Antimutagenic and Anticancer Effects of Leaf Mustard and Leaf Mustard Kimchi

  • In this study, we investigated antimutagenic and anticancer activities of leaf mustard (LM, Brassica juncea) and leaf mustard kimchi (LMK) during their fermentation period. Methanol extracts were prepared from raw mustard, brined leaf mustard in 10% Gueun salt solution for 2 hrs, leaf mustard fermented at 15$^{\circ}C$ for 5 days after brined in 10% Guenun salt solution for 2 hrs (Fr-LM), fresh leaf mustard kimchi (Fresh-LMK) and optimally ripened leaf mustard kimchi fermented at 5$^{\circ}C$ for 30 days (OR-LMK). OR-LMK showed the strongest inhibitory activities against the mutagenicities induced by aflatoxin B1 in Salmonella Typhimurium TA100. LMs and LMKs inhibited the survival or growth of AGS human gastric adenocarcinoma cells and HT-29 human colon carcinoma cells in MTT assay and growth inhibition test. Among the extracts, OR-LMK and FR-LM exhibited strong antiproliferative effect against cancer cells, especially HT-29 cells. DAPI staining assay showed that OR-LMK induced apoptosis cell death of HT-29 cells in a dose-dependent manner. These results suggest that leaf mustards and leaf mustard kimchi have chemopreventive activities.

돌산갓 전처리 중의 산도, 향균성 및 색도 변화 (Changes of Acidity, Antimicrobial Activity and Colors during Pretreatment of Leaf Mustard Dolsan(Brassica juncea))

  • In order to use leaf mustard Dolsan food preservative ingradient acidity, antimicrobial activity and colors were investigated during pretreatment of leaf mustard Dolsan. pH was remarkably decreased after 8 hours to pretreatment(extracted on shaking) of leaf mustard Dolsan, and no changes were observed after that time. pH of leaf part was higher than stalk after 8 hours storage. Titratable acidity was opposite tendency to the pH. pH was gradually decreased than initial stage during pretreatment of leaf mustard Dolsan at 30, 40 and 5$0^{\circ}C$. After 16 hours pretreatment, the higher pretreatment temperature was, the higher pH was, but after pretreatment for 48 hours, pH of sample pretreated at 5$0^{\circ}C$ was lower than that of sample pretreated at 40 $^{\circ}C$. Antimicrobial activity of leaf mustard Dolsan extract pretreated at 3$0^{\circ}C$ was the strongest of the samples pretreated at 30, 40 and 5$0^{\circ}C$, and that of stalk part was stronger than that of leaf part. L and time. L and b value was higher in the order of samples pretreated at 40, 50 and 3$0^{\circ}C$, and the lower pretreatment temperature was, the higher a value was. L value of stalk part was higher than that of leaf part, but a and b value of leaf part was higher than that of stalk part.

돌산갓의 Isothiocyanate류의 분석 (Analysis of Isothiocyanates in Dolsan Leaf Mustard(Brassica juncea))

  • The composition of isothiocyanates in Dolsan leaf mustard was investigated. Five major volatile isothiocyanates detected in leaf mustard were sec-butyl isothiocyanate, allyl isothiocyanate, 3-butenyl isothiocyanate, n-hexyl isothiocyanate and ${\beta}-phenylethyl$ isothiocyanate. In both leaf and leaf stalk allyl isothiocyanate and 3-butenyl isothiocyanate were the most abundant. The compositional difference of isothiocyanates between leaf and leaf stalk was that n-hexyl and ${\beta}-phenylethyl$ isothiocyanates were present more in leaf than leaf stalk.

  • 동결건조한 청갓과 담금당일 청갓김치, 15$^{\circ}C$에서 5일간 발효시킨 청갓 김치로부터 극성이 다른 용매들을 사용하여 각각의 용매별 추출물을 얻고 methanol(MeOH), hexane(C$_{6}$ $H_{14}$), dichlorometane(C $H_2$Cl$_2$), ethyl acetate(EtOAc), butanol(BuOH), water($H_2O$)의 각각의 용매분획별(DPPH) 1,1-diphenyl-2-picrylhydrazyl assay에 의한 항산화성을 측정하였고 항산화효과를 상호 비교하였다. 청갓과 담금당일 청갓 김치에서 EtOAc와 n-BuOH에서 얻은 분획층이 다른 분획층보다 항상화효과가 높게 나타났으며 15$^{\circ}C$ 5일간 발효시킨 청갓 김치에서는 C $H_2$Cl$_2$와 EtOAc분획층이 다른 분획층에 비하여 radical scavenger효과가 있음을 확인할 수 있었다. 결과의 차이는 다양한 재료에 의한 발효공정에 기인된다고 판단된다.

  • To furnish basic data for the utilization of leaf mustard as a raw material of salted and fermented vegetable food, the contents of carotenoids and chlorophylls of Dolsan Leaf Mustard(DLM) were investigated. Total carotenoid content of DLM was 4.75 mg%, and the compositions were ${\beta}-carotene(80.91%)$, lutein(13.07%), lutein epoxide(3.93%). The contents of chlorophyll a and b were 4.1 and 1.5 mg%, and leaf was 7.4- and 8.1-fold, respectively, higher than leaf stalk. The ratios of chlorophyll a/b in leaf (2.7:1) and leaf stalk(3.0:1) were similar to those of other vegetables.

적갓김치 Anthocyanins의 항산화 특성에 관한 연구 (Antioxidative Activities of Anthocyanins in Red Mustard Leaf Kimchi)

  • 재래종 적갓(Brassica juncea czerniak et coss)을 주재료로 담금하여 발효시킨 적갓김치(red mustard leaf kimchi, RML kimchi)의 crude anthocyanin(CA) 및 refined anthocyanin(RA) 획분들에 대한 항산화성을 규명하였다. 발효된 적갓김치는 산도 0.72(%), pH 4.6 그리고 crude anthocyanin 함량(건조물기준)은 49.4 mg/g이었다. 동결건조 적갓김치로부터 얻어진 조색소용액 (crude pigment solution)을 계통 추출분획하여 증류수 획분, methanol 획분, TFA-methanol (CA) 획분을 얻었으며, CA 획분을 계속 분획하여 정제된 5개의 정제된 RA 획분(Fr.1, Fr.2, Fr.3, Fr,4, Fr.5)을 얻었다. 이들의 항산화 활성을 linoleic acid의 autoxidation system에서 peroxide formation 억제능과 DPPH($\alpha$, $\alpha$'-diphenyl-$\beta$-picrylhydrazyl)의 free radical 소거능에 대하여 실험하였다. 그 결과 조색소용액 분획물 중 CA 획분이 항산화능이 높았고, CA 획분으로부터 얻어진 RA 획분 중 Fr.3이 가장 높은 항산화성을 보였다. 특히 Fr.3은 적갓김치의 대표적인 anthocyanin이었고 항산화 활성은 $\alpha$-tocopherel보다는 낮았으나 그것에 접근하는 유사한 특성을 보였다. 그리고 그 활성은 지질의 과산화 억제능과 수소공여성에서 다같이 비슷한 수준의 항산화성을 보였다.

  • 천연 식품보존료 개발의 일환으로 우리나라에서 옛부터 김치재료, 조미료 및 향신료 등으로 널리 이용되고 있는 십자화과 엽경채소류 중의 하나인 갓(Brassica juncea Coss.)을 대상으로 물과 에탄올로 항균성물질을 추출하여 몇 종의 병원균과 식중독균, 식품과 관련이 있는 세균 및 효모 등 15균주에 대하여 항균활성을 검토한 결과는 다음과 같다. 갓 에탄올 추출물은 실험에 사용된 대부분의 균주에서 항균효과가 나타났는데, 특히 Bacillus subtilis와 Bacillus natt의 최소 생육저해 농도가 10mg/ml로 가장 낮게 나타났다. 그러나 물 추출물은 세균과 효모에 대해 항균활성을 나타내었으나 최소 생육저해 농도는 40~60mg/ml로 나타나 항균효과는 에탄올 추출물에 비해 1/3~1/6 정도에 지나지 않았다. 또한 갓 에탄올 추출물 중의 항균활성 물질은 $121^{\circ}C$에서 15분간 가열한 후에도 활성이 유지되었으며, pH의 변화에도 영향을 받지 않았다.

식물공장에서 광주기 및 광강도가 십자화과 3종의 생육과 글루코시놀레이트 함량에 미치는 영향 (Effects of Photoperiod and Light Intensity on the Growth and Glucosinolates Content of Three Brassicaceae Species in a Plant Factory)

  • 본 연구는 같은 일적산광량(DLI) 조건에서 각각의 광강도와 광주기 조합이 십자화과 식물 3종의 생육 및 글루코시놀레이트(GSLs) 함량에 미치는 영향을 살펴보고자 수행되었다. 갓(Brassica juncea (L.) Czern.), 적겨자(Brassica juncea L.), 케일(Brassica oleracea L. var. acephala (DC.) Alef.) 종자를 암면 배지에 파종하여 3주간 육묘하였다. 육묘 후 DLI를 10mol·m-2·d-1로 설정하여 10h-280, 14h-200, 18h-155, 22h-127µmol·m-2·s-1 처리 조합으로 3주간 처리하였다. 14h-200µmol·m-2·s-1 처리한 결과, 적갓과 케일은 지상부 생체중/건물중, 엽수, 엽면적이 증가하였지만, 유의적 차이는 관찰되지 않았다. 적갓의 GSLs 함량에서 14h-200µmol·m-2·s-1 처리가 10h-280, 18h-155, 22h-127µmol·m-2·s-1 처리보다 각각 유의적으로 139.95, 135.87, 154.03% 증가하였으며, 케일의 경우 14h-200µmol·m-2·s-1 처리가 다른 처리구보다 각각 132.96, 122.06, 131.78% 유의하게 증가하였다. 적겨자는 18h-155µmol·m-2·s-1 처리에서 지상부 생체중/건물중, GSLs 함량이 증가하였으며, 14h-200µmol·m-2·s-1 처리에서 엽수가 10h-280, 18h-155, 22h-127µmol·m-2·s-1 처리보다 각각 15.62, 12.12, 32.14% 유의적으로 증가하였다. 유사한 십자화과 작물이라도 종에 따라 DLI 반응이 다르기 때문에 최적의 DLI 조건 구명과 최소의 전력 소비량 및 최대 효율을 내기 위한 광질 연구를 병행하여 더세밀한 결과를 도출해야 할 것으로 사료된다.

갓 추출물의 견직물에 대한 염색성 (The Dyeing Properties of Silk Fabric of Leaf Mustard (Brassica Juncea) Extract)

  • Dyeing properties of silk fabric with leaf Mustard was investigated the relation with pH variation, mordants, mordant method. K/S value shows the following sequence: pH 3>pH 5>pH 7>pH 9. K/S value shows high in post mordant conditions. Silk fabric shows the green, blue tone according to mordants(Al, Cu, Fe mordants). The tannic acid treatment silk fabric shows higher than the nontannic acid treatment silk fabric in K/S value. Lighting fastness of tannic acid treatment silk fabric shows better than that of nontannic acid treatment in Cu, Fe mordants. Water fastness of tannic acid treatment silk fabric was wholly improved. Tannic acid treatment improved dyeing properties of silk fabric with leaf Mustard.

Assessing persistence of cruciferous crops in the field

  • We assessed the persistence of eight major cruciferous crops-leaf mustard, oilseed rape, cabbage, broccoli, cauliflower, Chinese cabbage, turnip, and radish-growing in the field. In the first part of our experiment, we tested the viability of seeds that had been buried at two different soil depths for up to 16 months. We then broadcast seeds over the soil surface and left them undisturbed to investigate the survivorship of the resultant plants over two years. Seed viability was significantly affected by plant taxa and burial depth, but not substantially affected by the duration of burial. Although seeds of leaf mustard had the greatest viability among all crops examined here, the viability rates were significantly lower at 2 cm depth than at 15 cm. Seeds of leaf mustard, oilseed rape, broccoli, turnip, and Chinese cabbage remained viable throughout the 16-month period. A study of plant demography revealed that only leaf mustard and oilseed rape succeeded in producing seeds and overwintering in the undisturbed field. However, neither of those species competed well with other plants long-term and their overall growth and survival rates declined during the evaluation period. In addition, insect herbivory severely decreased the growth of all of these crops. Our results suggest that populations of leaf mustard and oilseed rape do not tend to persist in the field for more than a few years without disturbance and external seed inputs.
