This study was conducted to investigate the effect of protein kinds and levels on lipid metabolism in hypercholesterolemia rats. Male Sprague Dawley rats were administrated 1% cholesterol and 0.25% sodium cholate to induce hypercholesterolemia and were fed on diet containing two levels(7, 20%) of casein and soy protein respectively. The rats were sacrified after 5 weeks of feeding periods. Total lipid content in serum was decreased in proportion to level of protein and soy protein was more effective than casein in lipid-lowering effect. Serum triglyceride content was significantly decreased in proportion to level of protein and did not show significance by protein kind. Serum phospholipid content was more significantly decreased in 20% protein groups than that of 7% protein groups. Total cholesterol content in serum was significantly decreased by protein and were decreased in proportion to level of soy protein. Free cholesterol content did not show significance by protein level. The degree of decrease predominated in soy protein groups. Serum HDL-cholesterol content and RL-cholesterol were increased in proportion to level of protein and effect of soy protein was not more significantly than that of casein. Atherogenic index was significantly decreased in proportion to level of protein and soy protein groups were more effective. Serum LDL-, VLDL-cholesterol contents were more significantly decreased in 20% protein groups than that of 7% protein groups. Serum $\beta$-lipoprotein and chylomicrone-cholesterol contents also showed significantly decrease by soy protein.

This study was designed to evaluate lipid component of serum and organ in matured rats fed oil extract from ducks. Male Sprague-Dawley rats weighing 595g(52-week-old) were divided into 3 groups and fed experimental diets for 8 weeks. Rats were fed soybean oil(control group) diet, duck oil(DO group) diet and duck oil supplemented with 1% cholesterol(DOC group) diet. Serum triglyceride concentration was significantly lower in DO and DOC groups than that in the control group. Serum total cholesterol concentration was significantly higher in DOC group than those in the other groups, while serum HDL-cholesterol concentration was significantly lower in DOC group than those in the other groups. Hepatic total lipid, triglyceride and total cholesterol concentrations in DOC group were significantly higher than those in the other groups. These studies indicate that the duck oil decreases the concentrations of triglyceride and shows almost same concentrations of total cholesterol compared to the soybean oil supplemented group in serum and liver. But hypercholesterolemic effect of duck oil was not improved.

Lipid peroxidation is one of the main manifestations of oxidative damage and has been found to play an important role in the toxicity and carcinogenesis of many carcinogens. This study was carried out to determine the effects of vitamin C on lipid contents and fatty acid compositions of serum and liver in male rats treated with radiation or aflatoxin $B_1\;(AFB_1)$. Six week-old male Sprague-Dawley rats were randomly divided into 7 groups; control group, radiation exposed group, $AFB_1$ treated group, X-ray and $AFB_1$ co-treated group. Three groups, except control group, were each further divided into vitamin C administered group and not administered groups. For this study, vitamin C was injected with 10 mg/kg of body weight by intraperitoneal injection and 1 hr later, 0.4 mg/kg of $AFB_1$ was injected by the same method. These administrations were repeated every 3 days over a period of 15 days. Only one time, X-ray was irradiated on whole liver with 1,500 cGy. Then vitamin C and AFB1 were administered by the same level and same method described above. On the 16th day of treatments, the animals were sacrificed. From the analysis of the serum lipid patterns, significant decrease (p<0.01) in triglyceride (TG) and total cholesterol levels were observed in X-ray and $AFB_1$ co treated group administered with vitamin C (group 7). In liver lipids, the levels of free cholesterol and total cholesterol were also decreased in X-ray and $AFB_1$ co treated group administered with vitamin C (group 7). The levels of serum free cholesterol and hepatic TG were not significantly different among all groups according to vitamin C administrations. The high density lipoprotein (HDL)-cholesterol level of serum was significantly (p<0.01) increased while the low density lipoprotein (LDL)-cholesterol level was decreased in X-ray and $AFB_1$ co treated group administered with vitamin C (group 7). In the phospholipid fatty-acid compositions of serum and liver tissue, group 3, 5 and 7 showed an increase in polyunsaturated fatty-acid (PUFA) but a decrease in saturated fatty acid (SFA) when compared to the control group. The composition ratio of fatty acid varied according to vitamin C administration. These results suggested that vitamin C has partly suppressive effects on lipid contents and fatty acid composition of serum and liver in rats treated by radiation and $AFB_1$.
This study was performed to investigate the effects of concentration and degree of unsaturation of fatty acids on cellular proliferation and lipid peroxide production, using primary skin fibroblasts from neonate rats Fibroblasts (CPD : 2.8-5.4). Cells were cultured either in control medium (Dulbecco's modified Eagle's medium supplement with 10% fetal bovine serum) or in media supplemented with various kinds (stearic, oleic, linoleic, arachidonic, linolenic, eicosapentaenoic acid) and amounts (5, 10, 25, 50, 100, 150uM)of fatty acids. Cellular proliferation ratio and lipid peroxice production were measured and morphological changes were observed. Cellular proliferation was inhibited and morphological changes were observed. Cellular proliferation was inhibited and morphological changes were observed in cells grown in stearic containing media. Oleic, arachidonic, and eicosapentaenoic aicd tend to stimulate cellualar proliferation, and linolenic acid had no effects. Lipid peroxide concentrations in fibroblasts increased in proportion to the contents and unsaturation of fatty acids in media. Especially supplementation of arachidonic acid accelerated cellualr lipid peroxidation. Free radicals may cause severs damage to biological molecules, so lipid peroxidation probably contributes cellular membrane damages. However there were little relationship between lipid peroxide production and cellular proliferation in this study. (Korean J Nutrition 29(2) : 159~165, 1996)
We examined the associations between adiponectin or leptin and serum ICAM-1 levels in seventy-six hypercholesterolemic patients (mean age 59 yrs, 25 males and 51 females, LDL-cholesterol>=130mg/dL at screening). Blood lipid profiles and HOMA-IR derived from fasting glucose and insulin concentrations were determined. Serum levels of adiponectin, leptin and ICAM-1 were analyzed using ELISA The results showed that serum levels of leptin were positively associated with serum levels of ICAM-1 independent of age, sex and BMI (r=0.392, p<0.001). Serum levels of adiponectin were negatively associated with serum levels of ICAM-1 independent of age, sex and BMI (r=-0.343, p<0.005). Stepwise multiple linear regression analysis showed that serum leptin was an independent factor to be associated with serum ICAM-1 levels after adjusting for age, sex, BMI, alcohol intake, smoking status, blood lipids such as total cholesterol, triglyceride, HDL cholesterol and LDL cholesterol and HOMA-IR (p<0.001). With respect to adiponectin, its association with serum ICAM-1 was attenuated but still significant when further adjustments were made for age, sex, BMI, alcohol intake, smoking status, blood lipids such as total cholesterol, triglyceride, HDL cholesterol and LDL cholesterol and HOMA-IR (p<0.005). In conclusion, this study suggests that adiponectin and leptin are associated with endothelial derived inflammation.
To study the relation between serum fatty acids and coronary artery disease(CAD), 194 subjects were randomly selected and divided into three groups(control, single vessel disease group(SVD), muliple vessel disease group(MVD)) according to the angiographic results. Total serum levels of fatty acids and serum phospholipid(PL)-fatty acids(FAs)were analysed using gas chromatography and their associations with CAD were examined. Different patterns of total serum fatty acid levels were found in men and women. Levels of most fatty acids of SVD and MVD were significantly lower in men, while those of MVD were significantly higher in women. In terms of PL-FAs in both men and women, the levels of PL-FAs follow the order of control < SVD < MVD and in women, the difference was significant. Various ratio(including ratios denoting the activites of desaturase and elongase) of total serum-and PL-FAs were similar in the three groups. In the relation of serum fatty acids to serum lipid profiles, PUFA & LDL showed a negative correlation, while, SFA & LDL-cholesterol and PUFA and HDL-cholesterol showed positive correlatons. correlations. Systolic blood pressure and alcohol intake levels negatively affected the levels of serum LA, AA, EPA and DHA in the risk factor analysis. These findings are consistent with other evidence indicating that fatty acid compositions are changed in CAD, especially on the concentration base and the change was related to the severity of the disease. Therefore, for the purpose of disease prevention and therapeutic use, balanced intakes of various fatty acids must be seriously considered.

To examine the effect of Codium fragile on blood cholesterol and lipid metabolism, hyperlipidemia was induced in experimental animal rats through the administration of a hypercholesterolemic diet. Codium fragile powder was then administered to the rats for 5 weeks, after which, blood biochemical changes such as blood cholesterol, Aspartate Aminotransferase (AST: serum SGOT) and Alanine Aminotransferase (ALT: serum SGPT) enzyme activity, etc. were determined. And histological changes in liver cells were examined using an electron microscope. Codium fragile treatment resulted in a significant reduction of the levels of total cholesterol, blood triglyceride and low-density cholesterol (LDL. Chol) compared to the control rats. In contrast the expression levels of high-density cholesterol (HDL. chol.) were increased. The AST value of the Codium fragile administration group was significantly reduced and the blood ALT value of the Codium fragile group showed a significant decrease in comparison to the negative control group. In summary, this study demonstrated the beneficial possibilities of Codium fragile in improving the abnormality of lipid metabolism caused by liver cell damage and hyperlipidemia.
Purpose: The study investigated the effects of 9 weeks of group music rope-jumping training on health-related physical fitness and blood lipid in obese elementary school boys and girls. Method: Subjects were randomly assigned to either a training group (37 boys and 18 girls) or control group (36 boys and 19 girls). The training group exercised for 1 hour, 2 days per week during the 9-week supervised music rope-jumping training program. The control group was asked to maintain their normal daily physical activities. The effects of the interventions on physical fitness and blood lipids were analyzed by two-way repeated measures ANOVA (group ${\times}$ time). Results: There were significant group ${\times}$ time interaction effects on body weight (p<.023), %body fat (p=.09), body mass index (p=.018), and body fat mass (p=.019) in school girls. However, there was not an interaction effect on serum lipids in both genders. Conclusion: The 9-week music rope-jumping training program used was effective for improving body composition in obese elementary school girls.
Effects of ${\alpha}$-tocopherol and perilla oil on the toxicity of polychlorinated biphenyls (PCB) in male rat were studied. Rats were fed ad libitum for 6 weeks with the animal diet which contains PCB 30 ppm and 100 ppm. Perilla oil (0.5 g/kg body weight) and ${\alpha}$-tocopherol (30 mg/kg body weight) were administered intraperitoneally twice a week for 6 weeks. Rats fed with PCB showed enlargement of liver and spleen, increase in aspartate aminotransferase, alkaline phosphatase, sereum lipid and cytochrome P 450 and decrease in body weight and glutathione. When perilla oil was administered to rats fed with PCB increase in aspartate aminotransferase, alkaline phosphatase, serum lipid and cytochrome P45O and decrease in body weight and glutathione were significantly augmented, compared to rats fed with PCB alone. This means that perilla oil potentiates the toxicity of PCB. On the other hand when ${\alpha}$-tocopherol was administered to rats fed with PCB increase in aspartate aminotransferase, alkaline phosphatase, serum lipid and cytochrome P45O and decrease in body weight and glutathione were signigicantly reduced, compared to rats fed with PCB alone. This means that u-tocopherol reduces the toxicity of PCB. From the above results, it may be concluded that PCB is metabolized by microsomal mixed function oxidase and the metabolite causes the toxicity and microsomal glutathione plays a role of protection on the toxicity of PCB.
Doxorubicin is a powerful anticancer antibiotic extensively used in the treatment of several types of cancers. Long-term administration of this drug results in cumulative dose related cardiotoxicity due to enhanced production of free radicals leading to oxidative stress. Our earlier investigations have demonstrated significant antioxidant, anti-inflammatory and antitumour properties of Ganoderma lucidum extracts. We extended our investigations to evaluate the protective effect of Ganoderma lucidum extract against doxorubicin-induced cardiotoxicity. Administration of 3 doses of doxorubicin, 6 mg/kg body weights, i.p. per each dose, alternative days, showed dear signs of cardiotoxicity in rats. The drug enhanced serum creatine kinase (CK) activity and lipid peroxidation in tissue drastically. The drug also induced significant decrease in GSH level and activities of CAT, SOD and GPx. Administration of methanolic extract of G.lucidum (500 and 1,000 mg/kg body weight) significantly increased the level of GSH and activities of CAT, SOD and GPx. Activity of CK was significantly lowered in a dose dependent manner. The treatment also caused significant decrease in lipid peroxidation (MDA). The results thus indicated that methanolic extract of G.lucidum prevented oxidative stress caused by doxorubicin administration and the increase in serum CK activity and lipid peroxidation in the tissue. The experimental findings suggest the therapeutic potential of G.lucidum as adjuvant in cancer chemotherapy.
