Damage Analysis of Korean White Pine Stands in which the Black-tipped Sawfly was Chemically Controlled (잣나무넓적잎벌 방제림분(防除林分)에 대(對)한 잣나무 피해해석(被害解析))

  • To obtain basic information for establishing a pest control strategy for insect pest management system, changes in the population densities of the black-tipped sawfly(Acantholyda posticalis posticalis Matsumura) and damage patterns in tree growth were investigated in national forests in Hoigok-ri, Kapyung-gun, Kyunggi-do, where the pest control measures were taken. The results obtained were as follows ; 1. The larval density in the soil of the forests where the insecticides were applied have been kept below economic threshold for about 7 years. The density was the highest in the middle of slopes and similar to the level of the early stage of the insect outbreak. 2. After the pest control by insecticides, reduction in tree height and diameter growth lasted for 2-3 years in trees defoliated by over 70%. 3. The diameter growth of the trees damaged by black-tipped sawfly recovered faster in upper stem than in the lower. 4. volume growth of the trees defoliated over 70% by the insect decreased for three to four years. The volume loss of trees defoliated by 70% and 90% was 19.6% and 54.0%, respectively. 5. Maintaining the rate of defoliation below 50%, which is the economic threshold, by chemical control measures had an effect of reducing the tree volume loss by $40m^2/ha$ as compared with a stand defoliated by 90%.

Cytotoxic Effects of Tenebrio molitor Larval Extracts against Hepatocellular Carcinoma (갈색거저리 유충 추출물의 간암세포에 대한 세포독성 효능)

  • Various natural products or their derivatives, mostly originating from plants, fungi, and bacteria, have been exploited as therapeutic drugs to treat various human diseases. In addition to previously explored organisms, research on natural compounds has now expanded into unexamined living organisms in order to identify novel bioactive substances. Here, we determined whether or not the larval form of the mealworm beetle Tenebrio molitor, a species of darkling beetle, contains cytotoxic substances that exclusively affect cancer cell viability. Ethanol extract and its solvent partitioned fractions, hexane and ethyl acetate fractions, showed anticancer effects against various human cancer cells derived from the prostate (PC3 and 22Rv1), cervix (HeLa), liver (PLC/PRF5, HepG2, Hep3B, and SK-HEP-1), colon (HCT116), lung (NCI-H460), breast (MDA-MB231), and ovary (SKOV3). Cell death induced by the fractions was a mix of apoptosis, necrosis, and autophagy. The hexane fraction was administered intraperitoneally to nude mice bearing a hepatocellular carcinoma SK-HEP-1 and showed inhibition of tumor growth in vivo. Therefore, we concluded that worm extracts contain cytotoxic substances, which can be enriched by proper fractionation protocols, and further separation and purification could lead to the identification of novel molecules to treat human cancers.

Characteristics of Enterobacteria from Harmonia axyridis and Effects of Staphylococcus spp. on Development of H. axyridis (무당벌레(Harmonia axyridis ) 장내세균의 특성 및 Staphylococcus spp. 장내세균이 무당벌레의 발육에 미치는 영향)

  • Enterobacteria were isolated in the gut of the predacious multicolored Asian ladybird beetle, Harmonia axyridis, and their effects to the development of H. axyridis were examined. Populations of H. axyridis in this experiment were collected from Kimjae at Cheonbuk province (JK population), Geumsan at Chungnam province (CK population) and laboratory population at Laboratory of Insect Physiology in Chungnam National University, Daejeon. Thirty-four enterobacteria isolates were purified and isolated from the digestive tract of H. axyridis, and a total of 4 strains were classified into group by analysis of 16S rRNA gene sequences. About 70% of total isolates were phylogenetic groups of Bacillus genus and Staphylococcus genus, and they were commonly separated from the digestive tract of H. axyridis. After investigating their susceptibility against antibiotics with 18 representative enterobacteria isolates, ofloxacin and penicillin were selected for examination in this study of their ability to inhibit the growth of all of isolates. In order to remove the enterobacteria from the aphids, ofloxacin and penicillin were given to the green peach aphid, Myzus persicae, and the turnip aphid, Lipaphis erysimi. These aphids were provided to H. axyridis as prey. The weight of pupa, developmental periods of each larval instar, the number of eggs and their hatching ratio of H. axyridis with treatment aphids were lower compared with non-treatment aphids. Staphylococcus saprophyticus is a representative enterobacteria and commonly isolated from the digestive tract of H. axyridis. In the absence of S. saprophyticus, the developmental periods of each larval instar increased; however, the weights of pupa, the number of eggs, and their hatching ratio decreased.

  • Gametogenes, reproductive cycle, first sexual maturity(biological minimum size), sex ratio and larval development of the marsh clam Corbicula japonica were investigated monthly by histological observations. Samples were collected in brackish water of Gokgang stream, Kyungsangbuk-Do, Korea, from August 1997 to July 1998. Sexuality of Corbicula japonica is dioecious and the species are an oviparous clam. The gonads are irregularly arranged from the sub-region of mid-intestinal gland in visceral cavity to reticular connective tissue of foot. The ovary is composed of a number of ovarian sac which are branched arborescent. Oogonia actively proliferate along the germinal epithelium of ovarian sac, in which young oocytes are growing. The testis is composed of a number of testicular tubules, and the epithelium of the tubule has function of germinal epithelium, along which spermatogonia actively proliferate. A great number of undifferentiated mesenchymal tissue and eosinophilic granular cells are abundantly distributed between developing oocytes and spermatocytes in the early developmental stages. With the further development of the ovary and testis these tissue and cells gradually disappear. Then the undifferentiated mesenchymal tissue and eosinophilic granular cells are considered to be related to the growing of the oocytes and spermatocytes. The spawning period is from July to September, and the main spawning occur between July and August when seawater temperatures reach above 22$^{\circ}C$. The reproductive cycle of this species can be divided into five successive stages; early active (February to April), late active (May to July), ripe (June to September), partially spawned (July to September), degenerative (September to October) and resting stage (October to February). Percentages of first sexual maturity of female and male clams ranging in length from 10 mm to 12 mm are over 50% and 100% for clams over 16.0 mm in shell length. Fertilized eggs or Corbicula japonica were 80-90 ${\mu}{\textrm}{m}$ in diameter. In the early embryonic development of C. japonica, the appearance of polar body, trochophore and D-shaped veliger were observed around 40 min., 27 hours and 4 days after spawning, respectively, at a water temperature of 26.5-28.$0^{\circ}C$. The size of larvae of early umbo stage was about 185-210 ${\mu}{\textrm}{m}$ in shell length, 160-180 ${\mu}{\textrm}{m}$ in shell height around 7 days after fertilization. The correlation of relative growth between the culture day (D) and shell length (SL) was expressed by the following simple formula from D-shaped veliger to metamorphosing stage; SL = 13.300D + 209.36($r^2$= 0.9078).

  • Clownfish are important and very popular fish in the ornamental aquarium industry. Demand for the fish is increasing dramatically. The present study was conducted to verify methods of broodstock management, patterns of spawning, rates of egg hatching and estimates of larval growth fur the saddleback clownfish, Amphiprion polymnus. Spawning occurred 8 times between August 2002 to June 2004 with 2 females and 1 male participating. Fertilized eggs were separated by an adhesive matrix and were oval in shape. The eggs were $2.46{\pm}0.13mm$ in size as measured along the longest axis. The percentage of fertilized eggs was 96.7%. Hatching was observed seven days post-spawning and hatching rate was 85.5%. The sizes of the newly-hatched larvae were $4.58{\pm}0.21mm$ TL (total length). Larvae had an open mouth and anus, and an oval yolk sac. At the 1 st day after hatching, the sizes of the larvae were $4.90{\pm}0.35mm$ TL. The larvae began to eat rotifers after complete yolk absorption. On the 5th day post-hatch, larvae were $5.88{\pm}0.31mm$ TL with complete fins and the survival rate was 48.6%. At 8 days after hatching, a band began to appear on head and back of the larvae indicating the beginning of metamorphosis. Metamorphosis was completed at an average TL of $15.00{\pm}2.12mm$ on the 23rd day after hatching. By the 45th day after hatching, juveniles averaged $22.76{\pm}3.22mm$ TL and survival rate was 28.4%.

Egg Development and Morphology of Larva and Juvenile of Liparis tanakae in the Coastal Waters off Yeosu (여수 연안산 꼼치(Liparis tanakae)의 난발생 및 자치어 형태발달)

  • This study aims to contribute to the research on resource recovery for the rapidly declining population of Liparis tanakae by observing the larval development process and the morphology of juveniles based on their growth. Natural spawning eggs collected in Yeosu were used for observing the process of egg development and larval morphology. The water temperature during the rearing process was maintained at 12.3~13.5℃ (average 12.7℃). The fertilized eggs had an egg diameter ranging from 1.57 to 1.79 mm (average 1.71 mm) and were spherical and adhesive. Within 4 hours 35 minutes after fertilization, they reached the two-cell stage, and after 74 hours 10 minutes, the formation of the yolk sac began. At 106 hours post-fertilization, a caudal fin appeared at the tail tip. Hatching began at 526 hours, and the larvae developed with the yolk sac positioned just behind the eyes. The newly hatched larvae had both the mouth and anus open. Melanophores appeared inside the lower jaw and around the tail on the third day after hatching. By the 16th day after hatching, most of the yolk was absorbed, and melanophores were visible in the head region. Finally, on the 63rd day after hatching, the head region significantly developed, and the body shape and mouth were similar to those of an adult fish, signifying the transition to the juvenile stage. This study will serve as valuable data for aquaculture techniques related to the conservation and restoration of fish species based on the hatching and juvenile morphology of Liparis tanakae.

Toxicity Evaluation of Chemicals using Tree Frog Embryos, Hyla japonica (청개구리 배아를 활용한 화학물질의 독성평가 연구)

  • In this experiment, I investigated toxicity evaluation of chemicals using domestic frog embryos, along FETAX (Frog Embryo Teratogenesis Assay-Xenopus) protocol. I investigated $Cu^{2+}$ and Tebuconazole effect on the tree frog, Hyla japonica, embryos by probit analysis. Mortality and malformation rates increased and larval body length decreased depending on the concentrations of $Cu^{2+}$ and Tebuconazole. The teratogenic concentration ($EC_{50}$) of $Cu^{2+}$ and Tebuconazole were 0.05, 5.0mg/${\ell}$, respectively and the embryo lethal concentration ($LC_{50}$) of $Cu^{2+}$ and Tebuconazole were 0.16, 38.5, respectively. The teratogenic index (TI) appeared 3.0 in $Cu^{2+}$ and 7.7 in Tebuconazole, which showed teratogenicity in embryonic development of Hyla japonica. These results reveal that $Cu^{2+}$ and Tebuconazole in this experiment suppressed the development of embryos at relatively low concentration. Much of Hyla japonica embryos can be secured, and easy to incubate. In addition, mortality, malformation ratios, malformation patterns and growth rates are similar to the results from the other assay systems. Therefore, the Hyla japonica embryo teratogenesis assay system could be a useful tool to evaluate toxicity of pollutants in environment.

Morphological Development of Eggs and Larvae of the Pacific Cod, Gadus macrocephalus (대구(Gadus macrocephalus)의 난 발생 및 자어 형태발달)

  • Embryonic and larval developments on Pacific cod, Gadus macrocephalus are described from laboratory-reared specimens. The eggs (1.075~1.168 mm in diameter) were weakly adhesive, almost spherical in shape, transparent and unpigmented, showing a pale yellow yolk without oil globules. Hatching was occurred 220 hrs after fertilization at $9.8^{\circ}C$. The newly hatched larvae (3.72~4.40 mm in total length, TL) had 43~47 (11~13+32~34) myomeres, mouth and anus which was not yet opened. The yolk was completely absorbed at 4.58 mm TL, and transformed to postlarval stage. Locations of the anus and second dorsal fin origin, and distinctive growth of the larvae head were observed at 5.48 mm TL with melanophores in the gill cover and upper region of the occipital. In addition, melanophores were distinctive in the ventral region and caudal fin base at 6.88 mm TL. All fins were formed with a complete set of fin rays having the following counts (D. 3~7-12~14-13~14; A. 13-15) at 11.54 mm TL. Both lateral line and barbel of lower jaw were observed at 15.07 mm TL.

Developmental Changes in Digestive Organ and Digestive Enzyme Activity of Filefish Thamnaconus modestus (말쥐치 Thamnaconus modestus 자치어의 성장에 따른 소화기관 및 소화효소 발달)

  • Larvae and juveniles of the filefish Thamnaconus modestus were reared for 64 days after hatching (DAH) in order to determine the activity of four enzymes (trypsin, pepsin-like enzyme, lipase, amylase) during ontogeny. Larvae were fed on rotifer Brachionus plicatilis from 2 to 26 DAH, Artemia nauplii from 10 to 64 DAH, and then gradually changed to pelleted feed from 40 DAH. Temperature was kept between $21.5{\sim}24.2^{\circ}C$ Activity of trypsin and lipase was found in larvae 4 DAH ($6.0{\pm}1.4unit$) and 6 DAH ($4.5{\pm}1.4unit$), respectively. The evolution of activity in both enzymes showed a profile marked by drastic increases between late larval and early juvenile stages. Pepsin-like enzyme activity was found at 10 DAH and drastically increased from 28 DAH, corresponding with the early juvenile stage of T. modestus. Interestingly, developmental changes in the pepsin-like enzyme activity coincided well with increases in the number of gastric glands. Amylase activity was found at 10 DAH and was maintained at a low level up to 28 DAH, followed by a drastic increase from 28 DAH to 40 DAH. It might be concluded that a drastic increase in trypsin and pepsin-like enzyme activities, and a corresponding increase in the number of gastric glands reflects a higher somatic growth of T. modestus during the early juvenile period.

Evaluation of Three Species of Diatoms for Rearing Larvae of Sea Urchin, Strongylocentrotus intermedius (북쪽말똥성게, Strongylocentrotus intermedius 유생에 대한 3종 규조류의 먹이효과)

  • In order to evaluate three species of diatoms as the live food sources for rearing larvae of sea urchin, Strongylocentrotus intermedius, six different feed experiments were conducted for 35 days. We measured sizes, survival rates on elapsed day after hatching and required days of each larval stages of sea urchin in the different feeding experiments. Sizes of the larvae of sea urchin cultured with Isochrysis + Chaetoceros, Isochrysis, Chaetoceros and Pavlova were $735{\mu}m$, $718{\mu}m$, $701{\mu}m$ and $642{\mu}m$, respectively. The highest daily growth rates of the larvae of sea urchin showed Isochrysis within 6 armed and Chaetoceros after 6 armed. Survival rates of the larvae of sea urchin cultured with Chaetoceros, Isochrysis + Chaetoceros, Isochrysis and Pavlova were $37.2\%,\;35.0\%,\;32.4\%,\;and\;18.4\%$, respectively. The required days from fertilization to the 8 armed (Senior) stage in the larvae of sea urchin, fed Chaetoceros, Isochrysis, Paviova, chaetocerost+pavlova, pavloua + Isochrysis and Isochrvsis + chaetoceros were 24, 27, 33, 28, 30 and 25 days, respectively.
