소ㆍ돼지 도축지육 표면의 세균분포조사 (Survey on Microbiral Incidence of Meats in Slaughtered Cattles and Pigs)

  • Microbiological culture was conducted in the meat surface of cattles and pigs that was slaughtered in the Chung -Ju area and the result are as follows : 1. The number of bacteria in antemortem meat surface was higher ten times in March and ten to one thousand times in June to August compared with postmortem meat surface. 2. Microbes isolated in the meat surface, at the time of slaughter, was Stahylococcus spp., E. coli, Fungus and Streptococcus spp. 3. Bacteria was isolated in every parts of meat surface regardless to their location and many Fungus was isolated during summer.

국내 박물과 환경에 분포하는 미생물의 분리 (Distribution of Microorganisms in Domestic Museum Environments)

  • We isolated and identified microorganisms from the aerial environment of domestic museums. The fungi, Penicillium spp., Alternaria spp., and Cladosporium spp. were isolated in many museums. It seems that these fungi are related to biological degradation of textile remains. A total of 14 kinds of bacterial strains were isolated: Acinetobacter spp., Pseudomonas spp., Neisseria spp., Alcaligenes spp., Shigella spp., Klebsiella spp., Corynebacterium spp., Aerococcus spp., Bacillus spp., Micrococcus spp., Citrobacter spp., Erwinia spp., Salmonella spp., and Providencia spp. Acinetobacter spp., Pseudomonas spp., Neisseria spp., and Alcaligenes spp. were the predominate bacteria found in samples with a variety of bacteria. This suggests that there is a relationship between bacteria and the damage of textile remains. In the museum, we isolated Alternaria spp, Geotrichum spp., Penicillium spp. Acinetobacter spp., Pseudomonas spp., Alcaligenes spp. from the entrance, exhibit hall and storage, but they were found in smaller number and species in the exhibit cases and paulownia cases. We concluded that paulownia cases were not influenced by the microorganisms because of quality of care provided by the museum staff. Corynebacterium spp., and Bacillus spp. were not detected at the entrance and exhibit hall but were detected in paulownia cases. It is presumed that those bacteria did not flow in from outside, but resulted from contaminants in paulownia cases. In the distribution of microorganisms associated with textile remains, more fungi were detected than bacteria. Acinetobacter spp., Pseudomonas spp., and Neisseria spp., were isolated from silk items. Penicillium spp. and Cladosporium spp. were isolated in the silk and hump items. Aspergillus spp. and Penicillium spp. were isolated from the cotton items. On the other hands, there were no fungi strains in the wool items. Most of the isolated strains from textile remains were aerial microorganisms from the museum environment. These results suggest that textile remains were apt to contaminated by contact with the air.

군산부근 조간대에서의 해양종속영양 세균의 계절적 분포와 특성 (Seasonal Distribution and Characteristics of Heterotrophic Marine Bacteria in the Intertidal Zone Near Kunsan of Yellow Sea, Korea)

  • Annual distribution of hiterotrophic marine bacteria and seasonal characteristics were investigated in the intertidal waters and sediments in the vicinity of Kunsan of Yellow Sea, Korea. Annual distribution of heterotrophic marine bacteria ranged from $ 7.5*10^{2}$ to $1.1*10^{5}$ c.f.u./ml in water and from $1.6*10 ^{4}$ to $4.8*10^{6}$ c.f.u.per g dry sediments. As for the morphological distribution measured by epifluorescence microscopy, rod-form bacteria were distributed more than 74% of total observed bacteria during surveying periods. Average biovolume of sampled bacteria ranged from $3.19\;+-\;0.59*10^{-2}$ to $6.19\;+-\;0.76*10^{-2}$ $\mu$$m^{3}$ for coccus-form bacteria. Isolated bacteria showed various utilization of carbon sources such as glucose, maltose, lactose, xylose and arabinose, and showed tolerance at various range of salinity. It was isolated 82 genus in seawater and 114 genus in sediments. Dominant genera were Pseudomonas, Vibrio, Flavobacterium and Acinetobacter in seawater, and Pseudomonas, Acinetobacter, Vibrio, and Mycobacterium in sediments.

A Study on the Rapid Hydrolysis of Fish Using Proteolytic Bacteria Isolated from Anchovy Jeotkal

  • A study on the hydrolysis of anchovy using proteolytic bacteria isolated from anchovy jeotkal (a salt-fermented fish) was carried out to develop a rapid process of liquefied anchovy jeotkal. Five kinds of proteolytic bacteria, such as Staphylococcus sp.-l, Photobacterium sp., Volcaniella sp., Staphylococcus sp.-2 and Bacillus sp., were isolated from the anchovy jeotka1 that fermented with $20\%$ NaCl at room temperature for 2 months. Those grew well at $40^{\circ}C$, pH 7.0 on TPY broth with $2.0\%$ NaCl. The optimal hydrolysis temperature, pH, time and proteolytic bacteria densities for hydrolysis of minced anchovy were$40^{\circ}C$, 7.0, 6 hours and $1.8\times10^8$ cells/g raw anchovy, respectively.

물김치로부터 분리된 유산균의 항미생물 활성 (Antimicrobial Activities of Lactic Acid Bacteria Isolated from Mul-Kimchi)

  • 전남 목포시의 각 가정에서 담근 물김치에서 총 24균주의 유산균을 분리하였다. 이들 분리 유산균 중 5균주가 인공 위액에서 내성을 나타내었다. 이들 유산균은 Lactobacillus plantarum으로 동정하였고, 특히 물김치로부터 동정된 유산균 MUL-2, MUL-4, MUL-18은 인공위액에서 3시간 배양 후에도 초기 생균수에 비해 변화가 거의 없어 가장 강한 내성을 나타내었다. 이들 분리 유산균은 병원성 미생물인 Listeria monocytogenes, Escherichia coli, Staphyrococcus aureus, Vibrio parahaemolyticus, Samonella typhimurium에 대해 뚜렷한 항미생물활성을 나타내었다.

Pathogenic bacteria causing rot in commercial soybean sprout cultivation

  • Soybean sprout pathogenic bacteria were isolated from the large, deep containers of a commercial factory. Over a period of one year, 40 pathogenic-like bacteria were isolated among a total of 732 isolates. In addition to bacteria previously reported to be associated with rotting, such as Pseudomonas putida and Erwinia carotovora, several other genera were also identified: Acinetobacter spp., Chryseobacterium spp., Klebsiella sp., Pantoea agglomerans, Bacillus sp. Fatty acid methyl ester (FAME) analysis using the Microbial ID (MIDI) system, and 16s rRNA sequence analysis, yielded identical results, confirming the identities of these microorganisms. Several types of selective media were not good for identification and determination of population structure in commercial environments, as colony type was not specific to the genus. There was no dominant bacterium, and we were not able to find the main bacterium responsible for soybean spout rot. Even though we did not identify a major target for controlling rot or screening for resistant cultivars, the results of this study indicated that bacterial rot of soybean sprout is endemic. In addition, it emerged that factory epidemics in summer are not caused by the bacteria isolated in this study.

Susceptibility of Oral Bacterial to Sophoraflavanone G isolated from the Root of Sophora flavescens

  • Objective: The aim of this work is to investigate the antibacterial activity of the Sohporaflavanone G isolated from Sophora flavescens (S. flavescens), as the development of microbial resistance to antibiotics make it essential to constantly look for new and active compounds effective against pathogenic bacteria. Method : Sophoraflavanone G was isolated from the dried roots of Sophora flavescens Aiton (Leguminosae) by bioassay?guided fractionation. We investigated the effect of sophoraflavanone G on oral bacterial at various concentrations after incubation of 24 h in strains in the dose?dependent manner. Results: The structure of active compound, Sophoraflavanone G having a lavandulyl group at C?8, was elucidated on the basis of spectral data especially 1H?NMR and I3C?NMR. The antimicrobial activity showed that Sophoraflavanone G exhibited antimicrobial activilies against all the bacteria tested (MICs, 0.39 - 6.25 ㎍/ml). Sophoraflavanone G showed the strong antimicrobial activity against all the facultative bacteria and microaerophilic bacteria (MICs, 0.78 - 1.56 ㎍/ml) and also Sophoraflavanone G showed the strong antimicrobial activity against obligate anaerobic bacteria (MICs, 0.39 - 6.25 ㎍/ml).

Isolation and Identification of Noble Lactic Acid Bacteria

  • In this study, noble strains of lactic acid bacteria were isolated and identified by genetic analysis of 16s rRNA. Also, pH-dependent growth curve, cholesterol assimilation ability and sugar production efficiency were measured. Lactic acid bacteria were identified to inhabit in the milks from various animals. Results of sequence analysis showed that there were differences in 16S rRNA sequence among strains and part of gene deletion was also recognized. Growth rates were varied, too, depending on the pH of the medium. Lactobacillus rhamnosus LOCK908 isolated from cow milk showed the highest growth rate and high cholesterol assimilation ability. Results of sugar fermentation tests were relatively consistent with the sequencing results. So, we propose newly isolated Lactobacillus rhamnosus LOCK908 as useful candidate for a starter of fermented beverage and probiotics. Results of this study will contribute to the isolation and identification of noble Lactic acid bacteria and to the public health.

A report of 31 unrecorded bacterial species isolated from freshwater

  • A total of 31 bacterial strains were isolated from the Geum River basin in the Republic of Korea during our investigation of indigenous prokaryotic species. The isolated bacterial strains had high 16S rRNA gene sequence similarity (>98.7%) with those of validly published bacterial species, which have not been reported in Republic of Korea. The 31 bacterial strains were phylogenetically diverse and assigned to 4 phyla, 8 classes, 18 orders, 21 families, and 27 genera. At the genus level, the unreported species were affiliated with Kineococcus, Pedococcus, Rhodoluna, Salinibacterium, Rhodoluna, Arthrobacter, Williamsia, Nakamurella, Nocardioides of the class Actinobacteria, Patulibacter of the class Thermoleophilia, Pontibacter, Hymenobacter of the class Cytophagia, Flavobacterium of the class Flavobacteriia, Geomicrobium of the class Bacilli, Brevundimonas, Gellertiella, Rhizobium, Paracoccus, Taonella, Sphingomonas of the class Alphaproteobacteria, Burkholderia, Polaromonas, Hydrogenophaga, Chitinilyticum, Azospira, Zoogloea of the class Betaproteobacteria, and Pseudomonas of the class Gammaproteobacteria. The unreported bacterial species were further characterized by examining their morphological, cultural, physiological, and biochemical properties. The detailed descriptions of the 31 bacterial strains were provided.

후코이단과 후코이단 이용 박테리아의 멜론 성장 촉진 효과 검증 (Melon Growth Enhancement by Fucoidan and Fucoidan Decomposing Bacteria)

  • BACKGROUND: Marine algae is a productive organism that is consumed as a nutritious food. However, large amounts of unused portions of the algae are incinerated as trash or dumped in the sea, causing pollution. Recycling algae is important for saving resources and conserving the environment. In this study, the fucoidan which is a major carbohydrate of marine algae was tested as a source of fertilizer for farming. METHODS AND RESULTS: The growth rate of the melon was examined after treating fucoidan and the melon growth factors, weight and length of stem were measured. To discover the mechanism of melon growth promotion of fucoidan, bacteria that decomposed fucoidan were isolated from soil and abalone. Bacillus wiedmannii and Stenotrophomonas pavanii were isolated from terrestrial soil and Pseudomonas sp. was isolated from abalone. Among these three bacteria, Pseudomonas sp. had the highest and most specific fucoidan-decomposing activity. When Pseudomonas sp. was treated with fucoidan on melon-growing soil, the growth of melon was relatively improved compared to the treatment with fucoidan alone. CONCLUSION: We found that fucoidan, the main carbohydrate of marine algae, promoted melon growth. Fucoidan-decomposing microorganisms were isolated from terrestrial soil and marine organism, and we found that these bacteria stimulated the effect of melon growth promotion of marine algae. This is the first report that confirms the fertilizer effect of marine algae and shows the use of bacteria with marine algae.
