Preparation of Black Soybean Peptide and Its Effect on Weight Reduction in Rats

  • The large-scale preparation method for black soybean peptide (BSP; about 70% less than 10,000 Da; highly enriched with glutamic acid, aspartic acid, and arginine) was developed, and its effect on weight reduction and lipid profiles in rats was investigated. Sprague-Dawleymale rats were assigned to four dietary groups (high-fat diets containing 0, 2, 6, and 10% BSP) and fed four weeks to examine the effects of BSP. During the experiment, food intake was measured every two days and body weight was monitored two times a week. After the supplementation of BSP, liver and adipose tissues (epididymal, retroperitoneal and perirenal adipose tissue) in the rats were weighted and the lipid profiles in serum, liver, and feces were analyzed. At the results of body weight gain, liver and epididymal adipose tissue weight, BSP groups were more decreased than HF group (0% BSP), with greater decreases at higher BSP levels. The same patterns were shown in lipid profiles of serum, as BSP was increased, triglyceride and total cholesterol concentration decreased. The serum HDL-cholesterol level was increased with increasing at BSP levels. Total cholesterol concentration of liver and feces were decreased and increased, respectively, as BSP increased. The results confirm that BSP is involved in reducing the body weight and the improvement of lipid composition in serum and liver of rats and that BSP can be applied in weight reduction in the food products industry.

  • To investigate effects of dietary fish oil and vitamin E level on the tissue levels of vitamin E and vitamin A and to see which tissue is sensitive to lipid peroxidizability, male Sprague-Dawley rats were fed experimental diets composed of either menhaden oil or soybean oil nad either low(equivalent to 17 mg $\alpha$-tocopherol) or high (equivalent to 140mg $\alpha$-tocopherol) vitamin E level for 4 weeks. Palsma TBARS per mg lipid was significantly elevated in rats fed fish oil with low vitamin E level compared to soybean oil-fed rats. TBARS levels of liver, heart, kidney and liver microsomes were also increased by feeding fish oil with low vitamin E level. Plasma TBARS level was significantly correlated with TBARS levels of liver, heart, kidney and liver microsome. Plasma vitamin E level of groups with vitamin E supplementation was elevated significantly as compared to the those without vitamin E supplementation, whereas vitamin E levels of liver, heart and kidney were not changed significantly. Plasma TBARS was negatively correlated with plasma vitamin E(r=0.5763, P<0.001) and A(r=-0.4523, P<0.01) and seems to be a good indicator of in vivo lipid peroxidative stress.

마디풀(Polygonum aviculare L.) 성분의 지질과산화억제 및 간보호에 미치는 효과 (Anti-lipid Peroxidation and Liver Protective Effects of Polygonum aviculare L.)

  • The ethylacetate fraction of the overground portion of Polygonum aviculare L. exhibited the anti-lipid peroxidation and the liver protective effect in intoxicated rats. Through silica gel chromatography of the ethylacetate fraction monitered by bioassay, two flavonoids, avicularin and juglanin were isolated as active components. Avicularin and juglanin remarkablely inhibited the lipid peroxidation of rat liver induced by 50% ethanol. Especially avicularin exhibited the stronger anti-lipid peroxidation effect than juglanin. Avicularin as a main principle of Polygonum aviculare L. significantly exhibited liver protective activities by decreasing s-GOT and s-LDH levels which represent for the hepatotoxicity induced by $CCl_4$ in rats. In addition, avicularin significantly decreased not only s-LDH but also s-bilirubin levels in intoxicated rat induced by ${\alpha}-naphthylisothiocyanate\;(ANIT)$. These results suggest that avicularin has the protective effects against the hepatoxicity induced by $CCl_4$ and ANIT in rats.

  • Juvenile visceral steatosis (JVS) mouse is an animal model of the systemic camitine deficiency. JVS mice first develop fatty liver following cardiac hypertrophy. hyperammonemia, etc. To clarify the relationship between fatty liver and other symptoms. lipid hydroperoxides levels of peripheral oragans in JVS mice at 1 month were determined by the use of phosphine derivatives. We also report here a new method to quantitate the lipid components level in fatty liver of JVS mice.

  • As a part of pharmacological studies of saikosaponins, which were reported to exhibit diverse biological activities especially concerning with liver function, effects of saikosaponin on metabolizing enzymes and lipid peroxide contents in liver were examined. As the result, UDP-glucose dehydrogenase activity and lipid peroxidation which were due to acetaminophen were inhibited by saikosaponin treatment. But other metabolizing enzyme activities were not modified.

당뇨쥐의 간 Microsome에서 Phopholipase A_{2} 활성과 지질과산화 (Phospholipase $A_{2}$ Activity and Lipid Peroxidation in Liver Microsome of Streptozotocin Induced Diabetic Rats)

  • The purpose of this study was to investigate phospholipase $A_{2}$ activity and lipid peroxidation I streptozotocin induced diabetic rats. Sprague-Dawley male rats weighting-Dawley male rats weighting 300$\pm$10gm were randomly assigned to normal and STZ-induced diabetic group. Diabetes was induced by intravenous injection of 55mg/kg of STZ in sodium citrate buffer(pH 4.3). Animals were sacrificed at the 6th day of diabetic states. Body weight gains were lower in DM group. Phosphatidylcholine hydrolysis in liver was not significantly different between two groups, whereas phosphatidylethanolamine hydrolysis in liver was increased by 69% in DM group comparing with that of normal group. Liver microsomal phospholipase $A_{2}$ activity and level of TBARS was increased by 91%, 109% in DM group compared with that of normal group, respectively. The present results indicate that phospholipase $A_{2}$ activity is specific to PE hydrolysis, leading to lipid peroxidation process in STZ induced diabetic rats.

무, 양파의 시료제조 방법에 따른 흰쥐의 지방대사와 항산화능에 관한 연구 (Effect of Dry Powders, Ethanol Extracts and Juices of Radish and Onion on Lipid Metabolism and Antioxidative Capacity in Rats)

  • This study was performed to investigate the effects of dry powders, ethanol extracts and juices of radish and onion on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in rats. Forty-nine male Sprague-Dawley rats weighing 157$\pm$6g were blocked into seven groups according to body weight and raised for four weeks with diets containing 5%(w/w) dry powders of two different vegetables consumed frequently by Korean-radish(Raphanus sativus L.) and onion(Allium cepa L.), ethanol extracts and juices from equal amount of each dry powder. All the powders, ethanol extracts and juices of radish and onion decreased total lipids, triglycerides and total cholesterol concentrations in plasma and liver. Above all, onion ethanol extract decreased them most remarkably. It was thought that organosulfur compounds and flavonoids extracted from onion by ethanol inhibited biosynthesis and absorption of lipid and promoted degradation of lipid. Radish powder also decreased them by increasing fecal excretions of total lipids, triglycerides and total cholesterol most effectively. Catalase and glutathine peroxidase(GSH-px) activities in red blood cell(RBC) were most remarkably increased by radish powder and onion powder respectively. Superoxide dismutase(SOD), catalase and GSH-px activities in liver were most remarkably increased by onion ethanol extract, radish powder and onion ethanol extract respectively. Xanthine oxidase(XOD) activities in liver were most effectively decreased by ethanol extracts of radish and onion. Thiobarbituric acid reactive substance (TBARS) levels in plasma and liver of experimental groups were significantly lower than those of controls. Above all, onion powder decreased them most effectively. It was thought that vitamin E and high flavonoids in onion powder inhibited lipid peroxidation, promoting liver and RBC SOD, catalase and GSH-px activities and inhibiting XOD activities effectively. Flavonoids in onion ethanol extract inhibited lipid peroxidation, promoting three antioxident enzyme activities and inhibiting XOD activities most remarkably. Also flavonoids and high vitamin C in radish powder inhibited lipid peroxidation, promoting liver and RBC catalase most remarkably and inhibiting XOD activities. In conclusion, radish and onion were effective in lowering lipid levels and inhibiting of lipid peroxidation in animal tissue. From these data, radish and onion can be recommended in the treatment and prevention of diseases such as cardiovascular disease and cancer and in delaying aging. As ethanol from onion were most effective in lowering lipid level and promoting three antioxident enzymes, and inhibited lipid peroxidation as did we should try to utilize onion skin which is discarded though reported to have abundant flavonoids. $\alpha$-carotene 첨가식이가 고지혈증 쥐의 지질과산화물과 항산화효소 활성에 미치는 영향 (Effect of $\alpha$-carotene Supplementation on Lipid Peroxides and Antioxidative Enzyme Activities in Hyperlipidetmic Rats)

  • This study was performed to investigate the effect of dietary $\beta$-carotene supplementation on lipid metabolism and antioxidant enzyme activities in hyperlipidemic rats. Fifty Sprague-Dawley male rats aging 7 weeks were fed the control diet (CD,5% corn oil) and the high fat diet (HFD,15% beef tallow +1% cholesterol) for 4 weeks and then 0.02% $\beta$-carotene was supplemented to CD and HFD group for 8 more weeks. Serum lipid compositions, lipid peroxides and antioxidative enzymes in liver were analyzed at 4, 8 and 12week of the experiment. Serum levels of total lipid, total cholesterol, triglyceride, LDL-cholesterol, VLDL-cholesterol were higher in HFD groups than in CD groups (p < 0.001), Serum levels of HDL-cholesterol were higher in CD groups than in HFD groups (p < 0.01) . The effect of $\beta$-carotene supplementation was not significant in all groups but tended to be lower in total lipid, total cholesterol and Triglyceride. Thiobarbituric acid reactive substances (TBARS) levels in plasma and liver were showed significantly higher in HFD groups (p < 0.001, p < 0.05). The effects of $\beta$-carotene supplementation on the level of plasma and liver TBARS were not found except HFD groups at 12 week. Liver conjugated diene levels in HFD groups were higher than in CD groups (p < 0.01), but the effect of $\beta$-carotene supplementation did not show any differences. Liver lipofuscin levels were not significantly different among all groups. The activities of superoxide dismutase (SOD) and catalase were significantly lower in HFD groups at 8 week (p < 0.001) but were not significantly different at 4 and 12week. The activity of SOD in $\beta$-carotene supplemented HFD group was significantly higher at 8 week (p < 0.01). Glutathione peroxidase (GSH-Px) activity was significantly lower in HFD groups (p < 0.01) and was significantly increased in groups supplemented $\beta$-carotene (p < 0.05). It is suggested that $\beta$-carotene supplementation partly decreases the serum lipid and lipid peroxide levels and increases the activities of antioxidant enzymes in hyperlipidemic rats.

  • Albio rats right after weaning, weighing $50{\sim}55g$, were divided into the control, high-carbohydrate-, high-lipid-, and high-protein-fed groups. and were fed for 12 weeks with the respective diets to observe the increase in body weights as well as changes in the chemical constituentes and enzyme activities in the liver tissue, with the following results. (1) There was little difference in the rate of increase in the body weights among the groups, showing normal growth, except the high-protein-fed group which showed decrease in rate of body weight increase from the 7th week after feeding. (2) The liver weight was either increased after 12 weeks of feeding with the high-carbohydrate and high-Lipid diets, or showed no difference with the high-protein diet, as compared to the control weight. (3) The liver cytosol protein content was increased when fed with the high-protein diet, but decreased when fed with the high-carbohydrate and high lipid diets, as compared to the control content. (4) The triglyceride and cholesterol contents in the liver were decreased in the high-protein-fed group, but increased markedly in the high-carbohydrate- and high-lipid-fed groups as compared to the control values. (5) The hepatic glucokinase, G6PD, LDH, and fatty acid synthetase activities were increased in the high-carbohydrate and high-lipid-fed groups, and GOT and CPT activities were increased in the high-protein-fed group. From the above results. it was known that the high-carbohydrate and high-lipid diets stimulated the hepatic lipid metabolism, giving rise to lipogenesis, but the high- protein diet could prevent the lipogeuesis leading to the body weight increase.

감귤파괴로부터 분리한 Hesperidin이 흰쥐의 지방과 Cadmium 대사 및 항산화능에 미치는 영향 (Effect of Hesperidin Extracted from Tangerine Peel on Cd and Lipid Metabolism, and Antioxidative Capacity in Rats)

  • This study was performed to investigate the effets of hesperidin extracted from tangerine peel on Cadmium (Cd) and lipid metabolism lipid peroxide formation, and antioxidative enzyme activities in rats. Forty-eight male Sprague-Dawley rats weighing 158.3$\pm$3.5g were blocked into eight groups according to body weight. Rats were raised for three weeks with diets containing 0 or 0.04%(w/w) cadmium chloride and 1%(w/w) extracted hesperidin from tangerine peel, commercial hesperidin or naringin. Food intake, weight gain and food efficiency ratio were significantly lower in the Cd-administered groups. The Cd concentrations in blood and liver and the Cd excretions in urine and feces were significantly higher in the Cd-administered groups. Among the Cd groups, blood Cd concentrations were decreased, fecal Cd excretions were increased, and Cd retenition ratios were decreased by feeding flavonoid diets. Plasma total lipid concentrations were significantly lower in the extracted hesperidin group, plasma triglyceride concentrations were significantly lower in the extracted hesperidin and naringin groups. Plasma HDL-cholesterol concentrations and HDL : total cholesterol ratios were increased by feeding flavonoids. Among the Cd groups, liver total lipid concentratons were decreased by feeding flavonoids. Fecal total lipid, fecal cholesterol, and fecal triglyceride excretions were significantly higher in the naringin group, and they were increased by feeding flavonoids among Cd groups. Thiobarbituric acid reactive substance concentrations in plasma and liver were higher in Cd groups, and were significantly decreased by feeding flavonoids. The activities of erythrocyte catalase, superoxide dismutase and glutathione peroxidase showed a tendency to increase by feeding. The activities of liver catalase, superoxide dismutase and glutathione peroxidase were not significantly affected by administering Cd or flavonoids. In conclusion, all flavonoids that were used in this experiment inhibited lipid peroxide formation in plasma and liver, but this effect was not caused by the increased in the activities of antioxidative enzymes.
