxidative capacity of Liriopis Tuber by steaming process and to compare the effects in hyperlipidemia induced rats fed high cholesterol diet between Simvastatin and Liriopis Tuber by steaming process. Methods : The SD rats were divided into six groups: normal diet (Nor), high cholesterol diet (Veh), high cholesterol diet plus Simvastatin 5 mg/kg (Sim), high cholesterol diet plus LT0 extract 200 mg/kg (LT0), high cholesterol diet plus LT6 extract 200 mg/kg (LT6) and high cholesterol diet plus LT9 extract 200 mg/kg (LT9). We compared the total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL), low density lipoprotein cholesterol (LDL) contents and reactive oxygen species (ROS) from each serums. Protein expression in liver tissues related to antioxidant and cholesterol was analyzed. Results : The Antioxidant activity of Liriopis Tuber increased by steaming process. In vivo, TC, TG, LDL-c, atherogenic index (AI) and cardiac risk factor (CRF) decreased and HDL-c increased with increasing steaming frequency. aspartate aminotransferase (AST), alanine aminotransferase (ALT), creatinine, and blood urea nitrogen (BUN) decreased with increasing steaming frequency. ROS decreased only in LT9, and SOD, catalase and glutathione peroxidase (GPx) increased with increasing steaming frequency. phospho-AMP-activated protein kinase (p-AMPK) increased and sterol regulatory element-binding protein 2 (SREBP-2), Phospho-Acetyl-CoA Carboxylase (p-ACC) and HMG-CoA reductase (HMGCR) decreased with increasing steaming frequency. Liver staining showed a decrease in hepatic fat accumulation of LT9. LT9 showed significant results in all experiments. Conclusions : LT9 showed significance of anti-lipid effect and improved fatty liver of hyperlipemia induced rats fed on high cholesterol diet, In conclusion, LP9 can be effectively used for the treatment of hyperlipidemia.

  • Objectives : This research was performed to investigate the effect of acupuncture using invasive low level laser therapy (LLLT) at Yolgyol (LU7) + Yogu (LR5) on weight gain, food intake, food efficiency, lipid metabolism, atherogenic index, HTR (HDL-cholesterol to total cholesterol ratio) and morphological change of hepatic tissue in hyperlipdemia rats. Methods : Experimental groups were divided into high fat diet group (Control group), high fat diet and acupuncture therapy group at LU7 + LR5 (AT group), high fat diet and acupuncture group using 10 mW LLLT at LU7 +LR5 (LA10 group), high fat diet and acupuncture group using 20 mW LLLT at LU7+LR5(LA20 group), high fat diet and acupuncture group using 60 mW LLLT at LU7 + LR5 (LA60 group), once per 3 days during 9 weeks. Results : Body weight was decreased significantly in AT and LA20 groups compared with Control group. Food intake was increased significantly in LA60 group compared with Control group. Food efficiency was decreased significantly in LA10, LA20 and LA60 groups compared with control group. In the lipid metabolism, total cholesterol was decreased significantly in AT, LA10, LA20 and LA60 groups, triglyceride was decreased significantly in LA10, LA20 and LA60 groups, TG/HDL-cholesterol ratio was decreased significantly in LA 60 group compared with control group. In the morphological change, hepatic tissue were not showed balloning degeneration and irregular arrangement of hepatic cell in LA10 and LA20 groups with control group. Conclusions : Acupuncture using LLLT at LU7+LR5 can manage hyperlipemia by controlling body weight, food intake, food efficiency ratio and lipid metabolism.

Influence of Functional Food Containing Bacillus polyfermenticus SCD on Lipid and Antioxidant Metabolisms in Rats Fed a High-Fat and High-Cholesterol Diet

  • We evaluated the effects of newly developed functional food containing Bacillus polyfermenticus SCD as the main material on the lipid and antioxidant metabolisms of hypercholesterolemic rats. Thirty male SD rats were divided into three groups after a 1-week adaptation period and were fed with a high fat-cholesterol diet (control), or with a high fat-cholesterol diet supplemented with low or high doses ($3.1\;{\times}\;10^6\;cfu/day$ or $3.1\;{\times}\;10^8\;cfu/day$) of B. polyfermenticus SCD and other physiological active materials for 6 weeks. Both doses of B. polyfermenticus SCD significantly reduced hepatic total cholesterol and triglycerides, while increasing the fecal excretion rates of total lipid, total cholesterol and triglycerides. B. polyfermenticus SCD increased the total radical trapping antioxidant potential (TRAP). The erythrocytic glutathione peroxidase activity in the B. polyfermenticus groups was significantly lower than that in the control group. Plasma TRAP levels exhibited a highly significant negative correlation with hepatic total cholesterol while a significant positive correlation was detected between fecal total cholesterol and plasma TRAP. This hypolipidemic and antioxidative effect of B. polyfermenticus SCD seemed to be unrelated to its dosage. These results suggest that functional food containing B. polyfermenticus SCD can improve oxidative stress and hepatic lipid profiles by enhancing the excretion of cholesterol and triglycerides in feces of rats fed with high fat-high cholesterol diet.

Effect of Platycodon Grandiflorum Aqueous Extract on Lipid Levels in Rats

  • This study investigated the effects of Platycodon grandiflorum aqueous extract on lipid concentration of serum and liver in rats fed high cholesterol diet Male Sprague-Dawley rats were assigned to three groups (control, low dose of extract, high dose of extract) for four weeks. The serum total cholesterol concentration was significantly lower in the low and high doses of extract groups than in the control group. There was a significant decrease in the free cholesterol, cholesterol ester, LDL-cholesterol, and triglycerides concentrations in serum, and the total cholesterol and the triglycerides contentin liver in the low and high doses of extract groups compared to the control group. When the serum phospholipid concentration was compared among the groups, it was significantly lower in high dose of extract group than in control and low dose of extract group. It can be postulated that Platycodon grandiflorum aqueous extract may possess substantial hypolipidemic properties in rats.

식이의 총지방량과 P/S Ratio가 Plasma HDL-Cholesterol과 혈장 및 조직내의 지질함량에 미치는 영향 (Effects of Dietary Fat Level and P/S Ratio on HDL-cholesterol, Total Cholesterol and Triglyceride in Plasma and Selected Tissues of Rats)

  • The present studies were designed to compare the effects of both dietary fat levels and P / S ratio on lipid components in plasma and tissues. Changes in plasma HDL-cholesterol, cholesterol and TG, and also in tissue cholesterol and TG were determined in young rats fed diets providing total dietary fat as 10%, 25% or 45% of calories and P / S ratio as 0.2 or 4.0. Plasma cholesterol levels were getting higher as dietary fat levels increased at P / S 0.2. Plasma cholesterol was lower in rats fed dietary fat either 25% or 45 %, each with P / S 4.0. But at 10% no change in plasma cholesterol were observed by P / S 4.0 because of a possible insufficiency of the absolute amount of PUFA. HDL-cholesterol was rather less sensitive to the modification of dietary fat level, but was reduced in rats fed diets of P / S 4.0 at either 25% or 45% fat, even though HDL-cholesterol were increased in the group of 10% with P / S 0.2. Total cholesterol per g- liver were significantly increased as dietary fat levels increased. Liver cholesterol levels were higher in rats fed diets of P / S 4.0 at higher fat levels (25% or 45%) which possibly suggested that a reduction of plasma cholesterol by high PUFA diet was not at least from a decreased synthesis of cholesterol in liver. However, in muscle no significant differences were found by feeding high P / S ratio at each levels of fat. At 10% fat level, compared to 25% or 45%, cholesterol level was lower in g-liver but higher in g- muscle. Plasma TG was decreased as more dietary fat were supplied at P / S ratio, but no consistant response obtained at low P / S ratio. TG per g-liver were reduced by feeding P / S 4.0 diet at 10% or 45% fat level but no differences were found in muscle. P / S 4.0 diet was more efficient in lowering plasma cholesterol TG and HDL-cholesterolt levels only if fat level was more than 25% of the total calories And young rats were more resistant to dietary fat modification.

Efficacy of nobiletin in improving hypercholesterolemia and nonalcoholic fatty liver disease in high-cholesterol diet-fed mice

  • BACKGROUND/OBJECTIVES: Nobiletin (NOB), a citrus flavonoid, is reported to have beneficial effects on cardiovascular and metabolic health. However, there is limited research investigating the effect of long-term supplementation with low-dose NOB on high-cholesterol diet (HCD)-induced hypercholesterolemia and non-obese nonalcoholic fatty liver disease (NAFLD). Therefore, we investigated the influence of NOB on hypercholesterolemia and NAFLD in HCD-fed mice. SUBJECTS/METHODS: C57BL/6J mice were fed a normal diet (ND) or HCD (35 kcal% fat, 1.25% cholesterol, 0.5% cholic acid) with or without NOB (0.02%) for 20 weeks. RESULTS: HCD feeding markedly reduced the final body weight compared to ND feeding, with no apparent energy intake differences. NOB supplementation suppressed HCD-induced weight loss without altering energy intake. Moreover, NOB significantly decreased the total cholesterol (TC) levels and the low-density lipoprotein (LDL)/very-LDL-cholesterol to TC ratio, and increased the high-density lipoprotein-cholesterol/TC ratio in plasma, compared to those for HCD feeding alone. The plasma levels of inflammatory and atherosclerosis markers (C-reactive protein, oxidized LDL, interleukin [IL]-1β, IL-6, and plasminogen activator inhibitor-1) were significantly lower, whereas those of anti-atherogenic adiponectin and paraoxonase were higher in the NOB-supplemented group than in the HCD control group. Furthermore, NOB significantly decreased liver weight, hepatic cholesterol and triglyceride contents, and lipid droplet accumulation by inhibiting messenger RNA expression of hepatic genes and activity levels of cholesterol synthesis-, esterification-, and fatty acid synthesis-associated enzymes, concomitantly enhancing fatty acid oxidation-related gene expression and enzyme activities. Dietary NOB supplementation may protect against hypercholesterolemia and NAFLD via regulation of hepatic lipid metabolism in HCD-fed mice; these effects are associated with the amelioration of inflammation and reductions in the levels of atherosclerosis-associated cardiovascular markers. CONCLUSIONS: The present study suggests that NOB may serve as a potential therapeutic agent for the treatment of HCD-induced hypercholesterolemia and NAFLD.
