백서장기내(白鼠臟器內) 핵산(核酸)및 단백질대사(蛋白質代謝)에 관(關)한 연구(硏究) (The Metatolism of Nucleic Acid and Protein in Organs of the Albino Rats)

  • Some effects of dietary conditions on the metabolism of nucleic acid and protein in organs of the Albino Rats have been studied. The young rats to be examined were fed on the control diet and the diets deprived of one component among protein, carbohydrate, and lipid, such as protein free diet (PF: 432 kcal/100g) carbohydrate free diet (CF: 432kcal/100g), and lipid free diet (LF: 392kcal/100g) for three, seven, and fifteen days, respectively. The contents of DNA and RNA in the liver and the brain, and also those of protein-nitrogen(PN) and nonprotein-nitrogen (NPN) in the live, the brain, and the serum have been measured. The results are as followe: 1. The contents of DNA per gram of liver were increased by feeding on protein free diet. It is concluded that the critical factor for the result is not the increase in the rates of DNA syntheses, but the decrease in the turnover rates of DNA. 2. The metabolism of DNA in the liver showed the normal status by feeding on carbohydrate free diet. On the other hand, the rates of DNA syntheses were increased by feeding on lipid free diet. 3. The rates of DNA syntheses in the brain were decreased by feeding on the unbalanced diet, such as protein free, carbohydrete free, and lipid free diet. 4. In the liver and the brain, the rates of DNA syntheses were decreased by feeding on protein free diet. But the rates showed the normal status by feeding on the carbohydrate free diet, and also showed the similar metabolism to that in the case of the control group by feeding on lipid free diet. 5. In the liver, the rates of protein syntheses were decreased, whereas the contents of nonprotein-nitrogen were increased by feeding on protein free diet. 6. In the liver and the brain, the protein syntheses showed the more increasing rates than the rates in the case of the control diet by feeding on lipid free diet. 7. In the serum, the contents of protein did not change in a short period, also the insufficient feeling on protein was examined. It is clear that in the liver the rates of protein syntheses are decreased and the rates of protein catabolism are increased, since the rates of nucleic acid syntheses are decreased by feeding on the protein free diet. On the other hand, it is considered that in the brain the turnover rates of protein does not have correlation with the rates of nucleic acid syntheses, also these are decreased by feeding on protein free diet. And also it is believed that the phenomena of homeostasis for carrying the normal metabolism of nucleic acid and protein in the liver and the brain are operated in a short period as possible, by feeding on carbohydrate free and lipid free diets.

비타민 C 보충과 요가가 중년여성의 혈청 지질과산화물가 개선에 미치는 영향 (The Effects of Vitamin C Supplementation and Yoga on the Improvement of Serum Lipid Peroxidation in Middle-aged Women)

  • 비타민 C 섭취 및 요가가 중년 여성의 총항산화능, 지질과 산화 및 혈중지질에 미치는 영향을 알아보기 위하여 B광역시 H구에 거주하는 중년여성을 대상으로 12주간 실험을 실시하였다. 요가 프로그램은 주 3회, 1일 60분, 운동 강도 RPE $13{\sim}15$로 실시하였다. 비타민 C는 1일 1 g을 섭취시켰다. 12주 후 총항산화능의 변화는 정상군 28.23%, 비타민 C군 30%, 요가군 26.58%, 비타민 C 섭취와 요가군은 43.66%가 증가하였으나 군간 유의적인 차이가 없었다. 지질과산화물가의 변화는 정상군 10%, 비타민 C군 -25%, 요가군은 -32.14%, 비타민 C 섭취와 요가군은 -33.87%로 정상군에 비해 프로그램군이 유의적으로 감소하였으나 프로그램군간의 유의적인 차이는 없었다. 혈중지질농도 변화는 모든군에서 유의적인 차이가 없어 요가 또는 비타민 C 섭취에 따른 지질개선효과를 관찰할 수 없었다. 이상의 결과를 통해 중년여성에게 비타민 C 섭취나 규칙적인 요가 프로그램은 지질과산화물가를 낮춤으로써 체내 산화적 스트레스를 개선하는 효과가 있는 것으로 생각된다.

Serum Levels of Minerals, Ceruloplasmin, and Ferroxidase Activity in Female Adolescent Smokers

  • The purpose of this study was to investigate the nutritional status of serum minerals, ceruloplasmin, and ferroxidase activity in female adolescent students according to their smoking status. The subjects were 87 non-smokers and 88 smokers, who were female high school students. The smokers were divided into tow groups by smoking status, 35 light smokers(pack-year<1) and 53 smokers(pack-year$\geq$1). The serum concentrations of 6 minerals were determined by ICP emission spectroanalyzer. The serum concentration of ceruloplasmin and ceruloplasmin ferroxidase activity were determined. All data were statistically analyzed by SAS PC package program. The serum minerals concentrations of all subjects were in a normal range. There were significantly differences in the concentrations of serum copper, zinc, magnesium, and phosphorus by smoking status while there were no significant differences in the concentrations of serum iron and manganese by smoking status. Furthermore there were no significant differences in the ceruloplasmin concentration and ferroxidase activity by smoking status. In conclusion, smoking status altered the serum levels of some minerals in healthy young women. The minerals levels in light smokers with relatively short pack-year(pack-year<1) were altered compared to those in non-smokers or smokers. This finding seemed to be consistent with the results of previously published data related to antioxidant vitamin and lipid peroxide levels. However further research is needed to clarify these findings in female adolescent smokers.

Efficacy of Brown Seaweed Hot Water Extract Against Hcl-ethanol Induced Gastric Mucosal Injury in Rats

  • Effect of pre-treatment with hot water extract of marine brown alga Sargassum polycystum C.Ag. (100 mg/kg body wt, orally for period of 15 days) on HCI-ethanol (150 mM of HCI-etha-not mixture containing 0.15 N HCI in $70\%$ v/v ethanol given orally) induced gastric mucosal injury in rats was examined with respect to lipid peroxides, antioxidant enzyme status, acid/pepsin and glycoproteins in the gastric mucosa. The levels of lipid peroxides of gastric mucosa and volume, acidity of the gastric juice were increased with decreased levels of antioxidant enzymes and glycoproteins were observed in HCI-ethanol induced rats. The rats pre-treated with seaweed extract prior to HCI-ethanol induction reversed the depleted levels of antioxidant enzymes and reduced the elevated levels of lipid peroxides when compared with HCI-ethanol induced rats. The levels of glycoproteins and alterations in the gastric juice were also maintained at near normal levels in rats pre-treated with seaweed extract. The rats given seaweed extract alone did not show any toxicity, which was confirmed by histopathological studies. These results suggest that the seaweed extract contains some anti-ulcer agents, which may maintain the volume/acidity of gastric juice and improve the gastric mucosa antioxidant defense system against HCI-ethanol induced gastric mucosal injury in rats.

글루포시네이트 암모늄 중독환자에서 경련 예측인자로서의 혈중 지질 농도의 유용성 (The Usefulness of Serum Lipid Concentration as a Predictor of Convulsion in Patients with Glufosinate Ammonium Poisoning)

  • Purpose: Glufosinate ammonium (GA; phosphinothricin) can induce neurological complications such as altered mental status, amnesia, and convulsions. This study was conducted to evaluate whether blood lipid profiles can help predict convulsions in patients with GA poisoning. Methods: This study was a retrospective review of data acquired at a tertiary academic university hospital from March 2014 to July 2016. Independent t-test, Mann-Whitney test and Analysis of covariance (ANCOVA) of demographic and laboratory findings of 50 patients with GA poisoning were performed to identify correlations of general characteristics and laboratory findings, including blood lipid profiles of GA-poisoned patients between with and without convulsions. Results: Convulsion as a GA complication showed a significant association with poison volume, age, white blood cell count, and creatine phosphokinase (CK), albumin, lactate dehydrogenase (LDH), low-density lipoprotein (LDL), and high-density lipoprotein (HDL) content in blood according to an independent t-test and Mann-Whitney test. However, ANCOVA demonstrated significant association with LDL and triglyceride. Conclusion: Blood lipid profiles, especially serum LDL and triglyceride, were useful in predicting convulsions in patients with GA poisoning.

내당능 장애 모델 흰쥐에서 Vitamin E의 과잉섭취가 지질 분포와 혈청 Insulin 농도변화에 미치는 영향 (Effect of Vitamin E Supplementation on Lipid Distribution and Serum Insulin in Experimental Model Rats with Impaired Glucose Tolerance)

  • The aim of the study was to determine whether vitamin E supplementation in three experimental model rats with impaired glucse tolerance could change serum insulin concentration and lipid distribution. The three groups were adult(AS) and neonatal (NS) streptozotocin-induced groups, and a high sucrose diet(HS) group. Each group was divided into control and vitamin E supplementatino groups at the age of 9 weeks. The level of vitamin E supplementation was 5g/kg diet. Blood and organ samples were taken at 5 and 10 weeks and were examined for changes in the level of serum insulin, glucose, lipids, liver lipids, and oxidative status. Vitamin E supplementation significantly reduced serum insulin in the HS group and caused the significant beneficial changes in serum lipids and triglycerides in As grouop at 10 weeks . In all groups, serum vitamin E was increased and malondialdehyde(MDA) in serumand liver were decreased significantly by vitamin E supplementation. The results suggest that vitamin E supplementation improves lipid distribution in adult streptozotocin-induced rats and serum insulin concentration in high sucrose diet-induced rats. Vitamin E might prevent on reduce oxidative injury in all experimental model rats with impaired glucose tolerance.

동과인의 보충이 당뇨 유발 흰쥐의 글리코겐과 지질과산화에 미치는 영향 (Effects of Benincasa Hispida Seed Supplementation on Glyeogen status and Lipid Peroxidatin in Streptozotecin-Induced Diabetic Rats)

  • This study was designed to examine the effects of diets containing different levels of seeds of Benincasa hispida(wax gourd) on glycogen, protein levels and lipid profiles as well as malondialdehyde (MDA) in streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley rats were induced diabetes mellitus by STZ injection (45 mg/kg) into the tail vein and were divided into four groups: normal, STZ-control, and two experimental groups. Normal and STZ-control groups were fed the AIN-93 diet and the two experimental groups were fed a modified diet containing 2.5% and 5.0% of wax gourd seed powder for four weeks. The liver, muscle, lung, kidney, and pancreas were excised after sacrifice, then the glycogen, protein, and lipid peroxidation products were measured. The rats fed 2.5% wax gourd seed group showed higher levels of liver glycogen compared with that of the STZ-control group. The levels of kidney protein were significantly increased in the 2.5% and 5.0% wax gourd seed groups. There were no significant difference cholesterol and liver triglyceride levels of the liver and MDA concentration in the liver, lung, and kidney among all four groups. These results show that wax gourd seed treatment of 2.5% and 5.0% doses did not exhibit profound anti-lipid peroxidation properities.

Effect or Cornus officinalis Sieb. et Zuccha Extracts on Physiological and Antioxidative Activities in Streptozotocin Induced Diabetic Rats

  • This investigation was performed to study the antioxidant activities of Cornus officinalis Sieb extracts and the effect of Cornus officinalis Sieb extracts on glucose, lipid metabolism in diabetic rats. DPPH free radical scavanging activity and superoxide anion radical scavenging of Cornus officinalis Sieb extracts were 94.7% and 92.1%, respectively. Streptozotocin (45 mg/kg body weight, i.p.) induced diabetic rats showed a significant increases of plasma glucose, triglyceride and total cholesterol, concomitantly significant decrease of plasma high density lipoprotein. Glutathione level were decrease in cytosol of liver, lung and brain tissue of rats. Lipid peroxide were increase in microsome of liver cells. Group 1 and 2 were treated with Cornus officinalis Sieb extracts 200 mg/kg body weight and 100 mg/kg body weight for 24 days, individually. Group 1 and 2 rats showed decreased plasma glucose, triglyceride, total cholesterol and lipid peroxide in microsome of liver, and increased plasma high density lipoprotein and glutathione in cytosol of liver, lung and brain. The result suggest that Cornus officinalis Sieb extracts may normalize the Impaired antioxiants status in streptozotocin induced diabetic rats. Cornus officinalis Sieb extracts were used to improve the imbalance between free radicals and antioxidant system due to the diabetes.

여자노인에서 항산화 비타민 보충이 혈장 지질 개선에 미치는 영향 (Effects of Antioxidant Supplementation on Lipid Profiles in Elderly Women)

  • Antioxidant vitamin supplementation focuses one's attention on the prevention of age-related diseases. This study was conducted to investigate the antioxidant status and lipid profiles and to look into the antioxidant vitamin supplementation that affects lipid metabolism in 20 elderly non-smoking Korean women (placebo group: n = 6, vitC suppl: n = 7, vitE suppl: n = 7). Age, height, weight, muscle, percent of fat and WHR were not significantly different among the groups, however $\%$ of fat was above $33\%$ and WHR was above 0.9. And blood pressure of the placebo group was 131.7/81.7 (border line hypertension), that of vitamin C supplement was 141.4/87.1 (hypertension) and that of vitamin E supplement was 151.4/92.9 (hypertension). Although nutrient intakes of all groups were poor, antioxidant status (blood vitamins C, E, A, and beta-carotene) and lipid profile (TG, total-cholesterol, VLDL-cholesterol, LDL-cholesterol, HDL-cholesterol) were normal. For nutritional intervention, the vitamin C supplement group received L-ascorbic acid 1,000 mg, and vitamin E supplement group received d-alpha-tocopherol 400IU for 4 weeks, showing the effects of vitamin E supplementation. Response total cholesterol of HDL-cholesterol (T-Chol/HDL) in vitamin E supplement group was significantly decreased from 4.3 to 3.2. And response LDL-cholesterol of HDL-cholesterol (LDL/HDL) in the vitamin E supplement group was also significantly decreased from 2.6 to 1.7. In addition, after the adjustment for plasma lipids (TG, total cholesterol), plasma vitamin A levels in vitamin E supplement group were significantly increased from 7.89 mg/g to 14.91 mg/g. And systolic blood pressure in vitamin E supplement group was significantly reduced. These results suggested that vitamin E supplementation affects the lipid profiles and blood pressure in elderly non-smoking women. So various nutrition programs must be implemented against age-related diseases and further studies are needed regarding sorts and amounts of antioxidant nutrients and supplementation periods.

Vitamin E improves antioxidant status but not lipid metabolism in laying hens fed a aged corn-containing diet

  • Objective: The objective of this study was to determine whether a dietary vitamin E (VE) supplement could alleviate any detrimental effects of aged corn on lipid metabolism and antioxidant status in laying hens. Methods: The experiment consisted of a 2×3 factorial design with two corn types (normal corn and aged corn (stored for 4 yr) and three concentrations of VE (0, 20, and 100 IU/kg). A total of 216 Lohmann laying hens (50 wk of age) were randomly allocated into six treatment diets for 12 wk. Each treatment had 6 replicates of 6 hens per replicate. Results: The results show that aged corn significantly decreased the content of low-density lipoprotein cholesterol (p<0.05), and reduced chemokine-like receptor 1 (CMKLR1) mRNA expression (p<0.05) in the liver compared to controls. Diet with VE did not alter the content of crude fat and cholesterol (p>0.05), or acetyl-CoA carboxylase, lipoprotein lipase, fatty acid synthase or CMKLR1 mRNA expression (p>0.05) in the liver among treatment groups. Aged corn significantly increased the content of malondialdehyde (MDA) (p<0.05) and decreased superoxide dismutase (SOD) activity (p<0.05) in the liver. The VE increased the content of MDA (p<0.05) but decreased glutathione peroxidase (GSH-Px) activity in serum (p<0.01) and in the ovaries (p<0.05). Adding VE at 20 and 100 IU/kg significantly increased GSH-Px activity (p<0.05) in liver and in serum (p<0.01), 100 IU/kg VE significantly increased SOD activity (p<0.05) in serum. Aged corn had no significant effects on GSH-Px mRNA or SOD mRNA expression (p<0.01) in the liver and ovaries. Addition of 100 IU/kg VE could significantly increase SOD mRNA expression (p<0.01) in the liver and ovary. Conclusion: Aged corn affected lipid metabolism and decreased the antioxidant function of laying hens. Dietary VE supplementation was unable to counteract the negative effects of aged corn on lipid metabolism. However, addition of 100 IU/kg VE prevented aged corninduced lipid peroxidation in the organs of laying hens.
