Bioconversion of progesterone by immobilized aspergillus phoenicis (고정화된 aspergillus phoenicis를 이용한 progesterone 전환)

  • Progestrone bioconversion by immobilized Aspergillus phoenicis was studied. Progesterone was converted into 11$\alpha$-hydroxyprogesterone and 3-minor byproducts. Whole cells of A. phoenicis were immobillized by enreappment with calcium-alginate, K-carrageenan, or polyacrylamide. Of these materials tested, cell immobilized in $Ca^{2+}$ -alginate gels showed the highest activity for 11$\alpha$-hydroxylation of progesterone. In the case of mycelia immobilized in $Ca^{2+}$-alginate, futher progressing hydroxylation of 11$\alpha$-hydroxyprogesterone was greatly reduced. Spores of A. phoenicis which were immobillized with $Ca^{2+}$-alginate and germinatedin situ for 25 hours showed higher 11$\alpha$-hydroxylase activity than those of entrapped whole mycelia and maintained initial enzyme activity for all 8 times of repeated use. After 16 times of reuse, the activity was declined 30% or more. When culture media and $Zn^{2+}$ were introduced into the reaction media, the activity of the immobilized mycelia which had been lowered due to many times of reuse was effectively reactivated.

Synthesis and Antiinflammatory Activity of Novel Indazolones

  • In this study, a series of new $N^2$ substituted 1,2-dihydro-3H-indazol-3-ones (3a-d) as well as their condensed pyrazolo, pyridazino derivatives such as pyridazino[1,2-a]indazole-6,9,11-triones (4a-h) and 3,9-dioxo-3H,9H-pyrazolo[1,2-a]indazole (7) were synthesized. The antiinflammatory activity of some synthesized compounds was determined by carrageenan-induced rat paw edema technique using diclofenac as reference drug. The pharmacological data showed that most of the tested compounds exhibited a significant long lasting antiinflammatory activity, which in the case of compound 3b was superior to that of diclofenac.

Anti-inflammatory and Analgesic Activity of Alnus japonica Cortex Ethanol Extract (오리나무 수피 엑스의 소염 및 진통 활성)

  • An extract of Alnus japonica (Betulaceae) cortex has been traditionally used for purifying blood, and curing feces containing blood, enteritis, diarrhea, alcoholism and cut wounds. Alnus japonica cortex extract significantly inhibited carrageenan-induced paw edema at 1, 2 and 3 hrs after oral administration by 32.8, 24.4, and 46.9%. It also inhibited acetic acid-induced vascular permeability in mice by 15.3 and 28.0% at oral doses of 500 and 1,000 mg/kg, and it significantly reduced the volume of hindpaw of adjuvant-induced arthritis rats at the day 6 and 10 by 22.5 and 18.7% after the sample administration. Alnus japonica cortex extract increased threshold on inflamed paw (Randall-Selitto assay) at 3 hr by 137.5% compared to the control group. It also reduced the number of writhing syndrome dose- dependently.

Effects of Milk Proteins and Gums on the Dough Characteristics and Staling of Bread Made from Frozen Dough during Storage (우유단백질과 검류가 밀가루 반죽의 특성과 냉동반죽으로 제조한 식빵 저장중의 노화에 미치는 영향)

  • Rheological properties of the dough added with milk proteins and gums was studied to investigate the possibilities as anti-staling agents. Also, physical properties of the resultant bread baked from the frozen dough after 8 weeks of storage at $-20^{\circ}C$ were examined. The 4 sets of their combinations of milk proteins and gums, $casein-{\kappa}-carrageenan$ (CK), casein-sodium alginate (CA), $whey-{\kappa}-carrageenan$ (WK), and whey-sodium alginate (WA), were added to dough to examine their possible anti-staling effects. Rheological properties of dough were evaluated, and physical properties of resultant bread baked from frozen dough after 8 weeks storage at $-20^{\circ}C$ were examined. Addition of all treatments increased gelatinization temperature and water absorption, and lowered miximum viscosities and extension of doughs, compared to the control. Doughs added with CA and WA showed longer development times than that of the control. Addition of WK and WA resulted in lowest dough extensions. Treated bread showed lower moisture content decrease during storage at $5^{\circ}C$ for 4 days. Breads baked with frozen doughs after 6 weeks storage at $-20^{\circ}C$ showed similar results. Although textural hardness of breads increased with storage at $5^{\circ}C$, CA- and WA-added breads were less affected, showing they effectively retarded staling of breads.

A Study on Preparation of Wanjajun for Cook/Chill System II Quality Characteristics of Wanjajun Containing Hydrocolloids (Cook/Chill System에서의 고기완자 제조에 관한 연구II. Hydrocolloid 첨가 고기완자의 품질 특성)

  • Low-fat, precooked high quality Wanjajun was prepared by treating natural antioxidant herbs and hydrocolloids for quantity production of cook/chill system in foodservice. The proposed Wanjajun was formulated under the consideration of flavor and fat level. Wanjajun was prepared with hydrocolloid such as sodium alginate, car-boxymethyl cellulose (CMC), carrageenan and xanthan gum. Cooking yield, fat retention and water holding capacity of Wanjajun containing hydrocolloidal gums were higher than those of control. L-value of raw product was low in comparison with cooked product, but a-value was higher than that of other cooked product. Colors of raw and cooked products were not different among five kinds of products. pH value of cooked product was higher than that of raw product pH range of cooked products revealed to 6.10~6.23. There was no considerable change in pH value during storage. The coliform group and Staphylococus aureus were not detected during storage. Total bacteria1 counts were close to microbial guidelines for safe condition. Hardness, springness, cohesiveness, gumminess and brittleness of low-fat products were generally lower than those of control product. Hardness, gumminess and brittleness of reheated products were higher than those of preheated products. Flavor, tenderness, juiciness and overall quality were not different between precooked and reheated products. Textural characteristics of products were superior in the order of sodium alginate, xanthan gum, CMC, carrageenan and control product.

Production of Bio-ethanol from Red Algae by Acid Hydrolysis and Enzyme Treatment (산 및 효소 가수분해를 이용한 홍조류로부터 바이오 에탄올 생산)

  • Bio-ethanol production research using various material has been problemed for solving problems of environment pollution caused by fossil fuels. Red-algae consists of agar, carrageenan, and porphyran. If it is treated by acid, it is able to change useful bio-mass for bio-ethanol. In this study, we found an optimal condition for bio-ethanol production from acid hydrolysate in red-algae. To produce bio-ethanol, Saccharomyces cerevisiae KCCM1129 inoculated to acid hydrolysate of Gelidium amansii. The optimal condition for Gelidium amansii hydrolysis was found to be 30 min reaction at $H_2SO_4$ concentration of 1.5% and $121^{\circ}C$. At this condition, its produced to 7.04 g/L galactose and 1.94 g/L glucose. And acetic acid concentration of 2.0% in agar produced 0.75 g/L galactose. In contrast, Pachymeniopis elliptica was treated with $H_2SO_4$concentration of 1.5%, it produced 6.38 g/L galactose. And Pachymeniopis elliptica treated with acetic acid concentration of 2% produced 0.368 g/L galactose. The optimal condition of ethanol production was found to be 96 h reaction at $H_2SO_4$concentration of 1.0% and $30^{\circ}C$, which produced 3.77 g/L ethanol.

Prevention of Arthritic Inflammation Using an Oriental Herbal Combination BDX-1 Isolated from Achyranthes bidentata and Atractylodes japonica

  • An oriental herbal combination (BDX-1) was isolated from Achyranthes bidentata and Atractylodes japonica. We previously tested the clinical effectiveness of BDX-1 in rheumatoid arthritis (RA) patients and found that it has a beneficial therapeutic effect. Here, we provide experimental evidence for the effectiveness of BDX-1 on RA in murine models. The oral administration of BDX-1 was found to markedly inhibit collagen-induced arthritis, adjuvant-induced arthritis, and zymosan-induced inflammation. It also inhibited carrageenan-induced acute edema and acetic acid-induced writhing response. In addition, the biological activity of BDX-1 was found to be strongly increased by fermentation. Our results suggest that BDX-1 could be useful for the treatment of rheumatoid arthritis.

Chemical Investigations and Anti-inflammatory Activity of Fixed Oil of Butea monosperma Seeds

  • The fruit and seeds of Butea monosperma (Lam) Kuntze (Fabaceae) are useful in piles, anthelmintic, eye diseases, and inflammation in the Indian system of medicine. Hence, we have evaluated the anti-inflammatory activity of the fixed oil, mixed fatty acids, and unsaponifiable matter of B. monosperma against carrageenan-induced paw oedema and cotton pellet-induced granuloma in rats. The fixed oil, mixed fatty acids, and unsaponifiable matter of the oil exhibited significant anti-inflammatory activity on the tested experimental animal models. The unsaponifiable matter of the oil produced higher protection compared to fixed oil and mixed fatty acids. Phytochemical analysis of the fixed oil revealed the presence of steroids and terpenoids while unsaponifiable matter of the oil showed the presence of ${\beta}-sitosterol$. Also, four fatty acids were identified in the fixed oil by gas liquid chromatography. The anti-inflammatory activity of the fixed oil may be due to unsaponifiable matter or combination of unsaponifiable matter and mixed fatty acids.

Effects of Sophoraflavanone G, a Prenylated Flavonoid from Sophora Flavescens, on Cyclooxygenase-2 and In Vivo Inflammatory Response

  • Previously, several prenylated flavonoids having a C-8 lavandulyl moiety were found to inhibit cyclooxygenase-1 (COX-1) as well as 5-lipoxygenase (5-LOX), and sophoraflavanone G was the most potent inhibitor against these eicosanoid generating enzymes among 19 prenylated flavonoids tested. In this investigation, effects of sophoraflavanone G on COX-2 induction from RAW 264.7 cells and in vivo inflammatory response were studied. Sophoraflavanone G inhibited prostaglandin $E_2{\;}(PGE_2)$ production from lipopolysaccharide (LPS)-treated RAW cells by COX-2 down-regulation at 1-50 uM. Other prenylated flavonoids including kuraridin and sanggenon D also down-regulated COX-2 induction at 10-25 uM, while kurarinone and echinoisoflavanone did not. In addition, sophoraflavanone G showed in vivo anti-inflammatory activity against mouse croton oil-induced ear edema and rat carrageenan paw edema via oral (2-250 mg/kg) or topical administration (10-250 ug/ear). Although the potencies of inhibition were far less than that of a reference drug, prednisolone, this compound showed higher anti-inflammatory activity when applied topically, suggesting a potential use for several eicosanoidrelated skin inflammation such as atopic dermatitis.

In Vivo Anti-Nociceptive and Anti-Inflammatory Effect of the Two Triterpenes, Ursolic Acid and 23-Hydroxyursolic Acid, from Cussonia bancoensis

  • Triterpenoids, ursolic acid (1) and 23-hydroxyursolic acid (2) were obtained from the hydrolysis of BuOH fraction of Cussonia bancoensis extract to test anti nociceptive and anti-inflammatory effect of C. bancoensis (Araliaceae). Compound 1 and 2 exhibited anti-nociceptive effects, which were determined by acetic acid-induced writhing test and hot plate test. The effect of 2 was much more potent in acetic acid-induced writhing test than in hot plate test. Compound 1 and 2 significantly inhibited 1%-carrageenan-induced edema in the rat. These results suggest that the two triterpenes, ursolic acid and 23-hydroxyursolic acid, are responsible for the antinociceptive and anti-inflammatory effect of C. bancoesnsis.
