The Effects of Supplementation of Ascidian Tunic Shell into Laying Hen Diet on Egg Quality (산란계 사료에서 우렁쉥이껍질 첨가가 계란 품질에 미치는 영향)

  • This study was conducted to evaluate the feeding value of ascidian tunic shell the effects of its dietary supplementation on laying performance, egg-yolk pigmentation, egg-shell strength and egg taurine content. A total of 168 brown layers at the age of 29wks in commercial cage were fed for 4 wks with 7 different diets containing ascidian tunic shel1(AST) at varying levels of 0$\sim$5% Dm or 0% AST with 100ppm carophyll red. No differences were found in egg production and weight among the treatments indicating that ascidian tunic shell did not adversely affect the laying performances. Adding the ascidian tunic shell to the diets increased egg-yolk pigmentation compared to the control and resulted in simillar or better effect on egg-yolk pigmentation compared to 100ppm carophyll red. The data suggest that ascidian tunic shell may be used as feed ingredients in layer diet enrichment of egg-yolk pigmentation in the place of carophyll red(chemical pigment). Specific gravity and breaking strength of egg shell were significantly increased by the adding ascidian tunic shell to the diet, suggesting that ascidian tunic shell may be used as feed ingredients for increasing egg shell strength. Also taurine content of egg was significantly increased with increasing supplementation of ascidian tunic shell to the diet(p<0.05). Therefore, ascidian tunic shell may be used as feed ingredients in laying hen diet to improve egg quality such as egg-yolk pigmentation, egg-shell strength and egg taurine enrichment.

Swell Effect Correction for the High-resolution Marine Seismic Data (고해상 해저 탄성파 탐사자료에 대한 너울영향 보정)

  • The seismic data quality of marine geological and engineering survey deteriorates because of the sea swell. We often conduct a marine survey when the swell height is about 1 ~ 2 m. The swell effect correction is required to enhance the horizontal continuity of seismic data and satisfy the resolution less than 1 m. We applied the swell correction to the 8 channel high-resolution airgun seismic data and 3.5 kHz subbottom profiler (SBP) data. The correct sea bottom detection is important for the swell correction. To detect the sea bottom, we used maximum amplitude of seismic signal around the expected sea bottom, and picked the first increasing point larger than threshold value related with the maximum amplitude. To find sea bottom easily in the case of the low quality data, we transformed the input data to envelope data or the cross-correlated data using the sea bottom wavelet. We averaged the picked sea bottom depths and calculated the correction values. The maximum correction of the airgun data was about 0.8 m and the maximum correction of two kinds of 3.5 kHz SBP data was 0.5 m and 2.0 m respectively. We enhanced the continuity of the subsurface layer and produced the high quality seismic section using the proper methods of swell correction.

Three-dimensional magnetotelluric surveys for geothermal development in Pohang, Korea (포항지역 지열 개발을 위한 3 차원 자기지전류 탐사)

  • A three-dimensional (3D) magnetotelluric (MT) survey has been carried out to delineate subsurface structures and possible fractures, for development of low-temperature geothermal resources in Pohang, Korea. Quite good quality MT data could be obtained throughout the survey region by locating the remote reference in Kyushu, Japan, which is ${\sim}480\;km$ from the centre of the field site. 3D modelling and inversion are performed taking into account the sea effect in MT measurements near the seashore. The nearby sea in the Pohang area affects MT data at frequencies below $1\;Hz{\sim}0.2\;Hz$, depending on the distance from the seashore. The most severe sea effects were observed in the south-east parts of the survey area, closer to Youngil Bay. 3D inversion with and without the seawater constraint showed very similar results at shallow depths, roughly down to 2 km. At greater depths, however, a strong sea effect seems to form a fictitious conductive structure in ordinary 3D inversion, especially in the south-eastern part of the survey region. Comparison between drilling results and the resistivity profiles from inversions showed that five layered structures can be distinguished the subsurface beneath the target area. They are: (a) semi-consolidated mudstones with resistivity less than $10\;{\Omega}m$, which are ${\sim}300\;m$ thick in the northern part and ${\sim}600\;m$ thick in the southern part of the survey area; (b) occasional occurrence of trachybasalt and lapilli tuff within the mudstone layer has resistivity of a few tens of${\Omega}m$, (c) intrusive rhyolite ${\sim}400\;m$ thick has resistivity of several hundreds of ${\Omega}m$, (d) alternating sandstone and mudstone down to 1.5 km depth shows resistivity of ${\sim}100\;{\Omega}m$, (e) a conductive structure was found at a depth of ${\sim}3\;km$, but more geological and geophysical study should be carried out to identify this structure.

  • This study was to determine the fluctuation in phytoplankton assemblages with regarding to environmental conditions and nutrients, which were surveyed around Mokpo waters in the southwestern waters, Korea. Sampling was carried out on the Mokpo, Sinan, and Wando coasts from March to November 2003. The maximum sea surface temperature was recorded in August, and it ranged around $25^{\circ}C$ regardless of sampling sites. However, salinity in Mokpo waters showed a great variation, which ranged from 5-30 psu and recoded the minimum of 5 psu in July and the maximum of 30 psu in November. Moreover, in Mokpo waters, the chlorophyll a and SS concentration of the surface layer were also the highest values of $20\;{\mu}g\;l^{-1}\;and\;40\;{\mu}g\;l^{-1}$, respectively than those of Sinan and Wando waters. The concentrations of $NH_4-N,\;NO_2-N,\;NO_3-N,\;and\;PO_4-P$ were also he highest values of $0.018\;{\mu}mol\;^l{-1},\;0.062\;{\mu}mol\;l^{-1},\;1.2\;{\mu}mol\;l^{-1}\;and\;0.078\;{\mu}mol\;l^{-1}$, respectively in Morpo waters than those of Sinan and Wando waters. During the period of this study, the majority of the taxa were diatoms; Thalassiosira rotula, Rhizosolenia setigera, Prorocentrum minimum, Chaetoceros curvisetus, Leptocylindrus danicus, Pseudonitzschia pungens, and Chaetoceros spp. were detected in the dominant species of phytoplankton. The dinoflagellates were relatively abundant during the summer season in Wando waters, which attained an abundance of $10-20\%$. In Mokpo waters, DIN/DIP was the highest value of 700 in March, whereas the lowest was shown in Wando waters. However, DIN/DIP value in summer at Wando waters was extremely reversed, which appeared to be associated with the development of dinoflagellates. On the bais of factor analysis using SYSAT 6.0, nutrient showed somewhat correlation with chlorophyll a. Consequently, the process of discharge of fresh water in Mokpo waters plays an important role in extremely fluctuation in nutrients and conditions. Although Wando waters maintains a lack of nutrients, it should be influenced by different water current and may be associated with a concentration of nutrients.

Studies on Lipid and Fatty acid Composition of Korean Perilla Leaves(Penilla frutescens var. japonica HARA) (한국산 들깻잎의 지방질 및 지방산조성에 관한 연구)

  • The difference in content and composition of total lipid, lipid fractions and fatty acids of perilla leaves being used fresh vegetable was investigated in relation to the color of undersurface, i.e. green perilla leaves(GPL) and violet perilla leaves(VPL), by column- and thin layer- and gas chromatography. Total lipid(TL) content was of little difference between green leaves(GPL) (5.24%dw) and violet one (VPL) (5.02%dw), while neutral lipid(NL) content was higher In VPL(36.4% of TL) than GPL(34.7%). The major components were sterol ester and hydrocarbons(58.5%) and trigylcerides(14.9%) in NL, $mono-(42{\sim}45%)$ and $di-(13{\sim}15%)$ galactosyl digylceride in glycolipids(GL) and phosphatidyl ethanolamine$(40{\sim}45%)$ and phosphatidyl glycerol(13%) in phospholipids(PL) for both GPL and VPL. The number of component was 10 in all three fractions. The similarity of component between GPL and VPL was in decreasing order of NL(r=1.00), GL(r=0.997) and PL(r= 0.968). Major fatty acids were linolenic $(62{\sim}64%)$, palmitic$(10{\sim}12%)$ and linolic$(9{\sim}10%)$ for TL, linolenic, palmitic, myristic(43, 15, 14%) for NL, linolenic, oleic, palmitic(79, 11, 8%) for GL and linolenic, linoleic, palmitic(36, 25, 23%) for PL. Unsaturated fatty acid percentage was higher only in GL of VPL than GPL. The similarity of fatty acid composition between GPL and VPL was least in PL and so it was among other fraction with PL.

The Influence of Field Compaction Method on the Efficiency of Compaction and the C. B. R. Value (전압방법(轉壓方法)이 전압효과((轉壓效果) 및 C. B. R. 값에 미치는 영향(影響))

  • This study is intended to find out the degree of compaction in field compaction, with soil type, the thickness of soil layer and the number of roller passing through the field density test and the field C. B. R. test by comparing vibration and non-vibration compactions. The results in this study are summarized as follows. 1. When the number of roller passing is few, it shows that the efficiency of the compaction by vibratory compaction is greater than that by non-vibratory compaction, the difference of the compaction ratio between vibratory and non-vibratory compaction is decreased according as the number of roller passing is more frequent. 2. Mechanizing on a large scale it is possible for a large equipment to be able to reach the point of A-1 compact ion method with three to five times of roller passing. To provide for mechanizing on a larger scale it is advisable to fix the standard by the D-2 compaction method. 3. As dry-density increases, the C. B. R. value increases, but the increasing ratio of C. B. R. value showed greater in vibrating compacting. 4. According as the number of roller passing increases, the increasing tendency of the C. B. R. Value is slow and the difference of the C. B. R. value between vibration and non-vibration compaction is large, the C. B. R. value showed greater about 20% in vibration compaction than in non-vibration compaction. 5. In C-5 soil type, with increasing the thickness of compaction, the degree of compaction is decreased. When the thickness of compaction is increased from 20cm to 30cm, the degree of compaction is decreased slowly, while the thickness of the compaction is increased from 30cm to 40cm the degree of compaction is decreased remarkably. Therefore it is advisable to compact the ground under the thickness of 30cm.

  • The calcium-binding proteins (CaBP), parvalbumin (PV) and calbindin-D 28K (calbindin) are particularly abundant and specific in their distribution, and present in different subsets of neurons in many brain regions. Although their physiological roles in the neurons have not been elucidated, they are valuable markers of neuronal subpopulations for anatomical and developmental studies. This study is designed to characterize dorsal lateral geniculate nucleus (dLGN) neurons and axon terminals in terms of differential expression of immunoreactivity (IR) for two well-known CaBPs, PV and calbindin. The experiments were carried out on 6 adult monkeys. Monkeys were perfused under deep Nembutal anesthesia with 2% paraformaldehyde and 0.2% glutaraldehyde in 0.1M phosphate buffer. After removal, the brains were postfixed for 6-8 hr in 2% paraformaldehyde at $4^{\circ}C$ and infiltrated with 30% sucrose at $4^{\circ}C$. Thereafter, they were frozen in dry ice. Serial sections of the thalamus, at $20{\mu}m$, were made in the frontal plane with a sliding microtome. The sections were stained for PV and calbindin with indirect immunocytochemical methods. For electron microscopy, after infiltration with 30% sucrose the blocks of thalamus were serially sectioned at $50{\mu}m$ with a Vibratome in the coronal plane and stained immediately by indirect ABC methods without Triton X-100 in incubation medium. Stained sections were postfixed in 0.2% osmium tetroxide, dehydrated and flat-embedded in Spurr resin. The block was then trimmed to contain only a selected lamina or interlaminar space. The dLGN proper showed strong PV IR in fibers in all laminae and interlaminar zones. Particularly dense staining was noted in layers 1 and 2 that contain many stained fibers from optic tract. Neuronal cell body stained with PV was concentrated only in the laminae. In these laminae staining was moderate in cell bodies of all large and medium-sized neurons, and was strong in cell bodies of some small neurons together with their processes. Calbindin IR was marked in the neuronal cell body and neuropil in the S layers and interlaminar zones whereas moderate in the neuropil throughout the nucleus. Regional difference in distribution of PV and calbindin IR cell is distinct; the former is only in the laminae and the latter in both the S layer and interlaminar space. The CaBP-IR elements were confined to about $10{\mu}m$ in depth of Vibratome section. The IR product for CaBP was mainly associated with synaptic vesicle, pre- and post-synaptic membrane, and outer mitochondrial membrane and along microtubule. PV-IR was noted in various neuronal elements such as neuronal soma, dendrite, RLP, F, PSD and some myelinated or unmyelinated axons, and was not seen in the RSD and glial cells. Only a few neuronal components in dLGN was IR for calbindin and its reaction product was less dense than that of PV, and scattered throughout cytoplasm of soma of some relay neurons, and was also persent in some dendrite, myelinated axons and RLP. The RSD, F, PSD and glial elements were always non-IR for calbindin. Calbindin labelled RLP were presynaptic to unlabeled dendrite or dendritic spine and PSD. Calbindin-labeled dendrite of various sizes were always postsynaptic to unlabeled RSD, RLP or F. From this study it is suggested that dLGN cells of different functional systems and their differential projection to the visual cortex can be distinguished by differential expression of PV and calbindin.

Structural Features of the Glandular Trichomes in Leaves of Carnivorous Drosera anglica Huds. (식충식물 긴잎끈끈이주걱 (Drosera anglica Huds.) 분비모의 구조적 특성)

  • Carnivorous plants vary in their unique features of morphology, ultrastructure and biochemical properties by species. Furthermore, prey-capturing mechanism as well as structural and physiological adaptations have been used for grouping various carnivorous species. In Drosera plants, glandular trichomes, which develop in the leaf epidermis, are known to play the most important role during the prey capturing process. The present study examined such trichomes, focusing on the glandular type, in leaves of Drosera anglica using scanning and transmission electron microscopy. Three types of rudimentary glandular trichomes were found to develop within the folded leaf primordia and immature leaf during early development. The first type, stalked glandular trichomes (Type I), occurred on the margin and upper epidermis of the leaf. With maturation, the longest glandular trichomes having lengthy stalks, ca. $2.2{\sim}5.1\;mm$, developed along the margin, while shorter stalked trichomes, ca. up to $200\;{\mu}m$, were found on the inner leaf blade. The shorter ones consisted of a globose head having two layers of secretory cells, parenchyma bell cells and tracheids and a multicellular stalk. The stalks gradually decreased in length in centripetal fashion. The second type, Type II, having ca. $15{\sim}30\;{\mu}m$ short stalks, also developed along the inner blade. Both types secreted mucilage from the secretory cells which had a thin cell wall and cuticle layer. The sessile six-celled glandular trichomes were the third type, Type III, and were $25{\sim}40\;{\mu}m$ in length. They were distributed most commonly throughout the upper and lower epidermis, petiole and even on the stalk surfaces of the first two types of trichomes. The third type was also found to be involved in the active secretion. In prey capturing leaves, all trichome types secreted substances through thin cuticles in the head cell wall, which exhibited relatively loose wall components.

Effect of Nandrolone Decanoate on Disuse Muscle Atrophy and Bone Beating in Dogs (개에서 불용성 근위축과 골절 치유에 대한 Nandrolone decanoate의 효과)

  • Anabolic steroid compounds are widely used for the increase of muscle mass, density of bone and athletic ability. The present study conducted to evaluate the effects of nandrolone decanoate (ND), one of the anabolic steroid compounds, on disuse muscle atrophy and healing process of bone in dogs. Twenty physically healthy dogs of both sexes were used in this experiment and divided into three groups: group A (control), group B (low dose M-1.5 mg/kg) and group C (high dose ND-7.5 mg/kg). One-mm strip of full thickness bone was excised from the radius below the pronator teres muscle for the artificial fracture and then the fractured ends were fixed in apposition with bone plate leaving 1mm gap, and finally immobilized externally by Robert John's bandage for 4 weeks. ND was administered intramuscularly once a week far 8 weeks. Body weight, muscle mass change and fracture gap of the bone were evaluated immediately after surgery, and 2, 4, and 8 weeks after surgery. The rates of muscle mass change 8 weeks after surgery were $-2.75\pm0.16\%,\;1.68\pm0.11\%\;and\;1.74\pm0.48\%$ in groups A, B and C, respectively. The significant increase (p<0.05) of muscle mass increments were found in the treated groups. The fibrous connective tissue layer in the fracture gaps of the treated groups increased more than the control, especially in the group C at 4th week. More dense fibrous connective tissue were found in the treated groups at 8th week. Collectively, our results suggested that ND was an effective anabolic agent for the immobilized disuse muscle and bone healing.

Assessment on the Content of Heavy Metal in Orchard Soils in Middle Part of Korea (중부지역 과수원 토양중의 중금속 함량 평가)

  • Objectives of this study were to monitor the distribution of heavy metals, to compare extractable heavy metal with total content and to investigate the relationships between soil physico-chemical properties and heavy metals in orchard soil. Sampling sites were 48 in Gyeonggi, 36 in Gangwon, 36 in Chungbuk, and 44 in Chungnam, Soils were collected farm form two depths, 0 to 20 and 20 to 40 cm (here after referred to as upper and lower layers) from March to May in 1998. Total contents of heavy metal in soils were analyzed by ICP-OES after acid digestion ($HNO_3$:HCl:$H_2O_2$) whereas extractable contents were measured after successive extraction of 0.1N-HCl, 0.05 M-EDTA, and 0.005 M-DTPA. Mercury was analysed by mercury atomizer. The average contents of Cd Cu, and Pb in the extractant with 0.1N-HCl at upper layer were 0.080, 4.23, and 3.42 mg/kg, respectively. As content in the extractant with 1N-HCl was 0.44 mg/kg, and total contents of Zn, Ni and Hg were 78.9, 16.1, and 0.052 mg/kg, respectively. The ratios of concentrations of heavy metals to threshold values (Cd 1.5, Cu 50, Pb 100, Zn 300, Ni 40, Hg 4 mg/ke in Soil Environmental Conservation Act in Korea (2001) were low in the range of $1/2.5{\sim}1/76.9$ in orchard soils. The ratios of extractable heavy metal to total content ranged $5.4{\sim}9.21%$ for Cd, $27.9{\sim}47.8%$ for Cu, $12.6{\sim}21.8$% for Pb, $15.8{\sim}20.3%$ for Zn, $5.3{\sim}6.3%$ for Ni, and $0.7{\sim}3.6%$ for Zn, respectively. Cu and Pb contents in 0.05 M-EDTA extractable solution were higher than those in the other extractable solution. Total contents of Cd, Ni and Ni in soils were negatively correlated with sand content but positively correlated with silt and clay contents. Ratios of extractable heavy metal to total content were negatively correlated with clay content but ai and Ni contents were positively correlated with soil pH, organic matter, and available phosphorous. Therefore, the orchard soil was safe because the heavy metal contents of orchard soil were very low as compared to its threshold value in the Soil Environmental Conservation Act. However, it need to consider the input of agricultural materials to the agricultural land for farming practices for assessment of heavy metals.
