The Effects of Melatonin on Cisplatin-Induced Renal Cortical Cell Injury in Rabbits

  • Melatonin, a pineal gland hormone, is believed to act as an antioxidant via the stimulation of radical detoxifying enzymes and scavenging of free radicals. In this study, effects of in vitro and in vivo treatments of melatonin on the cisplatin-induced lipid peroxidation, LDH release and plasma creatinine were determined in rabbit renal cortical cells. The level of malondialdehyde (MDA) was assayed as an index of lipid peroxidation and the level of LDH release as an indicator of cellular damage. In in vitro studies, cisplatin increased the levels of MDA and LDH release in a concentration-and time-dependent manner. Melatonin inhibited the cisplatin-induced lipid peroxidation and LDH release in a concentration-dependent manner. The minimal effective concentration of melatonin that significantly reduced the $300\;{\mu}M$ cisplatin-induced lipid peroxidation and LDH release was 1 mM. In in vivo studies, the levels of lipid peroxidation and LDH release in renal cortical cells increased significantly 24 or 48 hours after a single injection of cisplatin (6 mg/kg). When the cisplatin-injected rabbits were pretreated with 10 mg/kg of melatonin, a significant reduction in both lipid peroxidation and LDH release was observed. The plasma creatinine level increased from $0.87{\pm}0.07$ mg/dl in control to $6.33{\pm}0.54$ mg/dl in cisplatin-injected rabbits (P<0.05). Melatonin partially prevented the increase in serum creatinine level $(1.98{\pm}0.11\;mg/dl)$ by cisplatin (P<0.05). In the proximal tubules from cisplatin-treated group, tubular cells had microvilli of variable heights. Necrotic debris was seen in tubular lumens. In most of cells, the mitochondria and lysosomes were increased in frequency. The endocytic vacuoles were not prominent and distribution of the brush border was irregular and shortened. These cisplatin-induced morphological changes were moderate in the melatonin-pretreated group. These results suggest that the toxicity of cisplatin is associated with the generation of reactive oxygen free radicals and that melatonin is a powerful antioxidant, which prevents some of the adverse effects of cisplatin.

(호도약침액)胡桃藥鍼液 독성물질(毒性物質)에 의한 간조직(肝組織) 손상(損傷)에 미치는 영향(影響) (Effect of Juglandis Semen Herbal Acupuncture on Toxic Agent-Induced Liver Cell Damage)

  • Objectives : This study was carried out to determine whether Juglandis Semen herbal acupuncture (JSA) exerts the protective effect against toxic agent-induced live. cell damage. Methods : The cell damage was estimated by measuring lactate dehydrogenase (LDH) release, and lipid peroxidation was estimated by measuring maiondialdehyde (MDA), a product of lipid peroxidation, in rabbit liver slices. Results : When tissues were incubated with 0.5 mM Hg for $10{\sim}120\;min$, LDH release and lipid peroxidation were increased as a function of incubation time, and these effects were significantly prevented by addition of 0.1% JSA. Hg increased LDH release and lipid peroxidation in dose-dependent manner over the range of $0.1{\sim}l\;mM$ concentrations, which were reduced by 0.1% JSA. When tissues were treated with 0.5 mM Hg in the presence of $0.05{\sim}l\;%$ JSA, LDH release and lipid peroxidation induced by Hg were prevented by JSA in a dose-dependent fashion. JSA at 0.5 and 1% prevented completely effects of 0.5 mM Hg. When tissues were treated with 0.5 mM Hg for 60 min, LDH release and lipid peroxidation were increased, which were significantly prevented by addition of 0.1 % JSA. tert-Butyl hydroperoxide (tBHP) increased LDH release and lipid peroxidation, which were significantly reduced by 0.1 % JSA. Such protective effects were similar to those of N,N'-diphenyl-p-phenylenediamine (DPPD), a potent antioxidant. When tissues were treated with 0.5 mM Hg, activities of catalase and glutathione peroxidase were inhibited, and glutathione content was also reduced. Such effects were prevented by JSA, but not by DPPD. JSA prevented Hg-induced morphological changes. Conclusions : These results indicate that JSA exerts the protective effect against liver cell injury induced by toxic agents through antioxidant action, and this effect may be attributed to an increase in activities of endogeous anitoxidant enzymes and GSH content. However, antioxidant effect of JSA is different from that of a well-known potent antioxidant DPPD.

Effect of Conjugated Linoleic Acid on Intestinal and Hepatic Antioxidant Enzyme Activity and Lipid Peroxidation in Broiler Chickens

  • The present study was designed to define whether dietary conjugated linoleic acid (CLA) could affect antioxidant enzymes including superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), catalase (CAT), and glutathione S transferase (GST), and the level of malondialdehyde (MDA), a marker of lipid peroxidation, in the small intestine and liver from broiler chickens. A total of twenty-four 3 wk-old male broiler chickens were assigned to three dietary treatments (1.5% corn oil, 0.75% corn oil plus 0.75% CLA, and 1.5% CLA, isocalorically), and fed a grower-finisher diet from 22 to 35 days. In the small intestinal mucosae, the specific activities of SOD, GSH-Px, CAT, and GST, and the level of MDA were not substantially influenced by dietary CLA. In the liver, the specific activities of SOD, GSH-Px, and GST, and the level of MDA were also unaffected by dietary CLA at the level of either 0.75% or 1.5% compared with corn oil at the level of 1.5%. However, the broiler chickens fed the diet containing 1.5% CLA resulted in a significant increase in peroxisomal CAT activity and a marked decrease in total lipid and non-esterified fatty acids (NEFA) from liver tissues compared with those fed the diet containing 1.5% corn oil. In conclusion, ability of CLA to increase hepatic CAT activity suggest that dietary CLA may affect, at least in part, antioxidant defense system as well as lipid metabolism in the liver of broiler chickens.

Effects of Arsenic (AsIII) on Lipid Peroxidation, Glutathione Content and Antioxidant Enzymes in Growing Pigs

  • This experiment was conducted to investigate the effect of arsenic ($As^{III}$) on lipid peroxidation, glutathione content and antioxidant enzymes in growing pigs. Ninety-six Duroc-Landrace-Yorkshire crossbred growing pigs (48 barrows and 48 gilts, respectively) were randomly assigned to four groups and each group was randomly assigned to three pens (four barrows and four gilts). The four groups received the same corn-soybean basal diet which was supplemented with 0, 10, 20, 30 mg/kg As respectively. Arsenic was added to the diet in the form of $As_2O_3$. The experiment lasted for seventy-eight days after a seven-day adaptation period. Malondialdehyde (MDA) levels, glutathione (GSH) contents and superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione reductase (GR) and glutathione-S-transferase (GST) activities were analyzed in serum, livers and kidneys of pigs. The results showed that pigs treated with 30 mg As/kg diet had a decreased average daily gain (ADG) (p<0.05) and an increased feed/gain ratio (F/G) (p<0.05) compared to the controls. The levels of MDA significantly increased (p<0.05), and the contents of GSH and the activities of SOD, CAT, GPx, GR and GST significantly decreased (p<0.05) in the pigs fed 30 mg As/kg diet. The results indicated that the mechanism of arsenic-induced oxidative stress in growing pigs involved lipid peroxidation, depletion of glutathione and decreased activities of some enzymes, such as SOD, CAT, GPx, GR and GST, which are associated with free radical metabolism.

함초 보충시 당뇨유발 흰쥐의 지질과산화와 무기질 함량에 미치는 영향 (Effects of Salicornia herbacea L. Supplementation on Lipid Peroxidation and Mineral Levels in Streptozotocin-Induced Diabetic Rats)

  • This study was designed to examine the effects of Salicornia herbacea L. (glasswort: GW) on the lipid peroxidation and mineral levels in diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats weighing 200-220 g by an injection of streptozotocin (STZ) dissolved in a citrate buffer into the tail vein at a dose of 45 mg/kg of body weight. Sprague-Dawley rats were fed an AIN-93 recommended diet and the experimental groups were fed a modified diet containing 10% and 20% of glasswort powder for 4 weeks. The experimental groups were divided into 6 groups which consisted of normal (N)-control group, N-GW 10% and N-GW 20% treated groups, STZ-control, STZ-GW 10% and STZ-GW 20% treated groups. The rats' liver and muscle glycogen, liver and kidney protein, cholesterol and triglyceride (TG) in liver, malondialdehyde (MDA) in liver and kidney values were measured, along with the hepatic of chromium (Cr), iron (Fe), and zinc (Zn) content. The liver glycogen levels was significantly affected in N-GW 20% group among all the experimental groups. The liver MDA levels of the STZ-GW 10% and STZ-GW 20% groups were significantly lower than for the STZ-control group. There were significant differences between the N-control group and the STZ-control group in the hepatic of Zn levels. The hepatic of Cr levels in the N-GW 20% and STZ-GW 10% and STZ-GW 20% groups were significanly higher than for the each control groups. These results exhibited dose related effect of glasswort and it may have favorable influence on lipid peroxidation in the liver.

루테인과 후코이단 병용 처리에 의한 AAPH 유도 세포 손상 억제 (Protective Effect of Co-treatment of Lutein and Fucoidan Against AAPH-Induced Damage in THP-1 Cells)

  • This study was designed to investigate the protective effect of the combination of fucoidan and lutein against AAPH-induced oxidative stress in THP-1 cells. The combination of fucoidan and lutein existed significant antioxidant effect on AAPH-damaged THP-1 cells by using lipid peroxidation and cellular antioxidant capacity assay. Fucoidan($1\;{\mu}g/m{\ell}$) and lutein($10\;{\mu}g/m{\ell}$) did not affect at all the viability of THP-1 cells, but protected the AAPH-damage of THP-1 cells at the same concentration. The viability of THP-1 cells was 0% with 1 mM AAPH alone, the protective effect of fucoidan($1\;{\mu}g/m{\ell}$) and lutein($10\;{\mu}g/m{\ell}$) was 37% and 36%, respectively. The combination of fucoidan($1\;{\mu}g/m{\ell}$) and lutein($10\;{\mu}g/m{\ell}$) exhibited significant inhibitory effect of lipid peroxidation using TBARS assay and cellular antioxidant capacity using DCFH-DA assay. In lipid peroxidation, the TBARS value of 1 mM AAPH alone was $0.8{\pm}0.03\;nM$ MDA, its of the combination of fucoidan($1\;{\mu}g/m{\ell}$) and lutein($10\;{\mu}g/m{\ell}$) was $0.2{\pm}0.05\;nM$ MDA. In cellular antioxidant capacity, the combination of fucoidan($1\;{\mu}g/m{\ell}$) and lutein($10\;{\mu}g/m{\ell}$) exhibited significant cellular antioxidant capacity of 76%, whereas quercetin($10\;{\mu}M$) as positive control exhibited the cellular antioxidant capacity of 32%. These results indicate that the cotreatment of fucoidan and lutein protects against AAPH-induced THP-1 cell damage by inhibiting lipid peroxidation, increasing cellular antioxidant capacity.

동과인의 보충이 당뇨 유발 흰쥐의 글리코겐과 지질과산화에 미치는 영향 (Effects of Benincasa Hispida Seed Supplementation on Glyeogen status and Lipid Peroxidatin in Streptozotecin-Induced Diabetic Rats)

  • This study was designed to examine the effects of diets containing different levels of seeds of Benincasa hispida(wax gourd) on glycogen, protein levels and lipid profiles as well as malondialdehyde (MDA) in streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley rats were induced diabetes mellitus by STZ injection (45 mg/kg) into the tail vein and were divided into four groups: normal, STZ-control, and two experimental groups. Normal and STZ-control groups were fed the AIN-93 diet and the two experimental groups were fed a modified diet containing 2.5% and 5.0% of wax gourd seed powder for four weeks. The liver, muscle, lung, kidney, and pancreas were excised after sacrifice, then the glycogen, protein, and lipid peroxidation products were measured. The rats fed 2.5% wax gourd seed group showed higher levels of liver glycogen compared with that of the STZ-control group. The levels of kidney protein were significantly increased in the 2.5% and 5.0% wax gourd seed groups. There were no significant difference cholesterol and liver triglyceride levels of the liver and MDA concentration in the liver, lung, and kidney among all four groups. These results show that wax gourd seed treatment of 2.5% and 5.0% doses did not exhibit profound anti-lipid peroxidation properities.

Interactions between Oxidative Stress, Lipid Profile and Antioxidants in Breast Cancer: A Case Control Study

  • Oxidant/antioxidant balance has been suggested as an important factor for initiation and progression of cancer. The objective of this study was to determine changes in the levels of malondialdehyde (MDA), nitric oxide (NO), total cholesterol, triglycerides, LDL-cholesterol, HDL-cholesterol, total antioxidant capacity (TAC), glutathione peroxidase (GSH-Px), and superoxide dismutase (SOD) activities in serum samples of breast cancer patients (n=30) and healthy subjects (n=100). MDA and NO levels were found to be increased in breast cancer patients compared to the healthy subject group (p<0.05). Total cholesterol and triglycerides were elevated; and HDL-cholesterol level was found to be decreased in the cancer patients as compared to the healthy subjects (p<0.05). Compared to the healthy group, both serum TAC levels (p<0.001) and activity of SOD and GSH-Px (p=0.05) were found to be decreased in the breast cancer patients as compared to the healthy controls. Considering the data presented in this study, we suggest that free radicals induce lipid eroxidation and peroxidation of unsaturated fatty acid with decreased activity of enzymatic antioxidants in breast cancer.

The Inhibitory Effects of the Methanolic Fraction of Pueraria Radix on Hydrogen peroxide-induced Lipid peroxidation and Cadmium-induced cytotoxicity

  • The effects of the methanol subfraction of Pueraria radix on hydrogen peroxide-induced lipid peroxidation and Cd-induced cytotoxicity were investigated in NIH3T3 fibroblasts. After the methanol subfraction treatment, the content of MDA induced by 600${\mu}$M H2O2 significantly decreased in proportion to the subfraction concentrations as well as 50${\mu}$M CdCl2-induced cytotoxicity. Especially, 200$\mu\textrm{g}$/mg concentration of methanol subfraction was strongly shown inhibition of lipid peroxidation and detoxification of Cd. These results suggest that the methanol subfraction of Pueraria radix retains a potential antioxidant and protective effect against cadmium.

콜레스테롤 및 타우린 첨가식이가 흰쥐 혈장과 간의 지질과산화물 농도와 항산화효소 활성에 미치는 영향 (Changes in Lipid Peroxidation Level and Antioxidant Enzyme Activities of Rats Supplemented with Dietary Cholesterol and/or Taurine)

  • 본 논문은 타우린 및 콜레스테롤이 체내 항산화체계 및 지질과산화물농도에 미치는 영향을 평가하기 위해 흰쥐를 대상으로 콜레스테롤(1.5%) 및 타우린(1.5%)이 첨가된 식이로 5주간 사육하였으며, 그 결과를 요약하면 다음과 같다. 첫 번째로 혈장의 MDA 농도는 콜레스테롤 또는 타우린 첨가에 의해 유의한 영향을 받지 않은 반면, 간조직의 MDA 농도는 HCD군에서 CD군에 비해 70% 유의하게 증가하였다(p<0.05). 고콜레스테롤식이에 타우린을 첨가해 준 결과 간조직의 지질과산화물 농도가 HCD군에 비해 유의하게 감소한 반면(p<0.05),콜레스테롤을 섭취하지 않는 상태에서 타우린을 첨가한 HTD군의 경우에는 간조직 MDA 농도가 대조군(CD)과 유의적인 차이를 나타내지 않아 식이내 타우린 첨가가 간의 지질과산화물 농도에 미치는 영향은 콜레스테롤 섭취수준에 의해 그 결과가 상이하게 나타남을 알 수 있었다. 두 번째, 혈장 및 간조직 의 GSH-Px 활성 은 식이내 콜레스테롤 및 타우린 첨가에 의해 유의한 영향을 받지 않았다. 간조직의 총SOD활성은 타우린 첨가에 의해 감소되었으며 (p<0.05), HTD군에서 CD군에 비해 유의하게 감소하였고 (p<0.05), HCHTD군의 경우에는 HCD군에 비해 감소하는 경향을 보였으나, 통계적 유의성은 나타나지 않았다. 세 번째, 혈장 및 간조직의 지질과산화물농도, 항산화효소 활성, 콜레스테롤 및 타우린농도와의 상관관계를 평가한 결과, 혈장 총콜레스테롤농도는 간조직의 콜레스테롤농도와 매우 유의한 양의 상관관계를 보였다(r=0.712 p<0.001). 혈장(r=0.399, p < 0.05) 또는 간조직의 콜레스테롤농도(r=0.429, p <0.05)는 간조직의 MDA농도와 유의한 양의 상관관계를 나타냈다. 혈장 타우린농도는 간조직의 GSH-Px 활성과는 유의하지는 않지만 음의 상관관계(r=-0.188, p > 0.05)를, 그리고 간조직의 SOD활성과는 유의적인 음의 상관관계(r=-0.481,p < 0.01)를 나타냈다. 한편, 간조직의 총 SOD 활성은 간조직의 GSH-Px 활성과 유의한 양의 상관관계 (r=0.554, p<0.001)를 나타내, 두가지 항산화효소의 활성이 서로 밀접하게 연관되어 있음을 알 수 있었다. 콜레스테롤이 산화촉진제로 작용하여 조직의 지질과산화가 촉진된 상황에서 타우린의 보충섭취가 간조직의 SOD 활성을 감소시키고 MDA 수준을 유의하게 감소시킨 본 연구의 결과로 미루어 볼 때, 산화적 손상을 초래하지 않는 안전한 혈중 지질저하제로서 타우린의 이용 가능성이 전망된다.
