The aim of this study was to investigate whether a water extract of L. cladonioides (LC) has an anti-obesity effect in 3T3-L1 cells and obese mice. Treatment of differentiated 3T3-L1 adipocytes with LC caused a significant increase in glycerol release and reduced the protein expression of the adipogenic transcription factors, $PPAR{\gamma}$ and C/$EBP{\alpha}$. In an animal model, obese mice were artificially induced by a high fat diet for 10 weeks. Experimental groups were treated with LC (100 mg/kg/day) by gavage for the next 10 weeks. At the end of experiment, the body weight of the LC group mice was reduced by 14.2% compared to the high fat diet (HFD) group. The treatment also decreased liver (31.0%), epididymal (18.0%) and retroperitoneal (19.3%) adipose tissue, and kidney (6.7%) weights, respectively, compared with those of the HFD group. LC prevented diet-induced increases in the serum level of TC (22.6%), TG (11.6%), and glucose (35.0%), respectively, compared with the HFD group. However, the HDL-C level was higher in the LC group (26.1%) than the HFD group. The results of this study thus suggest that LC suppressed lipid accumulation and expression of adipogenic transcription factors, and increased the amount of glycerol release. LC also indicated an anti-obese and anti-hyperlipidemic effect.
Chitosan, hydroxycitrate and L-carnitine have been known to be antiobesity components. The purpose of this study was to evaluate the combined effects of chitosan, hydroxycitrate and L-carnitine mixture as a potential antiobesity supplement in overweight women. Pre-menopausal healthy females who were overweight (percent ideal body weight > 110) were included in this study. Forty-nine subjects randomly received a placebo (n = 25) or antiobesity-supplement (n = 24), which was a mixture of chitosan, hydroxycitrate, and L-carnitine. Before and after the eight-week experimental period, anthropometric parameters, blood components and computerized tomography were measured. At baseline, the two groups were well matched in terms of age, body mass index and lipid profile. After the eight weeks of potential antiobesity supplementation, the subjects' body fat percent had decreased significantly (p < 0.001) by 5.6% (39.1 $\pm$ 1 vs 36.9 $\pm$ 1%) while lean body mass increased (p < 0.01). Vsceral fat area at the L4 vertebra decreased significantly (p < 0.01) by 8.6% in the supplemented group and the total fat area at the L4 vertebra showed a tendency to decrease (p = 0.051) by 2.4%. Also, in the group given the antiobesity-supplement rather than the placebo, the fasting triglyceride level decreased significantly (p < 0.05) by 10.0%. In addition, serum total cholesterol levels in the antiobesity-supplement group showed a tendency to decrease (p=0.159) by 2.7% (194 $\pm$ 6 vs 189 $\pm$ 6 mg/dl). No side effects were found in either group during the intervention. In conclusion, the present study demonstrated that taking a mixture of chitosan, hydroxycitrate, and L-carnitine as a potential antiobesity supplement for eight weeks produced advantageous changes in the weight and visceral fat accumulation of overweight women without any side effects. To investigate the effect of Feelch on alcohol metabolism, we measured both blood alcohol concentration in human and hepatic alcohol metabolizing enzyme activity in rats. The blood alcohol concentration in Feelch-ingested group was significantly lower than that in water-ingested group at 0, 40, and 80 minute after alcohol intake. The blood alcohol concentration between male and female taken 300ml of $21\%$ alcohol showed the significant differences; the peak value of blood alcohol concentration in male and female were $0.083\pm0.014\%\;and\;0.108\pm0.018\%$, respectively. In alcohol-fed rats, aldehyde dehydrogenase (ALDH) activity was significantly increased, whereas alcohol dehydrogenase (ADH) activity was not changed. In both Feelch-fed group and Feelch plus alcohol-fed group, ADH and ALDH activity were significantly increased as compared with each control group. Feelch decreased phospholipase $A_2$ activity and lipid peroxidation in hepatic tissue and activities of serum aminotransferases as compared with control. These results suggest that Feelch may have a hepatoprotective effects and this is likely due to lower blood alcohol concentration via the increment of hepatic ADH and ALDH activity.
This study was made to understand a hypoglycemic action of the fat soluble fraction of Panax ginseng C.A. Meyer in streptozotocin induced diabetic rats by determining the activities of several enzymes related to carbohydrate and lipid metabolism as well as several blood component levels such as glucose and ketone bodies, and non-esterified fatty acids. Albino rats (Sprague Dawley, 170-200g, 3) were injected once with 70mg streptozotocinhg body weight intraperitoneally and fed with ordinary diet for 7 days, and then the fat soluble fraction (5 mg~20 mg/day/rat) was injected intraperitoneally once a day for three days to rats having high blood glucose level over 340 mg/100ml. After a final injection of the fat soluble fraction, rats u.ere starved for 16 hours followed by the analysis of blood serum and liver enzymes. It was found that increased levels of glucose, ketone bodies and free fatty acids in streptozotocin induced rats were decreased appreciably by administration of the fat soluble fraction. However, the amount of administered fat soluble fraction did not show any significantly different hypoglycemic action. Decreased activities of glucokinase, phosphofructokinase, pyruvate kinase, 6-phosphogluconate dehydrogenase and acetyl CoA carboxylase of the liver of streptozotocin induced diabetic rats were greatly modified suggesting that a hypoglycemic action of the fat soluble fraction was also appreciable as ginseng saponin fraction. We also compared a hypoglycemic action of the fat soluble fraction prepared from American ginseng and Chinese ginseng with that of Korean pain ginseng. 핀o significant difference of the hypoglycemic activity was observed between the above ginseng fat soluble fractions, suggesting that a study of the fat soluble fraction might be one of the most interesting subjects relating to diabetic hyperglycemia in the near future.
Objectives : This study was designed to protect effect on atherosclerosis through regulation of low density lipoprotein(LDL) by 70 % ethanol extract Ganoderma lucidum (GL) in LDL receptor knockout mouse (LDLr ko mice) fed Western diet.Methods : The LDLr ko mice were divided into 3 groups ; Control, GL100, and GL300. After grouping, LDLr ko mice were fed Western diet. The GL (100 or 300 mg/kg body weight/day, p.o.) was administered every day for 8 weeks. The body weight and food intake were measured every day. The changes in the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), creatinine, triglyceride (TG), total cholesterol (TC), and high-density lipoprotein (HDL) in serum were analyzed after experiment.Results : The LDLr ko mice fed Western diet were increased body weight gain and blood biochemistry parameters such as ALT, AST, TG, TC, and LDL. However GL300 group significantly reduced the body weight. Also TG, TC, and LDL level did not increase. The levels of ALT, AST, HDL were not changed. Also, LDLr ko mice model liver were observed lipid drop, but GL groups did not appear. Futhermore, histological analysis of GL groups aorta tissue were similar to NOR groups.Conclusions : We confirmed that whether GL administration is protect atherosclerosis or not. As the results, blood biochemistry and histological analysis did not changed much in GL administration groups. This study provides scientific evidence that GL protect the atherosclerosis through the reduction of LDL cholesterol. Therefore GL has potential medicine inhibition of atherosclerosis.
실험동물에 있어서 $CCl_4$에 의한 급성 간손상시 피부조직의 oxygen free radical 대사효소 활성 변동을 검토하기 위해 흰쥐에 $CCl_4$와 olive oil의 동량 혼합액을 체중 100g당 0.1 ml씩 복강으로 투여하여 처치하였다. $CCl_4$ 투여로 인한 혈청 alanine aminotransferase 및 xanthine oxidase (XO) 활성은 현저히 증가되었으며 체중당 간무게 (%)및 간조직의 malondialdehyde함량 역시 유의하게 증가되었다. 그리고 병리조직 검사에서도 $CCl_4$투여군에서 간조직의 괴사성 병변이 관찰되었다. 따라서 $CCl_4$를 투여한 실험동물이 급성 간손상 모델로 확인되었다. 이와 같은 간손상 실험동물의 피부조직중 oxygen free radical의 생성 효소인 XO 활성은 대조군과 별다른 차이를 볼 수 없었으나 oxygen free radical의 scavenging 효소인 superoxide dismutase, glutathione peroxidase 및 catalase 활성은 대조군에 비하여 유의하게 감소되었다. 또한 세포화학적 검사에서 cerrous perhydroxide과립이 간손상 실험동물의 피부조직에서 많이 나타났다. 이상 실험결과는 $CCl_4$에 의한 급성 간손상 유도 실험동물의 피부조직에 $H_2O$$_2$의 축적이 나타나는 현상을 시사해주고 있다.
Acetaminophen, which causes acute liver min in humans and animals, has made useful inducer of hepatoxicity for studying hepatopreventive drugs. Injinhotang is known as one of the hepatopreventive drugs. However, its mechanism of recovery of hepatoxicity treated with acetaminophen is poorly understood. this study was performed to observe the antioxidative effect of injinhotang extract and its several combination groups. The results were obtained as follows:1. In the study on free radical scavenging effect in vitro(the suppressing effect on peroxidation of linoleic acid on concentration, the scavenging effect of DPPH radical, inhibitory effect of superoxide in xanthine-xanthine oxidase system and the inhibitory effect on lipid peroxidation reaction by hydroxy radical in H2O2-Fe2+system, injinhotang have more effect than its components groups relatively. 2. In the study on antioxidants system in vivo(the level of serum LPO, the level of hepatic LPO, catalase, GSH, GST), only injnhotang has a significant effect. 3. In the study on hepatotoxicity(GOT, GPT, $\gamma$-GTP, ALP, LDH, b ilirubin), only injinhotang has a significant effect. These results suggest that injinhotang has the protective effect on acetaminophen-induced hepatoxicity. The mechanisms of these are supposed to be involved in antioxidant and three drugs' cooperative synergy effect.
This study was carried out to examine the protective effect of Cultured Wild Ginseng(CWG) on the acute and subacute toxicities induced by doxorubicin(Doxo) in mice. Heart and liver weight was decreased following Doxo administration. In contrast, such a decrease was significantly attenuated by CWG administration. The value of serum CPK in Doxo group was increased compared with normal group. But the value of CWG group were decreased significantly compared with the values of Doxo group in the liver of the Doxo group, cloudy swelling of hepatic cells and narrowing of sinusoids were observed. Whereas in the CWG group, well oriented hepatic cell cords and sinusoids were observed. In the testis of the Doxo group, necrotic and degenerative changes of the seminiferous tubules, especially beneath testicular membrane were observed. But those lesions were alleviated in CWG group. Cross sectional area of testis and diameter of semineferous tubule were significantly increased in CWG group compared with Doxo group. Body weight was reduced in Doxo group compared with normal group. In contrast, such a decrease was significantly attenuated by CWG administration atwa5th day. Spermatogenetic cells in seminiferous tubules were necrotic and desquamated and the cellularity of seminiferous epithelia was reduced in Doxo group. But those lesions were attenuated by CWG administration. Cross sectional area of testis and diameter of seminiferous tubule were significantly increased in CWG group compared with Doxo group. In addition, the increase in lipid peroxidation(LPO) in testis was inaddition, the, iout such a increased was significantly inhibited in CWG group. BrdU labelled cells in the seminiferous tubules were remarkably decreased in Doxo group. Whereas the number of seminiferous tubules labelled with BrdU in spermatogonia was increased by CWG administration. The obtained results suggest that CWG has protective effect on doxorubicin-induced toxicity. This effect might be mediated through the supplementation of vital energy.

Sepsis remains common surgical problems with high morbidity and mortality despite improvement in the management for septic patient. Although hepatocellular dysfunction occurs during sepsis, the mechanism responsible for this remains unclear. In sepsis, a state of severe oxidative stress is encountered, with host endogenous antioxidant defenses overcome. Therefore, the aim of this study was to determine the effect of a-tocopherol (AT) vasoregulatory gene expression during polymicrobial sepsis. Rats were subjected to polymicrobial sepsis by cecal ligation and puncture (CLP). AT (15 mg/kg) was intraperitonealy injected for 3 days prior to CLP. Blood samples were taken 24 h after CLP for measurement of the extent of hepatocellular damage. Liver samples were taken for RT-PCR analysis of mRNA for genes of interest: endothelin-l (ET-l), its receptors $ET_{A}$ and $ET_{B}$, nitric oxide synthases (iNOS and eNOS), cyclooxygenase-2 (COX -2), heme oxygenase-l (HO-l), and tumor necrosis factor-$\alpha$ (TNF-$\alpha$). The activities of serum alanine aminotransferase and lipid peroxidation level were significantly increased; an increase which was prevented by AT pretreatment. CLP significantly increased the mRNA levels of ET-1 and $ET_{B}$; an increase that was prevented by AT pretreatment. However, the level of $ET_{A}$ mRNA significantly decreased after CLP; a decrease that was not prevented by AT pretreatment. There were significant increases in the mRNA expression of iNOS, HO-l and COX -2 in CLP groups. This increase was prevented by AT pretreatment. The expression of eNOS and TNF-$\alpha$ mRNA significantly increased in CLP, which was not prevented by AT pretreatment. Our findings suggest that there was an imbalanced vasoregulatory gene expression in sepsis, and AT ameliorates this change through its free radical scavenging activity.
The present was undertaken to establish a model for the control of adipocytes differentiation by using antibody from egg yolk. The emulsion of membrane protein of 3T3L-1 cell membrane protein with the complete Freund's adjuvant was firstly immunized in layer. Second and third boosting were undertaken with two weeks intervals by injection of the emulsion of the same antigen with the incomplete Freund's adjuvant. After 4 week of the first immunization, eggs were collected and antibody (IgY) was purified from egg yolk. The purity of IgY was 60-98% determined by single radial immunodiffusion (SRID) methods. Titer value of the antibody showed high reactiviy for the preadipocytes membrane protein measured by ELISA. When the IgY was added in the test media containing either 2.5% porcine serum or 10% FBS(control), the differentiation of 3T3L-1 cells and Glycerol-3-phosphate dehydrogenase(GPDH) activities was significantly decreased compared to the control cells(p〈0.05). When mice were subcutaneously injected with IgY raised against membrane protein of 3T3L-1 cells for 3 weeks, adipose tissue mass around ovary was tended to be decreased in female mice compared to those of control mice. It is suggested that a potential for manipulating of lipid accumulation through decrease in 3T3L-1 cell differentiation and fat accumulation in female mice by IgY treatment.
