Production of Reactive Oxygen Species and Nitric Oxide by Anticancer Agents in Rat Polymorphonuclear Leukocytes (항암제에 의한 흰쥐 다형핵백혈구의 활성산소종(reactive oxygen species) 및 산화질소(nitric oxide)의 생성)

  • The production of reactive oxygen species (ROS) and nitric oxide (NO) by anticancer agents in rat polymorphonuclear leukocytes (PMN) was examined. PMN treated for short term (< or = 4 h) with cyclophosphamide, cisplatin, tamoxifen and doxifluridine, respectively, exhibited an enhanced respiratory burst upon formylmethionylleucy1-phenylalanine (FMLP) stimulation. In the long term (> 4h), the production of ROS was suppressed in a concentration-dependent manner. The production of superoxide anion (${O_2}^-$) from the FMLP-stimulated PMN was enhanced by the treatment (for 1 hr) of cyclophosphamide, cisplatin, tamoxifen and doxifluridine, respectively. While 1 hr-treatment with cyclophosphamide, cisplatin, tamoxifen, and doxifluridine, respectively, suppressed the production of NO from the FMLP-stimulated PMN, while 8 hr-treatment enhanced the production of NO. Neomycin suppressed chemiluminescence in cisplatin-, tamoxifen- and doxifluridine-pretreated PMN, however near suppression of chemiluminescence by ethanol and genistein was observed in PMN pretreated with these agents. Staurosporine and bisindolylmaleimide suppressed chemiluminescence in cisplatin- and doxifluridine- pretreated PMN. Wortmannin has shown a slight suppression in cyclophosphamide-, cisplatin- and tamoxifen-pretreated PMN, but a strong suppression in doxifluridine-pretreated PMN. Methionine strongly suppressed in cyclophosphamide and cisplatin-pretreated PMN. In conclusion, these results indicate that long term treatment of PMN with cisplatin and doxifluridine inhibit respiratory burst through protein kinase C (PKC) translocation, phospholipase C (PLC), D (PLD) and tyrosine phosphorylation kinase (TPK) activation. Tamoxifen inhibits respiratory burst through PLC, PLD, TPK. Cyclophosphamide inhibits respiratory burst through myeloperoxidase (MPO) activity.

  • A routine hematological observation in the course of starvation was carried out on eight experimentel1y starved rabbits. They were strictly selected and restricted all of food intake with the exception of optional water intake until death. The body weight of each rabbit on the day before starvation was about 2 kilograms. The results are summarized as follows. 1. The average decrememt ratio of body weight on the terminal day before death was $34.3{\pm}7.5$ per cent with the range from 24.5 to 46.3 per cent. The average life duration until death was $10.25{\pm}2.6$ days, the range being from 6 to 14 days. 2. The decrease in number of reticulocytes with a parallel disappearance of polychromatic erythrocytes in peripheral blood in the course of starvation Was the most remarkable change in erythrocytic series, an evidence suggesting marked restriction of the erythropoietic function on 3rd to 4th day and almost complete suspension in about a week of starvation. 3. Erythrocyte count, hemoglobin content and haematocrit value of peripheral blood, were normal or indicative of slight hemoconcentration. 4. Mean Corpuscular Hemogloin Concentration was slightly higher than normal and Mean Corpuscular Volume tended to be low and no appreciable shifts were observed in Mean Corpuscular Diameter and Price-Jones curve of erythrocytes, while fewer macrocytes than normal were seen. These changes were considered to have resulted from a marked decrease in young erythrocytes in peripheral blood in the course of starvation. 5. Neither poikilccytoses or anisosytosis was observed. 6. Leukopenia was observed in all of 8 starved rabbits. The decrement ratio on the terminal day of starvation was between 13 to 64 per cent. The leukopenia was mainly due to fall of lymphocytes in 6 cases and to fall of neutrophilic leukocytes in the other 2 cases. In many cases, irregular fluctuation of neutrophilic leukocytes in its biological curve were seen in contrast to the relatively smooth changes of lymphocytes. Eosinophilic leukocytes tended to decrease in absolute number especially in later stage of starvation. Little significance in regard to monocytes and basophilic leukocytes in this study was discussed. 7. Proplasma cells, rarely plasma cells, appeared with a tendency to increase in number at later stage of starvation. 8. The most characteristic changes on circulating blood cells in complete starvation of rabbits were the leukoponia and failure of regeneration of erythroctes. These changes were considered as adaptive phenomena in response to the catabolic consumption of body constituents.

Severe Hemorrhage Induced Expressions of Ferritin and Heme Oxygenase-1 In Leukocytes (출혈로 인한 폐 염증세포에서의 ferritin과 heme oxygenase-1의 발현)

  • Serum ferritin levels are elevated in subjects with acute lung injury (ALI), and abnormalities in plasma and lung iron chemistry have also been demonstrated in ALI and acute respiratory distress syndrome (ARDS). Stress-inducible heme oxygenase-1 (HO-1), as well as ferritin, had shown anti-inflammatory actions. Biomarkers for early detection in patients who are likely to develop ARDS would give several therapeutic chances to the patients. In order to verify the predictability in severe hemorrhage-induced ALI in rats, we measured serum ferritin and HO-1 concentrations before and after hemorrhage. Severe hemorrhages significantly increased the number of leukocytes in bronchoalveolar lavage (BAL) fluid and lung tissue myeloperoxidase activity. Both serum ferritin and HO-1 levels increased following hemorrhage, but ferritin levels were elevated earlier than HO-1. In BAL cell immunohistochemical studies, ferritin and HO-1 expressions increased after hemorrhage and localized in the cytoplasm of leukocytes. These findings suggest that inflammatory leukocytes in BAL fluid can secrete ferritin and HO-1, and serum ferritin levels might be more valid factor in predicting ARDS than HO-1 levels in hemorrhage-induced ALI.

  • Background: The chemical characteristics of the exo-secretion from Phellinus linteus (referred to as exo-secretion) including the compositions of amino acids and monosaccharides were investigated. In addition, cytotoxicity of the exo-secretion on 5 tumor cell lines derived from human cancers and its antitumor activity against ascitic sarcoma-180 cells were examined. Methods: The antitumor activity of exo-secretion from Phellinus linteus was determined by measuring parameters including tumor weight, life span of mice, chemotatic activity of leukocytes, counts of immune cells, and activity of cytokines. Results: The exo-secretion from Phellinus linteus showed no direct cytotoxicity to the five tumor cell lines tested, but it had a strong antitumor activity against sarcoma-180 cells in ICR mice as measured by tumor weight and life span of mice. The exo-secretion stimulated the chemotaxis of leukocytes and production of immune cells and cytokines. Conclusion: These results suggest that the exo-secretion from Phellinus linteus do not act as a direct cytotoxic substance to cancer cells but as an immunomodulator.

Comparison of Efficiency between Pre-storage and Post-storage filtration by Leukoreduction Blood Filter

  • Leukocytes in blood components are involved in diverse adverse transfusion reactions such as febrile non-hemolytic transfusion reactions. Therefore, leukocyte reduction is required to reduce these adverse reactions. The objective of this study was to compare the efficiency of pre-storage filtration and post-storage filtration. Filtration time, residual leukocyte count, RBC recovery, and hemolysis were assessed after pre-storage or post-storage filtration. Compared to pre-storage filtration, filtration time was prolonged and hemolysis was dramatically increased when post-storage filtration was performed. Residual leukocytes count and RBC recovery after post-storage filtration were similar with those obtained after pre-storage filtration. These results suggest that pre-storage filtration has better efficiency than post-storage filtration. These are thought to contribute to the production of better quality of leukoreduction blood components.
