The Effect of Cuprophane Hemodialyzer Treated with Plasma and Albumin on Leukocyte Count (인공신에 의한 백혈구 수의 변화 혈장 및 알부민의 전처치가 이에 미치는 영향)

  • It is now well established that transient granulocytopenia is common in patients during the initial phase of hemodialysis with new cuprophane membrane, but not with reused nembrane, or some synthetic membranes. In this study, new cuprophane dialyzer was treated with plasma and albumin, respectively, then, peripheral blood leukocytes were counted at the onset and after 15 minute of hemodialysis. In the plasma-treated group, the percent of leukopenia was less (41.3 %) than control group (71.3 %). In the plasmatreated group, the degree of leukopenia was close to that ot'control grou p. The protective factor, therefore is thought to be present in the plasma other than albumin fraction.

  • We studied the protective effect of Echinacea purpurea against radiation by evaluating changes in the peripheral blood cell count and peripheral blood antioxidant activity. Echinacea purpurea administration had a suppressive effect on radiation-induced leukopenia, especially on lymphocytes and monocytes and resulted in a faster recovery of blood cell counts. Mouse peripheral blood antioxidant activity was increased by Echinacea purpurea, and a relationship between the suppressive effect on radiation-induced leukopenia and the antioxidant effect was suggested.

  • In order to indentify the effect of Astragali Radix(A.R) on cyclophosphamide(C.Y) induced leukopenia, A.R. extracts(EAR) were treated to mice orally, and blood sampling was done by periods. For the in vivo experiments, mice were divided into 4 groups, which treated EAR before, or after C.Y injection, or both, or none. Rapid normalization in the peripheral blood count of WBC, neutrophils, lymphocytes, RBC, and platelets observed in every EAR treated group regardless of the treatment periods of EAR. These studies suggest that, A.R. premedication can be effective in protection of bone marrow suppression during anticancer therapy.

Immunomodulatory Effect of a New Herbal Preparation (HemoHIM) in Cyclophosphamide-treated Mice

  • The immunomodulatory effect of a new herbal preparation, HemoHIM, on the recovery from leukopenia induced by cyclophosphamide treatment was investigated. The HemoHIM was made up with an addition of the ethanol-insoluble fraction to the total water extract of Angelica Radix, Cnidii Rhizoma and Paeonia Radix. Daily oral administration of 100 mg/kg BW or 500 mg/kg BW HemoHIM accelerated the recovery from cyclophosphamide-induced leukopenia. HemoHIM increased the number of leukocytes and lymphocytes in the peripheral blood when compared with the cyclophosphamide-treated control. Moreover, the suppressed natural killer (NK) cell activity and interferon $(IFN)-{\gamma}$ secretion in the cyclophosphamide-treated mice were restored by the administration of HemoHIM. HemoHIM significantly reduced the abnormally heightened ratio of interleukin $(IL)-4/IFN-{\gamma}$ and immunoglobulin (Ig)E/IgG2a in the cyclophosphamide-treated mice. These results suggest that HemoHIM accelerates the recovery from leukopenia and alleviates the imbalanced T helper (Th)l/Th2 responses in the cyclophosphamide-treated mice. Additionally, HemoHIM was found to stimulate normal splenocytes to secrete not only Thl type cytokines such as $IFN-{\gamma}$ and IL-2, but also Th2 type cytokine IL-4. In conclusion, our results show that HemoHIM certainly has an influence on the balanced recovery of immune cells and the activation of their activities in the cyclophosphamide-treated mice.

Classifying Host Susceptibility Using Porcine Circovirus Type 2 Viral Load and Antibody Titer (돼지 써코바이러스 2형 감염량과 항체가를 이용한 자돈의 저항성군 선발법)

  • Porcine circovirus type 2 (PCV2) is a notorious and ubiquitous virus in the swine industry. The susceptibility of the host to PCV2 infection is considered to be one factor associated with the dynamics of PCV2. The objective of this study was to verify the criteria for host susceptibility to PCV2, using blood parameters of post-weaned pigs naturally infected with the virus. The PCV2 DNA viral load, antibody titer, and leukopenia characteristics were measured in the serum extracted from the pigs at the 10th week. We classified the pigs into high (>5.0), intermediate (3.0 to 5.0), and low (<3.0) groups on the basis of the PCV2 viral load (log copies/ml), or as positive (${\leq}0.50$) and negative (>0.50) groups on the basis of antibody titer (sample-to-negative corrected ratio). Moreover, using these two categorized parameters, we suggested the criteria for classification into the susceptible and resistant groups. Statistical analyses revealed that pigs in the susceptible group had a significantly higher viral load (p<0.001) and negative antibody titer (p<0.001), as well as significantly lower leukocyte counts (p=0.018) and lower amounts of several leukocyte components (p<0.05), than pigs in the resistant group. We concluded that the susceptible group could be considered to have PCV2-induced leukopenia. Therefore, we suggest that the combined classifications of viral loads and anti-PCV2 antibodies can be used to determine PCV2-induced leukopenia in the subclinical PCV2 infection of post-weaned pig populations.

  • Enterotoxemia caused by Clostridium perfringens(Cl. perfringens)has occurred sporadically with devastating effect in Korean cattle. The disease is confirmed by detecting bacterial toxin the interstinal contents. however diagnosis has been challenging since the toxin is short lived and it must be demonstrated within one hour following death. The purpose of this paper is to provide additional clinicall and pathological information derived from observation of cattle administered with Cl. perfringens type A and/or its toxins isolated from natural case. Clinically cattle died suddenly in lateral recumbency with dyspnea tachycardia and muscle tremor without increased rectal temperature. hematology showed leukopenia with absolute decrease of all leukocytes. Blood glucose AST and ALT were increased. Grossly lungs were hyperemic haemorrhagic and edematous. Small intestine especially ileum was hyperemic and hemorrhagic with diffuse areas of necrosis. This study demonstrated that death without increased rectal temperature combined with leukopenia increased serum glucose pulmonary haemorrhage and edema plus necrotizing ileitis are important features associated with enterotoxemia due to Cl. perfringens type.   • Cerebellar atrophy was found that a patient was taking oral phenytoin for 3 years. 53 years old female patient with General tonic clonic(GTC) type seizure was prescribed phenytoin. In the process, she developed ataxic gate, dysarthria. Brain magnetic resonance imaging(MRI) finding was revealed differential diagnosis cerebellar atrophy. She was prescribed epileptol instead of phenytoin. But leukopenia, thrombocytopenia occurred. As a result, phenytoin restarted. Development of medical state decreased abuse of anticonvulsants. Considering various convulsive disorders, we must give attention to using anticonvulsants.

  • A protein-polysaccharide fraction was isolated from the mycelial culture of a Korean basidiomycetous fungus, Hebeloma crustuliniforme, and was named HCA (Hebeloma crustulineforme fraction A). When intraperitoneally injected at 200 mg/kg/day once daily for 10 days, HCA inhibited the growth of sarcoma 180 solid tumor in ICR mice by 65.1% (p<0.05). HCA also exerted protective effect against leukopenia induced by cyclophosphamide with the protection ratio of 39.7% (p<0.01).
