A nucleoside diphosphate-linked moiety X (Nudix) hydrolase-like gene, YSA1, has been identified as one of the gromwell plant extract-responsive genes in Cryptococcus neoformans. Ysa1 is known to control intracellular concentrations of ADP-ribose or O-acetyl-ADP-ribose, and has diverse biological functions, including the response to oxidative stress in the ascomycete yeast, Saccharomyces cerevisiae. In this study, we characterized the role of YSA1 in the stress response and adaptation of the basidiomycete yeast, C. neoformans. We constructed three independent deletion mutants for YSA1, and analyzed their mutant phenotypes. We found that ysa1 mutants did not show increased sensitivity to reactive oxygen species-producing oxidative damage agents, such as hydrogen peroxide and menadione, but exhibited increased sensitivity to diamide, which is a thiol-specific oxidant. Ysa1 was dispensable for the response to most environmental stresses, such as genotoxic, osmotic, and endoplasmic reticulum stress. In conclusion, modulation of YSA1 may regulate the cellular response and adaptation of C. neoformans to certain oxidative stresses and contribute to the evolution of antifungal drug resistance.
Environmental stresses caused by climate change, such as high temperature, drought and salinity severely impact plant growth and productivity. Among these factors, high temperature stress will become more severe during summer. In this study, we examined physiological and molecular responses of maize plants to high temperature stress during summer. Highest level of $H_2O_2$ was observed in maize leaves collected July 26 compared with June 25 and July 12. Results indicated that high temperature stress triggers production of reactive oxygen species (ROS) in maize leaves. In addition, photosynthetic efficiency (Fv/Fm) sharply decreased in leaves with increasing air temperatures during the day in the field. RT-PCR analysis of maize plants exposed to high temperatures of during the day in field revealed increased accumulation of mitochondrial and chloroplastic small heat shock protein (HSP) transcripts. Results demonstrate that Fv/Fm values and organelle-localized small HSP gene could be used as physiological and molecular indicators of plants impacted by environmental stresses.
Estimation of fatigue strength on the spot welded joint is very Important for strength design of spot welded steed sheet structures. In this paper, the effect of residual stress on the fatigue life of resistance spot weldment was studied. Residual stress fields of weldment were calculated by using thermo elastic plastic finite element analysis and equivalent fatigue stress considering residual stress effect was obtained. And then we predicted fatigue life, which included the effect of the residual stresses and the actual loading stresses. The calculation and experimental results were in good agreement. Therefore, the proposed calculated model can be considered to be sufficiently powerful for the prediction of fatigue life.
Creep tests for Titan were carned out using constant-load at $600^{\circ}C$, $650^{\circ}C$ and $700^{\circ}C$. Material constants necessary to predict creep life were acquired from the experimental creep data. And the applicability of Monkman-Grant(M-G) and modified M-G relationships was discussed. It was discovered the log-log plot of M-G relationships between the rupure time(tr) and he minimum creep rate(${\varepsilon}_m$) was conditional on test temperatures. The slop of m was 2.75 at $600^{\circ}C$ and m was 1.92 at $700^{\circ}C$. However; the log-log plot of modified M-G relationships between $t_r/\varepsilon_r$ and $\varepsilon_m$ was indpendent on stresses and temperatures. That is the slop of m' was almost 3.90 in all the data. Thus, change M-G relationships to creep life prediction could be vtilized more reasonably than that of M-G relationships for type Titan. It was divided that the constant slopes never theless of temperatures of practical stresses in the modified relationship were due to an intergranular break grown by wedge-type cauities.

Evolution of adaptive mechanisms to abiotic stress is essential for plant growth and development. Plants adapt to stress conditions by activating the abscisic acid (ABA) signaling pathway. It has been suggested that the ABA receptor, clade A protein phosphatase, SnRK2 type kinase, and SLAC1 anion channel are important components of the ABA signaling pathway. In this study, we report that the shaker type potassium (K+) channel, GORK, modulates plant responses to ABA and abiotic stresses. Our results indicate that the full length of PP2CA is needed to interact with the GORK C-terminal region. We identified a loss of function allele in gork that displayed ABA-hyposensitive phenotype. gork and pp2ca mutants showed opposite responses to ABA in seed germination and seedling growth. Additionally, gork mutant was tolerant to the NaCl and mannitol treatments, whereas pp2ca mutant was sensitive to the NaCl and mannitol treatments. Thus, our results indicate that GORK enhances the sensitivity to ABA and negatively regulates the mechanisms involved in high salinity and osmotic stresses via PP2CA-mediated signals.
Cold expansion of fastener holes has been successfully used for many years to impart beneficial compressive residual stresses. Beneficial compressive residual stress of fastener hole that has been cold expanded before is reduced by using of materials for a long time. As a result, fatigue life of material is reduced. So, compressive residual stresses of material have to regenerate by re-cold expansion method. In this paper, it was carried out a finite element analysis about variation of residual stress due to tensile stress and residual stress distribution that was regenerated by re-cold expansion method in the fastener hole. Here, a diversity tensile stress was used. Also, it was performed a finite element analysis according to cold expansion rate of re cold expansion in order to obtain a beneficial compressive residual stress.
Abiotic stress is the primary cause of crop loss worldwide, reducing average yields for most major crop plants by more than 50%. In addition, future agricultural production and management will encounter multifaceted challenges from global climate change. Therefore, it is necessary to study the molecular response of crop plants to the stresses in order to develop appropriate strategies to sustain food production under adverse environmental conditions. We carried out a large scale proteomic analysis of soybean plants in response to various abiotic stresses, including drought, salinity, waterlogging and their interactions. Proteins were analyzed by two dimensional polyacrylamide gel electrophoresis followed by matrix-assisted laser desorption/ionization time of flight (MALDI-TOF) mass spectrometry. The identified proteins are involved in a wide range of cellular functions. In addition to the well known stress-associated proteins, we identified several novel proteins, which were not reported before. In many cases our proteomic data bridges the gap between mRNA and metabolite data. Our studie provides new insights into identification of abiotic stress responsive proteins in soybean, and demonstrates the advantages of proteomic analysis in dissecting metabolic and regulatory networks.
A lysine histidine transporter (LHT) cDNA was isolated and characterized from the roots of Panax ginseng, designated PgLHT. The cDNA is 1,865 bp with an open reading frame that codes for a protein with 449 amino acids and a calculated molecular mass of 50.6 kDa with a predicted isoelectric point of 8.87. Hydropathy analysis shows that PgLHT is an integral membrane protein with 9 putative membrane-spanning domains. Multiple sequence alignments show that PgLHT shares a high homology with other plant LHTs. The expression profile of the gene was investigated by real-time quantitative polymerase chain reaction during various chemical treatments. PgLHT was up-regulated in the presence of abscisic acid, salicylic acid, methyl jasmonate, NaCl, and amino acids. To further explore the function of PgLHT gene, full-length cDNA of PgLHT was introduced into P. ginseng by Agrobacterium rhizogenes A4. The overexpression of PgLHT in the hairy roots led to an obviously increase of biomass compared to the controls, and after addition of the amino acids, the overexpressed-PgLHT hairy roots grew more rapidly than untreated controls during early stage of the culture cycle. The results suggested that the PgLHT isolated from ginseng might have role in the environmental stresses and growth response.
내연기관의 냉각기 고무호스에 사용되는 EPDM 고무는 열과 기계적 하중을 받는 동시에 국부적으로 형성된 전기적 스트레스를 받아 복합적인 노화가 발생한다. 본 연구에서는 EPDM 고무의 전기화학적 복합노화 시험을 하고, 아레니우스 관계식을 이용하여 노화온도($60^{\circ}C$, $80^{\circ}C$, $100^{\circ}C$)에 따른 가속수명 예측식을 구하였다. 수명에 영향을 미치는 인장변형률(5%, 10%)을 반영하여 예측식의 기울기($E_a/R$) 및 아레니우스 상수(C)를 고려한 수정된 수명예측 식을 제안하였다. 이를 통해 노화온도와 인장변형률에 영향을 받은 EPDM 고무의 수명을 예측하였고, 수정된 수명예측 시간이 실제 실험결과의 편차 범위 이내에서 일치함을 확인하였다.
