Rapid Gas Chromatographic Profiling and Screening of Acidic Non-Steroidal Antiinflammatory Drugs in Biological Samples

  • The solid-phase extraction (SPF) with subsequent tert-butyldimethylsilyl (TBDMS) derivatization was investigated for the rapid profiling and screening of various carboxylated non-steroidal antiinflammatory drugs (NSAIDs) simultaneously in biological fluid samples. Compared to the conventional SPF in adsorption mode using Chromosorb 102, Chromosorb 107, Carbopak B and Thermosorb, the SPF in partition mode using Chromosorb P as the adsorbent, and ethyl acetate/methylene chloride as the eluting solvents provided hightest overall recovenies of the NSAIDs from aqueous solutions with good precision. The solid-phase extracted NASIDs were silylated with N-methyl-N-(tert-butyldimethylsily)trifuoroacetamide to TBDMS derivatives and directly analyzed by capillary gas chromatography and gs chromatography-mass spectrometry. The usefulness of the present method was examined for the profilling and screening of saliva, serum and urine samples for various NSAIDs simultaneously.

Genomic Profiling of Liver Cancer

  • Development of liver cancers is driven largely by genomic alterations that deregulate signaling pathways, influencing growth and survival of cancer cells. Because of the hundreds or thousands of genomic/epigenomic alterations that have accumulated in the cancer genome, it is very challenging to find and test candidate genes driving tumor development and progression. Systematic studies of the liver cancer genome have become available in recent years. These studies have uncovered new potential driver genes, including those not previously known to be involved in the development of liver cancer. Novel approaches combining multiple datasets from patient tissues have created an unparalleled opportunity to uncover potential new therapeutic targets and prognostic/predictive biomarkers for personalized therapy that can improve clinical outcomes of the patients with liver cancer.

전기비저항 영상법에 의한 거제도의 지반조사 (Applicability of Resistivity Image Profiling to Geologic Survey in the Keoje-do Area)

  • Resistivity Image Profiling has been applied to a geological survey in the Keoje-do area. Survey lines are located near the KD-02 and KD-06 wells in the area, where we have already sampled all rock cores and carried out several kinds of geophysical logs. In each site a resistivity cross-section is obtained by inverting pole-pole apparent resistivities. Comparing the reconstructed resistivity section with the rock cores and logging data obtained in the well provides a detailed picture of subsurface geology. The geology of KD-02 site is composed of conglomerate, sandstone and shale with fractures. The reconstructed resistivity image is useful for assessing the grade of weathering of these rocks. The KD-06 site is mainly underlain by granitic rocks, and its fresh basement can be delineated by resistivities over $1000{\Omega}{\cdot}m$.

Distribution and Features of the Six Classes of Peroxiredoxins

  • Peroxiredoxins are cysteine-dependent peroxide reductases that group into 6 different, structurally discernable classes. In 2011, our research team reported the application of a bioinformatic approach called active site profiling to extract active site-proximal sequence segments from the 29 distinct, structurally-characterized peroxiredoxins available at the time. These extracted sequences were then used to create unique profiles for the six groups which were subsequently used to search GenBank(nr), allowing identification of ~3500 peroxiredoxin sequences and their respective subgroups. Summarized in this minireview are the features and phylogenetic distributions of each of these peroxiredoxin subgroups; an example is also provided illustrating the use of the web accessible, searchable database known as PREX to identify subfamily-specific peroxiredoxin sequences for the organism Vitis vinifera (grape).

  • The genome is almost identical in all the cells of the body. However, the functions and morphologies of each cell are different, and the factors that determine them are the genes and proteins expressed in the cells. Over the past decades, studies on epigenetic information, such as DNA methylation, histone modifications, chromatin accessibility, and chromatin conformation have shown that these properties play a fundamental role in gene regulation. Furthermore, various diseases such as cancer have been found to be associated with epigenetic mechanisms. In this study, we summarized the biological properties of epigenetics and single-cell epigenomic profiling techniques, and discussed future challenges in the field of epigenetics.

Gene Expression Profiling of Liver and Mammary Tissues of Lactating Dairy Cows

  • Gene expression profiling is a useful tool for identifying critical genes and pathways in metabolism. The objective of this study was to determine the major differences in the expression of genes associated with metabolism and metabolic regulation in liver and mammary tissues of lactating cows. We used the Michigan State University bovine metabolism (BMET) microarray; previously, we have designed a bovine metabolism-focused microarray containing known genes of metabolic interest using publicly available genomic internet database resources. This is a high-density array of 70mer oligonucleotides representing 2,349 bovine genes. The expression of 922 genes was different at p<0.05, and 398 genes (17%) were differentially expressed by two-fold or more with 222 higher in liver and 176 higher in mammary tissue. Gene ontology categories with a high percentage of genes more highly expressed in liver than mammary tissues included carbohydrate metabolism (glycolysis, glucoenogenesis, propanoate metabolism, butanoate metabolism, electron carrier and donor activity), lipid metabolism (fatty acid oxidation, chylomicron/lipid transport, bile acid metabolism, cholesterol metabolism, steroid metabolism, ketone body formation), and amino acid/nitrogen metabolism (amino acid biosynthetic process, amino acid catabolic process, urea cycle, and glutathione metabolic process). Categories with more genes highly expressed in mammary than liver tissue included amino acid and sugar transporters and MAPK, Wnt, and JAK-STAT signaling pathways. Real-time PCR analysis showed consistent results with those of microarray analysis for all 12 genes tested. In conclusion, microarray analyses clearly identified differential gene expression profiles between hepatic and mammary tissues that are consistent with the differences in metabolism of these two tissues. This study enables understanding of the molecular basis of metabolic adaptation of the liver and mammary gland during lactation in bovine species.

  • Objective : The purpose of this study was to investigate molecular basis of neuroprotective effect in total ginsenosides. After H202 induced neurotoxicity, gene expression profiling of SH-SY5Y neuroblastoma cells treated by total ginsenosides is analyzed. Method : After SH-SY5Y cells were cultured, they were damaged by H202 induced oxidative stress. After twenty four hours, experimental group is treated by total ginsenosides and control group is treated by 0.9% saline. A high density cDNA microarray chip is used to analyze the gene expression profiling of SH-SY5Y cells. The Significance Analysis of Microarray method is used for identifying genes on a microarray. Results : 1. According to the results of microarray experiment, 17 genes were up-regulated, 38 genes were down-regulated. 2. Expression of OPHNl, KTANl, ATM, PRKCE, MAPKs genes associated with cell proliferation, neural growth, and the prevention of apoptosis were increased. 3. Change of EPX gene was the greatest among all genes. EPX gene associated with oxidative stress, and tumor suppressor gene ADAM11 were decreased. Conclusion : According to this study, molecular basis of neuroprotective effect of total ginsenosides is as followings: the increase of gene expression associated with cell proliferation, neuron growth, the prevention of apoptotsis and decrease of gene expression associated with oxidative stress and tumor suppressor.

사례기반추론기법을 적용한 해킹메일 프로파일링 (Hacking Mail Profiling by Applying Case Based Reasoning)

  • 방어기법은 계속 진화하고 있지만, 여전히 이메일을 통한 APT 공격의 경우에는 탐지와 방어가 어렵다. 국내의 공공기관 및 많은 민간기업들이 지능적이고 지속적인 해킹메일 공격으로 인해 피해가 지속되고 있다. 본 논문에서는 최근 몇 년간 공공기관을 대상으로 유입된 실제 해킹메일들을 데이터베이스로 구축한 뒤, 해킹메일들의 특징을 추출한 뒤 해킹메일을 사례기반추론기법을 이용하여 유사한 사례들을 쉽게 탐색할 수 있도록 사례벡터를 설계하였다. 또한 해킹메일을 이용한 과거 공격사례들을 프로파일링 해 본 결과 특정 지역과 집단에 공격이 집중되었음을 확인할 수 있었다. 이를 통해 사례추론기법이 신규 이메일이 유입될 경우, 과거 해킹메일과의 유사성을 비교하여 악성 여부를 탐지하는데 응용할 수 있을 뿐 아니라, 침해사고 대응에 있어서도 효율적으로 활용 가능함을 제시하였다.

Gene Expression Profiling in Hepatic Tissue of two Pig Breeds

  • Microarray analyses provide information that can be used to enhance the efficiency of livestock production. For example, microarray profiling can potentially identify the biological processes responsible for the phenotypic characteristics of porcine liver. We performed transcriptome profiling to identify differentially expressed genes (DEGs) in liver of pigs from two breeds, the Korean native pigs (KNP) and Yorkshire pigs. We correctly identified expected DEGs using factor analysis for robust microarray summarization (FARMS) and robust multi-array average (RMA) strategies. We identified 366 DEGs in liver (p<0.05, fold-change>2). We also performed functional analyses, including gene ontology and molecular network analyses. In addition, we identified the regulatory relationship between DEGs and their transcription factors using in silico and qRT-PCR analysis. Our findings suggest that DEGs and their transcription factors may have a potential role in adipogenesis and/or lipid deposition in liver tissues of two pig breeds.

양산단층 남부 상천리 일대의 천부 전기비저항 및 VLF 탐사 (Shallow Eelectrical Resistivity and VLF Profiling at Sangchon-ri Area along the Southern Par of Yangsan Fault)

  • 단층의 존재 가능성과 파쇄대에 대한 지전기적 구조를 밝히기 위하여 양산단층 남부 상천리 지역에 대하여 전기 비저항 탐사(쌍극자 탐사)와 VLF 탐사를 수행하였다. 양산탄층 추정선을 가로질러 4개의 측선에 걸쳐 실시한 전기 비저항 탐사 결과 단층선을 경계로 주로 동측에는 고비저항대가 서측에는 저비저항대가 나타난다. 이는 수직운동성분이 포함된 주향이동 단층운동에 의해 동측 기반암 블록이 상대적으로 상승한 결과로 사료된다. 파쇄대와 밀접하게 관련되어 나타나는 VLF 자료의 0점 통과점과 지전기적 저-고 비저항대 경계부는 양산단층의 제 4기 단층운동 추정선과 거의 일치하게 나타난다. 저비저항대가 지표에서 거의 수직으로 나타나는 것으로 보아 본 연구 지역의 예상 단층선을 통과하는 양산단층은 단층 양측이 10m 이상의 기반암의 깊이 차이를 나타내며 고각의 경사를 지닌 단층으로 추정된다.
