Anti-atherogenic Effects of Curcumin in Hypercholesterolemic Rabbits (고콜레스테롤혈증 토끼에서 Curcumin의 항동맥경화 효과)

  • Curcumin, the yellow pigment in turmeric, curry and mustard, has anti-inflammatory and anti-carcinogenic activities. In this study; we investigated the hypocholesterolemic and anti-atherogenic effect of curcumin in rabbits. Hypercholesterolemia was induced by feeding high cholesterol diet to male rabbits for 30 days, and the animals were then fed high cholesterol diet containing 0.1% (w/w) or 0.5% (w/w) curcumin for additional 30 days. Supplementation of 0.l% curcumin tended to lower serum total cholesterol and low density lipoprotein (LDL)-cholesterol levels and inhibit serum lipid peroxidation. In the 0.5% curcumin-supplemented group, serum total cholesterol was significantly lowered by 11.7%, LDL-cholesterol by 12.8% and lipid peroxidation by 47.9% compared to the control group. Hepatic cholesterol and triglyceride contents were also significantly lowered by 50.6% and 37.4%, respectively compared to the control group. Lipid staining of the arteries isolated from the curcumin-treated rabbits showed that curcumin significantly decreased formation of fatty streaks and atheromatous plaques on the intima of the arteries. These results demonstrated that curcumin lowered serum cholesterol concentration, hepatic cholesterol and triglyceride contents, and accumulation of cholesterol in the artery These cholesterol lowering effects of curcumin, together with its anti-oxidative and anti-inflammatory activities, may play some important roles in preventing atherosclerosis.

Troglitazone Lowers Serum Triglycerides with Sexual Dimorphism in C57BL/6J Mice

  • Thiazolidinediones (TZDs) are widely used antidiabetic drugs that activate the nuclear peroxisome proliferator-activated receptor ${\gamma}(PPAR{\gamma})$, and thereby improve the metabolic abnormalities linking hypertriglyceridemia to diabetes, hyperglycemia, insulin resistance, and cardiovascular disease. To determine whether the $PPAR{\gamma}$ ligand troglitazone regulates lipid metabolism with sexual dimorphism, we examined the effects of troglitazone on circulating lipids, body weight and the expression of hepatic genes responsible for lipid metabolism in both sexes of C57BL/6J mice. Compared to mice fed a low fat control diet, both sexes of mice fed a troglitazone-treated low fat diet for 14 weeks did not exhibit changes in body weight gain, serum total cholesterol, HDL-cholesterol and LDL-cholesterol levels. However, serum triglycerides were significantly reduced in both sexes of mice, although these effects were more pronounced among males. Furthermore, troglitazone regulated the expression of hepatic genes critical for lipid and lipoprotein metabolism, the magnitudes of which were much higher in males compared to females, as evidenced by results for increased acyl-CoA oxidase and decreased apolipoprotein C-III mRMA levels. These results suggest that $PPAR{\gamma}$ activator troglitazone may exert sexually dimorphic control of serum triglycerides in part through the differential activation of $PPAR{\gamma}$ in liver between male and female mice.

Serum Lipids Can Convert Bovine Myogenic Satellite Cells to Adipocytes

  • Serum lipid (SL) is a commercially available cholesterol-rich, proteinaceous compound extracted from bovine serum. Here we investigated the adipogenic transdifferentiation potential of SL on bovine myogenic satellite cells. Exposure of satellite cells to SL could generate lipid droplets on day 2, and further exposure to SL increased cytoplasmic lipid accumulation giving adipocyte morphology. The expression analysis of PPAR gamma and GPDH adipocyte markers along with Oil-red-O staining results confirmed the transdifferentiation potential of SL. When cells were treated at different concentrations (5, 10, 20, $40{\mu}l$/ml) of SL, the results indicated that even levels as low as $5{\mu}l$ SL /ml could induce transdifferentiation, and maximum induction was obtained at $20{\mu}l$ SL/ml. After treatment with SL at different concentrations the expression levels of PPAR gamma varied significantly (p<0.05), whereas the expression of other adipogenic transcription factors showed no difference, indicating that SL acts through PPAR gamma. The combined effect of SL and troglitazone proved to be the best combination for induction of transdifferentiation compared to the individual effect of SL or troglitazone. Thus, overall results clearly show that SL induces transdifferentiation of bovine myogenic satellite cells to adipocytes.

Alterations in the blood glucose, serum lipids and renal oxidative stress in diabetic rats by supplementation of onion (Allium cepa. Linn)

  • This study examined the anti-diabetic effect of onion (Allium cepa. Linn) in the streptozotocin (STZ)-induced diabetic rats. Male Sprague-Dawley rats were divided into normal rats fed control diet or supplemented with onion powder (7% w/w) and diabetic rats fed control diet or supplemented with onion powder. Diabetes was induced by a single injection of STZ (60 mg/kg, ip) in citrate buffer. The animals were fed each of the experimental diet for 5 weeks. Blood glucose levels of rats supplemented with onion were lower than those of rats fed control diet in the diabetic rats. Onion also decreased the total serum lipid, triglyceride, and atherogenic index and increased HDL-cholesterol/total cholesterol ratio in the diabetic rats. Glutathione peroxidase, glutathione reductase and glutathione S-transferase activities were high in the diabetic rats compared to normal rats and reverted to near-control values by onion. These results indicate that onion decreased blood glucose, serum lipid levels and reduced renal oxidative stress in STZ-induced diabetic rats and this effect might exert the anti-diabetic effect of onion.

Effects of Sayeoksanhap-Pyeongweisan-Gamibang(SPG) on Hepatological Changes in Animals Damaged by $CCl_4$ (사역산합평위산가미방(四逆散合平胃散加味方)의 경구투여와 약침의 병용이 백서의 간손상에 미치는 영향)

  • Objectives: This study was designed to investigate protective effects of Sayeoksanhap-Pyeongweisan-Gamibang(SPG) on hepatological changes in rats damaged by $CCl_4$. Methods : We investigated the effects of SPG on hepatological changes in terms of AST/ALT levels, Total cholesterol, HDL-cholesterol and triglyceride in serum and lipid peroxidation in rat liver. Results : Combination treatment with acupuncture and oral administration of SPG decreased AST and ALT in serum. Combination treatment also decreased lipid peroxidation in liver and triglyceride in serum. In addition, HDL-cholesterol in serum was increased significantly by combination treatment. Contrary, level of total cholesterol was not changed by combination treatment. Conclusion : These results suggest that Combination treatment with acupuncture and oral administration of SPG was useful to protect liver against $CCl_4$ by its antioxidant action in liver tissues.

Relationships among Lifestyle, BMI, BP, and Serum Lipid Profiles in Working Men (성인 직장 남성의 생활습관과 체질량지수, 혈압 및 혈중지질농도의 상관성)

  • Purpose: The purpose of this study was to examine lifestyle, BMI, BP, and lipids profiles in male subjects and to explore the relationships among variables. Methods: A total of 148 male subjects were recruited from one life insurance company from December 1, 2005 to February 28, 2006. Data collection methods were structured questionnaire, anthropometry and serum analysis. The relationships among lifestyle, BMI, BP, and serum lipid profiles were assessed by descriptive analysis, t-test, ANOVA, and partial Pearson's correlation coefficient of variables after controlling for age, educational level, and economic status. Results: The mean BMI of the participants was relatively high as 25.38(range: 18.38 - 32.83). The differences of serum lipid profiles according to age, educational level, and economic status were significant. 'Use of caffeine and drugs'(r = -.187, p < .05) and 'consciousness of safety'( r= -.200, p < .05) was negatively related to BMI. Higher score of 'type of personality' domain was correlated with lower systolic BP(r = -.221, p < .01) and lower diastolic BP(r = -.195, p < .05) and was positively correlated with HDL(r = .191, p < .05). Conclusion: 'Use of caffeine and drugs', 'consciousness of safety' and 'type of personality' of lifestyle as well as 'dietary habit' and 'exercise' played a key role in circulatory disease.

A Study on the Growth Rate, Nutritional Effects and Serum Lipid Contents of Rats by Feeding with Leaf and Stem Extract of the Ginseng Radix (인삼잎과 줄기 혼합 추출액의 첨가급식이 흰쥐의 성장, 영양효과 및 혈청 지질에 미치는 영향)

  • This study was conducted to investigate the application possibility of leaf and stem extract(LSE) extracted from the mixture of leaf and stem of ginseng radix(Panax ginseng CA. Meyer). This study measured the intake levels and efficiency ratio, growth rate, absorption ratio of carbohydrate, lipid and protein of rat by feeding with LSE during 6 weeks. We analyse the hematological and serum metabolic variables, serum lipid concentrations. Total diet and protein intake levels were low, but efficiency ratios were significantly high in LSE administered groups than the control group. Weight gain, liver and kidney weight of LSE groups were significantly higher than the control group. Blood RBC, Hct, Hb, total protein and albumin concentrations were reasonable levels in LSE administered groups compared to the basal diet group. Also serum total cholesterol, LDL-cholesterol, triglyceride contents of LSE groups were low, but HDL-cholesterol level was higher than the basal diet group. These results imply that leaf and stem of ginseng radix could be used as possible food resources, functional food material and feed stuff.

Effects of the Zizyphus jujuba Seed Extract on the Lipid Components in Hyperlipidemic Rats

  • The purpose of this study was designed to observe the effects of Zizyphus jujuba seed extract on the concentrations of the lipids and blood glucose in the S.D. rats fed the experimental diets for 4 weeks. Concentrations of total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, triglyceride (TG), phospholipid (PL) and blood glucose in serum were significantly higher in the cholesterol administration groups (group 2 (cholesterol+water), group 3 (cholesterol+Zizyphus jujuba seed extract)) than those in the control group (group 1, basal diet+water). But the concentrations of total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, TG, PL and blood glucose in serum ware remarkably lower in the group 3 than those in the group 2. In the ratio of HIDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration, Zizyphus jujuba seed extract administration group was higher percentage than in the group 2. The activities of aspartate aminotransferase (AST), alanine amino-transferase (ALT), lactate dehydrogenase (LDH) and alkaline phosphatase (ALP) in serum were rather lower in the Zizyphus jujuba seed extract administration group (group 3) than in the cholesterol diet group (group 2). From the above research, Zizyphus jujuba seeds were effective on the improvement of the blood glucose, lipid compositions in serum of dietary hyperlipidemic rats. And particularly, Zizyphus jujuba seeds were more effective as a therapeutic regimen for the control of metabolic derangements in adult disease.

The Study of Serum lipid Profile and Food Behaviors in Healthy Offsprings of Korean NIDDM Patients (인슐린 비의존형 당뇨병의 가족력을 가진 정상 자녀의 혈청 지질 및 식습관에 관한 연구)

  • This study was to investigate the risk factors for the development of diabetes mellitus on healthy offsprings of Korea non-insulin dependent diabetes mellitus(NIDDM) patients. Forty four offsprings who have at least 1 parent with NIDDM and age, sex and body mass index(BMI) matched forty four healthy control subjects were collected for this study. BMI, skinfold thickness, waist/hip circumference ratio(WHR), fasting blood glucose, serum insulin, serum lipid profile, nutrient intakes and food behaviors were measured. There were no significant difference in BMI, WHR, fasting blood glucose, insulin, total cholesterol, triglyceride and daily nutrients intake between offsprings and controls. But skinfold thickness, LDL-cholesterol level and sweety food eating frequency were significantly higher in male offsprings than in male controls. And HDL-cholesterol was significantly lower in male offsprings than in male controls. Although there were no significant difference, offsprings had a overeating habit and ate more confectionery and greasy food than controls.

The Effect of Lycii fructus beer intake on serum lipid profiles and antioxidant activity in rats (구기자 맥주의 섭취가 흰쥐의 혈청 지질패턴 및 항산화효소 활성에 미치는 영향)

  • This study was performed to investigate the effect of Lycii fructus beer on serum lipid profiles and antioxidant activity in rat Sprague-Dawley (SD) rats weighting about 190g were divided into the following 5 groups ; distillate water (Control), 5% ethanol in distillate water (Ethanol), commercial beer (CB), Lycii fructus beer (LFB) and 5% alcohol red wine diluted with distillate water (RW). Body weight, total food intake, FER and percent organ (liver, kidney) weight per body weight were not significantly changed by Lycii fructus beer drinking. After 6 weeks, serum total cholesterol, triglyceride and HDL cholesterol level were not significantly different. But, Lycii fructus beer intake tended to decrease serum triglyceride level and atherogenic index. Also, GOT and GPT levels were expressed lower than Ethanol group. There was not significantly different in hepatic glutatiione (GSH) content and glutathione-S-transferase (GST) activities among 5 groups. Lipid peroxidation in the hepatic was decreased by Lycii fructus beer intake. The results demonstrated that Lycii fructus beer was potential and effective antioxidant that can protect the decrease associated with alcohol.
