  • An attempt has been made to investigate the possible occurrence of aflatoxins, a group of micotoxin which are toxic metabolites produced by Aspergillus flavus, and aflatoxin like substances in fungal fermented soybean products such as meju and soybean paste. Chloroform extracts from 15 samples which has been defated with petroleum ether are subjected to separations with thin-layer chromatography on silica gel washed with methanol prior to film coating, improves the chromatographic separation and ultraviolet absorption spectral identification. In addition to fluorescencing spot having an Rf value which is same as aflatoxin $B_1$, many of the fluorescencing spots have been occurred in every sample examined. Each fluorescencing spot is scratched and the ethanol extracts are subjected to further separation with thin-layer chromatography. Each fluorescence substance is eluted with ultra violet transparent absolute ethanol and the ultraviolet absorption spectra are checked. None of the absorption curve of eluates shsow accordance with the curve of aflatoxin. 2, 4-Dinitrophenyl hydrazine reagent, Tollen's reagent and ninhydrin reagent are applied on the chromatogram- The data show that aflatoxin is not present in any of the fungal preparations examined.

Quality Characteristics of Mealworm Seasoning Paste prepared by Chungukjang Recipe (청국장 제조 방법에 따른 갈색거저리 조미페이스트 개발 및 품질특성)

  • In this study, two kinds of mealworm seasoning pastes (naturally fermented or inoculation of Bacillus subtilis) were prepared by applying fermentation method of chungkukjang. The samples were labeled into soybean control (SC; no fermentation), mealworm control (MC; no fermentation), mealworm natural fermentation (MR) and mealworm inoculated fermentation of Bacillus (MB). Water content, color, pH, titratable acidity, salinity, amino acid nitrogen, reducing sugar and preference test were examined on the pastes. Moisture contents were 46.9 with soybean control (MC) and 54.5~73.3 with mealworm pastes. The L-value was not significantly different between the samples. In the case of the a-value and the b-value, SC showed the highest value whereas mealworm pastes showed lower values than SC. pH and salinity were higher in mealworm pastes. Amino nitrogen and reducing sugar showed the highest values of MR 326,67 and 63.33 mg%, respectively. In the preference test, the appearance and flavor were the highest values with SC, but texture, taste and overall acceptability were with MR, Therefore, soybean fermentation method with Bacillus species could be applied to use protein of mealworm and possibly edible insects as well.

Changes of Enzymatic Activities during the Fermentation of Soybean-Soypaste by Aspergillus spp. (Aspergillus spp.에 의한 콩된장 발효 과정중의 효소활성 변화)

  • This study was carried out as a preliminary test to investigate the improvement of soysauce and soybean paste for natural food. The soybean was treated on raw, soaked, roasted, and steamed condition and it was maked that rice koji was inoculated by Asp. oryzae, Asp. niger, Asp. awamori and on natural condition fermented. They were maked raw soybean-soypaste $(S_0)$, soaked soybean-soypaste $(S_1)$, roasted soybean-soypaste $(S_2)$, and steamed soybean-soypaste $(S_3)$ from soybean (60%), rice koji (30%) and salt (10%) respectively in order to investigate the changes of enzymes activity(amylase, protease, lipase and lipoxygenase activity) during fermentation of them. The results obtained were summarized as follows; Amylase activity was in the order of natural fermented microorganisms>Asp. oryzae>Asp. awamori>Asp. niger in the microorganisms, and $S_0>S_1>S_2>S_3$ in the soybean treatments. Protease activity was in the order of natural fermented microorganisms>Asp. niger>Asp. oryzae>Asp. awamori in the microorganisms, and $S_3>S_2>S_1>S_0$ in the soybean treatments. Lipase activity was a similar tendency in the microorganisms, but it was in the order of $S_0>S_1>S_3>S_2$ in the soybean treatments. Lipoxygenase activity was in the order of natural fermented microorganisms>Asp. oryzae>Asp. awamori>Asp. niger in the microorganisms, and $S_0>S_1>S_3>S_2$ in the treatments.

Metabolite Profiling and Microbial Community of Traditional Meju Show Primary and Secondary Metabolite Differences Correlated with Antioxidant Activities

  • Meju, a type of fermented soybean paste, is used as a starter in the preparation of various Korean traditional soybean-based foods. In this study, we performed Illumina-MiSeq paired-end sequencing for microbial communities and mass spectrometry analysis for metabolite profiling to investigate the differences between 11 traditional meju products from different regions across Korea. Even though the bacterial and fungal communities showed remarkable variety, major genera including Bacillus, Enterococcus, Variovorax, Pediococcus, Weissella, and Aspergillus were detected in every sample of meju. The metabolite profile patterns of the 11 samples were clustered into two main groups: group I (M1-5) and group II (M6-11). The metabolite analysis indicated a relatively higher amino acid content in group I, while group II exhibited higher isoflavone, soyasaponin, and lysophospholipid contents. The bioactivity analysis proved that the ABTS (2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)) radical-scavenging activity was higher in group II and the FRAP (ferric reducing antioxidant power) activity was higher in group I. The correlation analysis revealed that the ABTS activity was isoflavonoid, lipid, and soyasaponin related, whereas the FRAP activity was amino acid and flavonoid related. These results suggest that the antioxidant activities of meju are critically influenced by the microbiome and metabolite dynamics.

Quality Characteristics of Fermented Soybean Products by Bacillus sp. Isolated from Traditional Soybean Paste (전통장류 유래 Bacillus sp.를 이용한 콩 발효물의 품질 특성)

  • This study evaluated the quality characteristics of soybean fermented with several Bacillus sp., which were selected based on their high enzymatic and antimicrobial activities. Total aerobic bacterial counts of fermented soybeans with HJ5-2 ($3.00{\times}10^9CFU/mL$) were the highest among all strains. Lactic acid bacteria numbered $2.50{\times}10^2{\sim}7.30{\times}10^4CFU/mL$ in soybeans fermented with isolates. Amylase and protease activities of the RD7-7 sample were the highest among all strains. Reducing sugar and amino-type nitrogen contents of fermented soybeans with HJ18-4 (2.35%) and RD7-7 (227.96 mg%) were the highest. Total amino acid contents of the samples were 16.62~18.38%, and glutamic acid, aspartic acid, leucine, lysine, arginine were major amino acids. Oxalic acid (36.51~63.57 mg/100 g) and succinic acid (429.49~600.15 mg/100 g) were the predominant organic acid. These results provide useful information for development starter (single and complex) as well as for the production of high quality fermented soybean foods.

Isolation and Identification of Bacillus Strains with Antagonistic Properties against Film-forming Yeasts Overgrown in Low Salted Soybean Pastes (저염 장류에서 증식하는 산막 효모에 길항 작용을 갖는 Bacillus 균주의 분리)

  • Soybean pastes with 8% (w/w) salinity were prepared instead of soybean paste with 14% (w/w) salinity to meet the growing demands of Korean's low sodium diet. After aging, white films had appeared on the surface of all low-salted soybean pastes [8% (w/w) salinity] unlike high-salted soybean pastes [14% (w/w) salinity]. All of eight microbes isolated from the surface film were identified as Pichia kudriavzevii. Eleven Bacillus strains with good characteristics of fermentation were isolated from traditionally fermented soybean pastes in order to preserve their unique flavors and aromas after aging, and as a result of analyzing the biochemical characteristics and 16S rRNA sequences, those were identified as B. subtilis, B. licheniformis, and B. methylotrophicus. All of the Bacillus isolates had antagonistic activities against 8 isolates of the film-forming yeasts and harbored the genes for synthesis of antimicrobial surfactants including lichenysin and/or surfactin.

  • The studies on the color improvement of the Doenjang (two types of fermented soybean paste : soybean Doenjang and modified Doenjang) using various Aspergillus oryzae strains (6 strains : A, B, C, D, E, F) were conducted with the series of experiments of enzymatic activities (pretense, $\alpha$-and $\beta$-amylase), browning color formation (Lovibond color), major chemical components (amino nitrogen, reducing sugar and others) and sensory evaluation (color, taste and odor). Aspergillus oryzae 2157 (C strain) had a high potential for the color improvement of Doenjang products and was identified as non-browning strain during Doenjang fermentation and storage period. And also this strain was appeared to be in good enzyme activities and flavor characteristics.

Biochemical Characteristics of Whole Soybean Cereals Fermented with Aspergillus Strains. (Aspergillus속 균류들을 이용한 콩알메주 발효의 생화학적 특성)

  • Whole soybean cereal was fermented with four Aspergillus strains in pilot meju fermentation system. The pH range of the product was 7.40~7.98, the contents of reducing sugar and amino-nitrogen were 0.04~2.78%, 178~309 mg%, respectively and that of free fatty acid ranged 2.67~5.05%. The components of the amino acid, organic acid, free sugars and fatty acid showed distinctive patterns among four groups of fermented soybean cereals. Amylase activity and carbohydrate degradation rate of A. usami was higher than other strains. But protease and protein degradation rate, lipase and lipid degradation rate were similar in four strains. The odor concentrates of soybean cereals fermented with Aspergillus strains were different from Bacillus strains. Especially, pyrazine components, the main and common flavor chemicals in Bacillus strains, were not determined in this study and Aspergillus specific components were 9-methyl-acridine, dl-limonene and 2,3-butanediol. Soybean paste, made from A. oryzae fermented soybean cereal, showed excellent sensory evaluation.
