Fungicidal activities of leguminous seed extracts toward phytopathogenic fungi (콩과식물 종실 추출물의 살균활성)

  • Methanol extracts from 25 leguminous seeds were tested for their fungicidal activities toward six phytopathogenic fungi, using whole plant test in a greenhouse. The efficacy varied with both the plant pathogen and legume species used. At 5 mg/pot, potent fungicidal activities were produced from extracts of Cassia obtusifolia, Glycine max var. solitae, G. max var. yagkong, G. max var. hooktae, Phaseolus multiflorus, P. radiatus var. aurea, and Vigna sinensis against Botrytis cinerea, Puccinia recondita, and Erysiphe graminis. These seed extracts were highly effective against three B. cinerea strains resistant to carbendazim, procymidone, and diethofencarb. All leguminous seed extracts revealed weak or no fungicidal effect against Rhizoctonia solani, Pyricularia grisea, and Phytophthora infestans. As a naturally occurring fungicide, leguminous seed-derived materials described could be useful as new fungicidal products against various plant diseases induced by phytopathogenic fungi.

Phenolics Content and Antioxidant Activity of Sprouts in Several Legume Crops (두과작물 새싹의 폴리페놀 함량 및 항산화성 비교)

  • The study was conducted to determine the content of phenolics and flavonoids, antioxidant activity and antioxidant enzyme activity for the extract from 7 days old sprouts of cowpea (cv. "Seowon"), mungbean (cv. "Owool") and soybean (cv. "Pungsannamulkong"). Sprout length and weight of soybean sprouts were higher than those of cowpea and mungbean sprouts. Total phenolics content [mg ferulic acid equivalents (FAE) $kg^{-1}$ DW] was highest in soybean sprout extracts (82.2 mg $kg^{-1}$), followed by cowpea (32.2 mg $kg^{-1}$) and mungbean (24.5 mg $kg^{-1}$) sprout extracts (p < 0.05). The result of total flavonoid level [mg rutin equivalents $kg^{-1}$ DW] had same tendency to the total phenolics, showing lower amounts. The antioxidant activity of the methanol extracts from all the plant dose-dependently increased. DPPH (1,1-diphenyl-2-picryl hydrazyl radical) free radical scavenging activity was higher in cowpea (44%) and mungbean (42%) sprouts than in soybean sprouts (25%). Among antioxidant enzymes, APX and POX activities were highest in cowpea sprouts and CAT and SOD activities in soybean sprouts. The results showed that total phenolics content ($r^2$ = 0.5320 ~ 0.9032) and total flavonoids level ($r^2$ = 0.4672 ~ 0.9380) were highly correlated with antioxidant or with antioxidant enzyme activity, and that the level and activity of biologically active substances were different depending on plant species.

Agricultural Characters, Phenolic and Nutritional Contents, and Antioxidant Activities of Pigeon Pea (Cajanus cajan) Germplasms Cultivated in the Republic of Korea

  • Pigeon pea (Cajanus cajan) is an important legume species that produces seeds that are rich in phenolic compounds and dietary nutrients. The aim of the present study was to evaluate the agricultural characters, phenolic and nutritional contents, and antioxidant capacities of seeds from 82 pigeon pea germplasms cultivated in the Republic of Korea. The accessions exhibited compact (24.39%), semi-spreading (74.39%), or spreading (1.22%) growth habits and determinate (89.02%), indeterminate (8.54%), or semi-determinate (2.44%) flowering patterns. Days to 75% maturity ranged from 30 to 72 d, and yield per plant ranged from 6.00 to 148.60 g. Meanwhile, total phenolic, crude protein, crude fiber, and dietary fiber contents ranged from 16.42 ± 0.62 to 29.67 ± 0.43 mg gallic acid equivalent per g of dried extract, from 16.76 ± 6.74% to 22.61 ± 0.05%, from 4.70 ± 0.24 to 8.63 ± 0.02%, and from 12.98 ± 0.71 to 33.19 ± 1.50%, respectively. In addition, DPPH radical-scavenging capacity ranged from 1.61 ± 0.10 to 16.04 ± 2.30 mg ascorbic acid equivalent per g of dried extract, and Trolox equivalent antioxidant capacity ranged from 3.03 ± 0.86 to 42.24 ± 0.72 mg Trolox equivalent per g of dried extract. Phenolic content was correlated with both DPPH radical-scavenging capacity (r = 0.63) and Trolox equivalent antioxidant capacity (r = 0.29). Nine accessions (IT170290, IT170291, IT170270, IT170276, IT170379, IT170386, IT170388, IT170418, and IT170340) exhibited early maturity, compact and erect growth habits, and above average antioxidant activities and phenolic and protein contents. In addition, accessions IT170290 and IT170291 were especially promising pigeon pea germplasms to grow, owing to various favorable characteristics (e.g., high yield and dietary fiber content). Hence, these accessions could be useful cultivars to the Republic of Korea if considered in future agricultural systems.

Occurrence of Lepidopteran Insect Pests and Injury Aspects in Adzuki Bean Fields (팥 포장에서 나비목 해충의 발생과 피해 양상)

  • Three lepidopteran insect pests of the legume pod borer, Maruca vitrata (Lepidoptera: Pyralidae), the soybean pod worm, Matsumuraeses phaseoli (Lepidoptera: Tortricidae) and an Ostrinia spp. (Lepidoptera: Pyralidae) attacking adzuki bean, Vigna angularis, were confirmed as the major insect pests during the reproductive developmental stage of adzuki bean, and M. vitrata existed dominantly. Almost all plants in the adzuki bean field were injured by at least one of the three species, and $15{\sim}60%$ of reproductive organs were injured. Unbloomed inflorescence occurred continuously through the reproductive stage of adzuki bean. While the flower was many at the early stage and its number soon decreased, the pod was few at the early stage, but its number increased soon and maintained at a constant level. The results suggested that the adzuki bean injured by insect pests compensate the flower loss by developing new inflorescence, but the compensation do not cause the development of new pods. While M. vitrata and M. phaseoli were observed in flowers, pods and stem mainly during the first half of reproductive stage of adzuki bean, Ostrinia spp. was observed only in pods and stem during the second half. In addition, while all instars of larvae of M. vitrata were observed, larvae between the third and fifth instars for M. pahseoli and Ostrinia spp. were observed.

Effects of Scarification and Water Soaking Treatment on Germination of Hard-Seeded Legumes (두과 작물의 경실종자 발아촉진에 대한 종피연화처리의 효과)

  • The crops showing hard seed character have high seed viability after seed storage for long period. The low germination rate due to hard seed coat, however, cause a problem of low seedling establishment in field condition. Three legumes used in the experiment, lablab bean (Dolichos lablab L.), asparagus bean (Vigna sesquipedalis L. Fruwirth), and soybean (Glycine max L. Merr.), showed low germination rate (26, 17, and 5%, respectively) due to thick and hard seed coat. In this study artificial treatment for breaking dormancy was tested in hard seeds. The effect of proper treatment was various depending on species. The germination rate of lablab bean was highly improved up to 94% by soaking into water for 24 hours. In the case of asparagus bean, the rate was increased up to 90% by soaking for eight hours near boiling water until it cools. The germination rate of small hard seed soybean was increase to 96% by soaking into concentrated sulfuric acid for 10 minutes. Ultrastructural change revealed by scanning electron microscope (SEM) reflects that the structure of micropyle was changed and water uptake was facilitated with all treatments tested in the experiment. Especially, sulfuric acid treatment resulted in the degradation of micropylar tissue. These results demonstrate that the artificial treatment including sulfuric acid and (hot) water soaking treatment for promoting water uptake can be applied to improve seed germination in legume seed with thick and hard seed coat.

Suitability of Sainfoin (Onobrychis viciifolia) Hay as a Supplement to Fresh Grass in Dairy Cows

  • Two experiments were carried out to determine the utility of sainfoin hay, a temperate tanniferous forage legume, as a dietary supplement for grass-fed cows. The condensed tannins (CT) of sainfoin might counteract the typical metabolic ammonia load of cows in intensive grazing systems. Furthermore, the physical fibrousness of sainfoin might improve ruminal pH stability. In the preliminary experiment, the eating rate of non-lactating Holstein cows of two tanniferous hays, sainfoin and birdsfoot trefoil, was compared to that of a grass-clover hay after specific periods of time (n = 4). The eating rate of sainfoin was superior to that of the other forages. In the main experiment, designed as a replicated 3${\times}$3 Latin square, six ruminally-cannulated, lactating Red Holstein cows received grass, concentrate and either no supplementation, 3 kg/d of grass hay or 3 kg/d of sainfoin hay (n = 6). Measured intakes of the grass hay and the sainfoin hay were 2.0 and 1.5 kg DM, and two cows entirely refused to eat the sainfoin hay and had to be excluded from data analysis. Grass DMI was similar for cows supplemented with sainfoin hay and cows fed only grass whereas intake of concentrate was higher (p<0.01) for the latter treatment. Continuous measurement of ruminal pH showed that the minimum pH at night tended to be lower (p<0.10) with grass-only feeding compared to sainfoin supplementation, but pH did not decline below the threshold of subacute acidosis for a longer period of time. The slightly higher intake of nitrogen (N) for cows supplemented with sainfoin hay (413 g/d) compared to cows fed only grass (399 g/d) was accompanied by an increased (p<0.05) fecal N excretion and a tendency for an increased (p<0.10) urinary N excretion. Ruminal ammonia concentration, as well as plasma and milk urea, were not affected by sainfoin supplementation. In conclusion, the lack of positive effects typical for CT might be explained either by the limited CT content of this plant species (55 g/kg DM) or the relatively low proportion of sainfoin in the total diet or both. Moreover, due to the unexpected low grass quality, the general ammonia load might have been too low for CT to have an impact.

Inhibition Effect on Root Rot Disease of Panax ginseng by Crop Cultivation in Soil Occurring Replant Failure (윤작물 재배에 의한 인삼 뿌리썩음병 발생 억제 효과)

  • To study the effect of crop rotation on the control of ginseng root rot, growth characteristics and root rot ratio of 2-year-old ginseng was investigated after the crops of 18 species were cultured for one year in soil contaminated by the pathogen of root rot. Fusarium solani and Cylindrocarpon destructans were detected by 53.2% and 37.7%, respectively, from infected root of 4-year-old ginseng cultivated in soil occurring the injury by continuous cropping. Content of $NO_3$, Na, and $P_2O_5$ were distinctly changed, while content of pH, Ca, and Mg were slightly changed when whole plant of crops cultured for one year were buried in the ground. All of EC, $NO_3$, $P_2O_5$, and K were distinctly increased in soil cultured sudangrass, peanut, soybean, sunnhemp, and pepper. All of EC, $NO_3$, $P_2O_5$, and K among inorganic component showed negative effect on the growth of ginseng when they were excessively applied on soil. The growth of ginseng was promoted in soil cultivated perilla, sweet potato, sudangrass, and welsh onion, while suppressed in Hwanggi (Astragalus mongholicus), Deodeok (Codonopsis lanceolata) Doraji (Platycodon grandiflorum), Gamcho (Glycyrrhiza uralensis), Soybean. All of chicory, lettuce, radish, sunnhemp, and welsh onion had effective on the inhibition of ginseng root rot, while legume such as soybean, Hwanggi, Gamcho, peanut promoted the incidence of root rot. Though there were no significant correlation, $NO_3$ showed positive correlation, and Na showed negative correlation with the incidence of root rot.

Assessment of the Effects of Interactions between Climatic Conditions and Genetic Characteristics on the Agronomic Traits of Soybeans Grown in Six Different Experimental Fields

  • Soybean [Glycine max (L.) Merr.] is a species of legume native to East Asia. The interactions between climatic conditions and genetic characteristics are known to affect the agricultural performance of soybean. Therefore, the present investigation was conducted to identify the main elements affecting the agricultural performances of 11 soybean varieties/lines from China [Harbin ($45^{\circ}12^{\prime}N$), Yanji ($42^{\circ}53^{\prime}N$), Dalian ($39^{\circ}30^{\prime}N$), Qingdao ($36^{\circ}26^{\prime}N$)] and the Republic of Korea [Suwon ($37^{\circ}16^{\prime}N$), and Jeonju ($35^{\circ}49^{\prime}N$)]. The days to flowering (DTF) of soybeans with the e1-nf and e1-as alleles and the E1e2e3e4 genotype, except in 'Keumgangkong', 'Tawonkong', and 'Duyoukong', were relatively short compared to those of soybeans with other alleles. Although DTF of the soybeans was highly correlated with all climatic conditions [negative: precipitation, average temperature (AVT), accumulated temperature; positive: day-length (DL)], days to maturity and 100-seed weight of the soybeans showed no significant correlation with any climatic conditions. The soybeans with a dominant Dt1 allele, except 'Tawonkong', had the longest stem length (STL). Moreover, STL of the soybeans grown in the test fields showed a positive correlation with only DL; however, the results of our chamber test that was conducted to complement the field tests showed that STL of soybean was positively affected by AVT and DL. Although soybean yield (YLD) showed positive correlations with latitude and DL (except L62-667, OT89-5, and OT89-6), the response of YLD to the climatic conditions was cultivar-specific. Our results show that DTF and STL of soybeans grown in six different latitudes are highly affected by DL, and AVT and genetic characteristic also affect DTF and STL.

Effects of Temperature on the Development and Fecundity of Maruca vitrata (Lepidoptera: Crambidae) (콩명나방(Maruca vitrata) (나비목: 포충나방과) 발육과 산란에 미치는 온도의 영향)

  • Maruca vitrata is one of important pests in leguminous crops, especially red bean. We investigated the effects of temperature on development of each life stage, adult longevity and fecundity of M. vitrata for understanding the biological characteristics of the insect species at eight constant temperatures of 13, 16, 19, 22, 25, 28, 31, and 34℃. Eggs hatched successfully at all temperature subjected and larvae successfully developed to the adult stage from 16℃ to 31℃. The developmental period of egg decreased up to 31℃ and after then increased. The developmental period of larva and pupa, and adult longevity of M. vitrata decreased with increasing temperature. Lower and higher threshold temperature (TL and TH) were calculated by the Lobry-Rosso-Flandrois (LRF) and Sharpe-Schoolfield-Ikemoto (SSI) models. The lower developmental threshold (LDT) and thermal constant (K) from egg hatching to adult emergence of M. vitrata were estimated by linear regression as 12.8℃ and 280.8DD, respectively. TL and TH from egg hatching to adult emergence using SSI model were 14.2℃ and 31.9℃. Thermal windows, i.e., the range in temperature between the minimum and maximum rate of development, of M. vitrata was 17.7℃. In addition, we constructed the oviposition models of adult, using the investigated adult traits including survival, longevity, oviposition period and fecundity. Temperature-dependent development models and adult oviposition models will be helpful to understand the population dynamics of M vitrata and to establish the strategy of integrated pest management in legume crops.

Seed Coating Material and Seed Size Effects on Agronomic Characteristics of Over Sown Pasture Species (피복재료 및 종자의 크기가 겉뿌림 목초의 생육특성에 미치는 영향)

  • An experiment was conducted to evaluate the effects of lime, zeolite, and rock phosphate as coating materials and coated seed size on germination, establishment, and early growth of pasture species at the experimental livestock farm, Yonam College of Agriculture in 1997. Germinating energy and germination of coated tall fescue and orchardgrass seeds were lower than those of non-coated seeds. Coated seed of tall fescue with 70% lime and 15% rock phosphate was the best in germination. The germination of small size seed (< ${\varnothing}2$ mm) was better than those of medium (> ${\varnothing}2$ mm and < ${\varnothing}2$ mm) and large size seeds (> ${\varnothing}2$ mm). Germination of coated orchardgrass seed with 55% lime and 30% rock phosphate treatment was higher than that of other coated seeds, and germination of small size seed was higher than those of medium and large size seeds (p<0.05). Germination energy and germination of coated alfalfa and birdsfoot trefoil seeds were lower than those of non-coated seeds. Germination of coated alfalfa seed with 70% lime and 15% rock phosphate, and 45% lime and 55% rock phosphate was higher compared with other coated seeds. Germination of medium size seed was higher than those of small and large size seeds (p<0.05). Among coated birdsfoot trefoil seed, 45% lime and 40% rock phosphate treatment resulted in higher germinating energy and germination than other treatments. There was no significant difference in establishment and early growth of coated tall fescue and orchardgrass seeds. However, tall fescue and orchardgrass coated with 45 % lime and 40% phosphate rock showed higher establishment and early growth. There was also no significant difference in establishment and early growth of coated alfalfa among the treatments. Among coated treatments, establishment and early growth of alfalfa coated with 75% lime and 10% rock phosphate were the highest and showed 46.4% and $72.6\;g/\textrm{m}^2$, respectively. Establishment and early growth of coated birdsfoot trefoil with 55% lime and 30% rock phosphate were the highest among coated birdsfoot trefoil seed (p<0.05) and recorded 46.4% and $44.6\;g/\textrm{m}^2$, respectively. Results of this study indicate that the germination of coated grass and legume seed may be improved by increasing the percentage of rock phosphate and decreasing that of lime. With regard to seed size, the germination of pasture species with small seed size (< ${\varnothing}2$ mm) may be better than medium and large size seeds except alfalfa. Establishment and early growth of grasses can be also improved by increasing the percentage of rock phosphate and decreasing that of lime.
