The aim of the present study was to evaluate the effect of commercial monostrain and multistrain probiotics in diets on growth performance, intestinal morphology and mucin gene (MUC2) expression in broiler chicks. Three hundred seventy-eight 1-d-old male Arian broiler chicks were allocated in 3 experimental groups for 6 wk. The birds were fed on a corn-soybean based diet and depending on the addition were labeled as follows: control-unsupplemented (C), birds supplemented with Bacillus subtilis (BS) and lactic acid bacteria (LAB) based probiotics. Each treatment had 6 replicates of 21 broilers each. Treatment effects on body weight, feed intake, feed conversion ratio and biomarkers such as intestinal goblet cell density, villus length, villus width, and mucin gene expression were determined. Total feed intake did not differ significantly between control birds and those fed a diet with probiotics (p>0.05). However, significant differences in growth performance were found. Final body weight at 42 d of age was higher in birds fed a diet with probiotics compared to those fed a diet without probiotic (p<0.05). Inclusion of Bacillus subtilis based probiotic in the diets also significantly affected feed conversion rate (FCR) compared with control birds (p<0.05). No differences in growth performance were observed in birds fed different types of probiotic supplemented diets. Inclusion of lactic acid bacteria based probiotic in the diets significantly increased goblet cell number and villus length (p<0.05). Furthermore, diets with Bacillus subtilis based probiotics significantly increased gene expression (p<0.05), with higher intestinal MUC2 mRNA in birds fed diet with probiotics compared to those fed the control diet. In BS and LAB probiotic fed chicks, higher growth performance may be related to higher expression of the MUC2 gene in goblet cells and/or morphological change of small intestinal tract. The higher synthesis of the mucin gene after probiotic administration may positively affect bacterial interactions in the intestinal digestive tract, intestinal mucosal cell proliferation and consequently efficient nutrient absorption.
본 연구의 목적은 펄자임(갯벌 미생물 유래 단백질분해 효소제) 첨가가 육계의 생산성과 소화기관의 길이와 무게에 미치는 영향을 조사하는 것이다. 4일령의 암컷 Ross 240수를 각 15수씩 2개의 처리구(Control vs. Pearlzyme)로 나누어, 처리구당 8개의 펜에 무작위로 배치하였다. 육계는 그 후 32일령까지 4주 동안 2개의 사료(Control vs. Pearlzyme) 중에 하나를 섭취하였다. 그 결과, 펄자임 첨가사료는 급여 첫 한주 동안에 유의적인 증체효과를 보였지만(p<0.05), 시간의 경과와 함께 그 효과는 감소되어 마지막 한 주 동안에는 사료섭취량의 증가에도 불구하고, 증체량 및 사료효율이 유의하게 감소하였다(p<0.05). 대조구에서 4주간의 실험기간 동안 총 3.3%의 폐사율이 발생하였지만, 펄자임 첨가구에서는 폐사계가 전혀 발생되지 않았다. 그러나 펄자임 첨가사료에서는 육계의 소화기관 특히 직장과 맹장의 무게, 그리고 맹장의 길이가 유의하게 증가하였고(p<0.05), 근위의 무게는 증가하고(p=0.071), 소장의 무게는 감소하는(p=0.068) 경향이 있었다. 따라서 본 연구의 결과는, 육계 사료에 첨가된 펄자임은 급여 첫 한 주 동안에는 증체효과를 보였지만, 급여기간이 경과함에 따라 그 효과는 오히려 감소되었으며, 소화기관의 발달에 영향을 미친다는 것을 시사한다.
Alginate를 가열에 의해 저분자화하여 저분자화에 따른 소화생리 특성을 검토하기 위해, 랫드에 저분자 alginate인 HAG-10,HAG-50, HAG-100 및 alginate를 장기간 섭취시켰을 때, 체중, 각종 장기의 무게와 길이 및 췌장과 소장의 소화효소활성과 단백질, DNA 및 RNA 함량을 측정하였고 소장응모의 미세구조의 변화를 관찰하였다. 랫드의 체중은 $5\%$와 $10\%$ HAG-50 및 $10\%$ alginate에서 체중의 증가가 유의적으로 현저히 억제되었다. 간장의 무게는 유의적인 차이가 없었고, 위의 무게와 길이는 $10\%$ HAG-10, $1\%$, $5\%$ 및$10\%$ HAG-50, $1\%$와 $5\%$ HAG-100 그리고 $5\%$ alginate에서 증가하는 경향을 보였으며, 췌장의 무게와 길이는 모두 약간 증가하였다. 소장과 맹장의 무게와 길이는 $1\%$와 $5\%$ HAG-10을 제외하고 모두 증가하였으나 대장의 무게와 길이는 전반적으로 감소하였다. 췌장의 amylase 활성은 유의적인 차이는 보이지 않았고, lipase 활성은 HAG-50에서 약간 저하하였으며, protease 활성은 $1\%$ HAG-10만 제외하고 유의적으로 저하하였다. 췌장의 단백질 함량은 모두 증가하였으나, DNA와 RNA 함량은 유의 적 인 차이가 없었다. 소장의 단백질 함량은 $5\%$와 $10\%$ HAG-50에서 그리고DNA는 $5\%$ HAG-50에서 가장 높은 함량을 보였으며, RNA는 전반적으로 증가하였다 소장융모의 미세구조는 HAG-50에서 주름이 많고 표면적이 넓은 잎사귀 모양의 응모세포와 돌림주름 및 Boblet cell이 현저히 발달되어 있었다.
중국산 수입 미꾸리로부터 획득한 악구충 유충의 형태학적 특징을 관찰하고 종 동정하고자 하였다. Early third-stage larva($EL_3$)는 중국산 미꾸리의 내장에서 분리 수집하였고 advanced third-stage larva($AdL_3$)는 $EL_3$를 마우스에 실험감염시킨 후 4주에 회수하였다 미꾸리 및 마우 스로부터 획득한 충체들을 10% formalin으로 고정하였고 alcohol-glycerin 용액을 거쳐 glycerinfelly로 봉입한 후 관찰 및 계측하였다. 총 9 493마리의 중국산 미꾸리를 조사하였던 바 총 369마리의 $EL_3$가 검출되었다. $EL_3$는 평균 0.624{\;}{\times}{\;}0.101 mm 크기이었고 체표면에 약 190 줄의 가로주름(transverse striation)이 나 있었으며 각 주름에는 미세한 가시(cuticularspine)들 이 일정한 간격으로 배열되어 있었다. 충체의 전단에는 1쌍의 구순이 돌출되어 있었고 그 뒤로 근 육성의 인두(평균 0.229 mal)와 갈색의 장이 이어졌으며 후단 근처의 복측에 항문이 개구하였다 특징적인 head bulb(평균 0.034{\;}{\times}{\;}0.071 mm)에는 소구(hooklets)가 평균 36 7개, 39.5개, 41.6개 및 44.3개씩 4줄 배열되어 있었고 head bulb 바로 뒤에는 2쌍의 cervical sac(평균 0.133 mm)이 인두를 따라 길이로 배열되어 있었다. AdLB는 평균 $2.660{\;}{\times}{\;}0.346{\;}mm$ 크기이었고 충체 전단의 구순 특징적인 head bulb(평균 $0.097{\;}{\times}{\;}0.193{\;}mm$), 근육성의 인두(평균 0.755 mm), 2쌍의 cewicalsac(평균 0.355 mm). 갈색의 장 등을 가지고 있었다 대부분의 충체 head bulb에는 소구가 평균 39.0개, 41 9개, 43.9개 및 45.6개씩 4줄 배열되어 있었으나 몇 몇 충체에서는 5번째 줄에 3~7개의 소구가 배열되어 있었다. 이상의 형태학적 특징 및 계측치를 토대로 하여 중국산 수입 미꾸리와 실험감염 마우스로부터 획득한 악구충을 돼지악구충 (Gnathostoma hispidum)의 제3기 유충으로 동정하였다.
민물도요(Calidris alpina sakhalina)의 장에서 echinostome에 속하는 Himasthla(Dietz,1909) 속의 국내 미기록종을 발견하고 형태 및 분류학적 연구를 시도하였다. 새들은 경기도 강화군 여차리 일대에서 새 그물을 사용하여 포획하였다. 잡은 새들의 장에서 기생충을 분리하여 생리 식염수로 씻고 고정한 후, acetocarmine으로 연색한 후 현미경으로 관찰하였다. 발견된 Himasthla kusasigi(Yamaguti, 1939)는 작고, 길쭉한 형태로 전장 5.27mm 정도 였으며 31개의 가시가 달린 두관(0.27 mm) 을 갖고 있었다. 구홉반($0.057{\times}0.0684mm$)은 복홉반 보다 작았다. 복홉반($0.35{\times}0.29mm$)은 몸쪽 앞부분에 가깝게 위치하였다. 충체는 타원형의 전고환($0.47{\times}0.30mm$), 후고환($0.59{\times}0.27mm$) 두개의 고환을 갖고 있었다. 고환들은 충체 뒤쪽에 가깝게 위치하였다. 자궁은 길고 구불구불하였으며 충체 길이의 2/3 정도를 차지하였다. 난소($0.13{\times}0.14mm$)는 작고 둥글었다. 난소 아래에는 작고 둥근 수정낭이 보였다.
본 실험은 육계 사료 내 혼합 생균제를 첨가 급여하였을 때, 육계의 성장 성적, 소장의 길이와 무게, 맹장 내 균총 변화와 암모니아 농도에 미치는 영향을 평가하고, 항생제 대체제 혹은 생리 활성 물질로서의 활용 가능성을 알아보기 위하여 실시하였다. 1일령 Ross 육용종 수평아리를 공시하여, 개체별로 체중을 측정한 후 4개 처리에 7반복으로 반복당 25수씩, 총 700수를 선발하고, 완전 임의 배치하여 실험을 진행하였다. 항생제(Avilamycin)를 첨가한 대조구[(+) control]와 항생제를 첨가하지 않은 대조구[(-) control] 및 항생제를 첨가하지 않은 대조구에 혼합 생균제를 각각 0.1%, 0.2% 수준으로 첨가한 처리구로 나누어 5주간 급여하였다. 5주령 종료 체중 및 증체량에 있어서는 처리구 간에 통계적인 유의성 있는 차이를 보이지 않았으나, 평균치로 보아 혼합 생균제 첨가구와 항생제 첨가구의 생산 성적은 아주 유사하였다. 한편, 항생제를 첨가하지 않은 대조구[(-) control]의 성적은 나머지 3개 처리구에 비하여 제일 성장률이 저조하였다. 사료 요구율에 있어서는 항생제 또는 혼합 생균제 첨가시 이들을 첨가하지 않은 대조구[(-) control]에 비해 유의성 있게 향상된 결과를 보였다. 맹장 내 암모니아 농도는 혼합 생균제를 첨가하여 급여한 처리구에서 개선되는 결과가 나타나 질소의 이용률이 증가하고, 분변 내 유해가스의 생성을 감소시킬 수 있었음을 암시하였다. 십이지장, 공장 및 회장의 길이에서는 처리구 간에 큰 차이가 없는 것으로 나타났다. 그러나 공장의 상대적 중량은 항생제 무첨가 대조구에 비해 혼합 생균제 0.1% 및 0.2% 처리구에서 모두 유의하게 증가하는 결과가 관찰되었다(P<0.05). 장 내 균총은 항생제를 첨가 급여한 처리구에서 총 균수와 유산균이 감소하였고, 혼합 생균제의 첨가 급여에 의한 효과는 관찰되지 않았다. 본 연구 결과, 혼합 생균제를 첨가 급여하였을 때 육계의 생산성을 개선하고, 사육 초기의 사료 요구율 개선에 효과적이며, 암모니아 농도가 감소함으로써 장 내 환경 개선에 좋은 영향을 미치는 것으로 나타났다. 따라서 항생제 무첨가 사료에 성장 개선 목적의 사료 첨가제로서 이용 가치가 있었음을 나타내었다.
본 연구는 브로일러 사료에 일라이트와 은을 이온 결합시킨 $bio-silverlite^{(R)}$의 첨가가 육계의 육성 성적, 장기 변화 및 장내 미생물 성상에 미치는 영향을 검토하고자 실시하였다. 처리구는 negative control과 항생제 첨가구에 속하는 positive control, 사료에 0.5(S 0.5%) 및 1.5%(S 1.5%)를 첨가구인 4처리구 5반복으로 반복당 10수씩 총 200수로 시험하였다. 증체량은 항생제 첨가구, S 0.5%구 및 S 1.5%구가 대조구에 비하여 높았으며, 사료 효율은 $bio-silverlite^{(R)}$ 첨가 수준이 높을수록 우수하게 나타났다(p<0.05). 소장 길이는 +C구가 -C나 $bio-silverlite^{(R)}$ 처리구(S 0.5% 구와 S 1.5% 구)에 비하여 높았으나(p<0.05), 소장 무게는 $bio-silverlite^{(R)}$ 첨가 수준이 높을수록 높게 나타났다(p<0.05). 근위 중량은 $bio-silverlite^{(R)}$ 첨가구가 -C구와+C구에 비하여 낮았으며(p<0.05), 맹장 무게는 $bio-silverlite^{(R)}$ 처리구가 높은 경향을 나타냈다. Villi의 길이는 S 1.5%구가 3주와 6주 후에도 -C구 및 +C구에 비하여 길었다. TBC와 CBC는 주령 및 사료 첨가제의 첨가 형태에 따른 변화가 거의 없는 것으로 나타났다. 반면 LAB는 negative 대조구인 -C 구에 비하여 +C 구 및 S 0.5% 및 S 1.5% 처리구에서 높은 경향을 나타냈다.
Purpose: Anastomotic leakage (AL) is associated with high morbidity and mortality, high reoperation rates, and increased hospital length of stay. Here we investigated the risk factors for AL after anterior resection for rectal cancer with a double stapling technique. Patients and Methods: Data for 460 patients who underwent primary anterior resection with a double stapling technique for rectal carcinoma at a single institution from 2003 to 2007 were prospectively collected. All patients experienced a total mesorectal excision (TME) operation. Clinical AL was defined as the presence of leakage signs and confirmed by diagnostic work-up according to ICD-9 codes 997.4, 567.22 (abdominopelvic abscess), and 569.81 (fistula of the intestine). Univariate and logistic regression analyses of 20 variables were undertaken to determine risk factors for AL. Survival was analysed using the Cox regression method. Results: AL was noted in 35 (7.6%) of 460 patients with rectal cancer. :Median age of the patients was 65 (50-74) and 161 (35%) were male. The diagnosis of AL was made between the 6th and 12th postoperative day (POD; mean 8th POD). After univariate and multivariate analysis, age (p=0.004), gender (p=0.007), tumor site (p<0.001), preoperative body mass index (EMI) (p<0.001), the reduction of TSGF on 5th POD less than 10U/ml (p=0.044) and the pH value of pelvic dranage less than or equal to 6.978 on 3rd POD (p<0.001) were selected as 6 independent risk factors for AL. Background: Damage to the healthy intestinal epithelial layer and regulation of the intestinal immune system, closely interrelated, are considered pivotal parts of the curative treatment for inflammatory bowel disease (IBD). Plant-based diets and phytochemicals can support the immune microenvironment in the intestinal epithelial barrier for a balanced immune system by improving the intestinal microecological balance and may have therapeutic potential in colitis. However, there have been only a few reports on the therapeutic potential of plant-derived exosome-like nanoparticles (PENs) and the underlying mechanism in colitis. This study aimed to assess the therapeutic effect of PENs from Panax ginseng, ginseng-derived exosome-like nanoparticles (GENs), in a mouse model of IBD, with a focus on the intestinal immune microenvironment. Method: To evaluate the anti-inflammatory effect of GENs on acute colitis, we treated GENs in Caco2 and lipopolysaccharide (LPS) -induced RAW 264.7 macrophages and analyzed the gene expression of proinflammatory cytokines and anti-inflammatory cytokines such as TNF-α, IL-6, and IL-10 by real-time PCR (RT-PCR). Furthermore, we further examined bacterial DNA from feces and determined the alteration of gut microbiota composition in DSS-induced colitis mice after administration of GENs through 16S rRNA gene sequencing analysis. Result: GENs with low toxicity showed a long-lasting intestinal retention effect for 48 h, which could lead to effective suppression of pro-inflammatory cytokines such as TNF-α and IL-6 production through inhibition of NF-κB in DSS-induced colitis. As a result, it showed longer colon length and suppressed thickening of the colon wall in the mice treated with GENs. Due to the amelioration of the progression of DSS-induced colitis with GENs treatment, the prolonged survival rate was observed for 17 days compared to 9 days in the PBS-treated group. In the gut microbiota analysis, the ratio of Firmicutes/Bacteroidota was decreased, which means GENs have therapeutic effectiveness against IBD. Ingesting GENs would be expected to slow colitis progression, strengthen the gut microbiota, and maintain gut homeostasis by preventing bacterial dysbiosis. Conclusion: GENs have a therapeutic effect on colitis through modulation of the intestinal microbiota and immune microenvironment. GENs not only ameliorate the inflammation in the damaged intestine by downregulating pro-inflammatory cytokines but also help balance the microbiota on the intestinal barrier and thereby improve the digestive system.
Scutellariae Radix (Scu.), one of the immune-regulatory substances, is recognized to play the role in the metabolic process of inflammation, allergy and immunity. It has been traditionally used in the Oriental medicine to treat inflammatory bowel diseases (IBD). The purpose of this study was to evaluate the effects of water extracts of Scutellariae Radix on the spleen lymphocyte immune function in the Balb/c female mice treated with dextran sodium sulfate (DSS) to induce colitis. Water extract of Scutellariae Radix (100 mg/kg) and sulfasalazine (50 mg/kg) were administrated orally for 2 weeks of experimental period. Mice were divided into three experimental groups randomly: DSS group (5% DSS was ad libitum for 5 days) as control group, DSS + Scu. (water extracts of Scutellariae Radix for 2 weeks after 5% DSS was ad libitum for 5 days) as experimental group, and DSS + Sulfasalazine group (Sulfasalazine for 2 weeks after 5% DSS was ad libitum for 5 days) as positive control group. Levels of Ig A, Ig E, CD4$^{+}$, CD8$^{+}$, TNF-$\alpha$ and other cytokines were measured. Treatment of DSS for 5 days induced bowel inflammation and the treatment with Scu. water exteract and sulfasalazine significantly recovered the damage. The length of intestine of DSS group was significantly shorter than that of other groups. The serum and fecal concentration of Ig A of SS + Scu group was higher than those of DSS group. The contents of CD4$^{+}$ T cells was higher in the DSS + Scu. group than the other groups and CD8$^{+}$ T cells was the lowest in DSS + Sulfasalazine group. The Ig A level of cultured supernatant of spleen lymphocyte was the highest, while the Ig E level was the lowest in SS + Scu group. The concentration of TNF-$\alpha$, cytokine secreted from the Th1 cell in the supernatant spleen lymphocyte, was the highest in the DSS group and the lowest in the DSS + Scu. group. The concentration of IFN-${\gamma}$ and ll...-12 was lower in the DSS + Scu. group than those of the other groups. The concentration of IL-4 in the supernatant of spleen lymphocyte was the lowest in the DSS + Scu. group but IL-10 was not significantly different. Based on these findings, water extract of Scutellariae Radix exhibited the inhibitory effect via IL-4 production thereby inhibited the production of Ig E and strengthened immune system, and alleviated injury in DSS- induced colitis mice model.

거쳐 즉시 처리하여야 합니다. 다만, 즉시 처리가 곤란한 경우는 회원에게 그 사유와 처리일정을 통보하여야
합니다.
제 11 조 (회원의 의무)
① 이용자는 회원가입 신청 또는 회원정보 변경 시 실명으로 모든 사항을 사실에 근거하여 작성하여야 하며,
허위 또는 타인의 정보를 등록할 경우 일체의 권리를 주장할 수 없습니다.
② 당 사이트가 관계법령 및 개인정보 보호정책에 의거하여 그 책임을 지는 경우를 제외하고 회원에게 부여된
ID의 비밀번호 관리소홀, 부정사용에 의하여 발생하는 모든 결과에 대한 책임은 회원에게 있습니다.
③ 회원은 당 사이트 및 제 3자의 지적 재산권을 침해해서는 안 됩니다.
제 4 장 서비스의 이용
제 12 조 (서비스 이용 시간)
① 서비스 이용은 당 사이트의 업무상 또는 기술상 특별한 지장이 없는 한 연중무휴, 1일 24시간 운영을

① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
