Evaluation of Morphological Characteristics and RAPD Analysis in Korean Landraces of Naked Barley (재래종 쌀보리의 형태적 특성 및 RAPD에 의한 유연 관계 분석)

  • Barley varieties collected from 1940 to 1951 allover the Korean peninsula by Dr. Takahashi Ryuhei were reintroduced from the Research Institute for Bioresouces in Okayama University, Japan, and the evaluation of morphological characteristics and RAPD analysis were performed. Totally, 493 varieties of Korean barley landraces were planted in the green house, from which seeds of 491 varieties were harvested and conserved in the seedbank of the Rural Development Administration. Majority of the naked barley varieties showed dense spikes with long awn, late heading, winter habits, and long plant height. However, variants having various phenotypes such as short awn, blue aleurone color, brachytic type and waxyness were also identified. Plant height, spike length, and cold-tolerance in the varieties were also highly variable among them. Homogeneity tests on the variation of growth habits, spike density, anthocyanin pigmentation on the seed coat, and hairiness on leaf sheath between naked and covered barley showed that the variations of naked barley were similar to those of covered barley. It maybe indicate that the most of naked barley landraces were mutated from the covered barley landraces. Korean landraces of naked barley were broadly divided into 4 groups by the dendrogram produced by morphological characteristics; however, the identities of the group were rather indistinct. Many varieties, belonged to the same group, were showed different band patterns in RAPD analysis using 5 different primer sets. These results indicate that the 112 varieties of naked barley landraces were different genotypes.

Review of Disease Incidence of Major Crops in 2001 (2001년 농작물 병해 발생개황)

  • Climate in the year of 2001 could be summarized as severe drought from March to May, unusually high temperature in whole plant growth periods and clear weather condition especially in harvesting time of September and October without any typoons. In rice, major diseases such as leaf and panicle blast, bacterial blight, sheath blight and bacterial grain rot developed little due to unfavorable weather conditions, however, brown spot occurred severely due partly to the reduced use of N-fertilizer, Rice stripe virus infection was unusually high mainly in the west coast areas with four times more diseaseD area campared to the previous year, In pepper phytophthora blight was extremely severe in Cheonnam and Cheonbuk provinces, where had frequent rainfalls during growing period. Incidence of major diseases of tomato and cucumber in 2001 was relatively mild. In watermelon, penicillium fruit rot, one of the peculiar disease, spread over major production areas, whereas CGMMV, usually severe in every years was much reduced. Watermelon plants growing in open-fields were more severely diseased than those in plastic houses. Major diseases of chinese melon and strawberry occurred slightly and in particular, sudden wilt syndrome of chinese melon which was severe in 2000 showed mild infections in 2001. Incidence of white rot of garlic and onion was much lower unlike other years due mainly to spring drought and high temperature in the growing period. Infected area of potato bacterial wilt tended to increase in 2001 while fusarium wilt of sweetpotato was decreased. Apple diseases were generally mild, but powdery mildew of pear increased sharply. Barley scab was not severe as seen in other years.

  • This experiment was conducted to clarify the effect of double layer mulching on reducing the labor required to weed control and Leaf sheath training of the garlic cultivation. Six mulching methods(non-mulching, transparent P.E., rice hull+transparent P.E., sawdust+transparent P.E., rice straw+transparent P.E., black P.E. film+ transparent P.E.) were used for the experiment, and transparent P.E film was removed on April 10. Shattering Grain Loss in Relation to the Impulse During Harvest and Year-Variation of Grain Tensile Strength in Rice (벼 수확시 충격에 의한 탈립 손실과 탈립성의 년차간 변이)

  • Tensile strength and shattering losses of rice grains in a binder harvest and in a free falling test were examined. Indica$\times$Japonica varieties, Milyang 23, Hangangchal, and Taebaek, and a Japonica variety, Jinheung were used for this study. The average tensile strength varied with varieties, showing 0.743N (Newton) in Milyang 23, 1.078N in Hangangchal, 1.573 N in Taebaek, and 1.775 N in Jinheung. Extrapolation of the average tensile strength for the zero field loss of grain indicated about 1.7 N. Moisture content of straw at the cutting position in a binder harvest was 76.0 to 86.7% in culm and 76.7 to 77.8% in leaf sheath. The percent loss of grains occurring in binder barvest in the field was equivalent to the degree of shattering in the panicle-falling test onto the concrete floor at the height of about 40cm. The grain shattering (Y) was closely related to the impulse (X), the change in momentum: Y=-25.73+4958.4X, ($r^2$=0.987) for Milyang 23, Y=-12.97+2258.3X, ($r^2$=0.979) for Hanagangchal, Y=-5.24+840.4X, ($r^2$ =0.976) for Taebaek. The tensile strengths of grains and their variance varied singnificantly yearly even in the same variety. The magnitude of year-variance was somewhat smaller for the easily shattering variety.

Determination and Effects of N and Si Fertilization Levels on Grain, Quality and Pests of Rice after Winter Green-house Water-melon Cropping

  • In Korea, rice cultivars have been changing to 'quality' rice rather than high yielding cultivars. However, more than 10% of paddy field has been changed to greenhouse in winter season for cropping of water-melon, oriental-melon, straw berry and et cetra. This experiment has been made to identify the usefulness of critical N and Si fertilization(SF) level to obtain high grain quality rice with reduced insect pest damage by N and SF combination. Before the experiment, watermelon-rice cropping system was maintained for three seasons by farmer from 1998 to 2001. The experiment of N and Si (silicate) fertilization levels was evaluated with Hwayoung-byeo (Oryza sativa L., medium-maturing variety) in 2002 and 2003 in Uiryeong, Korea. Nitrogen fertilization (NF) levels were three and five in 2002 and 2003, respectively, and three SF levels were compared for getting the valuable N/SF level in both years. TOYO-value was positively affected by Si application in N100% plot but it was negatively related with NF level. Normal grain percentage was positively related with TOYO-value and it was highest in 0N plot and Si plots in N100%. Other appearance qualities like powdered, damaged, and cracked grain, were decreased with increasing N fertilization level. SF improved appearance quality in N100% plots but no effects in other treatments. Leaf sheath related diseases were significantly decreased by SF but it was negatively related with NF. In conclusion, SF could be improve grain quality at the same yield levels of conventional fertilization and it also could be reduce the diseases damages of rice plant in all N treatments. NF treatment reduced grain quality and improved grain yield at N50% level, however NF above N50% could not get any kind of benefits. So, compared with conventional fertilizer, reduced NF level is recommended for high grain quality with reduced insect pest damage.

  • A tarsonemid mite. Stet~rotc~r.sot~rrrs~puin.sk i Smiley 1967. is an important rice pest in major rice producing countries of Thailand. Philippine. China, and Taiwan. S. spinki was found first time in Korea from rice grown in environment controlled greenhouse. Rice fed by the mite showed damage symptoms of deformed panicles and inflorescence, lesions on the inner surface of leaf sheath, and browning of rice hulls. Females of S. .spi~lkwi as 263.0 pni (246.5-284.6f 12.2) in body length and 92.4 pm (79.5- 104.9 t 7.6) in body width. Body was elongate and broadest in region of hysterosoma. Body color was pale brown. Legs were robust except for the legs IV which were typical tarsonemid female legs terminating in a whiplike seta two times the length of the leg. Male of S. spir~kiw as 196.5 pm ( 176.5-222.8 + 15.8) in body length and 109.3 ym (98.6- 1 17.7 + 6.4) in body width. Anterior ends of apodemes 111 were extended further than apodemes IV. Femur IV had large inner median lateral flange. and inner anterior and outer median setae were short about equal length. Tarsal claw was stout and curved ventrally.

  • Propanil [N-(3,4-dichlorophenyl) propanamide] which was applied at 4,200g ai/hapostemergence 7 days after seeding or transplanting, completely reduced the growth of shoot and root of barnyardgrass at 100% under dry condition while plant height, root length and shoot fresh weight of barnyardgrass at 63, 40 and 78%, respectively under water condition. On the other hand, the herbicide did not affect the growth of shoot and root of rice grown under water condition and transplanting condition, but reduced the plant height, root length and shoot fresh weight of broadcast rice on soil at 24, 18 and 28%, respectively, under dry condition. Microscopically, the epidermal cells of treated-barnyardgrasses under both conditions were severely constricted, chloroplasts in the cells of vascular bunble sheath were partially lacked, and mesophyll cells were often ruptured, whereas those of treated-rice were not affected. Histological observations showed that propanil reduced the thickness of leaf blade of barnyardgrass under both conditions at 36-48% due to mainly reduction and constriction of mesophyll cell, while it did not affect or even increased the thickness of leaves of rice under all conditions compared to control. These results indicate that broadcast rice on soil were more injured than drilled rice in soil under dry condition, however, in the other tested conditions ricer were not affected.

Occurrence of Freesia Basal Rot Caused by Sclerotium sp. (Sclerotium sp.에 의한 프리지아 균핵병 발생)

  • Basal rot of freesia caused by a Sclerotium sp. occurred at Incheon areas. Incidence of the disease reached up to 45% and averaged 17.0% in the fields. Typical symptoms consisted of sheath dry and leaf blight due to rots on basal leaves. The causal fungus was identified as Sclerotium sp. based on following mycological characteristics. The fungus formed sclerotia on cultural media and plant tissues, but did not produce asexual spores. On cultural medium, aerial mycelia of the fungus changed color from white to clay with cultural age and smelled musty odor. Numerous irregular and elliptical black microsclerotia of the fungus were formed on potato dextrose agar (PDA) after 5 days of incubation at $25^{\circ}C$ and sized $115{\sim}200{\times}95{\sim}150 (av. 145{\sim}126.5){\mu}m$. The fungus grew at $10{\sim}32^{\circ}C$ and $pH 4.0{\sim}8.5$. However, the optimal temperature and pH for mycelial growth of the fungus were $24^{\circ}C$ and 5.5 respectively. The isolate showed present pathogenicity to not only freesia but gladiolus in the pathogenicity test, and the symptoms were similar to those observed in the fields. Basal rot of freesia caused by Sclerotium sp. is firstly reported in Korea.

Characteristics of agronomy to vitamin A strengthening rice at large scale GMO field (대규모 GMO 포장에서 비타민 A 강화 벼의 농업특성 검정)

  • This study was carried out to develop of macro-protocol and the biosafety guide Golden Rice, modified vitamin A at large scale GMO field. Commercialization of genetically modified (GM) plants will be required the assessment of risks associated with the release of GM plants that should include a detailed risk assessment of their impacts in the environment and human health. Prior to GM plant release, applicants should provide the information on GM crops for approval. A total of $4,700m^2$ GM field, each of Golden Rice and 'Nakdong' were used for further analysis on agronomic traits and pathogenesis. Generally plant growth and panicle number in the transgenic lines were lower than in the wild type under field condition. The major agronomic traits such as plant growth, grain quality in Golden Rice were similar than in 'Nakdong'. The yield difference was due to reduced ripened rate and the 1,000 grain weight. The grain shape was similar donor plant, however, color of brown rice was different from segregation. Also the difference of pathogenesis reaction was not significant in the leaf blast and sheath blight between Golden Rice and 'Nakdong'. These results provided the agronomic data and pathogenesis for risk assessment analysis of Golden Rice and suggested that the macro-protocol could be useful to detect GM plants.

  • Rice is responsible for the stable crop of 3 billion people worldwide, about half of Asian depends on it, and rice is grown in more than 100 countries. Rice diseases can lead to devastating economic loss by decreasing yield production, disturbing a stable food supply and demand chain. The most commonly used method to control rice disease is chemical control. However, misuse of chemical control can cause environmental pollution, residual toxicity, and the emergence of chemical-resistant pathogens, the deterioration of soil quality, and the destruction of biodiversity. In order to control rice diseases, research on alternative biocontrol is actively pursued including microorganism-oriented biocontrol agents. Microbial agents control plant disease through competition with and antibiotic effects and parasitism against plant pathogens. Microorganisms isolated from the rice rhizosphere are studied comprehensively as biocontrol agents against rice pathogens. Bacillus sp., Pseudomonas sp., and Trichoderma sp. were reported to control rice diseases, such as blast, sheath blight, bacterial leaf blight, brown spot, and bakanae diseases. Here we reviewed the microorganisms that are studied as biocontrol agents against rice diseases.
