리포좀에 봉입된 5-플루오로우라실 프로드럭의 약물 동태 및 장기 분포 (Pharmacokinetics and Tissue Distribution of 5-Fluorouracil Prodrugs Entrapped in Liposome)

  • In cancer chemotherapy, it is necessary to control the phamacokinetic behavior of an antitumor drug for effective treatment. Therefore, two 5-fluorouracil derivatives synthesize d with N-a-cyloxycarbonyl derivatives {1-(N-t-butyloxycarbonyl)leucyloxymethyl-5-FU(BLFU) and 1-(N-t-carbobenzyloxymethyl)leucyloxymethyl-5-FU(CLFU)}. prodrugs of 5-fluorouracil, antitumor agent, were loaded into liposome of different lipid compositions. After liposomal drugs were injected intramuscularly, their pharmacokinetics and tissue distribution were assessed. The $AUC_{0{\to}{\infty}$ values were 1.29, 72.50, 85.57, 66.40 and 103.60${\mu}$g.hr/ml for 5-FU, BLFU, CLFU, BLFU- and CLFU-loaded liposome, respectively. 5-FU was distributed to spleen and liver with a maximal concentration after 1 hr and eliminated after 24 hr. But both prodrugs and dimyristoylphosphatidylcholine liposome entrapped prodrugs were distributed to spleen and liver at a lower concentration but maintained for a long time with a relatively high concentration in lung. Especially, liposome-entrapped CLFU was distributed to lung with a maximal concentration after 1 hr and redistributed to spleen increasingly, while the concentration of liposome-entrapped BLFU in lung reached a maximal level after 12 hr.

  • 공시균의 생육과 aflatoxin 생성에 미치는 temperature cycling의 영향을 조사하기 위해 temperature cycling($28^{\circ}C$에서 12시간, $7^{\circ}C$에서 12시간) aflatoxin 최대 생성 온도인 $28^{\circ}C$ 및 temperature cycling 할 때에 동일한 input를 가지는 $17.5^{\circ}C$의 세 가지 온도 시험구로 나누어 실험하였다. 그 결과 SLS 배지에서 temperature cycling을 행함으로써 $28^{\circ}C와\;17.5^{\circ}C$에서 일정하게 배양하는 것보다 공시균의 aflatoxin 생성이 증가하였다. 공시균의 aflatoxin 생합성은 색소 생성과는 밀접한 관계가 있는 반면 지질 생성과는 상반된 현상을 보였으며, 지방산 조성과는 뚜렷한 연관성을 보이지 않았다.

관상동맥질환자의 혈청 지방산 조성에 관한 연구 (Serum Fatty Acids in Patients with Angiographically-Documented Coronary Artery Disease)

  • To study the relation between serum fatty acids and coronary artery disease(CAD), 194 subjects were randomly selected and divided into three groups(control, single vessel disease group(SVD), muliple vessel disease group(MVD)) according to the angiographic results. Total serum levels of fatty acids and serum phospholipid(PL)-fatty acids(FAs)were analysed using gas chromatography and their associations with CAD were examined. Different patterns of total serum fatty acid levels were found in men and women. Levels of most fatty acids of SVD and MVD were significantly lower in men, while those of MVD were significantly higher in women. In terms of PL-FAs in both men and women, the levels of PL-FAs follow the order of control < SVD < MVD and in women, the difference was significant. Various ratio(including ratios denoting the activites of desaturase and elongase) of total serum-and PL-FAs were similar in the three groups. In the relation of serum fatty acids to serum lipid profiles, PUFA & LDL showed a negative correlation, while, SFA & LDL-cholesterol and PUFA and HDL-cholesterol showed positive correlatons. correlations. Systolic blood pressure and alcohol intake levels negatively affected the levels of serum LA, AA, EPA and DHA in the risk factor analysis. These findings are consistent with other evidence indicating that fatty acid compositions are changed in CAD, especially on the concentration base and the change was related to the severity of the disease. Therefore, for the purpose of disease prevention and therapeutic use, balanced intakes of various fatty acids must be seriously considered.

  • The comparative effects of gamma irradiation an ozone treatment on the microbiological and physicochemical qualities were investigated for the improvement of hygienic quality of aloe powder and bee pollen. Gamma irradiation at 7.5~10kGy could reduce total aerobic bacteria, molds and coliforms below detection levels, but ozone treatment up to 18 ppm for 8hr was not sufficient to eliminate the microorganisms from aloe powder and bee pollen. The physicochemical properties such as fatty acid an amino acid compositions, mineral content, TBA value, barbaloin and pigment contents were not significantly changed by gamma irradiation, whereas ozone treatment caused significant changes in fatty acid composition, lipid oxidation and destruction of barbaloin and natural pigments.

Comparative Studies of Panax ginseng and Panax quinquefolium on TCDD-induced Toxicity in Rats

  • One prominent characteristic of2,3,7,8-tetrachlorodibenzop-dioxin (TCDD) toxicity in rats is a reduction of body weight accompanied by an altered serum lipid profile such as hyperlipidemia. A single administration of TCDD (50 ug/kg) resulted in a decrease of body weight and increase of serum cholesterol in rats. TCDD-induced weight loss and serum cholesterol elevation was reduced in rats administered with water extract (100 mg/kg) or saponin fraction (40 mg/kg) of Panax ginseng C.A.Meyer. In contrast, the administration of Panax quinquefolium did not inhibit the TCDD-induced weight loss and serum cholesterol elevation. Histological examinations of liver and testis revealed the administration of saponin fraction of Panax ginseng attenuated the TCDD-induced hispathologicallesions whereas the administration of saponin fraction of Panax quinquefolium did not. High performance liquid chromatographic analysis demonstrated high percentiles of ginsenoside Rg and ginsenoside $Rh_1$ were evident in saponin fraction of Panax ginseng. Results indicate that the protective effects of Panax ginseng, not Panax quinquefolium, on the TCDD-induced toxicity might be resulted from different compositions of saponins in Panax ginseng.

유과의 저장기간에 따른 휘발성 향미성분 및 특성 분석 (Changes of Korean Traditional Yu-gwa Flavor and Characteristics during Storage)

  • The changes of the color, texture and volatile flavor compounds of Yu-gwa were investigated that affected by the oxidation during storage to characteristic Yu-gwa quality. Among the proximate compositions, carbohydrate was the most abundant component, and followed by lipid and moisture. Although the change of the color showed different pattern by the packaging materials during the storage period, the value of yellowness(b) increased but that of lightness(L) decreased dramatically after 3 month storage. In the textural properties reported closely related with the moisture content, hardness was fairly affected on the period of the storage rather than the type of packaging materials. The flavor compounds of Yu-gwa were analyzed to evaluate the change of distinct volatile compounds during storage. Of the twenty one separated volatile compounds, major volatiles were aldehydes, alcohols and alkenes. The results also showed that polyethylene(PE) contained less volatiles than polypropylene(PP) by the oxidation process during storage.2,4-Decadienal was gradually increased with the period of the storage, whereas octane and furan were decreased. The results provided that the change of the flavor distribution during the storage, and also the possibility of the volatiles such as hexanal, nonanal and 2,4-decadienal as the indicator for the oxidation process.

국내 양식 은어 지방의 지방산 조성 및 저장 중 지방 산화 (Fatty Acid Composition and Oxidation of the Lipids in Sweetfish Cultured in Korea)

  • 은어의 수분 함량은 72.5%, 지방 함량은 5.3%, 회분 함량은 1.1%였다. 은어 지방 중에 단일 불포화지방산은 34.0%였다. 그중 oleic acid가 21.1%, palmitoleic acid가 7.8%였다. 다가 불포화지방산은 28.2%였는데, 그중 DHA가 12.1%, linoleic acid가 8.1%, EPA가 5.7%였다. 통째인 은어를 12일 동안 냉장($2^{\circ}C$) 보관하며 TBA 값과 PV를 측정한 결과, ascorbic acid를 처리한 군과 진공포장한 군이 대조군과 비교하여 유의적인 차이가 없었다(p>0.05). FFA는 12일 동안 세 그룹간의 유의적인 차이가 없었다(p>0.05). 잘게 다진 상태의 은어를 12일 동안 냉장($2^{\circ}C$) 보관하면서 TBA와 PV를 측정한 결과, ascorbic acid를 처리한 군과 진공포장한 군이 대조군에 비하여 지방산화 정도가 유의적으로 낮았다(p<0.05). 이러한 연구 결과로 볼 때, 은어가 불포화지방산과 DHA와 EPA의 좋은 급원 식품이며, ascorbic acid 처리와 진공포장이 은어 지방의 산화를 억제하는데 도움이 될 것으로 판단된다.

녹두의 유리 및 결합지질의 조성에 관한 연구 (Composition of Lipid Glass and Fatty Acid in Free and Bound Lipids From Mungbean)

  • 녹두(Phasealus radiatus, L)로 부터 유리지질과 결합지질(저온 및 고온)을 얻은 다음, 이들을 다시 중성지질, 당지질, 인지질로 분획 정량하고 다시 이 분획된 성분을 정량하였으며 아울러 각 분획별 지방산 조성을 분석하였다. 녹두 유리지질의 중성지질, 당지질, 인지질의 함량비율은 각각 89.1, 7.1, 3.7%였으며 결합지질(저온)은 56.4%, 28.6, 15.0%였으며 결합지질(고온)의 경우는 49.2, 29.0, 21.8%였다. 유리지질 획분의 중성지질조성 중 주된 성분은 triglycerides (82.8%), 1,2-diglycerides(6.6%), esterified sterols(4.6%)이었고, 저온에게 추출한 결합지질은 triglycerides (32.5%), free fatty acid(28.2%), 1,3-diglycerides(26.3%)들이고 고온에서 추출한 결합지질은 triglycerides(57.8%), free fatty acid(21.6%)이었다. 한편 유리지질 획분의 주된 당지질 성분은 esteryl steryl glycosides(89.9%), monogalactosyl diglycerides(8.2%)였고 저온 또는 고온에서 추출된 결합지질의 당지질성분은 esteryl steryl glycosides가 주성분$(76{\sim}93%)$였고 monogalactosyl diglycerides도 함유되어 있었다. 유리지질 및 결합지질의 인지질조성을 보면 phosphatidyl ethanolamine, diphosphatidyl glycerides, phosphatidyl choline, phosphatidyl inositol+phosphatidyl serine 등이 주요성분으로 함유되어 있었다. 또한 각 분획된 지질의 지방산 조성은 유리지질에서는 linoleic acid(39.6%), palmitic acid(24.9%), linolenic acid(19.7%), 저온 또는 고온 결합지질에서는 linolenic acid, palmitic acid, linolenic acid이었고 유리지질에서는 결합지질에서 나타나지 않는 arachidic acid와 behenic acid가 함유되어 있었다.

Ob/Ob 마우스에서 경신강지환(輕身降脂丸)18의 비만조절 (Modulation of obesity by Gyeongshingangjeehwan18 in ob/ob mice)

  • Objectives : This study was undertaken to verify the effects of Gyeongshingangjeehwan18 (GGEx18) on obesity using ob/ob male mice. Methods : Eight-week old mice (wild-type C57BL/6J and ob/ob) were used for all experiments. Wild-type C57BL/6J mice were used as lean control and obese ob/ob mice were randomly divided into 5 groups: obese control, GGEx15, GGEx16, GGEx17, and GGEx18. After mice were treated with several kinds of GGEx for 11 weeks, body weight gain, feeding efficiency ratio, plasma lipid and glucose metabolism. Results : 1. Compared with obese controls, GGEx-treated mice had lower body weight gain and feeding efficiency ratio, the magnitudes of which were prominent in GGEx16 and GGEx18. 2. Consistent with their effects on body weight gain, GGEx16 and GGEx18 not only decreased plasma triglycerides levels, but also increased HDL-cholesterol concentration. 3. CT analysis revealed that visceral fat areas were decreased in all treatment groups compared with obese control mice. The decrease in visceral fat area was prominent in GGEx16 and GGEx18, although they were not statistically significant. 4. The size of adipocytes were significantly decreased by GGEx18, whereas the adipocyte number per unit area was significantly increased, suggesting that GGEx18 decreased the number of large adipocytes. Hepatic lipid accumulation was decreased by GGEx16 and GGEx18, and the inhibitory effect was most effective in GGEx18. 5. Plasma GOT and GPT concentrations were significantly lower following GGEx16 and GGEx18 treatment compared with obese controls. Organ weights were not changed by GGEx treatment, indicating GGEx do not show any toxic effects. Conclusions : These results suggest that GGEx may regulate obesity. Of the 4 compositions, GGEx18 seems to be most effective in improving obesity and lipid disorders.

뽕잎과 감잎분말 첨가가 유화형 소시지의 지방산화, 아질산염, 염기태질소화합물 및 지방산 조성에 미치는 효과 (The Effects of Emulsion-type Sausages Containing Mulberry Leaf and Persimmon Leaf Powder on Lipid Oxidation, Nitrite, VBN and Fatty Acid Composition)

  • 본 연구는 유화형 소시지에 뽕잎 및 감잎분말을 각각 0.04와 0.08% 수준으로 첨가하여 지방산화, 잔존 아질산염 함량, 휘발성 염기태질소(VBN)함량 및 지방산 조성에 미치는 영향을 비교하고자 실시하였다. 지방산화도를 나타내는 TB-ARS는 저장 45일까지 대조구와 뽕잎 및 감잎분말을 첨가한 소시지간에 차이가 없었지만, 저장 60일에는 뽕잎 및 감잎분 말 0.04% 첨가한 소시지가 대조구에 비해 유의적으로 낮은 TBARS를 나타내었다. 잔존 아질산염 함량은 뽕잎 및 감잎 분말을 첨가한 소시지가 낮게 나타났다. 휘발성 염기태질소 ;VBN)함량은 저장기간이 경과함에 따라 대조구와 뽕잎 및 감잎분말을 첨가한 소시지 모두 증가하였다. 지방산 조성은 뽕잎 및 감잎분말을 첨가한 소시지가 PUFA/SFA 비율이 높게 나타났다. 이상에서 뽕잎과 감잎분말을 첨가하여 유화형 소시지를 제조하였을 때 휘발성 염기태질소 함량은 큰 차이를 보이지 않았지만, 소시지의 지방산화를 감소시키면서 잔존 아질산 염을 낮추는 기능성 있는 제품 생산이 가능한 것으로 사료된다.
