Potential in vitro Protective Effect of Quercetin, Catechin, Caffeic Acid and Phytic Acid against Ethanol-Induced Oxidative Stress in SK-Hep-1 Cells

  • Phytochemicals have been known to exhibit potent antioxidant activity. This study examined cytoprotective effects of phytochemicals including quercetin, catechin, caffeic acid, and phytic acid against oxidative damage in SK-Hep-1 cells induced by the oxidative and non-oxidative metabolism of ethanol. Exposure of the cells to excess ethanol resulted in a significant increase in cytotoxicity, reactive oxygen species (ROS) production, lipid hydroperoxide (LPO), and antioxidant enzyme activity. Excess ethanol also caused a reduction in mitochondrial membrane potential (MMP) and the quantity of reduced glutathione (GSH). Co-treatment of cells with ethanol and quercetin, catechin, caffeic acid and phytic acid significantly inhibited oxidative ethanol metabolism-induced cytotoxicity by blocking ROS production. When the cells were treated with ethanol after pretreatment of 4-methylpyrazole (4-MP), increased cytotoxicity, ROS production, antioxidant enzyme activity, and loss of MMP were observed. The addition of quercetin, catechin, caffeic acid and phytic acid to these cells showed suppression of non-oxidative ethanol metabolism-induced cytotoxicity, similar to oxidative ethanol metabolism. These results suggest that quercetin, catechin, caffeic acid and phytic acid have protective effects against ethanol metabolism-induced oxidative insult in SK-Hep-1 cells by blocking ROS production and elevating antioxidant potentials.

새로운 항산화제 검색법에 의한 SOD Mimic 천연 약물의 개발-상백피의 항염증효과 (Development of the SOD Mimics from the Natural Product by a Novel Biosystem-Antiinflammatory Effect of Morus alba)

  • Aqueous extract of Morus alba L. blocked the toxic effect of paraquat on E. coli growth. The active components in the extract may be capable of crossing the cell membranes and protect against superoxide toxicity in E. coli, The extract inhibited $FeSO_4/H_2O_2$ induced lipid peroxidation in rat liver homogenate and protected against t-butyl hydroperoxide caused Ac2F cell damage. Moreover, the extract showed inhibitory effect on phospholipase $A_2$ activity in a dose dependent manner. Antiinflammatory effect of the extract was further investigated using the carrageenin-induced oedema model. A single adminstration of the extract (3g/kg body, p.o.) was more effective than indomethacin. These results suggest that the isolation and identification of the active components would have significant therapeutic application to inflammation associated with oxygen radicals.

카페인 첨가가 흰쥐 간의 항산화 활성에 미치는 영향 (The Effect of Caffeine on the Antioxidative Activities of Mouse Liver)

  • The purpose of this study was to examine the antioxidative activities and tissue cell of mouse liver added caffeine. The body weight of all experimental groups increased during experimental periods, but the body weight of caffeine-containing groups were lower than those of any other experimental groups. Superoxide dismutase and catalase activities tended to decrease significantly with caffeine-containing groups, but increased in control and ginseng-containing groups. Hydroperoxide contents were increase significantly with caffeine-containing groups. Lipid peroxidation levels decreased in ginseng-containing groups, but it increased significantly with caffeine-containing group. Protein contents were a tendency of similar between control and ginseng-containing groups, but it showed a increasing tendency in caffeine-containing groups. Microscopic observation of mouse liver cell were similar tissue in ginseng and caffeine-containing groups, but it showed somewhat more injuring only at the liver cell of anhydrous caffeine group, and became the suspicion in liver diseases. This results show that antioxidative activities are slightly higher in non-caffeine and ginseng-containing drinks than caffeine-cotaining drinks. From this standpoint, we suggest that too much drinking of caffeine-containing drinks for a long time is undesirable.

In vitro screening of anti-skin aging and antioxidant properties of aqueous/solvent extracts from distinctive stages of silkworm (Bombyx mori L.) pupae

  • Silkworm pupae, a by-product of the silk industry are known to be valuable resource of nutrients for humans as well as animals besides encompassing diverse bioactive constituents. However, there is a paucity of knowledge on their role in amelioration of oxidative stress and anti-skin aging properties. In the present study, we evaluated the inhibitory effect of aqueous as well as ethanolic (30% and 70%) extracts from distinct stages of male and female silkworm pupae belonging to two silkworm varieties on skin aging-related enzymes. The activities of collagenase, elastase and tyrosinase were effectively inhibited by 70% ethanolic silkworm pupal extracts (SPE), followed by 30% with aqueous extracts exhibiting meager inhibitory potential. SPE were also investigated for their antioxidant activity in oxidative-stressed murine fibroblasts (L929). The intracellular ROS and lipid peroxidation induced by tert-butyl hydroperoxide (t-BHP) in fibroblasts was better attenuated by pre treatment with ethanolic (30%) and aqueous extracts, respectively. The safety of the extracts was determined by studying their effect on fibroblast cell viability and it was found that none of the extracts were cytotoxic. Our findings indicate the potential utility of SPE as anti-aging components in cosmeceuticals.

실크 단백질 가수분해물의 간 손상에 대한 보호효과 (Protective effect of silk protein hydrolysates against tert-BHP induced liver damage)

  • 동물실험에서 실크단백질 산 가수분해물을 투여하고 t-BHP투여한 군의 혈액 생화학적 검사 결과 t-BHP만 투여한 군과 비교하였을 때 AST, ALT 그리고 LDH가 실크단백질 산 가수분해물의 투여 농도가 높아질수록 수치가 감소하는 것으로 나타났고 세포가 손상할 시에 증가하는 MDA를 간 조직을 대상으로 측정한 결과 실크단백질 산 가수분해물의 농도가 높아질수록 수치가 대조군과 유사한 정도로 감소하는 것으로 보아 간 손상에 관여하는 효소의 누출 억제효과가 있는 것으로 사료된다. HPLC로 간 조직에서의 GSH 측정결과 t-BHP만 투여한 군과 비교하였을 때 유의적으로 증가하였고 조직학적 검사 결과 t-BHP만 투여한 군과 비교하였을 때 실크단백질 산 가수분해물을 투여한 군이 대조군과 가까운 모습을 보이는 것으로 관찰되어 실크단백질 산 가수분해물이 산화적 스트레스로부터의 간 보호 효과가 있는 것으로 사료된다. 따라서 실크단백질 산 가수분해물의 기능적 소재로서의 이용가능성이 확대될 것으로 사료된다.

고도불포화지방산 함량이 높은 유지를 섭취시킨 흰쥐에서 양조간장과 멜라노이딘의 지질산화 억제효과 (Protective Effect of Soybean Sauce and Melanoidin on Lipid Oxidation in Rats Fed High PUFA Oils)

  • In vitro와 in vivo에서 양조간장과 glucose-lysine 모델 멜라노이딘의 항산화효과를 조사하여 다음과 같은 결과를 얻었다. In vitro에서 양조간장과 멜라노이딘의 항산화효과를 linoleic acid emulsion계를 이용하여 비교하였을 때 양조간장과 멜라노이딘은 대조군에 비해 우수한 항산화활성을 나타내었고 특히 양조간장의 효과가 더 우수한 것으로 나타났다. DPPH 라디칼 소거활성 역시 멜라노이딘에 비해 양조간장의 효과가 큰 것으로 나타났다. 지방산 조성이 상이한 옥수수기름과 어유를 섭취시킨 흰쥐에 10% 양조간장 및 멜라노이딘을 5주간 섭취시켰을 때 간의 지방산 조성은 섭취 지방산에 따라 차이가 뚜렷하였으나 양조간장과 멜라노이딘의 효과는 대조군과 유사하였다. 지질 과산화물 생성정도를 TBARS로 측정하였을 때 불포화지방산 함량이 높은 어유 섭취군에서 지질과산화가 증가하였으며 간장과 멜라노이딘은 지질과산화를 억제하는 효과를 나타내었다. CL(chemilurninescence)-HPLC 방법을 이용하여 간과 혈장의 막 인지질 PCOOH 함량을 측정한 결과, 어유의 섭취는 옥수수기름 섭취 군에 비해 지질 과산화를 월등히 증가시켰고 양조간장과 멜라노이딘의 섭취는 유의적으로 지질 과산화를 억제한 것으로 나타났다. 특히 양조간장의 효과가 멜라노이딘 보다 우수하여 이는 콩유래 항산화물질인 수용성 펩티드나 저분자 단백질들이 효과적으로 항산화활성을 나타낸 것으로 여겨지며, 불포화지방산이 높은 식품의 섭취시 간장을 소스로 이용하는 것이 지질의 과산화를 억제할 것으로 기대되어진다.

진두발, 김 및 굴의 지질에 있어서 광증감 산화에 관한 연구 (Studies on Photosensitized Oxidation in the Lipids of Irish moss, Laver and Oyster)

  • 김, 미역, 다시마, 톳 등의 해조류는 chlorophyll을 다량 함유하고 있고 굴은 소화관 내에는 미분해의 chlorophyll이 축적되어 있다. chlorophyll은 일중항 산소에 의한 광증감 산화를 촉매 한다는 것이 알려져 있다. 따라서 홍조류인 김과 진두발의 천일건조 및 굴의 자외선 조사시 광증감 산화가 지질산화에 어느정도 기여하는가를 밝히기 위해 광증감 산화 시킨시료의 총 지질의 과산화물가를 비교, 검토 하였고 과산화물의 위치이성체의 조성을 측정하였다. 굴의 총 지질 함량은 $2.7\%$였고, 또한 진두 발과 김의 총 지질 함량은 각각 $0.1\%$였다. 굴의 과산화물가는 생시료의 경우 56.7 meq/kg 이던 것이 4시간동안 자외선 조사 후에는 100.9 meq/kg으로 증가하였다. 진두발의 경우 원조의 140.2 meq/kg에서 3일간 천일건조후에는 213.4 meq/kg으로 증가 하였고, 김은 원조의 63.3 meq/kg에서 3일간 천일건조후에는 118.8 meq/kg으로 증가하여 산화가 진행되었음을 예측할수 있었다. 각 시료의 광증감 산화에 의해 생성한 과산화물의 위치 이성체의 조성을 GC-MS로 측정 한 결과 18:1 (n-9)와 18:2 (n-6)로부터 생성된 것으로 추정되는 위치 이성체를 분리하였다. 또한 9-OOH와 13-OOH의 비율이 비교적 높았으나 18:1(n-9)와 18:2(n-6)이외의 지방산으로부터 생성될 것으로 추정되는 위치이성체는 분리 확인할수 없었다.

랫드에서의 t-BHP 유발 산화스트레스에 대한 기능성 들깻잎 열수 추출물의 간 보호 효과 (Protective Effect of Functional Perilla frutescens Hot-water Extract Against tert-butyl hydroperoxide-Induced Liver Oxidative Damage in Rats)

  • t-BHP는 대표적인 산화스트레스에 의한 간 손상 모델이며, 본 연구자는 랫드에 기능성이 강화된 기능성 들깻잎 추출물(P. frutescens leaf extract, PLE)을 250, 500 또는 1000 mg/kg body weight (b.w.)으로 6일간 경구 투여하고 t-BHP의 복강 주사로 간 손상을 일으킨 후 기능성 들깻잎 추출물이 간에서 산화스트레스를 얼마나 억제하여 주는지를 혈액의 간 손상 지표인 lactate dehydrogenase (LDH), aspartate aminotransferase (AST) 그리고 alanine aminotransferase (ALT)를 측정하였으며, 간 조직에서 항산화 바이오 마커인 reduced glutathione (GSH), 지질과산화물의 척도인 malondialdehyde (MDA)를 통해서 측정하였다. 산화스트레스에 의한 간 손상시 GSH는 아무것도 처리하지 않은 정상 대조군($146.0{\pm}5.4$ mM GSH/g protein)에 비해 t-BHP로 산화스트레스를 유발한 그룹에서 $128.6{\pm}6.8$ mM GSH/g protein로 감소하였다. 반면 기능성 들깻잎 추출물을 250, 500 그리고 1000 mg/kg b.w. 을 투여한 그룹에서는 $129.3{\pm}2.6$ mM GSH/g protein, $151.9{\pm}6.8$ mM GSH/g protein, $171.9{\pm}5.2$ mM GSH/g protein로 농도 의존적으로 GSH 함량이 회복되는 경향을 나타내며, 500 mg/kg b.w. 투여군부터 정상 대조군의 GSH 함량과 비슷한 수준을 나타내었다. 또한, 간 조직에서 산화스트레스에 의하여 발생된 지질과산화물을 측정하였을 때 t-BHP에 의하여 산화스트레스만 유발한 그룹은 $834.0{\pm}154.7{\mu}M/g$ protein 로 정상 대조군의 $385.6{\pm}39.7{\mu}M/g$ protein 보다 2.17배 높은 MDA를 생성한 것으로 지질과산화가 많이 일어난 것을 확인하였으나, 기능성 들깻잎 추출물을 투여한 그룹의 MDA의 생성량은 $669.2{\pm}145.0$, $595.1{\pm}142.6$, $415.9{\pm}133.8{\mu}M/g$ protein 로 농도에 따라 감소하는 경향을 확인하였다. 랫드의 간 조직에서, 독성을 유발하는 t-BHP를 복강 투여 후 기능성 들깻잎 추출물을 경구 투여하였을 때 지질과산화의 지표인 MDA의 감소와 항산화의 지표인 GSH 함량이 증가하였다. 조직병리학적 확인을 위하여 간 조직을 H&E 염색 후 광학현미경으로 관찰하였을 때, t-BHP만 처리한 음성 대조군의 경우 간 세포의 괴사와 조직의 변형을 확인할 수 있었지만 기능성 들깻잎 추출물을 처리하였을 경우 모두 정상 상태의 조직을 관찰할 수 있었다. 위의 결과들을 종합하였을 때, t-BHP가 간에서 산화스트레스를 유발하여 간 손상을 야기시키고, 간 조직의 인지질 막 손상을 줄 수 있으며, 기능성 들깻잎 추출물은 간 손상에 대한 보호 효과가 있음을 확인하였다.

튀김유의 재사용에 의한 품질 특성: 전통 튀김기와 수유식 개량 튀김기의 비교 (Physicochemical Analysis in the Reuse of Deep-Frying Oil: Comparison of Traditional Fryer and Modified Fryer)

  • The property of deep frying oil is one of the important factors in fried food quality. The purpose of this study is to identify the quality of deep frying oil in continuous usages for 4 days by two types of fryers: traditional deep-fat fryer and modified oil-water fryer. After frying pork cutlets, the frying oil was kept not only for several physical analyses such as color, viscosity, and water content but also for the quality analyses of frying oil, such as free fatty acid, double bond changes and oxidative rancidity formation. The oil fried in a traditional deep-fat fryer was significantly increased in terms of physical values including moisture content, viscosity, and color, compared to those of the modified fryer, continuously for 4 days. The oil fried by a traditional deep-fat fryer exhibited a significant increase in its free fatty acid content compared to that fried by a modified oil-water fryer, while the iodine value was significantly decreased in the oil fried by a traditional deep-fat fryer when compared to control oil and oil fried by the modified oil-water fryer. In the peroxide value as an indicator of primary oxidation products, the oil fried by both fryers was significantly increased till the second day but decreased in value after the third day because of unstable hydroperoxide decomposition. The p-anicidine value is used as an indicator of secondary oxidation products, the oil fried in a traditional deep-fat fryer was significantly increased in value compared to that of a modified oil-water fryer.

Quercetin Prevents Hydrogen Peroxide-induced Necrotic and Apoptotic Cell Death in Human Colonic Epithelial Cells

  • Quercetin is one of the most distributed flavonoids in the plant kingdom and occurs naturally in a wide range of fruits and vegetables. This study was undertaken to determine whether quercetin exerts beneficial effect against necrotic and apoptotic cell death induced by hydrogen peroxide ($H_2O2$) in intestinal cells using the human-derived cultured T84 colonic epithelial cell line. Necrotic cell death was induced by exposing cells to 0.5 mM $H_2O_2$ for 2 h and apoptosis was induced by incubating cells in normal culture medium for 18 h following exposure of cells to 0.5 mM $H_2O2$ for 2 h. Cell viability was evaluated by the trypan blue exclusion assay and apoptosis was assessed by Hoechst 33258 staining and flow cytometry. $H_2O_2$ induced necrotic cell death in a time and dose-dependent fashion. Both necrotic and apoptotic cell deaths were not prevented by the antioxidants N,N'-diphenyl-p-phenylenediamine(DPPD) and Trolox, whereas both cell deaths induced by the organic hydroperoxide t-butylhydroperoxide (tBHP) were prevented by DPPD, suggesting that $H_2O_2$ induces cell death through a lipid peroxidation-independent mechanism. $H_2O2$-induced necrotic death was prevented by deferoxamine and 3-aminobenzamide, while the apoptotic cell death was not affected by these agents. Quercetin prevented both necrotic and apoptotic cell deaths induced by $H_2O_2$ in a dose-dependent manner. $H_2O_2$ caused activation of poly (ADP-ribose) polmerase (PARP), which was inhibited by deferoxamine, 3-aminobenzamide, and quercetin, but not DPPD. These results indicate that quercetin inhibits both necroticand apoptotic deaths of T84 cells. The anti-necrotic effect of quercetin may be attributed to its iron chelator activity rather than a direct $H_2O_2$ scavenging capacity and antioxidant. The present study suggests that quercetin may play a therapeutic role in the treatment of human gastrointestinal diseases mediated by oxidants.
