귀뚜라미의 수용성 및 알콜 추출물의 간보호 효과 (Protective Effects of Water/Methanol Extracts of Cricket on the Acute Hepatic Damages in the ICR-mice Induced by Administration of $CCl_4$)

  • 귀뚜라미의 곤충산업의 발달로 대량체계화 사육된 귀뚜라미의 새로운 용도 개발의 일환으로 귀뚜라미의 간보호 효과를 조사하였다. 사염화탄소로 6주령의 ICR 응성 생쥐에 급성간염을 유발시킨 후 귀뚜라미 물 추출물(20 mg/kg) 및 메탄올 추출물(20 mg/kg)을 각각 투여하여 GOT, GPT, LDH 간혈청 효소 활성을 조사한 결과 귀뚜라미 메탄올 추출물은 대조구에 비해 GOT, GPT, LDH치가 74, 50, 101% 감소하였다. 지질과산화 억제정도는 귀뚜라미 물추출물 및 메탄올추출물이 정상군 대비 40%이상의 억제 효과를 나타내었다. 또한 강제수영법에 의한 귀뚜라미 추출물의 항피로 효과를 측정시 대조군에 비해 메탄올 200 mg/kg에서 현저한 수영시간 증가를 보여 양성대조군인 토코페롤 100 mg/kg 수준의 유의성 있는 항피로 효과를 나타낸 것을 알 수 있었다. 이상의 결과로 보아 귀뚜라미의 수용성 및 알콜추출물은 간보호 효과를 가지는 건강보조식품으로서 개발 가능성을 가짐을 알 수 있다.

배무채 에탄올층의 D-galactosamine 간손상에 대한 보호효과 (Hepatoprotective Effect of Ethanol Extract of xBrassicaraphanus on Liver Injury in Rats Treated by D-galactosamine)

  • The protective effect of xBrassicoraphanus (BR) on liver inhury was evaluated in the rats with liver injury induced by i.p. injection of D-galactosamine (GalN) following 2 week oral treatment of ethanol extract of xBrassicoraphanus (EBR). EBR (200 mg/kg) significantly suppressed the levels of ALT, AST, SDH, ${\gamma}-GT$, ALP, LDH and lipid peroxidation compared with GalN treated control, while EBR at 100 mg/kg significantly suppressed AST and ${\gamma}-GT$. Similarly, EBR at 200 mg/kg significantly attenuated the levels of Phase I enzymes such as XO, AO, AH and AD as well as significantly increased the levels of Phase II enzymes such as SOD, catalase and GSH-Px in the GalN treated rats. Taken together, these results indicate that the ethanol extract of xBrassicoraphanus may have a hepatoprotective effect against GalN induced liver injury, suggesting the ethanol extract of xBrassicoraphanus can be applied as hepatoprotective functional food. However, its mechanism should be further studied in molecular and cellular view points.

  • This study was performed to investigate the effect of Angelicae gigantis Radix extract oil the hepatic detoxifying enzyme activities and lipids. Male sprague-dawley rats were administrated the extracts with benzo(a)pyrene, a hepatotoxic agent, inducing liver damages. Results obtained from this study were as follows: 1. The markedly increased enzyme activities (AST, ALT, LDH, ALP, ${\gamma}$-GTP, GSH-Px) in B(a)P induced groups tended to be decreased by the treatment of the Angelicae gigantis Radix extract. 2. Liver GSH content and lipid peroxide activity were decreased by the administration of the extracts. 3. It tended that the curative effects were better than the protective effects of the extracts.

  • The present study was undertaken to find a protective effect of Coptis Rhizoma Extract on liver injury induced by benzo(a)pyrene in rats. This crude drug significantly suppressed the increase of several biochemical parameters such as transaminase, lactate dehydrogenase and alkaline phosphatase in serum and liver and total cholesterol. But there were little changes in elevated serum phospholipid level. The results suggested that Coptis Rhizoma prevent bezo(a)pyrene-induced hepatic injury.

유기주석화합물의 독성에 대한 홍삼의 방어효과 - 면역 및 성 호르몬물질 중심으로 - (The protective effect of Red Giseng on the organotin compounds(TBTO) poison : Focusing on the Immunity and sex hormone)

  • This study was carried out effect of red ginseng extract(1.0g/kg) on organotin compounds (10, 20 and 40 mg/kg) which poisons against some organs like thyroid gland, liver, kindey, testis, ovary, serum immuinty and sex hormone activity of rats were examinned by gastric tubing for 3 weeks. The weight of each organ in TBTO treated group were significantly increased other organs which excepted kinedy in males and only liver in females.(p<0.05, p<0.01). In case of Immunity activity of each sex, IgM level was small change comparsion with that of control group in all sex. but IgG level was significantly decreased females rather than males comparsion with that of control group.(p<0.05, p<0.01) In case of sex hormone activity, the testosterone activity of males and the estradiol activity of females were significantly decreased rather than the control group. on the other hand, red singsong treated group was only significantly increased estradiol activity.( p<0.05, p<0.01)

The Effect of Petasites japonicus Extract on Hepatotoxicity in Rats

  • Petasites japonicus (Compositae) is a perennial herb which has been used as treatment of antitussive, expectorant, sedatives, paralysis, diurectics in folk remedies. The pharmacological studies of this natural drug have not yet estabilished. So, we examined anti-lipid peroxidative effects and liver protective effect on $CCl_4$ induced lipid peroxidation and hepatotoxicity in rats. In vivo liver lipid, MeOH Ex. revealed significant increase of anti-lipid peroxidative effects according to concentration dependently. In chemical parameters obtained from serum analysis, MeOH Ex. as a increase of medicine concentration (0.25 g/kg, 0.5 g/kg, 1.0 g/kg), GOT, GPT, AlP decreased. In 0.5 g/kg GPT administered group, there was relative in GOT, AlP. Cholesterol decreased in 1.0 g/kg and 1.0 g/kg administered group, BUN decreased relatively in 1.0 g/kg administered group.

기니피그에서 홍삼 사포닌의 2,3,7,8-TCDD 독성 방어 효과 (Protective Effects of Red Ginseng Saponins against to 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) Induced Toxicity in Guinea Pigs)

  • This study was carried out to investigate the protective effect of Red Ginseng Saponins on 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) induced toxicities in guinea pigs ($200{\pm}10$ g). Normal control (NC) group guinea pigs ($200{\pm}10$ g) received vehicle and saline, while the TCDD-treated (TT) group was given water-extract (WE), saponin fraction (SF) and non-saponin fraction (NSF). Korean red ginseng fractions were administered from 1 week before TCDD-exposure for 4 weeks. Body weight loss and deteriorated clinical parameters related to sugar metabolism and liver function such as lipase and AST, respectively, these were significantly reduced by both saponin and non-saponin fractions. However, increase of lipase was attenuated by the saponin fraction in a dose-dependent manner. Only AST was affected by the saponin fraction. The results suggest that saponins are active substances in the Korean red ginseng water extract against TCDD induced toxicities in Guinea pigs.

단삼(丹蔘)이 고지혈증 흰쥐의 혈중 지질 변화에 미치는 영향 (Effects of Salviae miltiorrhizae Radix (SMR) on Serum Lipid Level in Hyperlipidemic Rats)

  • Objective : Salviae miltiorrhizae Radix (SMR) has widely used to treat patients with cardiovascular diseases such as coronary arteriosclerosis, angina pectoris and hyperlipidemia. This study was designed to investigate the effects of SMR on changes in serum cholesterol and protective effects on liver tissue damage in Hyperlipidemic rats. Methods : The present author investigated changes in serum glucose, cholesterols, AST/ALT and histopathological changes of liver tissue by oral administration of SMR in Rats. Results : In this study, body weights of hyperlipidemic rats induced high fat diet did not changed, and treatment with SMR did not affect body weights in hyperlipidemic rats. For experimental period, Food and Water uptake in SMR administered group were the same as those in hyperlipidemic control group. In this experiment, treatment with SMR decreased total cholesterol and AST in serum which elevated by high fat diet respectively. In addition, SMR administration protected liver tissue from damage induced by induction of hyperlipidemia. Conclusions: These results suggest that SMR is useful to treat patients with disease related to cardiovascular diseases including hyperlipidemia, because SMR can decrease cholesterol and AST in serum and also have non-specific protective effect on tissues including liver.

Effect of Onion Extract on the Carbon Tetrachloride-induced Liver Injury in Mouse

  • The protective effects of onion extract (OE), onion powder extracted in ethanol for 2 days. on carbon tetrachloride ($CCl_4$)-induced hepatotoxicities and the possible mechanisms involved in this protection were investigated in mice. Pretreatment with OE prior to the administration of $CCl_4$ significantly reduced the increase in serum alanine and aspartate aminotransferase activities and hepatic lipid peroxidation in a dose-dependent manner. In addition, pretreatment with OE significantly prevented the depletion of reduced glutathione content in the liver of $CCl_4$-intoxicated mice. $CCl_4$-induced hepatotoxicity was also prevented, as indicated by a liver histopathologic findings. The effects of OE on the cytochrome P450 (P450) 2E1, the major isozyme involved in $CCl_4$ biotransformation were investigated. Treatment of mice with OE resulted in a significant decrease in P450 2E1-dependent p-nitrophenol and aniline hydroxylation in a dose-dependent manner. Consistent with these observations, the P450 2E1 expressions were also decreased, as determined by immunoblot analysis. OE also exhibited antioxidant effects in FeCl$_2$-ascorbate induced lipid peroxidation in rat liver homogenates and in superoxide radical scavenging activity. These results show that the protective effects of OE against the $CCl_4$-induced hepatotoxicity may be due to its ability to block bioactivation of $CCl_4$, mainly tty inhibiting the expression and activities of P450 2E1 and by scavenging free radicals.

흰쥐 장기내 카드뮴 독성에 대한 셀레늄이 방어효과에 관한 연구 (A Study on the Protective Effects of Selenium Against Cadmium Toxicity in Mice)

  • This study is performed to find out the effects of selenium against cadmium toxicity. The experimental mice were divided into 6 gruops such as control group, cadmium alone treatment group, selenium treatment groups and two simultaneous treatment groups of selenium and cadmium. Mice were given intraperitoneal administration with two dosage of sodium selenite such as 1.0 mg/kg, 2.5 mg/kg body weight and cadmium chloride was administered 3.0 mg/kg body weight. After giving the challenge dose, the concentration of cadmium and metallothionein and histopathological change of liver and kidney were determined. The results were summarized as follows on 1. The simultaneously administration of selenium and cadmium significantly more decreased cadmium concentration in kidney and iiver tissues compared to the administration of cadmium only(P<0.05). 2. The simultaneously administration of selenium and cadmium more increased metallothionein concentration compared to administration of cadmium only. 3. The simultaneously administration of selenium and cadmium more decreased cadmium concentration in urine compared to the administration of cadmium only. 4. When liver and kidney tissues were observed with optical microscope, no obvious changes were visible in those tissues.
