Free Radical Scavenging Activity and Inhibition of Linoleic Acid Peroxidation of Commercial Tocotrienol Fraction

  • Tocotrienols (T3) are minor plant constituents found abundantly in rice bran, which provide a significant source of vitamin E in animal feeds. T3 was reported to have an intrinsic hypocholesterolemic effect by inhibiting HMG-Co A reductase. It has similar antioxidative properties as tocopherols in food and biological system due to their similar chemical structures. However, the antioxidant activity and mechanism of T3 to scavenge free radicals and to inhibit the peroxidation of linoleic acid are less understood. The purpose of this study was to investigate the scavenging effect of T3 on free radicals and its inhibition of peroxide formation. Free radical scavenging activity was monitored by the DPPH (1,1-diphenyl-2-picrylhydrazyl) method whereas inhibition of linoleic acid peroxidation was evaluated using the thiocyanate method. Thiobarbituric acid (TBA) test was used to determine malonaldehyde formation from linoleic acid peroxidation. Free radical scavenging activity increased with increasing concentration levels of T3. T3 exhibited 38.2, 78.6, 92.7 and 96.2% radical scavenging activity at concentrations of 2, 8, 32 and 128 ppm, respectively. At 128 ppm, it was highly effective in inhibiting linoleic acid peroxidation. The activity of T3 evaluated by the thiocyanate method showed low absorbance values indicating a high level of antioxidant activity. All treatments showed similar trends in antioxidant activity when evaluated by both the thiocyanate method and TBA test.

연밥 조추출물의 항산화 효능에 관한 융합 연구 (Convergence study on the antioxidant effect of crude extracts of Nelumbo nucifera Gaertner)

  • 본 연구의 목적은 glutamic acid, linoleic acid, 연밥의 ethyl acetate 조추출물 및 ethyl alcohol 조추출물의 항산화효과를 알아보고자 하였다. 항산화능을 알아보고자 DPPH 라디칼 소거능, 지질과산화능 및 슈퍼옥사이드 디스뮤타제 유사활성을 측정하였다. Glutamic acid, linoleic acid, 연밥의 ethyl acetate 조추출물 및 ethyl alcohol 조추출물은 농도 의존적으로 DPPH 라디칼 소거능과 슈퍼옥사이드 디스뮤타제 유사활성을 증가시켰다. Glutamic acid, linoleic acid, 연밥의 ethyl acetate 조추출물 및 ethyl alcohol 조추출물은 대조군의 지질과산화의 증가에 비하여 시간 경과에 따른 반비례 양상으로 지질과산화가 감소하였다. 이상의 결과에서 연밥의 조추출물은 우수한 항산화능 물질이 존재 할 가능성을 제시하며 융합 연구를 통한 노화 방지 관련 의약품 개발 등에 응용되어질 수 있다.

  • Phenolics as antioxidant were added to linoleic acid to prevent lipid oxidation. Antioxidative effectiveness of them was measured by peroxie value at each 24hour interval in order to compare with 0.02% protocatechuic acid(PRL) and phloroglucinol(PHL) in linloleic acid, contrast tube at 37$^{\circ}C$for 96 hours blowing oxygen into specimen. Perocide values of oxidized linoleic acid, PRL, PHL for 96 hours were 78, 42, 30, From that the effect is more clearly demonstrated by NMR rather than UV and that the effect was dependent on the functional group and geometric molecular structure of phenolics.

  • 홍삼에서 분리한 수용성 갈변물질의 항산화 활성을 linoleic acid, Ox-brain autoxidant, Fe$^{2+}$ ADP/NAD 및 cumene hydoperoxide 시스템에서 조사한 결과를 다음과 같다. 1. Linoleic acid :투석내액의 갈변물질보다 투석외액의 갈변물질 쪽에서 hydroperoxy radical 저해율이 높은 것으로 나타났으며, 이때 S-2의 산화저해율은 S-1, L시험구보다 적은 농도에서 저해율이 높았다. S-2의 산화저해율은 49,52, 62.44 및 97.5%로 나타나 첨가농도가 증가함에 따라 linoleic acid 산화저해율은 높았다. 2. Ox-brain autoxidant : 자동산화에의한 항산화활성을 조사한 결과, 투석내액의 갈변물질보다 투석외액의 갈변물질 쪽에서 자동산화에 의한 항산화활성이 높은 것으로 나타났다. 특히S-2의 항산화 활성은 22.5, 31.7, 31.9 및 33.5%로 나타났다. 3. Fe$^{2+}$ ADP/NAD 및 cumene hydroperoxide : 횐쥐 간의 microsome 분획을 분리하여 수용성 갈변물질의 ADP/NABP 및 cumene hydroperoxide 항산화 활성을 조사한 결과, 모든 시험구에서 반응 초기에 항산화 활성이 있으나, 반응 시간이 길수록 항산화 활성은 감소하였다. 그 활성의 순서는 S-2>S-1>L순으로 나타났으나 항산화 활성의 강도는 전체적으로 약하게 나타났다.

  • Acornic powder was extracted with methanol and ethylacetate to obtain a crude acornic compounds. And they were examined concerning their antioxidant activities for linoleic acid. The results were as follows: 1. Hydrogen donating activity for DPPH was higher in 100ppm acornic compound than 100ppm BHT and 100ppm ${\alpha}$-tocopherol. 2. When the 100ppm acornic compound was added to linoleic acid, which was heated at $50^{\circ}C$ for 48 hours, antioxidant activities by POV and TBA was higher than that of 100ppm ${\alpha}$-tocopherol, but the effect was almost the same as the 100ppm BHT. 3. Antioxiodant activity of acornic compound showed synergistic effect along with malic acid, citric acid, tartaric acid, alanine, arginine, histidine, lysine-HCl, galactose, maltose, glucose and sucrose. 4. Acornic compound inhibited peroxidation of linoleic acid induced by heavy metals.

Occurrence of Glutathione Sulphydryl (GSH) and Antioxidant Activities in Probiotic Lactobacillus spp.

  • The antioxidative ability on the basis of reduced glutathione sulphydryl level, the inhibition activities of linoleic acid peroxidation of cell free extract of Lactobacillus spp. and the effects of types of media and growth phase of the cells on the cellular GSH level have been determined. Correlation between reduced glutathione sulphydryl level and antioxidative ability of Lactobacillus spp. was analyzed: Lactobacillus casei HY 2782 contained 25.15 $\mu$mole/g of GSH, the cellular GSH level of L. casei HY 2782 reached maximum after 24 h of cultivation and tended to decrease on further cultivation up to 72 h. There was a significantly higher level of cellular GSH when grown in de Man Rogosa and Sharpe (MRS) broth than in tryptone phytone yeast extract (TPY) broth or bromcresol pruple dextrose (BCP) broth (p<0.05). The antioxidant activity of cell free extract of Lactobacillus spp. have been shown to be significantly different among strains in the inhibition of linoleic acid peroxidation by thiobarbituric acid (TBA) test (p<0.01). L. casei HY 2782 and L. acidophilus ATCC 4356 revealed a high degree of antioxidative effect in linoleic acid oxidation system. Spearmans' rank correlation coefficient between inhibitory activity on linoleic acid peroxidation and cellular GSH levels of Lactobacillus spp. was 0.65, which means a significant positive correlation.

Antioxidant Activity of Medicinal Plant Extracts Used as Folk Remedies by Diabetic Patients

  • The aim of this study was to investigate the antioxidant effect of medicinal plants used by diabetic patients. Fourteen medicinal plants were selected and antioxidant activity such as 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, inhibition effect of linoleic acid autoxidation and low density lipoprotein (LDL) oxidation, thiobarbituric acid (TBA) value were measured. The Cornus officinalis had DPPH radical scavenging activity of 84.79%, which was higher than the 78.79% for $\alpha$-tocopherol. Rosa rugosa Thunberg, Pueraria thundergiana Bentham, Artemisia princeps var. orientalis and Sasamorpha purpurascens Nakai also had high values. Extracts with higher DPPH radical scavenging activities had higher total phenol concentrations, and positive correlations between these parameters were found. Inhibitory activities of linoleic acid autoxidation, LDL oxidation and TBA value, used as indices of oxidative stress, were observed in most of the selected medicinal plants. The highest inhibitory activity for TBA value was observed in the extract of Pueraria thundergiana Bentham. Rosa rugosa Thunberg (75.50 %), Sasamorpha purpurascens Nakai (74.00 %), and Cornus officinalis (73.00 %) all had high antioxidant activity against linoleic acid autoxidation, similar to that of $\alpha$-tocopherol. The same 3 plants also had high in inhibitory activity against LDL oxidation. Therefore, we demonstrated that medicinal plants used as folk remedy by diabetic patients had antioxidant activity. Especially, Rosa rugosa Thunberg, Cornus officinalis, Pueraria thundergiana Bentham, Arlemisia princeps var. orientalis, and Sasamorpha purpurascens Nakai had high phenol concentrations which resulted in high values for DPPH radical scavenging. These same plants exhibited high values for inhibitory activities for TBA value, linoleic acid autoxidation and LDL oxidation, indices associated with lipid peroxidation.

토코페롤류의 항산화작용과 Linoleic Acid Methylester에서 생성된 cis/trans-, trans/trans-Hydroperoxide Isomer (Antioxidant Effect of Tocopherols and Tocotrienols and cis/trans-, trans/trans-Hydroperoxide Isomer from Linoleic Acid Methylester)

  • Linoleic acid methylester와 토코페놀류$({\alpha}-,\;{\beta}-,\;{\gamma}-,\;{\delta}-tocopherol$${\alpha}-,\;{\gamma}-,\;{\delta}-tocotrienol)$를 사용한 model system에서의 자동산화과정에서 온도$(40,\;60,\;80^{\circ}C)$와 산소의 양 $(0,\;10,\;20%\;O_2)$에 따르는 항산화효과를 고찰하였다. Linoleic acid methylester에서 생성된 주요 산화생성물인 13-cis/trans-, 13-trans/trans-, 9-trans/cis-, 9-trans/trans-hydroperoxide를 HPLC를 이용하여 분석하였으며, 그 양적 변화로 항산화정도를 비교하였다. 모든 토코페롤류$({\alpha}-tocotrienol$제외)는 $60^{\circ}C$ 범위에서 항산화효과가 있었다. ${\alpha}-tocopherol$, ${\alpha}-tocotrienol$ and ${\delta}-tocotrienol$의 경우 $80^{\circ}C$ 범위에서 산화촉진효과가 관찰되었다. $40^{\circ}C$ 범위에서는 주로 cis/trans-hydroperoxide가 많이 생성되었고 $80^{\circ}C$ 범위에서는 trans/trans-hydroperoxide가 많이 생성되었다. $80^{\circ}C$ 범위의 hydroperoxide 생성량간의 오차가 심한 구간을 제외한 $40^{\circ}C/10%,\;O_2$에서부터 $60^{\circ}C/20%,\;O_2$까지의 산화구간에서 cis/trans-: trans/trans-hydroperoxides의 비율은 ${\alpha}-T>{\alpha}-T_3>{\gamma}-T>{\beta}-T>{\gamma}-T_3>{\delta}-T>{\delta}-T_3$의 순으로 고찰되어, 토코페롤류 중 ${\alpha}-tocopherol$이 가장 낮은 항산화효과가 있는 것으로 나타났다.

황금 열수추출물의 항산화 작용에 대한 연구 (Antioxidant Activity of Extract from Scutellaria baicalensis Georgi)

  • 한번의 생약제로 쓰고 있는 황금(Scutellaria baicalensis Georgi)에서 얻은 추출물에 대하여 유지에 대한 항산화 효과를 검토하였다. 즉 황금 추출물을 linoleic acid에 소정량 첨가하고 5$0^{\circ}C$에 일정기간 저 장하여 linoleic acid의 산화 안정성을 비교 검토하였으며, 또한 유지 자동산화의 촉진인자인 금속이온 (Fe3+)을 첨가하여 황금 추출물의 금속이온 봉쇄효과를 조사하였다. 황금 추출물의 항산화 효과는 전 농도별에서 높은 항산화 효과를 나타내었다. 그리고 시간 경과에 따른 항산화 효과에 있어서도 정도의 차이는 있지만 전 농도에서 유도기의 연장효과가 상당히 우수한 것으로 나타났다. 황금 추출물의 금속 이온 봉쇄효과에 있어서는 linoleic acid에 황금 추출물과 Fe3+을 첨가한 구가 황금 추출물의 무첨가구보 다 항산화 효과가 아주 우수한 것으로 나타나 금속이온 봉쇄력이 강한 것으로 나타났다.

쥐에서 식이에 보충한 Conjugated Linoleic Acid가 식이지방 종류에 따라 항산화작용에 미치는 영향 (Conjugated Linoleic Acid Supplemented to Dietary Fat Has an Antioxidant Activities, but It Depends on the Type of Fat in Diet)

  • The study was designed to observe an antioxidant activities of conjugated linoleic acid(CLA) in rat liver by determining the activities of antioxidative enzymes(superoxide dismutase, glutathione peroxidase, catalase) and the levels of tocopherol and thiobarbituric acid reactive substance(TBARS). Male Sprague Dawley rats at weeks-old were divided into groups according to the types of dietary fat(beef tallow and fish oil) and then each group was subdivided into groups depending on CLA supplement. All rats were fed experimental diet containing 12% total fat including 1% CLA by weight for 30 weeks. CLA supplemented to beef tallow diet did not have significant effect on the level of TBARS and tocopherol. The level of TBARS was significantly increased in fish oil diet(highly unsaturated fat diet), but its level was significantly reduced by increasing SOD and GSH-Px activities when CLA was supplemented to fish oil diet so that CLA showed a sparing action of tocopherol in tissue. CLA did not have significant effect on peroxisomal catalase activities, but its activity was significantly increased when TBARS production was high in the fish oil diet. CLA could be incorporated into phospholipid of microsomal membrane, and interfered the conversions of C18 : 0 into C18 : 1 and C18 : 2 into C20 : 4 in liver. In conclusion, CLA had an antioxidant activities depending on the type of fat in diet. Therefore, it could be recommended to use CLA when highly unsaturated fat was used in meal preparation.
