  • Pectobacterium carotovorum subsp. carotovorum is the causative agent of soft rot in crops such as potato and cabbages. Loop-mediated isothermal amplification (LAMP) is a simple DNA amplification method, as well as isothermal PCR technique. In this study, a new method for the rapid detection of Pectobacterium carotovorum subsp. carotovorum was developed using LAMP that named PCC-LAMP. Based on lytic murein transglycolase gene of Pectobacterium carotovorum subsp. carotovorum, a set of four primers for LAMP was designed. The optimal PCC-LAMP reaction temperature was established at $61^{\circ}C$. Under standard conditions, PCC-LAMP amplified $1{\times}10^3$ copies of clone PCC-pBX437 per reaction. Further, this method can also assay directly by SYBR Green I without electrophoresis. Amplification was not detected for five other bacterial species. In conclusion, PCC-LAMP may be a useful method for the detection Pectobacterium carotovorum subsp. carotovorum in the field.

  • The primary step for efficient control of viral diseases is the development of simple, rapid, and sensitive virus detection. Reverse transcription loop-mediated isothermal amplification (RT-LAMP) has been used to detect viral RNA molecules because of its simplicity and high sensitivity for a number of viruses. RT-LAMP for the detection of Potato virus X (PVX) was developed and compared with conventional reverse transcription polymerase chain reaction (RT-PCR) to demonstrate its advantages over RT-PCR. RT-LAMP reactions were conducted with or without a set of loop primers since one out of six primers showed PVX specificity. Based on real-time monitoring, RT-LAMP detected PVX around 30 min, compared to 120 min for RT-PCR. By adding a fluorescent reagent during the reaction, the extra step of visualization by gel electrophoresis was not necessary. RT-LAMP was conducted using simple inexpensive instruments and a regular incubator to evaluate whether RNA could be amplified at a constant temperature instead of using an expensive thermal cycler. This study shows the potential of RT-LAMP for the diagnosis of viral diseases and PVX epidemiology because of its simplicity and rapidness compared to RT-PCR.

  • LED lighting, considered to be a new growth industry, has attracted a great deal of attention due to its higher illumination and longer life time than existing light sources. In this study, high-efficiency and long-life LED fog lamps for automobiles were developed, which can substitute the existing 27 W halogen fog lamps for a used car market. For this purpose, the number of LED modules, the body, heat sink, and the output of the fog lamp were first optimized through a numerical analysis. Then, a 10 W-class LED fog lamp was prototyped based on the optimized numerical model, and the performance of the fog lamp was successfully verified through the experiments.

  • In this paper we present the method that improve alarm security system using the electronic circuits added the emergency lamp by our proposed Algorithms. In conventional method the emergency lamp of warning & alarm system was not executed perfectly its performance because of battery with short life. For improving this problem we, using Peripheral Interface Controller IC, designed the circuit added the emergency lamp for an warning & alarm system and for prevent from stopping the electric current, and compared our proposed method with conventional method. By designing the circuit to stop up over charge we can extend life of battery, use of a stoppage of electric current in emergency and according to the lightness around. Therefore we are very convenient and profitable in our life. In this future we will study the method to lower the cost of architecture for practical utilization.

  • The number of tunnel has fast increased with the rapid expansion of highway network. Tunnel should be designed to provide for drivers both safety and pleasant driving conditions. In this perspective, the design for tunnel lightning is very important in order to provide its safety, pleasantness, and cost-efficiency of maintenance, all of which should be considered and analyzed for a better tunnel lighting. This paper attempts to compare the low-pressure sodium lamp, which have usually been used for tunnel lighting, with the fluorescent lamp, which we consider as an alternative for the former. In an effort to determine the number of lamps to meet the required illuminance in the tunnel, this research employs a simulation technique which would allow us to conjecture, with the aid of basic model, the life cycle cost for illumination per each tunnel. This analysis is expected to provide a basic method and related information for tunnel development and design.

  • Loop-mediated isothermal amplification (LAMP) technique relies on autocycling strand displacement DNA sysnthesis without template denaturation steps under isothermal conditions. LAMP has more advantages than modern PCR, as it requires only basic laboratory equipment like an isothermal water bath, oven, and heating cabinet. Hence, in this study, five random primers were designed with Streptococcus parauberis, shikimate kinase Arok gene sequences (Genbank accession number: CP0024711). Two primers were selected based on the ladder pattern. Other optimum reaction conditions like temperature, time, and sensitivity were established and confirmed with conventional SYBR-green PCR. Results confirmed that the designed random primers were species specific, without any non-target DNA amplification under isothermal conditions. Hence, this study suggests that LAMP techniques could be used in the diagnosis of fish pathogen, specifically S. parauberis.

  • Electrodeless discharge lamps have been objects of interest and research for several decades, mainly because of their potential for extremely long life, high lamp efficacies and smaller than standard fluorescent lamps size. A series of measurements and observations concerning variables has yielded optical and electrical characteristics for electrodeless discharge lamp like incandescent lamp and circular lamp feature. Last experiment were carried out to determine the lamp temperature at several surface points during operation. Light output levels in excess over 60,000 cd/$m^2$ have been measured in electrodeless discharge lamp for a general of conditions. At lamp surface temperatures have been measured over $80^{\circ}C$, and starting current have been measured over ${\sim}A$.

  • The purpose of this paper is to increase the lamp efficacy by filling the optimal Nitrogen gas pressure in the outer enclosure of metal halide lamp. This study presents a new model of Langmuir in the outer enclosure of discharge lamp. In this paper, in the first place, the diameter of Langmuir sheath and the total gab loss were calculated. Secondly, the each gas loss was computed if the arbitrary gas pressure is filled, With the calculated data, flash over which affects the lamp efficacy, lamp life and color rendering was considered. In case of the established discharge lamp, the gab pressure has been filled by experience only roughly. The gas loss is converted into the radiation output. Therefore, the lamp efficacy is improved by reducing the gas loss.

  • In eukaryotes, RNA splicing, an essential biological process, is crucial for precise gene expression. Inaccurate RNA splicing can cause aberrant mRNA production, disrupting protein synthesis. To regulate splicing efficiency, some splicing factors are reported to undergo Ubiquitin-like Modifier (SUMO)ylation. Our data indicate that in Saccharomyces cerevisiae, the SUMO protease, Ulp2, is involved in splicing. In the ulp2Δ mutant, some ribosomal protein (RP) transcripts exhibited a significant increase in the levels of intron-containing pre-mRNA because of improper splicing. Moreover, we confirmed Ulp2 protein binding to the intronic regions of RP genes. These findings highlight a critical Ulp2 role in RP transcript splicing.
