Effects of Ascorbate and $\alpha$-Tocopherol Administration on Liver Function in Chronically Ethanol-Treated Rats (에탄올을 장기간 섭취한 흰쥐의 간기능에 미치는 아스코르빈산 및 알파토코페롤 투여의 영향)

  • These studies were carried out to investigate the effects of ascorbate and $\alpha$-tocopherol administration on the biochemical parameters of liver function and hydroperoxidation in liver of chronically ethanol-treated rats. Forty healthy Sprague-Dawley (SD) rats weighing about 120g were used for this experiment and divided into the following 5 groups: control group (CON), ethanol control group (ECON), ascorbate treated group (EASC), $\alpha$-tocopherol treated group (ETOC) and ascorbate.$\alpha$-tocopherol mixture treated group(EASC + ETOC). Ethanol was administered orally by 5ml per kg, body weight per day for 8weeks. Antioxidants treated groups were administered orally by 5mg per kg body weight per day in saline solution for 3 weeks. Lipid hydroperoxides were analyzed by using chemiluminescense-high performance liquid chromatography (CL-HPLC) method phosphatidylcholine hydroperoxide value (PCOOH) in liver tissues. Ethanol treatment significantly (p<0.05) resulted in an increase in GPT and GOT activities and liver hydroperoxide values comparing with the untreated control, while administration of $\alpha$-tocopherol and ascorbate+$\alpha$-tocopherol to the chronically ethanol-treated rats significantly (p<0.001) decreased GPT and GOT activities and liver hydroperoxide value. These results indicate that dietary $\alpha$-tocopherol and $\alpha$-tocopherol combined with ascorbate administration may inhibit the formation of liver lipid hydroperoxidation in vivo and were very effective in recovering the liver function in chronically ethanol-treated rats.

  • Sasa (S.) quelpaertensis Nakai (Korean name, Jeju-Joritdae), which has anti-oxidative and anti-inflammatory activities, is a type of bamboo grass distributed widely in Jeju Island, Korea. S. quelpaertensis leaves are used for therapeutic purposes in traditional Korean medicine. This study examined the hepatoprotective effects of the S. quelpaertensis ethyl acetate fraction (SQEA) in a mouse model to mimic alcoholic liver damage. The mice were administered orally with 30% alcohol (5 g/kg) once per day with or without SQEA treatments (100 and 200 mg/kg) for 14 days consecutively. Alcohol consumption increased the serum alcohol content and histopathological changes but reduced the liver weight. Moreover, the livers of the alcohol group exhibited the accumulation of malondialdehyde and cytochrome P450 2E1 (CYP2E1), and lipid droplet coating protein perilipin-2. On the other hand, SQEA dose-dependently attenuated the alcohol-induced serum ethanol content and liver histopathological changes but increased the liver weight. Moreover, SQEA attenuated the level of CYP2E1 and inhibited alcohol-induced lipogenesis in the liver via decreased perilipin-2 expression. These results suggest that SQEA can provide a potent way to reduce the liver damage caused by alcohol consumption.

A Case of Rhabdomyosarcoma in the Liver (간에서 발견된 횡문근육종 1례)

  • Pure primary hepatic rhabdomyosarcoma in adult is very uncommon. There have been only five previous case of primary rhabdomyosarcoma of the adult liver. A case of hepatic rhabdomyosarcoma was diagnosed in a 52 year-old female. She was admitted to the hospital due to the epigastric pain and weight loss. A CT scan of the abdomen showed a large hypodense mass with focal calcification occupies most of the both lobes of the liver. The liver biopsy showed massive liver tumor composed entirely of oval shaped cells showing light microscopic and immunohistochemical evidence of rhabdomyoblastic differentiation. We report a case of hepatic rhabdomyosarcoma with review of literature.

A Study on the Enzyme Activity in Liver and Kidney of Mouse Administered Mercury Chloride in the Drinking Water (무기수은이 마우스의 간장 및 신장내 효소반응에 미치는 영향)

  • The accumulations of mercury, lactate dehydrogenase and antioxidant enzymes activities of which are glutathione peroxidase, catalase and superoxide dismutase, and pathological changes were investigated in liver and kidney of mice which were fed the water supplemented with two levels (0.5 mM and 1.0 mM) of mercury chloride (HgCl$_2$). During the mercury feeding, the weight gain of mice in experimental groups was less than that of control group mice, while no overt signs related to mercury toxicity were noted in any experimental groups. Mercury concentrations in liver and kidney increased significantly in the early period (1~2 weeks) after mercury administration, which were measured as high as 100 times in liver and kidney in comparison to those of the control groups, but there were relatively stable for the levels of accumulation in following periods. The lactate dehydrogenase activities in liver and kidney were relatively increased in the period of 2~3 weeks of mercury administration in the experimental groups, there were normal levels in other periods of administration without the dose-dependencies. The glutathione peroxidase activities were not affected by the dosages of mercury chloride and the duration of ingestion. But the catalase activities significantly increased in 2~3 weeks after ingestion, and the superoxide dismutase activities of kidney also showed a peak in 3 weeks of ingestion while this peak was not found in the results measured in liver tissues.

Liver PPAR${\alpha}$ and UCP2 are Involved in the Regulation of Ovariectomy-Induced Adiposity and Steatosis by Swim Training

  • It is suggested that ovariectomy induces body weight gain primarily in the form of adipose tissue in rodents. Since liver peroxisome proliferator-activated receptor ${\alpha}$ (PPAR${\alpha}$) and uncoupling 2 (UCP2) are involved in the regulation of energy expenditure, it was investigated whether swim training regulates ovariectomy-induced adiposity and steatosis through liver PPAR${\alpha}$ and UCP2 activation in female ovariectomized mice, an animal model of postmenopausal women. Swim-trained mice had significantly decreased adipose tissue weights compared with sedentary control mice. Histological analysis showed that hepatic lipid accumulation was inhibited by swim training. Concomitantly, swim training significantly increased mRNA levels of PPAR${\alpha}$ and its target genes responsible for peroxisomal fatty acid ${\beta}$-oxidation, such as acyl-CoA oxidase, enoyl-CoA hydratase/3-hydroxyacyl-CoA dehydrogenase and thiolase in the liver. Moreover, swim training induced the mRNA expression of UCP2. These results suggest that swim training can effectively prevent adiposity and steatosis caused by ovariectomy, in part through activation of liver PPAR${\alpha}$ and UCP2 in female obese mice.

  • For the purpose of investigating whether the administration of sunflower pollen load has any influence upon liver cholesterol metabolism in mouse, lipids were isolated from sunflower pollen load, identified and quantitated by thin-layer and gas liquid chromatographies. We also studied changes in liver cholesterol level in mouse according to the amount and the period of pollen load administration. Lipids of sunflower pollen load were constituted 84.10f of neutral lipid, 10.50% of glycolipid and 5.40% of phospholipid. The main fatty acid contents of neutral lipid, glycolipid and phospholipid were ranged 28.48 to 33.70% of linoleic acid, 12.90 to 47.50% of palmitic acid ana 11.20 to 12.20% of oleic acid, however, phospholipid contained more palmitic acid than the other lipids. The body weight of the Pollen fed mouse significantly increased during experimental Period in comparison with control group. From the fact tat the ratio of liver weight to body weight of pollen fed mouse was smaller than that of control group, it was proved that liver lipid metabolism of pollen fed mouse was more active than that of control group. During early experimental period, liver cholesterol level had been increased according to pollen load administration(P.O), and then the level decreased rapidly to the similar level to that of control group at the end of the period.

Hepatoprotective Effects of Pig Placental Hydrolysates on Liver Damage-induced Rats by Injecting Carbon Tetrachloride (사염화탄소 주입으로 간손상을 유발시킨 백서에서 돈태반가수분해 물 투여가 간기능 보호에 미치는 영향)

  • The aim of this study was to determine hepatoprotective effects of pig placenta hydrolysates (PLA) on liver damage-induced rats. Sprague Dawley rats were subcutaneously injected with carbon tetrachlorides and divided into 4 groups: negative control (DMSO treated), two PLA treated groups, low dosage of PLA (LPLA; 300 mg/kg body weight) and high dosage of PLA (HPLA; 1000 mg/kg body weight), and silymarin treated group (80 mg/kg body weight). Rats in each group orally received assigned compounds for 3 weeks. For the normal control group vehicle (corn oil) was injected. After 3 weeks, liver and spleen weights were greater in the two PLA and silymarin groups than the negative control group. After bromosulfalein (BSP) treatment, the clearance rates were in the descending order of negative control > LPLA > HPLA = silymarin and serum BSP levels exhibited the opposite trends of the clearance rates. Serum levels of aspartate aminotransferase, alanine aminotransferase, triglyceride, and cholesterol lowered with PLA in a dose-dependent manner. The decrease with HPLA was similar to that with silymarin. In addition, triglyceride, cholesterol, lipid peroxides levels in the liver decreased in HPLA and silymarin compared to the negative control. The liver morphometry was improved in HPLA- or silymarin-treated rats in comparison to the negative control. In conclusion, HPLA administration ameliorated the hepatotoxicity as much as silymarin in rats injected with carbon tetrachlorides.

Methanol Extract of Agaricus blazei Murill Reduces Hepatic Damage Induced by $CCl_4$ and High Fat and Improves Liver Lipid Profile in Rats

  • The effect of methanol extract from Agaricus blazei Murill on the hepatotoxicity was investigated $CCl_4$ is one of the oldest and most widely used toxins for the induction of hepatic damages and fibrosis in experimental animals. In this study, male Sprague-Dawley(SD) rats were randomly divided into 6 groups of the control(C), $CCl_4(T),\;CCl_4$ and high fat group(TL) with matching sub-groups of Agaricus blazei Murill extract-fed groups of CA, TA and TLA. Methanol extracts of Agaricus blazei Murill were fed 50 mg/kg B.W daily via drinking water. A 1.2 mL of $CCl_4/kg$ body weight was administered by oral intubation twice a week for total of six times. The levels of total-cholesterol, TG, LDL and LDL-phospholipids were elevated by $CCl_4$ treatment as compared to the control(C). However, Agaricus blazei Murill methanol extract feeding in the group of TA and TLA significantly(p<0.05) decreased TG by 53.1 % and 17.9% compared to the internal control of T and TL, respectively. Triglyceride of TL was increased by 3.33 times(p<0.05) compared to the control(C) with $CCl_4$ and high fat administration from 3.78 mg/g to 12.60 mg/g liver. The extract(CA) also reduced kidney weight compared to the control(C). With the administration of high fat and $CCl_4$(TLA), the extract reduced the organ weight of both liver and kidney and further, significantly reduced TG, total cholesterol and GTP activity. Hepatoprotective effects of Agaricus blazei Murill on GOT, GPT, AP and LDH activities were enhanced by the extract feeding. Electronmicrograph showed that $CCl_4$ deteriorated the structure of cytoplasmic matrix with its uneven distribution. However, the extract reconstituted the damaged cytoplasm and stimulated mitochondriogenesis. The above results suggest that Agaricus blazei Murill may have a possible protective effect against chemically induced liver damage and further help to reduce the symptoms of fatty liver.

  • Objectives : Present study investigated the hepatoprotective effects and the optimal mixing ratio of fermented Schizandrae Fructus Pomace (fSFP) and Hoveniae Semen Cum Fructus (HSCF) extract combination in carbon tetrachloride (CCl4)-induced acute liver injury mice. Methods : ICR mice were orally administered with 200 mg/kg of fSFP, HSCF and mixtures of fSFP and HSCF [MSH (w:w); 1:1, 1:2, 1:4, 1:6, 2:1, 4:1, 6:1, and 8:1] for 7 consecutive days. Silymarin (100 mg/kg) was administered as a reference drug. 0.5 mL/kg of CCl4 was injected intraperitoneally to induce acute liver injury. Body weight gain, relative liver weight, serum chemistry, histopathological analysis, and hepatic endogenous antioxidants capacities were observed. Results : All diverse combinations of MSH significantly reduced relative liver weight increase by CCl4. In addition, MSH administrations significantly decreased the elevation of serum alanine aminotransferase and aspartate aminotransferase activities by CCl4. Histopathological observation indicated that all MSH treatments significantly reduced the increase of degenerated hepatocytes, inflammatory cell infiltration, and histological activity index score by CCl4. Moreover, all MSH administrations reduced the elevation of malondialdehyde contents, and ameliorated the reduction of hepatic glutathione contents, superoxide dismutase activity, and catalase activity. Among the various mixing ratio of MSH combinations, MSH 1:1 and 2:1 showed the most potent anti-oxidative stress, and hepatoprotective effect. Conclusion : Present results suggest that 1:1 and 2:1 combinations of MSH is promising herbal formulation with the hepatoprotective effect against oxidative stress.

A Baseline Study on the Choice of Optimal Screening Test Items among Workers with Abnormal Liver Function Tests on Workers' Periodic Health Examination (근로자 건강진단시 간기능 이상자의 정밀검사항목 개선을 위한 조사연구)

  • Workers' periodic health examination is the main tools used to manage the health problems of most workers in Korea. The most common health problem found in workers' periodic health examination is liver disorder. Liver disorder is also one of the most common health problems in general population and one of the leading causes of mortality in adult population. Regulation proposed by government (No. 207, Ministry of Labor, 1992) defines the criteria for selection of workers with the liver dysfunction for further evaluative examination and the examination items used for diagnosis of the workers with liver dysfunction. This study was designed to evaluate the proficiency of each examination items presently defined in Regulation and propose the optimal examination items for detection of the liver disorders found by workers' periodic health examination. Study subjects are 186 workers with abnormal liver function tests in screening examination of workers' periodic health examination. Questionnaire survey including past history of liver disorder, drinking history, height and weight was done. Physical examination by physician, routine test items defined by Regulation (SGOT, SGPT, $\gamma$-GTP, protein, albumin, total and direct bilirubin, alkaline phosphatase, $\alpha$-feto protein, HBsAg and anti-HBs), anti-HCV antibody test and liver ultrasonography were done. Results are as follows; 1. Result of evaluative examination utilizing only the items defined in Regulation was; There were 75 workers with suspected live. disorder(40.3%), 63 with no liver dysfunction (33.9%), 13 with suspected hepatitis B(7.0%), 10 workers with hepatitis B(5.4%), 10 workers with hepatitis B carrier state(5.4%), 10 with alcoholic liver disorders(5.4%), 5 with fatty liver(2.7%). When alternative diagnostic criteria applying additional examination items (drinking history, body mass index, anti-HCV antibody and ultrasonography) diagnosability of liver disorder was increased. When all four items were included, final results were; 23 workers (17.8%) with hepatitis B (10 carriers, 13 suspects and 10 hepatitis B), 10 (5.4%) with hepatitis C(4 carriers, 5 suspects and 1 hepatitis C), 13(7.0%) with alcoholic liver disorder, 45(24.2%) with fatty liver (40 suspects, 5 fatty liver), 410%) with suspected liver disorders and 44 (23.7%) with normal liver. 2. Of examination items defined by Regulation, only SGOT, SGPT, $\gamma$-GTP and HBsAg were significantly different in abnormal rate and mean value, and all other laboratory findings did not showed significant difference between two groups. Drinking history, body mass index and anti-HCV antibody test which are the items that authors included in this study, also showed significant difference between two groups. Utilization of body mass index (BMI) for abnormal liver function group in diagnosis of fatty liver had high specificity (97.6%) but sensitivity (22.3%) was low. Therefore we suggest that SGOT, SGPT, $\gamma$-GTP, HBsAg, alcohol drinking history, BMI and anti-HCV Ab were useful for diagnosis of liver disorders among worker's periodic health examination.
