A phytochemical study of Alphonsea cylindrica King (unreported) has led to the isolation of six alkaloids. The compounds were identified as kinabaline (1; azafluorenone alkaloid), muniranine (2), O-methylmoschatoline (3; oxoaporphine alkaloid), lysicamine (4), atherospermidine (5) and N-methylouregidione (6; 4, 5-dioxoaporphine alkaloid). The structures of the isolated compounds were determined based on the spectroscopic techniques and by comparison with data reported in the literature. Alkaloid 2 was isolated as a new derivative of azafluorenone while alkaloids 1, 3 - 6 were isolated for the first time from Alphonsea species. In addition, alkaloid 3 and 4 showed inhibition zone against Staphylococcus aureus, Pseudomonas aeruginosa and Bacillus cereus in disc diffusion test. The minimum inhibition concentration (MIC) values of lysicamine (4) against S. aureus, B. cereus and P. aeruginosa were found to be smaller than O-methylmoschatoline (3). Therefore, the reported antibacterial activity showed the potential of this plant as natural antibacterial agent and supported the documented traditional use of Alphonsea sp. in the treatment of diarrhea and fever.
The intestinal microflora of humans is an extraordinarily complex mixture of microorganisms, the majority of which are anaerobic microorganisms. The distribution of amylolytic microorganisms in the human large intestinal tract was investigated in various individuals of differing ages using anaerobic culture techniques. A large percentage of the amylolytic microorganisms present belonged to the Genus Bifidobacteria. The number of Bifidobacteria increased significantly at two years of age. Adults and children above 2 years old carried about $0.8{\times}10^9-2.0{\times}10^{10}$ colony forming units (CFU/gram) of amylolytic Bifidobacteria. Among these amylolytic Bifidobacteria, Int-57 was chosen for further studies. Between 65% and 85% of the amylase produced was secreted and the remaining amylase was bound to the cell wall facing the outside. Amylase production could be induced by starch in a stable form. When cells were grown on maltose or glucose, amylase production was much lower than on starch and amylase activity disappeared after 24 hours growth on these media. Partially purified enzymes showed optimum activity at a temperature of $50^{\circ}C$ and at an optimum pH of 5.5, respectively. Heat treatment at $70^{\circ}C$ for 30 minutes almost completely inactivated amylase. The hydrolysis products of starch were mainly maltose and maltotriose. Soluble starch, amylose, amylopectin, and $\gamma$-cyclodextrin($\gamma$-CD) were easily hydrolyzed. The rate of hydrolysis of $\alpha$-CD and $\beta$-CD was slower than that of $\gamma$-CD. Carboxymethyl cellulose, $\beta$-1, 3-glucan and inulin were not hydrolyzed.
As an effort to understand alienated gifted students, we investigated learning characteristics and learning tactics of a scientifically gifted student with economic difficulty and physical disadvantage. The student we studied is attending the Saturday Physics Class which is an after school science activity offered by our university. We adopted techniques of qualitative case study. Participant observation was carried out at the field and the interview was done with the participant, his mother, and his teacher of 5th grade. Field documents and self-reports were used to understand the student synthetically. As a result, learning characteristics of the participant could be summarized as a spontaneous learning which originated from the internal motivation and struggle for learning to overcome the sense of inferiority and isolation from the peers. The participant adopted a strategic method for learning to satisfy his learning desire given the circumstance of socioeconomic and physical disadvantage: the three tactics we found were various learning routes, meta-cognitive ability and fervent response.
Apple blotch, caused by Marssonina coronaria, induce early defoliation in apple and leading to critical economic losses in apple orchards in Korea. Since M. coronaria is difficult to culture, we developed isolation and cultural method. We collected M. coronaria isolates from Gyeongbuk Province and then constructed phylogentic tree based on ITS regions. As the results, phylogenetic relationship indicated that all Korean isolates formed a same cluster and closely related to Chinese isolates [1]. Ecological characteristic of M. coronaria have been observed in apple orchards which located in Gyeongbuk Province from 2011 to present. As the results, the typical apple blotch symptoms were observed from July, and then the infected leaves were discolored and formed acervuli on the leaves. After rainfall, severe infection of symptoms such as discoloration and early defoliation were continuously observed until October. Also overwintered conidia were observed in next March on the fallen diseased leaves [2]. In the last 5 years, ascopores of M. coronaria were not observed in apple orchards which were severely infected by M. coronaria in Korea. Thus, it is assumed that overwintered conidia could be a primary inoculum of M. coronaria. Meanwhile, apple blotch has long latent periods compare to other apple disease. During the latent period, early diagnosis of apple blotch is the most important to control the disease by spray fungicide. In this reason, we developed novel diagnostic method to detect M. coronaria during latent period using optical coherence tomography (OCT) and Loop-mediated isothermal amplification (LAMP) method [2, 3]. In this presentation, it will introduce ecological characterization of M. coronaria in Korea and unique detection technique of M. coronaria in apple. It will be helpful to develop new strategies to control apple blotch in Korea.

This paper describes a series of shaking table and pseudodynamic tests for evaluation of seismic performance of base-isolated structures subjected to various seismic earthquake inputs. The main objectives of this study are : (1) evaluation of the effectiveness of base-isolation systems for low-rise structures against severe seismic loads through shaking table tests, (2) verification of the substructuring pseudodynamic test method for the base-isolated structures in comparison with the shaking table test results. In the shaking table test, a quarter scaled three-story structure base-isolated by laminated rubber bearings is tested. In the pseudodynamic test, only the laminated rubber bearing s are tested using the substructuring technique, while the concurrent seismic responses of the superstructure are computed using on-line numerical integration. Comparison with the shaking table test results indicates that the substructuring pseudodynamic test method is very effective for determining the dynamic responses of the base-isolated structure.

Salmonellosis is one of the life-threating diseases of goat in Bangladesh. Therefore, the present study was designed to study the prevalence of Salmonellosis, and isolation and characterizations of the Salmonella spp. from apparently healthy and diarrheic goat. A total of 47 faces samples were collected from selected place and cultured onto different prescribed medium to isolate it. In this study, 12.76% (6/47) samples were found to be positive for Salmonella spp. During culture on SS agar medium, all of the Salmonella isolates produced round, smooth, opaque, translucent and black color colonies on SS agar media. All of the isolated Salmonella spp. fermented dextrose, maltose and mannitol with production of acid and gas but did not ferment sucrose and lactose. However, these isolates had showed Indole and Voges-Proskauer test negative, Methyl-Red test positive. All of these isolates were subjected to rapid plate agglutination test with polyvalent "O" (Poly 'O') and polyvalent "H" (poly 'H') antisera where positive agglutination were observed. They were highly sensitive to ciprofloxacin, spiramycin and gentamycin; moderately sensitive to oxytetracyline, streptomycin and amoxicillin; less sensitive to sulphamethoxazole and resistant to penicillin-G. Based on the present findings, it may be concluded that the investigated Salmonella spp. from goats might be S. typhimurium, S. enteritidis, S. brandenburg, S. salford, S. newbrunswick, S. newport or S. dublin. Further study will be needed, therefore it requires further characterization using other serological and molecular techniques.
This study was conducted to determine the herbicidal activity of herbicidal substances and identify them in bulbs of Lycoris flavescens. Methanol extract was purified by a series of chromatographic techniques including silica gel column chromatography, preparative TLC, and HPLC. The final HPLC gave two active fractions and two herbicidal substances were obtained. By GC/MS analysis, one was identified as galanthine (galanthan-1-ol) and the other was identified as montanine ($O^2$-methyl pancracine), an isoquinoline alkaloid. Montanine showed nearly 100% of growth inhibition on the shoot and root of barnyardgrass (Echinochloa crusgalli) seedlings at $20{\mu}g\;mL^{-1}$ as compared with the control. Meanwhile, methanol extract of L. flavescens bulbs showed only about 3.1% and 8.3% of growth inhibition on the shoot and root of rice cultivar, Hwayeongbyeo seedlings at $1,000{\mu}g\;mL^{-1}$ as compared with the control, respectively.

The growth of the older population is expected to further increase social problems associated with population aging, such as isolation, poverty, and depression. The emerging issues associated with the older population are also expected to provide further momentum on studies about the dwelling environment as factors that ensure the health of older people as well as improve their quality of life. Therefore, approaches for explaining the issues of the older age group should be diversified using a variety of factors and appropriate analytic tools. Studies on measuring depression have principally focused on assessing an objective self-report questionnaire, usually in a highly structured, textual form which may not reflect the cognitive impairment of older adults. The aim of this study was to define and measure dwelling depression among older adults in Korea. There are two specific hypotheses in this study as follows: (a) there will be statistically significant relationships with dwelling dissatisfaction and depression, and (b) dwelling depression tools containing text and images will be, respectively, assessment tools that have a good construct with content validity and reliability. In the first experiment, to define and measure dwelling depression, 301 people over 65 years old living in single and two-person households were surveyed using a text-based dwelling depression questionnaires from September 1-30, 2017. In the second experiment, to examine whether the projective image questionnaire could serve as a suitable replacement for the text-based questionnaires, the same participants were surveyed from January 22 to February 2, 2018. The results show that depression has a close correlation with dwelling dissatisfaction. In addition, the geriatric dwelling depression index (GDDI) based on the projective image was refined. Additionally, the projective image questionnaire has a close correlation with the text-based questionnaire. Finally, through ROC curve analysis, it was found that the projective image questionnaire can accurately predict a depression group. To this end, this preliminary study examined the validity of the projective image questionnaire in older adults to make this instrument feasible for older populations and to contribute to a profound understanding of geriatric depression due to the living environment. We hope they will provide a basis for further research on psychological diagnoses using projective images.

Background: Withania somnifera (Solanaceae), generally known as Indian ginseng, is a medicinal plant that is used in Ayurvedic practice for promoting health and longevity. This study aims to identify the bioactive metabolites from Indian ginseng and elucidate their structures. Methods: Withanolides were purified by chromatographic techniques, including HPLC coupled with LC/MS. Chemical structures of isolated withanolides were clarified by analyzing the spectroscopic data from 1D and 2D NMR, and HR-ESIMS experiment. Absolute configurations of the withanolides were established by the application of NMR chemical shifts and ECD calculations. Anti-adipogenic activities of isolates were evaluated using 3T3-L1 preadipocytes with Oil Red O staining and quantitative real-time PCR (qPCR). Results: Phytochemical examination of the roots of Indian ginseng afforded to the isolation of six withanolides (1-6), including three novel withanolides, withasilolides GeI (1-3). All the six compounds inhibited adipogenesis and suppressed the enlargement of lipid droplets, compared to those of the control. Additionally, the mRNA expression levels of Fabp4 and Adipsin, the adipocyte markers decreased noticeably following treatment with 25 µM of 1-6. The active compounds (1-6) also promoted lipid metabolism by upregulating the expression of the lipolytic genes HSL and ATGL and downregulating the expression of the lipogenic gene SREBP1. Conclusion: The results of our experimental studies suggest that the withasilolides identified herein have anti-adipogenic potential and can be considered for the development of therapeutic strategies against adipogenesis in obesity. Our study also provides a mechanistic rationale for using Indian ginseng as a potential therapeutic agent against obesity and related metabolic diseases.

The physical properties of biomaterials are important for their isolation and separation from body fluids. In particular, the precise evaluation of the multi-physical properties of single biomolecules is essential in that the correlation between physical and biological properties of specific biomolecule. However, the majority of scientific equipment, can only determine specific-physical properties of single nanoparticles, making the evaluation of the multi-physical properties difficult. The improvement of analytical techniques for the evaluation of multi-physical properties is therefore required in various research fields. In this study, we developed a motion-tracking algorithm to evaluate the multi-physical properties of single-nanoparticles by analyzing their behavior. We observed the Brownian motion and electric-field-induced drift of fluorescent nanoparticles injected in a microfluidic chip with two electrodes using confocal microscopy. The proposed algorithm is able to determine the size of the nanoparticles by i) removing the background noise from images, ii) tracking the motion of nanoparticles using the circular-Hough transform, iii) extracting the mean squared displacement (MSD) of the tracked nanoparticles, and iv) applying the MSD to the Stokes-Einstein equation. We compared the evaluated size of the nanoparticles with the size measured by SEM. We also determined the zeta-potential and surface-charge density of the nanoparticles using the extracted electrophoretic velocity and the Helmholtz-Smoluchowski equation. The proposed motion-tracking algorithm could be employed in various fields related to biomaterial analysis, such as exosome analysis.
