  • Global gene expression profile was analyzed by microarray analysis of rat liver RNA after acute acetaminophen (APAP) administration. A single dose of 1g/kg body weight of APAP was given orally, and the liver samples were obtained after 24, 48 h, and 2 weeks. Histopathologic and biochemical studies enabled the classification of the APAP effect into injury (24 and 48 h) and regeneration (2 weeks) stages. The expression levels of 4900 clones on a custom rat gene microarray were analyzed and 484 clones were differentially expressed with more than a 1.625-fold difference(which equals 0.7 in log2 scale) at one or more time points. Two hundred ninety seven clones were classified as injury-specific clones, while 149 clones as regeneration-specific ones. Characteristic gene expression profiles could be associated with APAP-induced gene expression changes in lipid metabolism, stress response, and protein metabolism. We established a global gene expression profile utilizing microarray analysis in rat liver upon acute APAP administration with a full chronological profile that not only covers injury stage but also later point of regeneration stage.

  • This study investigated the effects of soyoligosaccharide consumption on lipid profile of plasma, liver and feces and immune responses in Sprague-Dawley male rats. Sixty male Sprague-Dawley rats 4-wk-old were provided the soyo-ligosaccharide containing diets for 4 weeks (0, 100 g/kg diet); each of these diets was supplemented with either 70 or 200 g fat/kg diet, giving a total of 4 experimental groups. The effect of weight reduction was most significantly observed in the group fed low fat and soyoligosacchairde diet. The plasma total lipid and cholesterol contents were not changed by either fat proportion or soyoligosaccharide supplementation in the diets. Also the plasma triglyceride lowering effect by soyoligosaccharide was not observed in rats fed either low fat or high fat diet. However, the significant decrease in TG contents was found with rats fed high fat diets compared to the control/no soyoligosaccharide diet. Elevation of plasma LDL-cholesterol and reduction of HDL-cholesterol by feeding high fat diet was not altered by supplementing soyoligosaccharide. This was also applied to the liver lipid profiles. The significant increases in liver total lipid, trigly-cerides and cholesterol by high fat diet was not abolished by feeding soyoligosaccharide. However, the desirable effects of feeding soyoligosaccharide were found with total lipid and cholesterol excretion through feces in rats fed high fat diets. Immune organ weights and spleen cell proliferations did not affected by experimental diets. These results de-monstrated that soyoligosaccharide intakes increased the lipid output via feces, especially in rats fed the high fat diet, but more researches are needed on immune responses.

탄수화물 급원에 따른 HCA의 공급이 흰쥐의 체중 및 지질 대사에 미치는 영향 (Effect of Feeding with Different Source of Carbohydrate and HCA on Body Weight Gain and Lipid Metabolism in Rats)

  • This study was conducted to investigate the effects of feeding different carbohydrate sources and garcinia cambogia extract(HCA) on body weight and lipid metabolism. Fifty 10-month-old male Sprague-Dawley rats weighting $635{\pm}6g$ were randomly divided into 5 groups and fed different experimental diets for 4 weeks. The carbohydrate(CHO) sources of each group were cornstarch(control group, 100% of CHO), fructose(F group and FH group, 25% of CHO) and sucrose(S group and SH group, 25% of CHO). FH group and SH group were fed diets containing 1%(W/W) of HCA. Food intake, body weight gain, and calorie efficiency were not significantly different among the groups. Perirenal fat pad weight of FH group was significantly lower than F group, but epididymal fat pad weight was not different among the groups. Fasting glucose level were not significant among the groups. Plasma lipid profile of FH or SH group was slightly lower than F or S group, respectively. The degree of difference of plasma lipid level was greater between F and FH group than those of between S and SH group. In liver, total lipid, triglyceride and total cholesterol level were slightly higher in F group than S group, and tended to be lower in FH group than F group, but tended to be higher in SH group than S group. Liver citrate lyase activity were not significant among the groups. These results suggest that HCA is potential material for reduction of body weight and improvement of plasma lipid profiles. But, there was no difference between fructose intake with HCA and sucrose intake with HCA in reduction of body weight and lipid metabolism.

질량분석 시스템을 이용한 극지 토양 유래 신규 미생물의 지질 A 화학적 구조 분석 (Determination of Lipid A Profile of Gram-Negative Bacteria from Arctic Soils Using Mass Spectrometric Approaches)

  • For decades, the microorganisms in arctic soils have been newly discovered according to the climate change and global warming. In this study, the chemical structure of a lipid A molecule from Pseudomonas sp. strain PAMC 28615 which was newly discovered from arctic soils was characterized by mass spectrometric approaches such as matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) and MALDI multi-stage tandem mass spectrometry (MS). First, lipopolysaccharide (LPS) from Pseudomonas sp. strain PAMC 28615 was extracted and subsequently hydrolyzed to obtain the lipid A. The parent ion peak at m/z 1632 was determined by MALDI-TOF MS, which also can validate our lipid A purification method. For detailed structural determination, we performed the multiple-stage tandem mass analysis ($MS^4$) of the parent ion, and subsequently the abundant fragment ions in each MS stage are tested. The fragment ions in each MS stage were produced from the loss of phosphate groups and fatty acyl groups, which could be used to confirm the composition or the position of the lipid A components. Consequently, the mass spectrometry-based lipid A profiling method could provide the detail chemical structure of lipid A from the Pseudomonas sp. strain PAMC 28615 as an arctic bacterium from the frozen arctic soil.

Antidiabetic Potential of Kefir Combination from Goat Milk and Soy Milk in Rats Induced with Streptozotocin-Nicotinamide

  • The study aimed to evaluate the effect of kefir combination from goat milk and soy milk on lipid profile, plasma glucose, glutathione peroxidase (GPx) activity and the improvement of pancreatic β-cell in diabetic rats. Male rats were divided into five treatments: normal control, diabetic control, goat milk kefir, combination of goat milk-soy milk kefir and soy milk kefir. All rats were induced by streptooztocin-nicotinamide (STZ-NA), except for normal control. After 35 d experiment, the rats were sampled for blood, sacrificed and sampled for pancreatic tissues. Results showed that diabetic rats fed kefir combination had higher (p<0.05) triglyceride than the rats fed goat milk or soy milk kefir. Decreasing of plasma glucose in diabetic rats fed kefir combination was higher (p<0.05) than rats fed goat millk kefir. The activity of GPx in diabetic rats fed three kinds of kefir were higher (p<0.01) than untreated diabetic rats. The average number of Langerhans and β-cells in diabetic rats fed kefir combination was the same as the normal control, but it was higher than diabetic control. It was concluded that kefir combination can be used as antidiabetic through maintaining in serum triglyceride, decreasing in plasma glucose, increasing in GPx activity and improving in pancreatic β-cells.

Antihyperlipidemic effects of alcoholic extract of Pongamia pinnata Linn. leaves on high fat diet fed rats

  • The objective of the study was to investigate the antihyperlipidemic activity of alcoholic extract of Pongamia pinnata Linn. (PPAE) leaves in rats fed with high fat diet (HFD). PPAE was administered orally in the divided doses of 250 and 500 mg/kg/day for 30 days in HFD fed rats. Body weights were observed and the analysis of serum lipid profile was carried out on day 30. Marked decrease in the body weight, total cholesterol (TC), triglycerides (TG), low density lipoprotein (LDL) and very low density lipoprotein (VLDL) whereas significant increase in the levels of high density lipoprotein (HDL) were observed after treatment with PPAE. However, PPAE in a dose of 250 mg/kg did not show significant (P < 0.05) increase in HDL levels. The PPAE also lowered TC: HDL-c and LDL: HDL-c ratios significantly suggesting it's antihyperlipidemic and cardioprotective potential. The present work reveals that PPAE at the dose of 500 mg/kg/day exhibited significant (P < 0.01) antihyperlipidemic effects.

The effects of physical training on antioxidative status under exercise-induced oxidative stress

  • This study investigated the effect of physical training and oxidative stress on the anti oxidative activity and on plasma lipid profile. Forty eight rats were given either a physical training or no training for 4 weeks and were then subdivided into 3 groups: before-exercise (BE); during-exercise (DE); after-exercise (AE). The antioxidative activity was evaluated with the activities of catalase in plasma and superoxide dismutase (SOD), the ratio of reduced glutathione/ oxidized glutathione (GSH/GSSG) and the level of malondialdehyde (MDA) in liver. The plasma concentrations of triglyceride (TG), total cholesterol (TC), high-density lipoprotein-cholesterol (HDL-C)) were also compared. Compared to those of non-training group. catalase activities of training group were lower before exercise but higher during and after exercise. SOD activities were higher regardless of exercise. GSH/GSSG ratio was higher before exercise but was not significantly different during exercise and even lower after exercise. There were no differences between non-training group and training group in MDA levels regardless of exercise. Compared to those of non-training group, atherosclerotic index of training group was lower after exercise and there were no significant differences before and during exercise. There were no differences between non-training group and training group in HDL-C regardless of exercise. These results suggest that moderate physical training can activate antioxidant defenses and decrease the atherosclerotic index and this beneficial effect is evident under exercise-induced oxidative stress.

Biological Evaluation of Residual Malathion in the Meat of Dipped Hens: Influence on Lipid Profile of Erythrocytes and Brain and Pancreatic Lipase and Amylase Activity

  • Biological evaluation of residual malathion after 168 hrs of single dipping exposure of White Leghorn hens to different concentrations (0, 0.5, 1.0 and 1.5%) of pesticides was investigated. Thirty-two male albino rats divided into four groups of eight each were kept on 20% isoproteinous diet prepared from the meat of these malathion dipped hens. After 30 days feeding trial, the rats were killed by decapitation. No significant change was found in erythrocytes. However, the triglyceride concentration in brain tissue was increased significantly (p<0.05) when dose level of pesticide was 1% in dipping solution. Similarly, malathion exposed poultry meat failed in altering any significant change in the pancreatic amylase and lipase activities of rats. This study concludes the virtual absence of toxic accumulation of pesticide in the meat of birds after 168 hrs of exposure in usual concentration range upto 1.5%.

The G115 standardized ginseng extract: an example for safety, efficacy, and quality of an herbal medicine

  • Ginseng products on the market show high variability in their composition and overall quality. This becomes a challenge for both consumers and health-care professionals who are in search of high-quality, reliable ginseng products that have a proven safety and efficacy profile. The botanical extract standardization is of crucial importance in this context as it determines the reproducibility of the quality of the product that is essential for the evaluation of effectiveness and safety. This review focuses on the well-characterized and standardized ginseng extract, G115, which represents an excellent example of an herbal drug preparation with constant safety and efficacy within the herbal medicinal products. Over the many decades, extensive preclinical and clinical research has been conducted to evaluate the efficacy and safety of G115. In vitro and in vivo studies of G115 have shown pharmacological effects on physical performance, cognitive function, metabolism, and the immune system. Furthermore, a significant number of G115 clinical studies, most of them double-blind placebo-controlled, have reinforced the findings of preclinical evidence and proved the efficacy of this extract on blood glucose and lipid regulation, chronic obstructive pulmonary disease, energy, physical performance, and immune and cognitive functions. Clinical trials and 50 years of presence on the market are proof of a good safety profile of G115.

고지혈증 랫트를 이용한 작약의 수종 용매 추출물에 의한 항고지혈 효과 (Effect of Several Solvent Extracts from Paeoniae Radix on Experimental Hyperlipidemia in Rats)

  • Hexane, chloroform, methanol and water extracts of Paeoniae Radix were tested on the experimentally induced hypercholesterolemia in rats for lowering effect of serum lipoprotein contents. Hyperlipidemia was induced on male Wistar rats by feeding high cholestetrol diet for 7 days. Serum lipid profile was verified on these rats by measuring total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL), and low density lipoprotein cholesterol (LDL). Then, the diet was changed to normal. At the same time, hexane, chloroform, methanol and water extract of Paeoniae Radix were given orally on daily basis, and the changes in the serum lipid profile were assessed for 4 weeks. Methanol extract of Paeoniae radix decreased TC level at 1, 2, and 4 week point significantly, and water extract decreased TC level at 4 week point significantly comparing with the control group.
