철 결핍 조건에서 배양된 Edwardsiella tarda의 면역학적 특성 (Immunological characteristics of Edwardsiella tarda grown under iron-restricted condition)

  • E. tarda의 독성 비교를 위하여 일반 TSB, iron-chelate인 2,2‘-dipyridyl 첨가 TSB 및 Fe 첨가 TSB의 세 가지 조건에서 E. tarda를 배양하였다. 독성시험의 결과는 iron-chelate 첨가 시험구의 E. tarda에서 가장 낮은 독성을 나타내었다. 그리고 각각의 OMPs와 IROMPs을 분리하여 SDS-PAGE로 항원성을 비교한 결과, iron-chelate 첨가 시험구의 E. tarda IROMPs에서 68, 73 kDa 크기의 분자들이 더 많이 발현된다는 것을 확인할 수 있었다. 두 가지 vaccine 투여구 중에서 2,2‘-dipyridyl 첨가 배지로 배양한 균체로 제작한 DP-FKC 항원 투여구가 응집 항체가가 더 높게 나타났다. 면역반응의 변화를 알기 위한 응집 항체가 조사에서는 FKC 항원과 DP-FKC 항원 투여구에서 모두 대조구보다 높은 값을 나타내었고, 3주 째에 최고치를 나타내었다. ELISPOT을 이용한 항체 생성 세포 수의 검출에서도 응집 항체가와 유사한 pattern을 나타내었는데, 두 가지 vaccine 투여구에서 2주 째에 가장 많은 항체 생성 세포 수를 나타내었다. 공격실험에서는 3주 째에 FKC 항원 투여구가 50%, DP-FKC 항원 투여구에서는 80%의 생존율을 나타내었다. 전체적으로 DP-FKC 항원 투여구의 생존율이 높게 나타났다. 이상의 결과로부터 넙치에 대한 E. tarda의 vaccine으로는 현재 연구되고 있는 FKC vaccine보다 철 결핍 조건에서 배양된 균체의 항원인 DP-FKC vaccine이 동일한 조건으로 사용하였을 때 더 나은 방어력을 가진다는 것을 알 수 있었다.

The Effect of Dietary Supplementation of Fe-methionine Chelate and FeSO4 on the Iron Content of Broiler Meat

  • A broiler experiment was conducted to compare the effects of supplementary iron sources and levels on the iron content of broiler meat. Two hundred and fifty hatched Ross broiler chickens were randomly assigned to 5 dietary treatments. Each treatment had 5 replicates of 10 birds (5 males and 5 females). Birds were housed in raised floor batteries and fed traditional broiler diets ad libitum for 5 weeks. Dietary treatments were as follows: Control, Fe-Met 100 (100 ppm iron as Fe-methionine), Fe-Met 200, $FeSO_4$ 100 (100 ppm iron as $FeSO_4{\cdot}7H_2O$) and $FeSO_4\;200$. There were no significant differences among treatments in parameters related to production performance. Liver contained approximately 10 times more iron than the leg muscle which contained approximately 3 times more iron than either breast muscle or wing muscle. Significant differences in iron content in the broiler meat were observed. In the breast meat, Fe-Met treatments were significantly (p<0.05) higher than other treatments in iron content. In the leg meat, Fe-Met treatments and $FeSO_4\;200$ treatment were significantly higher than the control in iron content. In the wing muscle, Fe-Met 200 treatment was significantly higher than other treatments in iron content. Iron content in the liver was significantly influenced by source and supplementation level of iron. Fe-Met treatments were higher than $FeSO_4$ treatments and 200 ppm treatments were higher than 100 ppm treatments in iron content in the liver. It is concluded that iron-methionine chelate is more efficient than iron sulfate and 200 ppm iron supplementation as Fe-Met is recommended for maximum iron enrichment in broiler meat.

Production of Iron Enriched Eggs of Laying Hens

  • An experiment was conducted to investigate the efficiency of transfer of dietary iron sources to eggs of laying hens. Eighty ISA-Brown laying birds of 30 wk old were housed in 40 cages of 2 birds each. Eight birds in four cages were assigned to one of the following ten treatments: T1; control, T2; 100 ppm iron supplementation with iron-methionine chelate (Fe-Met-100), T3; Fe-Met- 200, T4; Fe-Met-300, T5; 100 ppm iron supplementation with iron sulfate ($FeSO_4$-100), T6; $FeSO_4$-200, T7; $FeSO_4$-300, T8; 100 ppm iron supplementation with Availa-$Fe^{(R)}$ (Availa-Fe-100), T9; Availa-Fe-200 and T10; Availa-Fe-300. Results of 40 d feeding trial showed that there were no consistent responses in laying performance by source and level of iron supplementation. However, eggshell strength and color were improved by Fe supplementation. Egg iron content was maximized at 10-15 days after feeding supplemental Fe. Fe- Met was the most effective source in enriching Fe of eggs followed by Availa-Fe and $FeSO_4$. Increasing supplementary Fe level more than 100 ppm was not effective in Fe-Met and Availa-Fe treatments. Average Fe enrichment of 18% was achieved after feeding Fe-Met-100 for 15 d. In conclusion, enrichment of Fe in egg could be effectively achieved by supplementation of Fe-Met-100 for 15 d.

The Effect of Level and Period of Fe-methionine Chelate Supplementation on the Iron Content of Boiler Meat

  • A broiler experiment was conducted to compare the effects of duration and level of iron-methionine chelate (Fe-Met) supplementation on the iron, copper (Cu) and zinc (Zn) content of broiler meat. Two hundred and fifty hatched Ross broiler chickens were randomly assigned to 5 dietary treatments. Each treatment had 5 replicates of 10 birds (5 males and 5 females) each. Birds were housed in raised floor batteries and fed traditional broiler diets ad libitum for 5 weeks. Dietary treatments were as follows: Control and two levels of Fe-Met (100 or 200 ppm in Fe) supplemented for either the whole period (0-5 wk) or grower period (4-5 wk). Production performance was not significantly affected by treatments. Iron content in the muscles (breast, leg and wing) and organs (liver and spleen) were significantly (p<0.05) increased as the level and duration of Fe-Met supplementation increased. The highest concentration of iron was shown in Fe-Met 200 fed for the whole period. Liver contained the highest amount of iron followed by spleen, leg muscle, wing muscle and breast muscle. Supplementation of Fe-Met 200 for the grower period resulted in higher iron concentration in liver and spleen than supplementation of Fe-Met 100 for the whole period. However, the same treatment resulted in lower iron concentration in muscles (breast, leg and wing) than the treatment of Fe-Met 100 for the whole period. In order to achieve the highest iron enrichment in the muscles, Fe-Met should be supplemented at 200 ppm in Fe for the whole period (5 wks). Fe-Met supplementation increased copper concentration in all muscles and organs except wing muscle. Zinc concentration decreased in breast and wing muscle but tended to increase in leg muscle, liver and spleen by Fe-Met 200 supplementation. Color of muscle was not significantly affected by Fe-Met treatments. However, redness of leg and breast muscle, and yellowness of leg and breast muscle tended to increase by supplementation of Fe-Met for the whole period. It was concluded that iron content of broiler meat can be effectively enriched by supplementation of 200 ppm of Fe as Fe-Met for 5 wks.

침출수 및 철킬레이트를 이용한 실규모 매립가스 내 황화수소 제거 (Hydrogen Sulfide Removal in Full-scale Landfill Gas Using Leachate and Chelated Iron)

  • 황화수소는 매립가스와 바이오가스 사용 전에 제거해야 하는 유해한 불순물이다. 본 연구에서는 공기 산화를 활용한 황화수소 저감 방법을 연구하였다. C시 매립장에서 발생하는 매립가스의 유량 조절이 가능한 실규모 황화수소 저감 장치를 운전하였다. 실험 결과, 세정액 내 용존 철 농도는 황화물 산화 효율에 유의한 영향을 미쳤다. 매립지 발생 침출수 내 철 성분은 매립가스 내 황화수소 제거를 위한 용도로 철킬레이트와 혼용할 수 있었다. 철 농도가 90 mM 이상인 경우 9 초 이내의 접촉 시간에서 83 % 이상의 $H_2S$가 제거되었다. 따라서 촉매 산화 흡착법은 매립가스 및 바이오가스 정제를 위한 용도로서 충분히 가치가 있는 것으로 판단되었다.

산세수와 게껍질을 이용한 신기능성 철분 비료의 상추 생육 촉진 효과 (Growth-promoting Effect of New Iron-chelating Fertilizer on Lettuce)

  • 작물 재배에서 빈번히 발생되는 철분 부족 현상은 식물의 생육 저하와 생산성 감소를 야기한다. 철분의 흡수는 철분 킬레이트제를 이용해서 높일 수 있다. 본 연구에서는 제철산업의 부산물인 산세수 유래의 2가 철 이온을 게껍질(CSP)에 킬레이트시킨 신기능성 철분 비료(FCSP)를 제작하고, 상추 재배에 적용하여 생육 촉진 효과를 분석하였다. 제조한 철 비료의 효과를 검증하기 위해, 연구실 내 소규모 시험에서 무처리구, 게껍질 처리구, 그리고 여러 농도의 Fe-게껍질 킬레이트 처리구에서 상추의 생장을 분석한 결과, 50 ppm Fe-게껍질 킬레이트 처리구에서 엽수, 엽중, 엽장 및 엽폭이 무처리구와 게껍질 처리구에 비해 증가하였으며, 상추 잎의 엽록소의 함량 또한 증가하였다. 포장 시험 역시 50 ppm Fe-게껍질 킬레이트 처리구에서 엽수, 엽중, 엽장 및 엽폭에서 화학비료와 퇴비를 혼합한 기존 재배 방식 처리구에 비해 최대 2배 이상의 증가를 보였고, 식물체내 Fe의 함량 또한 가장 높게 나타났다. 또한 철 이온을 게껍질에 킬레이트한 비료의 처리가 토양 내의 Fe와 Ca의 함량을 증가시켰으며 유효인산과 유기물의 함량 또한 증가시켜 토질을 개선시키는 효과를 보였다. 이러한 결과들을 종합할 때, Fe-게껍질 킬레이트 복합체는 기존의 비료 처리 방식인 화학비료와 퇴비 사용 보다 뛰어난 생육 증대 효과와 토질 개선 효과를 나타냄으로써 작물 재배에 있어서 효과적인 신기능성 철분 소재로서 활용 가치가 있을 것으로 사료된다.

  • Diverse p-terphenyl compounds, named curtisians, have been isolated from the fungus Paxillus curtisii, and degradation of wood by this fungus is thought to be progressed by iron chelation of p-terphenyl curtisians. In this study, the iron chelation ability of p-terphenyls has been proved by chrome azurol S (CAS) assay, reducing power, and UV-visible spectroscopic analyses. The catechol moiety of p-terphenyl is an essential factor for the potent iron chelation ability, and thus deacylated curtisian with a tetrahydroxyl moiety in the central ring of p-terphenyl is more effective than acylated curtisians.

대계 약침액의 항산화 효능 (Antioxidant Property of Aqua-Acupuncture Solution from Circium japonicum)

  • 대계는 전통 한의학에서 약용 물질로 널리 쓰이고 있으므로, 대계의 약침을 조제하여 항산화적 활성을 검증해 보았다. 실험의 재료로 쓰인 대계는 동국대학교 한의과대학 부속 한방 병원에서 구입하여 사용하였고 (경상북도 경주), 이를 약침액으로 조제한 후 동결 건조 방법으로 5.1 %의 분말을 회수 하였다. 대계 약침액의 항산화 능력을 측정하기 위하여 DPPH자유기의 소거능은 Gyamfi et al. 의 방법을 따랐고, superoxide radicals $(O_2^-{\cdot})$ 소거능은 Gotoh et al.의 방법을 일부 수정하여 사용하였다. Hydroxyl radicals의 직접적인 소거능과 iron ions 사이의 관계는 deoxyribose assay를 통하여 검증하였다. 이상의 실험 결과에서 1) Non-site-specific scavenging assay (hydroxyl radicals, OH) 및 2) Site-specific scavenging assay (chelate iron ions) 그리고 3) 전산화 효과에서 모두 대계 약침액은 농도 의존적인 저해 효과를 보였으며, 특히 pro-oxidant 실험에서는 산화를 촉진하지 않는 것으로 나타났다. 끝으로 hydorxyl radical에 의하여 매개되는 DNA의 분절 효과를 적절하게 방어하는 것을 관찰할 수 있었다. 따라서 대계 약침액은 산화적 스트레스를 직접적으로 방어 하고, 다양한 산화적 손상에 의한 병증을 예방할 것으로 사료된다.

Relationship between liver iron concentration determined by R2-MRI, serum ferritin, and liver enzymes in patients with thalassemia intermedia

  • Background Iron overload is a risk factor affecting all patients with thalassemia intermedia (TI). We aimed to determine whether there is a relationship of serum ferritin (SF) and alanine aminotransferase (ALT) with liver iron concentration (LIC) determined by R2 magnetic resonance imaging (R2-MRI), to estimate the most relevant degree of iron overload and best time to chelate in patients with TI. Methods In this cross-sectional study, 119 patients with TI (mean age years) were randomly selected and compared with 120 patients who had a diagnosis of thalassemia major (TM). Correlations of LIC, as determined by R2-MRI, with SF and ALT levels, were assessed in all participants. A P-value <0.05 was considered statistically significant. Results SF and LIC levels were lower in patients with TI than in those with TM; only ferritin values were significant. We found a statistically significant relationship between SF and LIC, with cut-off estimates of SF in patients with TI who had splenectomy and those who entered puberty spontaneously (916 and 940 ng/mL, respectively) with LIC >5 mg Fe/g dry weight (P<0.0001). A significant relationship was also found for patients with TI who had elevated ALT level (63.5 U/L), of 3.15 times the upper normal laboratory limit, using a cut-off for LIC ${\geq}5mg\;Fe/g\;dry\;weight$. Conclusion We determined the cut-off values for ALT and SF indicating the best time to start iron chelation therapy in patients with TI, and found significant correlations among iron overload, SF, and ALT.

Effects of Fe-soy Proteinate Chelate Supplementation to Diets of Periparturient Sows and Piglets on the Fe Level in the Blood of Piglets

  • The objective of the present study was to investigate the effects of Fe-soy proteinate chelate (Fe-SP) on sows milk, piglet blood parameters and performance. A total of 15 sows of 3 wk before parturition and pigs after births to 3 wk were assigned to three dietary treatments: control (sow-basal diet, piglets with Fe injection); Fe-SP 100 (Fe 100 ppm as Fe-SP in sow and piglet diet); Fe-SP 200 (Fe 200 ppm as Fe-SP in sow and piglet diet). Each treatment had 5 replicates (sows) of six piglets per sow randomly selected from the same offspring. For this experiment, Fe-SP was manufactured. There were no significant differences among treatments in number of pigs born in total or alive per litter, birth weight, number of pigs weaned per litter and weaning weight. However, weight gain, feed intake and feed conversion ratio significantly (p<0.05) decreased as the supplementation level of Fe-SP increased. There were no significant differences among treatments in Fe content at 3 wk before parturition in sow blood. However, Fe content at 2 wk before parturition in sow blood significantly (p<0.05) increased as the supplementation of Fe-SP. While there were no significant differences among treatments in Fe content at 1 wk before parturition in sow blood, it tended to increase as the supplementation level of Fe-SP increased. There were no significant differences among treatments in Fe content of sow milk. However, it tended to increase as the supplementation level of Fe-SP increased. Iron content in the blood of piglets was significantly (p<0.05) higher in control (Fe injected) than Fe-SP 100 and Fe-SP 200 treatments at $1^{st}$ and $2^{nd}$ wk but it was significantly higher in Fe-SP 200 than others in $3^{rd}$ wk. Zinc content in the blood also significantly (p<0.05) increased as the Fe-SP supplementation level increased in $3^{rd}$ wk. In conclusion, Fe-SP supplementation significantly affected Fe content in the blood of piglets. Iron injection was more effective at $1^{st}$ and $2^{nd}$ wk, while Fe-SP 200 supplementation was effective at $3^{rd}$ wk in improving blood Fe level in piglets.
