Two New PVY Strains Isolated from Tobacco Plant in Korea (한국 잎담배에서 분리된 PVY계통)

  • Since 1980, burley tobacco plants grown mainly in the western hat of the Korea. have shown two new types of disease symptom. Both symptoms were found to be caused by two different PVY strains : the vein banding type by a PVY strain designated as PVY-VB and necrosis on leaf veins by a PVY strain designated as PVY-VN, Identification of the PVY strains was based on host range test. aphid( Myzus Persicae) transmission test, physical properties, serology, and observation of virus particle morphology. The virus particles were measured to be about 730 nm without any difference in shape or dimensions between the two strains. Both strains also gave a positive reaction to the PVY antiserum in SDS-agar gel double diffusion test. These strains, however, gave a negative reaction to the tobacco etch virus and tobacco vein mottling virus antisera.

Analysis of metabolites in wheat roots in response to salinity stress

  • Salinity stress is one of the most important abiotic stresses and severely impairs plant growth and production. Root is the first site for nutrient accumulation like as $Na^+$ in the plant. To investigate the response of wheat root under salinity stress, we executed the characterization of morphology and analysis of metabolites. Wheat seeds cv. Keumgang (Korean cultivar) were grown on the moist filter paper in Petri dish. After 5 days, seedlings were transferred to hydroponic apparatus at 1500 LUX light intensity, at $20^{\circ}C$ with 70% relative humidity in a growth chamber. Seedlings (5-day-old) were exposed to 50mM, 75mM, 100mM NaCl for 5 days. Ten-day-old seedlings were used for morphological characterization and metabolite analysis. Root and leaf length became shorter in high NaCl concentration compared to following NaCl treatment. For confirmation of salt accumulation, wheat roots were stained with $CoroNa^+$ Green AM, and fluoresce, and the image was taken by confocal microscopy. $Na^+$ ion accumulation rate was higher at 100mM compared to the untreated sample. Furthermore, to analyze metabolites in the wheat root, samples were extracted by $D_2O$ solvent, and extracted sample was analyzed by 1H NMR spectroscopy. Fourteen metabolites were identified in wheat roots using NMR spectroscopy. Methanol and ethanol were up-regulated, whereas formate, aspartate, aminobutyrate, acetate and valine were down-regulated under salinity stress on roots of wheat. Fumarate had no change, while glucose, betaine, choline, glutamate and lactate were unevenly affected during salinity stress.

Comparative Morphology of Plastids on Vegitative Tissue of Cannabis sativa L. (대마(Cannabis sativa L.) 영양조직의 색소체 비교)

  • Various plastids in leaf and bracteal tissues of Cannabis sativa L. were examined by electron microscopy. Young chloroplasts without starch grain in mesophyll cells were ellipsoidal, and osmiophilic globules within them were common in stroma. During the plastid differentiation, the mature chloroplasts in mesophyll were changed in shape depending on the numbers and sizes of starch grain in stroma. Electron-dense granular substances were occurred along the outer membrane of chloroplasts in mesophyll. Typical plastids with reticulate body were present in the glandular trichomes. Electron-grey material appeared along the surface of a plastid. A light area in reticulate body is considered to represent junction point of thylakoids.

Identification of Agrobacterium tumefaciens from Soil and Transformation of Maize (토양으로부터의 Agrobacterium tumefaciens의 분리, 동정 및 옥수수의 형질전환에 이용)

  • Several strains of Agrobacterium tumefaciens were isolated from soil in the Taegu area and characterized to develop some useful vector systems for higher plant genetic engineering. The selected colonies had a unique form, and strains from the colonies were capable of tumor formation on the sunflower leaf surface. They had a large plasmid. The restriction analysis showed that they were another kinds of Ti plasmic compared with C58 and Ach5. The isolated strains were identified as the nopaline type and also as biovar 1 A. tumefaciens, according to their tumor morphology, blophyslcal and biochemical characteristics. One of the isolated strains, AK204 was transformed with binary vector (pGA642), having selectable marker (Kmr, Tcr). Furthermore, maize tissue cells were transformed by cocultivation with AK204/pGA642, and the transformants were selected on the selective medium and identified using PAGE patterns of their soluble proteins.

  • Some morphological and anatomical characteristics of four Abies species in Korea were investigated to find the characteristics and the taxonomical relationship between the species. The results were summarized as follows: There was no significant difference in needle thickness and number of hypodermic cell layer among 18 morphological and anatomical characteristics of cone and needle between the species. A. koreana and A. nephrolepis were similar in above fourteen characters and A. holophylla and A. firma were also similar in the 14 characters. A. koreana and A. firma showed similarity in above two characters. Main resin canals of needles in A. koreana and A. nephrolepis were situated in leaf margin (L-LM), those of A. holophylla, in L, L-M and M, and that of A. firma fully in other locations except L.

  • Two naturalized allien species are reported from Jeju Island: Papaver setigerium collected from the ground along the road in lowland Veronica serpyllifolia collected from the roadside and ranches on alt. 400-800 m. P. setigerium is similar to P. somniferum, but different from the latter by plant height of 30-70 cm, shorter discoid stigma than fruit in diameter, somewhat sparsely bristly on stems, leaves and calyxes. V. serpyllifolia is similar to V. tenella in morphology, but different from the latter by short plant height, petiolated leaves on lower stem and serrated leaf margins.

  • Two Unrecorded species from Jeju Province are reported: Maesa japonica of Maesaceae which is unrecorded family and Cardamine tanakae of Brassicaceae. Monotypic family Maesaceae, is distinguished from Myrsinaceae and Primulaceae by having pedicels with a pair of bracteoles, flowers without staminodia, semi-inferior ovary and indehiscent fruits with many seeds. M. japonica is readily distinguishable from M. salicifolia in China and M. montana var. formosana in Taiwan and Japan by shapes of blades, corolla and bracteoles, the presence/absence of serrulation of leaf margins, ratio of length of corolla tube and corolla lobes, feature and color of fruit surface. C. tanakae is most similar to Korean endemic C. glechomifolia in morphology, however, the two taxa are readily distinguishable by the number of leaflets of cauline leaves, size of terminal leaflets, number of flowers per inflorescence, the presence of whitish hairs on ovary and fruits.

Morphology of Silica bodies on Cyperaceae in Korea (한국산 사초과(Cyperaceae) 식물의 규소체(Silica body) 형태에 의한 분류)

  • Many plants take up soluble silica from the soil, and they deposit in plant tissues. Silica bodies are most commonly found in the leaf epidermis. Silica body is important factor in growth and development of plant. It help to maintain rigidity in stem and linear leaves, and its content may be correlated with resistance to fungal disease. In this study, several morphologically distinctive forms of silica bodies on Cyperaceae in Korea are recognized, which are eventually classified into two groups. Silica body type of first group (Rhynchospora, Fuirena, Scleria, Carex, Eriophorum) is A-type, which is conical in shape. Silica body type of second group (Cyperus, Scirpus, Eleocharis, Fimbristylis, Kyllinga, Bulbostylis, Lipocarpha) is B-type, which is conical in shape with numerous satellite body. These types are consistent enough to use as characters in taxonomic studies within genus. Silica body type may be associated with habitat: the species in dry region have A-type, whereas the species in wet region have B-type. These results suggested that shapes and presence or absence of silica body might be valuable to systematic analysis.

Comparison of the morphology and distribution of the genus Megaleranthis Ohwi with those of its relative genera (Ranunculaceae) (모데미풀속과 근연속들의 형태 및 분포 비교 (미나리아재비과))

  • To elucidate the taxonomic status of the genus Megaleranthis Ohwi, a monotype in Korea, its distribution and morphological characteristics were studied and a cluster analysis was performed. The cluster analysis showed that Megaleranthis was separately clustered from Trollius and Calathodes. Megaleranthis is morphologically different from Calathodes Hook.f. et Thomson in having petals, and from Trollius L. in having an involucre instead of a caulescent leaf under the flower and follicles with simple veins. The three genera above are distributed independently within Asia, although they can be found together at the boundaries where their distributions overlap: Trollius and Calathodes occur together between the Sichuan and Yunnan provinces in China, and Megaleranthis and Trollius can both be found on the northern edge of the Korean Peninsula. From both a morphological and a distributional viewpoint, Megaleranthis should be recognized as an independent genus different from both Trollius and Calathodes.

First record of Sorbaria kirilowii (Rosaceae-Spiraeoideae) from Korea (쉬땅나무속(장미과-조팝나무아과)의 미기록 식물: 좀쉬땅나무)

  • We report an unrecorded species of the genus Sorbaria (Rosaceae) in Korea, namely Sorbaria kirilowii (Regel & Tiling) Maxim. This plant was discovered both on Pocheon-si, Naecheon-myeon and Yeoju-gun, Buknae-myeon in Gyeonggi Province. It is distinguishable from Sorbaria sorbifolia (L.) A. Braun var. stellipila Maxim. by characteristics such as presence of hair on leaf and rachis, morphology of inflorescence, length and width of petal, shape of sepal, number and length of stamens, presence of hair upon and size of follicles. This taxon was given the Korean common name 'Jom-swi-ttang-na-mu' based on the small size of floral characters.
