  • 본 연구에서는 현미로부터 tocoles 획분을 분리하여 고콜레스테롤 식이를 섭취한 흰쥐의 체내 지질대사에 미치는 영향을 조사하였다. 실험동물은 SD계 숫컷 5주령을 구입하여 정상군, 대조군 (1% cholesterol), rice oil 첨가군, tocoles 첨가군의 4군으로 나누어 AIN-76 diet 조성에 의거하여 식이를 조제하여 4주 동안 섭취시켰다. 실험기간이 끝난 후 복부 대동맥으로부터 채혈하고 각 조직을 적출하여 간, 신장, 비장 및 부고환지방 조직의 무게를 측정하였다. 또한 혈청 lipid profile과 TBARS 함량 및 ALT, AST 및 ALP 활성을 분석하였고 간장중 중성지방, 콜레스테롤 및 TBARS 함량과 G6PDH 및 malic enzyme을 분석하였다. 그 결과 간장 및 부고환지방의 무게가 대조군에서 유의적으로 증가하였으나, tocoles 첨가군은 정상군의 수준으로 감소하였다. 혈청 내의 중성지방과 총 콜레스테롤은 실험군 간에 유의적인 차이는 나타내지 않았으나 감소하는 경향을 보였다. 간장 조직 내 지질함량은 대조군에서 높게 나타났으며, tocoles 군에서 중성지방과 콜레스테롤 및 총 지방의 함량이 유의하게 감소하였다. 지질 과산화물 함량은 혈청과 간장 조직 모두 유의적으로 감소하였다.

Effectsof Garlic Vinegar Supplementation on Changes of Body Weight, Plasma Glucose, and Plasma Lipid Profile in High Cholesterol-fed Rats

  • This study was carried out to investigate the effects of garlic vinegar supplementation on changes in body weight, blood glucose and serum triglyceride and cholesterol. Rats were fed a cholesterol-diet with or without garlic vinegar (20% garlic juice )supplement for 28days. Body weights in rats fed a diet containing garlic vinegar were significantly lower than those of control rats. The concentration of plasma glucose, total cholesterol . LDL-cholesterol , and HDL-cholesterol were not significantly different between the two groups for a period of up to four weeks. However, the concentration of plasma triglyceride was slightly decreased in the garlic vinegar-supplemented group. Results suggest that the supplementation of garlic vinegar is beneficial for weight reduction in high-cholesterol fed rats.

  • Chrysanthemum zawadskii (CZ) is a perennial herb belonging to the Asteraceae family. CZ is used medicinally to treat inflammatory and uterine diseases in Asia. CZ was extracted with 50% ethanol and CZ extract (CZE; at 125, 250, and 500 mg/kg body weight) was administered orally every day for 5 or 6 weeks to investigate the anti-diabetic effects in streptozotocin (STZ)-induced rats and STZ + high-fat diet (HFD)-fed mice. CZE significantly decreased fasting blood glucose levels in STZ- and STZ + HFD-induced diabetic models. In addition, glucose tolerance and insulin tolerance were improved in the STZ + HFD + CZE group by increasing insulin levels and decreasing hemoglobin A1c (HbA1c) levels in serum. Furthermore, CZE supplements decreased components of the serum lipid profile such as triglyceride, total cholesterol, and low-density lipoprotein cholesterol levels. These results suggest that CZE may be a potential candidate for controlling hyperglycemia.

Effect of Dietary Selenium and Fish oil on Lipid Peroxidation and Fatty Acid Profile in the Rat (식이 셀레늄 수준과 식이 지방산 조성이 쥐의 지질과 산화 상태와 조직의 지방산 조성에 미치는 영향)

  • The influence of selenuium deficiency and fish oil on lipid peoxidation status and fatty acid composition of tissues(plasma aorta and liver) was studied. Male Sprague Dawley rats were fed for eight weeks semipurified diets containing 7% corn oil(by weight) or 5, 5% fish oil(MaxEPA) plus 1.5% corn oil with oil with or without selenium status (glutathione peroxidase activity and selenium levels) were significantly lower in the rats given inadequate selenium in plasma aorta (p<0.02 and p<0.001 respectively) gut not that in plasma Selenium supplementation decreased hepatic MDA levels(p<0.02) Increases in the levels of 20:5(n-3) 22:5(n-3), 22:6(n-3) 20:3(n-6) and a decrease in the level of 20:4(n-6) were observed in plasma total lipids and aortic and hepatic phospholipids when fish oil was fed. Though selenium supplementation increased the level of n-3 fatty acids(such as 22:6(n-3)) in plsama and the aorta is overall effect was smaller than the effect of fish oil feeding. These data suggest that selenium may play a significant but minor role in protecting against lipid peroxide-tion even when vitamin E intakes are in excess of current recommendations in both corn oil and fish oil diets.

Long Term Feeding with Soy Isoflavone and L-Carnitine Synergistically Suppresses Body Weight Gain and Adiposity in High-Fat Diet Induced Obese Mice

  • Objective: We investigated the efficacy of a 12-week supplementation of soy isoflavone with L-carnitine on the development of obesity in high fat-induced obese C57BL/6J mice, which are known as a good model of diet-induced obesity. Methods: We measured body weights, adipose tissue mass, serum/liver lipid profiles and fat cell size/number in C57BL/6J mice fed diets containing either low fat (4%) or high fat (35%), or high fat supplemented with soy isoflavone powder containing 10% isoflavone and L-camitine for 12 weeks. Results: Body weight gain, abdominal adipose tissue and liver weight were lower by 31% 78% and 31.4% respectively, in mice on high fat diet containing soy isoflavone+L-carnitine (SC mixture) compared with high fat diet group. Also, SC mixture improved serum lipid profiles such as total cholesterol (TC), triglycerides (TG), and liver lipid profiles such as total lipids and TG. As subsequent results, this SC mixture prevented high-fat diet from accumulating TG in the liver. The size of fat cell was also significantly decreased in SC mixture fed mice. At the end point of this experiment, our results showed that feeding with soy isoflavone for 12 weeks finally increased camitine palmitoyltransferase 1 (CPT 1) activity through elevating the level of CPT1 expression. Conclusions: This study suggests that long-tenn supplementation with dietary soy isoflavone and L-carnitine is more synergistically beneficial for the suppression of high-fat diet induced obesity by inhibiting liver TG accumulation and the gain in abdominal adipose tissue weight than that with soy isoflavone. The antiobesity effects of SC mixture might be attributed, at least in part, to the induction of fatty acid catabolism by soy isoflavone, genistein.

Effects of Spirulina Added Salad Dressing on the Antioxidant Index and DNA Protection in Mice (스피루리나 첨가 샐러드 드레싱 보충 식이가 마우스 혈장 항산화 지표 및 DNA보호에 미치는 영양)

  • The effects of spirulina-added salad dressing on lipid profiles and antioxidant biomarkers such as total glutathionine, TBARS value, carbonyl value, GPx, GR, SOD and paraoxonase activity in plasma or liver of mice were evaluated Sixteen male ICR mice weighing 20$\pm$2 g were divided into two groups and fed low fat ($5\%$ fat) diet (low fat control: LFC) and low fat control plus dressing diet (LFD) for eight weeks. Body weight, tissue weights of liver, heart and kidney, and the distribution of body fat deposition were not significantly different between two groups. Also, the profile of TG, TC, LDL and HDL cholesterol were similar between two groups. The DNA damage was determined using the comet assay (single cell gel assay) with alkaline electrophoresis and quantified by measuring tail length (TL). Spirulina salad dressing consumption resulted in significant decrease in lymphocyte DNA damage expressed by TL (LFC: $28.8{\mu}m$, LFD: $20.3{\mu}m$). Additionally, salad dressing consumption for 8 wks decreased the lipid peroxidation assayed by TBARS to $12.6\%$ compared with the control. The levels of antioxidant vitamins such as $\beta$-carotene were significantly higher in plasma of LFD group than those in LFC group based on HPLC method This study shows that spirulina-added salad dressing exerts degenerative disease-protective effects on oxidative DNA damage and lipid peroxidation possibly via a free radical levels.

Effect of Herb Distillate on Hepatic Xanthine Oxidase Activity and Serum Lipid Profiles in Carbon Tetrachloride-Administered Rats

  • In order to evaluate the hepatoprotective effect of an herb distillate, ie., a mixture of 28 traditional Korean herbs, germanium, tormarine and Gijangsoo (Gijang water), $CCl_4$ was intraperitoneally administered to rats before or after supplementation of the diluted herb distillate (HD) for 2 weeks. Then hepatic xanthine oxidase activity and serum lipid profiles were determined. The experimental groups had higher feed intake than the normal control (NC), but had lower weight gain. Water intake and the amount of feces were not significantly different, but urine was excreted in lower amounts in all the experimental groups compared to the NC. Liver weights in the HD-supplemented groups were lower than that of the distilled water-supplemented groups (DW-groups) after $CCl_4$-administration. Serum ALT activities in all the experimental groups were higher than that of the NC-group. However, the increasing activity of serum ALT in the HD-supplemented groups (HD-groups) was lower than that of the DW-groups. Total serum and LDL-cholesterol levels were higher in all the $CCl_4$-administered groups than in the NC-groups, and serum HDL-cholesterol levels were lower in all the experimental groups compared with the NC-groups. Meanwhile, the increasing rate of total serum and LDL-cholesterol levels and the decreasing rate of HDL-cholesterol in the HD-groups were lower than that of the DW-groups. But, levels of serum TG were similar among all the experimental groups. The activities of hepatic xanthine oxidase (XOD) type O of the $CCl_4$-administered rats showed a significant increase in and an increasing rate of XOD in the HD-groups, which was lower than that of the DW-groups. On the other hand, GST activities in all the experimental groups were significantly decreased, and the decreasing rate was lower in the HD-groups than in the DW-groups. The hepatic contents of GSH and LPO in all the rats were not changed by $CCl_4$ administration. These results suggest that the decreased liver damage in the HD-supplemented groups was due to the inhibition of XOD-type O activity by constituents of HD, as well as by a prevention/inhibition of serum lipid profile changes in $CCl_4$-treated rats. However, further detailed studies are needed to support this hypothesis.

Quality Characteristics of Chitosan-ascorbate Treated Kwamaegi Prepared by Vacuum Drying, and Lowering Effect of Serum Lipids in Rats Fed High Fat Diets (Chitosan-ascorbate 처리 감압건조 과메기의 품질특성과 고지방식이 흰쥐의 혈청지질에 미치는 영향)

  • Quality characteristics of Kwamaegi (semi-dried saury) prepared by treatment of chitosan-ascorbate (CA) and vacuum drying at $40{\sim}60^{\circ}C$(VDK), and the effect of the Kwamaegi on serum lipid profiles and anti-oxidation-related enzyme activity in rats fed high fat diets were investigated. The preparation periods were $4.5{\sim}8.3$ hr in VDK, while naturally dried Kwamaegi (NDK) took 360480 hr. Total microbe contents of VDK and NDK were $0.2{\sim}0.5$ and 8.2 log CFU/g, respectively. There was no significant difference in amino-nitrogen content. Compared with NDK, the acid and peroxide value, and fishy flavor of VDK40 (dried at $40^{\circ}C$) were significantly lower, and the texture, color and overall acceptability were higher. In animal experiments, weight gain, content of LDL-cholesterol and lipid peroxide, activities of total (T) and O type (O) xanthine oxidase, and the O/T ratio (%) were significantly lower in the VDK40 diet group than in the NDK diet group. The content of HDL-cholesterol in the VDK40 diet group was higher than in the NDK diet group. These results suggest that preparing CA-treated Kwamaegi with vacuum-drying at $40^{\circ}C$ can be applied throughout the year, and may shorten preparation time and improve its microbiological safety and nutritional values.

Lipid-Lowering Action of Powder and Water Extract of Mulberry Leaves in C57BL/6 Mice Fed High-Fat Diet (고지방식이를 급여한 C57BL/6 마우스에서 뽕잎분말과 열수추출물의 지질저하 작용)

  • This study was to investigate the hypolipidemic effect of powdered mulberry leaves (PML) and water extract of powdered mulberry leaves (WML) on high-fat fed mice. Male C57BL/6 mice were divided into four groups; a normal group (N), a high-fat (HF) group, a high-fat group supplemented with PML (HF-PML) and a high-fat group supplemented with WML (HF-WML). The PML or WML was added to a standard diet based on 1% dried mulberry loaves (1g PML/100g diet and 0.22g WML/100g diet) for 6 weeks. Body weight and organ weights were not different among thle groups in high-fat fed mice, whereas food intake and daily energy intake were significantly (p<0.05) lowered in the HF-PML group. In plasma and liver, the supplementation of PML and WML significantly (p<0.05) lowered cholesterol and triglyceride concentrations compared to the HF group. The HDL-cholesterol/total cholesterol ratio was significantly (p<0.05) higher in the HF-PML and HF-WML groups than in the B:.w group. The fecal triglyceride and cholesterol concentrations were significantly (p<0.05) increased in the HF-PML and HF-WML groups compared to the HF group. Hepatic lipid regulating enzyme activities, fatty acid synthase, fatty acid ${\beta}-oxidation$ and carnitine palmitoyl transferase were significantly lower in the HF group than in the N group. However, the activities of these hepatic lipid regulating enzymes activities were significantly (p<0.05) elevated in the HF-PML and HF-WML groups compared to the HF group. Accordingly, these results suggest that PML and WML improve plasma and hepatic lipid levels partly by increasing fecal lipid excretion and enhancing hepatic lipid regulating enzymes activities.

The Effect of Red-Yeast-Rice Supplement on Serum Lipid Profile and Glucose Control in Subjects with Impaired Fasting Glucose or Impaired Glucose Tolerance (공복 혈당장애 및 내당능장애자에서 홍국의 섭취가 혈중지질 및 혈당 조절에 미치는 영향)

  • This study was aimed at evaluating the effect of red-yeast-rice supplementation on cholesterol-lowering and glucose control in subjects with impaired fasting glucose (IFT) or impaired glucose tolerance (IGT). We conducted a doubleblind, placebo-controlled study with 3 groups; placebo, low dose group (red yeast rice 210.0mg/capsule, 2.52g/day) and high dose group (red yeast rice 420.0mg/capsule, 5.04g/day), which were randomly assigned to subjects with impaired fasting glucose or impaired glucose tolerance. We measured fasting serum concentrations of total-, LDL-, HDL-cholesterol, triglyceride, glucose, insulin, free fatty acid (FFA) and 2 h oral glucose tolerence test (OGTT) before and after the supplementation. Both low dose and high dose groups had significant decrease in LDL cholesterol and atherogenic index (AI) compared with placebo group (p<0.05). Additionally, total and HDL cholesterol improved significantly in high dose group compared with placebo group (p<0.05). Fasting serum glucose decreased in test groups and increased in placebo group after intervention. However, it was not significant differences. In subjects which fasting blood glucose is more than 110mg/dL, fasting glucose had a tendency to decrease in high dose group (p<0.1) and Hemoglobin A1c (HbA1c) had significant decrease in low dose group (p<0.05), while insulin and HOMA-IR had a tendency to increase in placebo group after intervention. Mean changes of glucose related parameters (fasting glucose, insulin, HOMA-IR) compared with placebo group did not show significant differences. In conclusion, subjects with impaired fasting glucose or impaired glucose tolerance were significantly improved in serum lipid profile by red yeast rice supplementation without serious side effects. These are more effective in the case of a high dose. The effects of red yeast rice supplementation on glucose control were insignificant.
