Development of an anti-obesity dietary supplement inhibiting the digestion of carbohydrate and lipid (비만 개선 효과를 지닌 탄수화물 및 지방 흡수 억제 기능성 식이조성물 개발)

  • In a previous study, we developed a new food additive as an egg yolk antibody (IgY) against carbohydrate digestion enzymer for the regulation of blood glucose level and weight control. The IgY delayed and decreased the increment if blood glucose level after administration of sucrose in human being by 30% in 20∼30 min. We also developed a lipase inhibitor as a water extract of two kinds of herb, Platycodon grandiflorum and Solanum Melongena, Twenty three volunteers were subjected to the intake of the egg yolk IgY Plus the herbal extracts for 50 days. In average, the treated subjects appeared to lose 1.96 kg of body weight and 3.4 kg of body fat mass during the treated period. Furthermore, Panniculus adiposus and breech size were significantly decreased during the experimental period. Above results suggested that the administration of the dietary additives composed of egg yolk IgY and natural herbal extract improve the obesity by the decrement of body weight and body fat mass.

Protective Effects of In Vitro Gastrointestinal Digests of Abalone (Haliotis discus hannai) Intestines against Oxidative Stress in RAW264.7 Macrophage Cells

  • Abalone (Haliotis discus hannai), mostly distributed and maricultured in southwestern coastal areas of South Korea, is recognized as an economically important species in the fishery industry. Abalone intestines are one of the by-products of abalone processing. To investigate abalone intestines as bioactive substances, abalone intestine gastrointestinal digests (AIGIDs) of various molecular weights (MWs) were prepared using in vitro gastrointestinal digestion and an ultrafiltration system, and tested for inhibitory effects against reactive oxygen species (ROS) and oxidative stress in macrophage cells treated with hydrogen peroxide ($H_2O_2$). In our results, among AIGIDs, AIGID-III (MW=5-10 kDa) showed potent inhibitory activities for lipid peroxidation and free radicals. Additionally, the results clearly indicated that AIGID-III treatment could prevent cytotoxic damage of macrophages by $H_2O_2$-induced oxidative stress due to its potent scavenging ability against cellular ROS. These results suggest that AIGIDs may have protective and therapeutic potential for oxidative stress syndromes and immune diseases through ROS inhibition in macrophage cells.

Effect of Lipid Sources with Different Fatty Acid Profiles on Intake, Nutrient Digestion and Ruminal Fermentation of Feedlot Nellore Steers

  • The present study was conducted to determine the effect of lipid sources with different fatty acid profiles on nutrient digestion and ruminal fermentation. Ten rumen and duodenal fistulated Nellore steers (268 body weight${\pm}27kg$) were distributed in a duplicated $5{\times}5$ Latin square. Dietary treatments were as follows: without fat (WF), palm oil (PO), linseed oil (LO), protected fat (PF; Lactoplus), and whole soybeans (WS). The roughage feed was corn silage (600 g/kg on a dry matter [DM] basis) plus concentrate (400 g/kg on a DM basis). The higher intake of DM and organic matter (OM) (p<0.001) was found in animals on the diet with PF and WF (around 4.38 and 4.20 kg/d, respectively). Treatments with PO and LO decreased by around 10% the total digestibility of DM and OM (p<0.05). The addition of LO decreased by around 22.3% the neutral detergent fiber digestibility (p = 0.047) compared with other diets. The higher microbial protein synthesis was found in animals on the diet with LO and WS (33 g N/kg OM apparently digested in the rumen; p = 0.040). The highest C18:0 and linolenic acid intakes occurred in animals fed LO (p<0.001), and the highest intake of oleic (p = 0.002) and C16 acids (p = 0.022) occurred with the diets with LO and PF. Diet with PF decreased biohydrogenation extent (p = 0.05) of C18:1 n9,c, C18:2 n6,c, and total unsaturated fatty acids (UFA; around 20%, 7%, and 13%, respectively). The diet with PF and WF increased the concentration of $NH_3-N$ (p<0.001); however, the diet did not change volatile fatty acids (p>0.05), such as the molar percentage of acetate, propionate, butyrate and the acetate:propionate ratio. Treatments PO, LO and with WS decreased by around 50% the concentration of protozoa (p<0.001). Diets with some type of protection (PF and WS) decreased the effects of lipid on ruminal fermentation and presented similar outflow of benefit UFA as LO.

Influence of Food Wastewater Loading Rate on the Reactor Performance and Stability in the Thermophilic Aerobic Process (음폐수 부하량에 따른 고온호기성 공정의 처리 양상)

  • In this study, the feasibility of a single-stage thermophilic aerobic process for the treatment of high-strength food wastewater produced from the recycling process of food wastes was examined to substitute anaerobic digestion process. Also, the removal and stability of thermophilic aerobic process were assessed according to the changes of hydraulic retention times (HRTs) and organic loading rates (OLRs). When the OLR increased from 9.2 to $37.2kgCOD/m^3d$, a pH value in R1 (HRT : 5 d) significantly decreased to 5.0, due to the organic acid accumulation. On the other hand, the pH value in R2 (HRT : 10 d) was stable and R2 showed the high removal of COD, organic acid and lipid, even though the OLR increased from 4.6 to $18.6kgCOD/m^3d$. In R1, the COD loading rates for COD removal was suddenly dropped, as the COD loading rate increased from 18.6 to $28.4kgCOD/m^3d$. In contrast, R2 showed that the COD loading rates for COD removal increased with regard to increment in the loading rates of 3.61, 7.05, 9.43 and $12.2kgCOD/m^3d$, indicative of the high COD removal efficiency. Therefore, the results demonstrated that over 10-d HRT, the high concentration of raw food wastewater was efficiently treated in the single-stage thermophilic aerobic process.

Ultrastructure of Oocytes During Oogenesis and Oocyte Degeneration Associated with Follicle Cells in Female Sinonovacula constricta(BIVALVIA: PHARIDAE) in Western Korea

  • The ultrastructure of oocytes during oogenesis and oocyte degeneration associated with follicle cells in female Sinonovacula constricta(Lamarck, 1818) were investigated by electron microscope observations. Ovarian follicles are surrounded by a matrix of vesicular connective tissue cells(VCT cells). VCT cells contain large quantities of glycogen particles and several lipid droplets in their cytoplasm. It is suggested that VCT cells act as a source of nutrients for vitellogenesis during oogenesis. In early vitellogenic oocytes, several coated vesicles, which appear at the basal region of the oocyte, lead to the formation of membrane-bound vesicles via endocytosis. The uptake of nutritive materials in coated vesicles formed by endocytosis appears through the formation of coated pits on the oolemma during vitellogenesis. During the late stage of oogenesis, yolk precursors(yolk granules), mitochondria and lipid droplets are present in the cytoplasm of late vitellogenic oocytes. In particular, proteinaceous yolk granules containing several different components are intermingles and form immature yolk granules. In the mature oocyte, small immature yolk granules are intermingled and form large mature yolk granules. Vitellogenesis occurs through a process of autosynthesis, involving combined activity of the Golgi complex, mitochondria and rough endoplasmic reticulum in the cytoplasm of vitellogenic oocytes. The process of heterosynthesis is where extraovarian precursors are incorporated into oocytes by endocytosis at the basal region of early vitellogenic oocytes before the formation of the vitelline coat. Follicle cells appear to play an important role in vitellogenesis and oocyte degeneration. The functions of attached follicle cells to the oocyte during oocyte degeneration are phagocytosis and digestion of phagosomes originating from oocyte degeneration. After digestion of phagosomes, it is assumed that the function of follicle cells can permit a transfer of yolk precursors necessary for vitellogenesis and allows for the accumulation of glycogen and lipid during oocyte degeneration, which can be employed by vitellogenic oocytes. Follicle cells of S. constricta may possess a lysosomal system for induction of oocyte breakdown and might resorb phagosomes in the cytoplasm for nutrient accumulation during oocyte degeneration.

  • Generally pancreas consist of lipid, water and protein, digestion enzyme complex (pro-tease, lipase, amylase). The sample used in this work was frozen dry and treated by a semi-batch flow type. In order to develop a supercritical fluid extraction process to rem-ove lipid from the pancreas, experiments were conducted at various operating conditions(pressure range $1500{\sim}2800psi$, temperature range $25{\sim}40^{cdot}C$, particle size$(0.25{\sim}1.0mm$, flow rate $20{\sim}80m{\ell}/min)$. Also cholesterol in the pancreas was removed. The highest extraction efficiency was 2500psi, $35^{\cdot}C$, 0.25mm of pancreas size. The enzyme activity of the pancreas produced from this work showed high value compared with imported pancreas.

Oocyte Degeneration Associated with Follicle Cells in Female Mactra chinensis (Bivalvia: Mactridae)

  • Ultrastructural studies of oocyte degeneration in the oocyte, and the functions of follicle cells during oocyte degeneration are described to clarify the reproductive mechanism on oocyte degeneration of Mactra chinensis using cytological methods. Commonly, the follicle cells are attached to the oocyte. Follicle cells play an important role in oocyte degeneration. In particular, the functions of follicle cells during oocyte degeneration are associated with phagocytosis and the intracellular digestion of products. In this study, morphologically similar degenerated phagosomes (various lysosomes), which were observed in the degenerated oocytes, appeared in the follicle cells. After the spawning of the oocytes, the follicle cells were involved in oocyte degeneration through phagocytosis by phagolysosomes. Therefore, it can be assumed that follicle cells reabsorb phagosomes from degenerated oocytes. In this study, the presence of lipid granules, which occurred from degenerating yolk granules, gradually increased in degenerating oocytes. The function of follicle cells can accumulate reserves of lipid granules and glycogen in the cytoplasm, which can be employed by the vitellogenic oocyte. Based on observations of follicle cells attached to degenerating oocytes after spawning, the follicle cells of this species are involved in the lysosomal induction of oocyte degeneration for the reabsorption of phagosomes (phagolysosomes) in the cytoplasm for nutrient storage, as seen in other bivalves.

Beneficial effects of body fat and obesity through the inhibition of the digestion of carbohydrate and lipid in gastrointestinal tract (장내의 탄수화물과 지방 흡수 억제를 통한 체지방 및 비만 개선 효과에 관한 연구)

  • In a previous study, a dietary supplement was developed in our lab using natural herbal extracts against digest enzyme activity in GI tract for weight control. This natural herbal extracts could regulate absorption of glucose and lipid by the inhibition of digest enzyme activity. In this study, we screened the natural herbs that inhibit glucoamylase activity and developed an water extract of cinnamon. The cinnamon extract delayed and decreased the increment of carbohydrate degradation through the inhibition of glucoamylase activity in vitro. Fifty volunteers were subjected to the intake of the herbal extracts by taking twice a day for 60 days. As a result, the treated subjects lost 3 kg of body weight and 3.5 kg of body fat mass after the treatment. Furthermore, the body mass index and waist size were significantly decreased during the experimental period. Above results suggested that the administration of the dietary additives composed of cinnamon and natural herbal extract improves the obesity by the decrement of body weight and body fat mass.

Stability of the enzyme-modified starch-based hydrogel model premix with curcumin during in vitro digestion (효소변형 전분기반 하이드로젤 모델 프리믹스 내 탑재된 커큐민의 소화과정 중 안정성)

  • In this study, the effect of enzyme-modified starch used in the preparation of filled hydrogel powder loaded with curcumin (FHP) on redispersibility, thermal and UV stability, and curcumin retention during in vitro digestion was investigated. FHP maintained stability without layer separation when redispersed and showed more stability against UVB than the emulsion powder (EMP). There was no significant difference in the chemical stability of curcumin between rice starch-based filled hydrogel powder (RS-FHP) and enzyme-modified starch-based filled hydrogel powder (GS-FHP). However, the gel matrix of GS-FHP maintained greater stability of lipid droplets in the stomach compared to RS-FHP, thereby improving the retention rate of curcumin after in vitro digestion. GS-FHP could be used as a novel material for developing premixes that require stable formulation and maintenance of functional substances, as it can increase the dispersion stability and retention rate of functional substances after digestion.

Effect of 1% Garlic Powder on Serum and Liver Lipid and Plasma Amino Acid Concentration in Rats Fed Cholesterol Diet (고콜레스테롤 식이 섭취시 1%마늘가루 첨가가 혈액 및 간조직 중 지질과 혈중 유리 아미노산 농도에 미치는 영향)

  • The serum lipid-lowering effect of garlic was examined in hypercholesterolemic rats. The food intake was not significantly different by garlic powder supplementation. The inclusion of 1% garlic powder in a diets containing 1% cholesterol brought a reduction in the serum cholesterol and triglyceride. Liver lipids were not affected by the addition of l% garlic Powder in hypercholesterolemic rats. Earlier studies have suggested that the effect of garlic on serum cholesterol may be related to the sulfur-containing amino acids of products released during protein digestion. However, our results in rats fed garlic powder diet showed no specific effect on plasma free amino acid concentrations. In conclusion. this study clears shows that garlic intake is able to reduce the levels of serum cholesterol in hypercholesteroolemic rats. However, further studies have to be done in order to elucidate the mechanism underlying this phenomenon.
