Cryopreserved Marine Microalgae Grown Using Different Freezing Methods

  • Marine microalgae are a key diet component in finfish and shellfish aquaculture. Cryopreservation of the microalgae is suggested by many other studies as the best method for long-term storage. To test cryopreservation efficacy, 19 taxas of marine microalgal species were examined. In the first experiment we compared dimethylsulfoxide ($Me_2SO$) and glycerol, which are most widely used as cryoprotectant agents (CPAs). The cryopreservation comprised two freezing procedures. Firstly, the samples containing the CPAs were kept at $4^{\circ}C$ for 10 min before being plunged into liquid nitrogen ($-196^{\circ}C$). Secondly, samples containing CPAs were pre-cooled ($-1^{\circ}C$ $min^{-1}$ to $-80^{\circ}C$ before being plunged into liquid nitrogen. Most of the species were successfully cryopreserved using $Me_2SO$, whereas the Prasinophyceae (T. striata and T. suecica) were successfully cryopreserved using glycerol. In general, the cooling method had no influence on the survival of the microalgae except in the case of the Tetraselmis species. In the second experiment, the cultured solution was divided before cryopreservation into concentrated and non-concentrated groups to identify the effect of cell density during cryopreservation. After 12 months of storage, the samples were again divided into centrifugation and non-centrifugation groups to learn the effect of $Me_2SO$ on the culture. Viability and growth of the microalgae were not influenced by cell density or the centrifugal removal of the $Me_2SO$ after thawing.

죽초액의 첨가가 재래닭의 생산성 및 육질에 미치는 영향

  • This experiment was conducted to investigate the effects of Bamboo Vinegar Liquid(BVL) on performance and meat quality of Korean native chicken(KNC) for 8 weeks. One hundreds KNC were divided into 4 groups with 5 replicates per treatment and placed in pens. Dietary levels of food waste 0%, 1%, 2% and 4% were included in experimental diets. The growth performance and carcass weight of KNC fed BVL supplement diet were significantly different(P<0.05) from the control group. The lipid content, cholesterol content and TBA in breast meat were significantly(P<0.05) decreased by addition of BVL. However, the addition of BVL did not affect the meat color. The saturated fatty acid composition of breast meat was decreased(P<0.05), whereas unsaturated fatty acid was increased(P<0.05) by addition of BVL 1% and 2%. Taste of sensory evaluation was improved by addition BVL. The results of this study indicate that dietary supplemental BVL 1% and 2% may improve the performance and meat quality of KNC. The prevention effect of KNC which challenged with Salmonella gallinarum (1${\times}$10$\^$9/ CFU/ml) was evaluated. A reisolation of S.gallinarum in liver, pathological index of villus and colon were evaluated. There was no reisolation of S.gallinarum in liver and a few pathological index of colon in 4% BVL supplement fed KNC than 1 and 2%. These results indicate that dietary supplemental BVL 4% may improve the prevention effect to salmonellosis in chickens.

나트륨 섭취량과 관련된 식행동의 성별 및 연령별 비교 (Comparison of Dietary Behaviors Related to Sodium Intake by Gender and Age)

  • This study was performed to understand recognition and behaviors related to sodium intake of Korean adults. The data were collected from subjects including 267 male and 285 female adults in nationwide and compared by gender and by age. We found that the male group showed significantly higher smoking, alcohol drinking, and exercise does the female group. The older group (40 to 59 years) revealed significantly higher exercise and lower alcohol drinking; however general disease and hypertension prevalence, diet therapy practice, and meditation for hypertension were higher. Recognizing sodium levels of foods containing high-sodium, and sodium-nutriture labels when purchasing foods, and knowing differences between salt and sodium of the male group or recognizing sodium levels of foods containing high-sodium of the older group was worse than the other. Among the 32 food behaviors, only 12 were identified as significantly correlated to sodium intake levels including behaviors of preferring Chinese and Japanese foods to Western foods, preferring kimchi to raw vegetables, completely consuming soup, stew, noodle liquid, liking of dried fish and salted mackerel, frequent eat-outs or delivered foods, and so forth. There were significant differences between gender or age groups in terms of sodium intake-increasing behaviors; the male group showed higher behaviors of preferring salty taste and eating all broths. And the older group revealed higher behavior of adding table salt as well as the previous two, however, the younger group showed more behaviors of eat-outs or delivered foods and not the liquid of kimchi.

쥐 우유중의 Myo-Inositol 생성에 관한 연구 (Myo-Inositol Synthesis in the Milk of Lactating Rats)

  • 우유중에 상당량 존재하는 myo-Inositol의 생성을 알아보기 위하여 3% SE-52를 충진시킨 gas-liquid chromatography에 의해 myo-Inositol의 정량을 행하고 이 myo-Inositol의 유래를 feeding 실험과 $(^{14}C)-glucose$와 (3H)-myo-Inositol을 쥐에 주사한 실험으로 알아 븐 결과 다음과 같은 결과를 얻었다. 1. 쥐우유를 G.L.C. 방법으로 분석하여 우유 100gm.당 61-91mg.의 myo-Inositol (free)을 얻었다. 2. Feeding 실험 결과 우유 myo-Inositol의 85%는 diet에서 흡수되었고 나머지는 mammary gland에서 합성된 것으로 나타났다. 3. $(^{14}C)-glucose$에서 $(^{14}C)-myo-Inositol$ 생성은 lactose의 생성과 비슷하게 한시간이내에 최고에 달하였다. 4. $(^3H)-myo-Inositol$을 lactating rat에 주사하여 실험한 결과 우유 myo-Inositol의 대부분은 혈액 myo-Inositol에서 유래되었다. $(^{14}C)-glucose$를 주사한 후 우유중에 나타난 myo-Inositol의 비방사능(比放射能)은 4시간 후에 혈액 glucose의 8%였다.

사료에 오미자 가공부산물 분말의 첨가 급여가 계란의 리그난 함량에 미치는 영향 (Production of Lignan-Rich Eggs as Functional Food by Supplementing Schisandra chinensis By-Product in Laying Hens)

  • 산란계는 식단의 건강 증진 성분을 계란에 '생체 축적'이 가능한 것으로 알려져 있다. 본 연구의 목적은 오미자 가공 부산물의 분말을 사료에 첨가하여 산란계에 급여가 리그난이 풍부한 기능성 계란의 생산 가능성을 조사하기 위하여 수행하였다. 실험에 사용된 사료는 황옥수수, 쌀겨, 대두박, 어분, 육골분, 가금류분, 프리믹스, CaCO3 등이 첨가된 기본 사료에 오미자 가공 부산물 분말을 첨가하여 제형화하였다. 실험의 설계는 대조구와 오미자 가공부산물을 1%, 3%, 5% 및 7% 첨가한 실험군으로 분류하여 완전임의 배치법으로 배치하였다. 계란의 생산성, 평균 무게, 난황과 난백의 리그난 함량은 7일 간격으로 조사하였다. 계란 생산과 평균 계란 무게는 오미자 가공부산물 분말을 사료에 5% 이하로 첨가하여 급여하였을 경우에는 차이가 없었으나, 7%의 고농도로 첨가하여 사료를 급여하였을 경우에는 처리 3주후에는 유의하게 감소하였다. 계란내의 리그난 함량을 조사하기 위하여 액체 크로마토그래피(HPLC)로 분석한 결과, 오미자 가공부산물 분말의 첨가 농도가 높을수록 난황의 gomisin N 과 schisandrin C의 함량을 유의하게 증가시켰으나(p<0.05), 난백에서는 gomisin N은 농도 의존적으로 증가하였으나 schisandrin C는 검출되지 않았다. 이러한 결과는 오미자 가공 부산물 분말을 사료여 첨가하여 산란계에 급여할 경우에는 계란의 품질을 향상시키고 리그난이 함유된 계란의 생산이 가능한 것으로 나타났다. 결론적으로 오미자 가공 부산물 분말을 5% 이하로 사료에 첨가하여 급여한다면 오미자의 주요 생리활성인 gomisin N 와 schisandrin C가 풍부한 기능성 계란의 생산이 가능할 것으로 판단된다.

Comparison of Methodologies to Quantify Phytate Phosphorus in Diets Containing Phytase and Excreta from Broilers

  • The use of a suitable methodology to quantify the phytate phosphorus ($P_{phy}$) content in both the feed and the excreta from broilers is required to enable accurate calculation of the catalytic efficiency of the phytase supplemented in the feed. This study was conducted to compare 2 analytical methodologies (colorimetry and also high-performance liquid chromatography with a refractive index detector) in order to calculate the phytase efficiency by utilizing the results from the methodology that was shown to be the most appropriate. One hundred and twenty broilers were distributed in a $(4+1){\times}2$ factorial arrangement, corresponding to 4 diets that were equally deficient in P supplemented with increasing levels of phytase (0, 750, 1,500, and 2,250 units of phytase activity - FTU - per kg of feed) plus 1 positive control diet without phytase, supplied to male and female birds. The result indicated that the colorimetric methodology with an extraction ratio of 1:20 (mass of sample in g:volume of the solvent extractor in mL) was shown to be the most adequate. There was no interaction between the phytase level and the sex of the broilers (p>0.05). Males consumed 12% more $P_{phy}$ than did females (p<0.01), but the sex of the broilers did not affect (p>0.05) the excretion and retention coefficient of $P_{phy}$. The increase in the phytase level of the diet reduced (linear, p<0.01) the $P_{phy}$ excretion. The greatest $P_{phy}$ retention was estimated at 87.85% when the diet contained 1,950 FTU/kg (p<0.01), indicating that it is possible to reduce the inorganic P in the formulation at an amount equivalent to 87.85% of the $P_{phy}$ content present in the feed, which, in this research, corresponds to a decrease in 2.86 g of P/kg of the feed.

목초 사일리지 급여 시 보리와 채종박 보충급여에 의한 제 3위 소화액내 Soluble Non-ammonia Nitrogen Fraction의 Flow 패턴 변화 (Diurnal Patterns in the Flow of Escapable Soluble Non-Ammonia Nitrogen Fractions in Omasal Digesta as Influenced by Barley and Rapeseed Meal Supplementation in Cows Fed Grass Silage Based Diet)

  • 본 연구는 목초 사일리지를 기초사료로 급여한 젖소에 있어서 보리와 유채박 급여가 제 3위 소화액 내 soluble non-ammonia nitrogen fractions(SNAN fractions; 아미노산, 펩티드, 용해성 단백질 및 total SNAN)의 flow 패턴변화를 측정하기 위하여 실시하였다. 반추위 캐뉼라가 장착된 4마리 젖소는 4×4 라틴방각법에 의해 기초사료로 grass-red clover silage를 자유급여하여 사일리지 단독급여구(GS), 기초사료 + 보리6.0kg/d 급여구, 기초사료 + 유채박2.1kg/d 급여구 및 기초사료 + 보리6.0kg/d + 유채박2.1kg/d 급여구로 배치하였다. 제 3위 소화액은 제 3위 소화액 채취기구를 이용하여 사료급여 후 1.5h 간격으로 채취하였고, 그 소화액 내 SNAN fractions 정량은 ninhydrin assay를 이용하여 분석하였다. 보충사료 급여는 GS구에 비해 비록 통계학적 유의성은 없었지만 total SNAN flow를 증가시켰다. 제 3위 소화액 내 펩티드 flow 패턴은 12시간 feeding cycle 내내 SNAN fractions 중 양적으로 가장 높았으며, GS구에 비해 보충사료급여구에서 높은 펩티드 flow 패턴을 보였다. GS구를 제외한 보충사료처리구간에는 SNAN fractions flow 패턴 차이가 없었다. 15N을 이용하여 분석된 사료유래 SNAN fractions flow 패턴에서는 보충사료급여구가 GS구에 비해 높게 나타났다. 이상의 결과 제 3위로 유입되는 펩티드 flow는 전체 SNAN flow 중 양적으로 가장 중요한 N 이며, 보충사료 급여 시 그 펩티드 flow의 증가 가능성을 시사한다.

Supplementing Rhodobacter sphaeroides in the diet of lactating Holstein cows may naturally produce coenzyme Q10-enriched milk

  • Objective: To examine the effects of Rhodobacter sphaeroides (R. sphaeroides) supplementation as a direct-fed microbial (DFM) on rumen fermentation in dairy cows and on coenzyme Q10 (CoQ10) transition into milk, an in vitro rumen simulation batch culture and an in vivo dairy cow experiment were conducted. Methods: The characteristics of in vitro ruminal fermentation were investigated using rumen fluids from six cannulated Holstein dairy cows at 2 h post-afternoon feeding. A control treatment was included in the experiments based on a typified total mixed ration (TMR) for lactating dairy cows, which was identical to the one used in the in vivo study, plus R. sphaeroides at 0.1%, 0.3%, and 0.5% TMR dry matter. The in vivo study employed six ruminally cannulated lactating Holstein cows randomly allotted to either the control TMR (C-TMR) treatment or to a diet supplemented with a 0.5% R. sphaeroides culture (S-TMR, dry matter basis) ad libitum. The presence of R. sphaeroides was verified using denaturing gradient gel electrophoresis (DGGE) applied to the bacterial samples obtained from the in vivo study. The concentration of CoQ10 in milk and in the supernatant from the in vitro study was determined using high performance liquid chromatography. Results: The results of the in vitro batch culture and DGGE showed that the concentration of CoQ10 significantly increased after 2 h of R. sphaeroides supplementation above 0.1%. When supplemented to the diet of lactating cows at the level of 0.5%, R. sphaeroides did not present any adverse effect on dry matter intake and milk yield. However, the concentration of CoQ10 in milk dramatically increased, with treated cows producing 70.9% more CoQ10 than control cows. Conclusion: The CoQ10 concentration in milk increased via the use of a novel DFM, and R. sphaeroides might be used for producing value-added milk and dairy products in the future.

남녀 대학생의 지방에 대한 영양지식과 실천도 (College Students' Nutrition Knowledge and Practices toward Dietary Fat)

  • This study was made for male and female university students regarding the nutrition knowledge about fat, fat contents in food, and practice toward fat. The result is as follows;1.Out of 13 questions inquiring the knowledge about fat a significant difference was observed between female and male students in 10 questions. Male students' nutrition knowledge about fat was $6.33{\pm}0.16$, and that of female students' was $6.33{\pm}0.16$ and there was no significant difference. The ratio of correct answer was 49% and 59% respectively. Female students marked high ratio of correct answer such items as ${\omega}-3$ fatty acid, the relation between high fat diet and atherosclerosis(coronary disease), the relation between obesity and fat, the relation between ${\omega}-3$ fatty acid and heart disease, cholesterol, fatty acid in butter, degree of unsaturation in liquid oil, margarine, chicken fat, rancity of fat. 2.Male students' total score of knowledge about fat quantity in food was $4.91{\pm}0.10$ and that of female students was $5.58{\pm}0.10$. There was significant difference(p<0.001) and the ratio of correct answer was 55% and 62% respectively. Out of 9 food items, significant difference was observed in 7 items according to student's majoring subject. The ratio of correct answer for the questions about the quantity of fat in food female showed high figures in such items as white meat(p<0.01), whole milk(p<0.0001), skim milk(p<0.01), potato chips(p<0.001), biscuit(p<0.001). However, male students showed high figures on the question about the quantity of fat in vegetable margarine. 3. Out of 12 items observing the degree of diet practice on fat, significant difference was observed between male and female students in 6 items. Female students practice the question items positively with high ratio: intake of fish instead of meat(p<0.01), removing visible fat in meat(p<0.0001), removing chicken skin(p<0.0001), removing oil during cooking (p<0.05), selection of low fat milk or skim milk(p<0.05), selection of meat part(p<0.001). There was significant difference between male and female students in practicing diet on fat (p<0.001) and the total score was $31.52{\pm}0.52$ and $34.65{\pm}0.41$ respectively.

곤드레 (Cirsium setidens) 에탄올 추출물의 알코올성 지방간 손상 억제 효과 (Protective effects of Cirsium setidens ethanolic extracts against alcoholic fatty liver injury in rats)

  • 본 연구에서는 Sprague-Dawley 종 흰 쥐 수컷을 정상 대조군 (C), 알코올 군 (E), 알코올 + 100 mg/kgBW 곤드레 에탄올 추출물군 (E+LCS), 알코올 + 500 mg/kgBW 곤드레 에탄올 추출물군 (E+HCS)으로 나누어 Lieber-DeCarli control diet 혹은 Lieber-DeCarli ethanol diet를 8주간 공급하였으며, 이때 곤드레 에탄올 추출물은 액상 사료에 직접 섞어 공급하였다. 알코올과 곤드레 에탄올 추출물의 식이 공급 종료 후 간 조직의 지방구 축적 정도를 살펴본 결과, E+LCS군과 E+HSC군은 E군에 비해 지방간 발생이 유의적으로 억제되었으며, 정상 대조군인 C군과 유의적인 차이가 없었다. 이와 비슷하게, 곤드레 에탄올 추출물의 공급은 알코올에 의해 증가된 간 조직과 혈청의 중성지방 농도를 유의적으로 낮추었으며, 혈청 AST와 혈청 ALT 활성도 정상 대조군 수준으로 회복시키는 것으로 나타났다. 한편, 곤드레 에탄올 추출물의 공급은 p-AMPK과 p-ACC 단백질 수준을 농도 의존적으로 증가시켰으며, 두 단백질 모두 E군에 비해 E+HSC군에서 유의적으로 높게 나타났다. 또한 FAS mRNA와 SCD1 mRNA 수준은 E군에 비해 E+HSC군에서 유의적으로 낮게 나타났다. 곤드레 에탄올 추출물은 간 조직에서 알코올 공급에 의해 증가된 $NF{\kappa}B$의 활성을 유의적으로 낮추었으며, $NF{\kappa}B$의 표적 단백질인 $TNF{\alpha}$ 단백질 수준을 농도의존적으로 낮추었다. 본 연구 결과를 통해 곤드레는 알코올에 의한 지방간 발생 및 관련된 간 손상을 유의적으로 억제할 수 있음을 확인하였으며 이 과정에서 AMPK 활성 증가와 $NF{\kappa}B$ 활성 억제가 관여함을 제시하였다.
