  • Four compounds were isolated from the rhizome of Belamcanda chinensis (Iridaceae) by repeated column chromatography and were elucidated as iristectorigenin A, irigenin, tectorigenin and tectoridin by spectral analysis. The estrogenic activity of these isoflavones was tested using Ishikawa cells. Irigenin, tectorigenin and tectoridin were highly estrogenic ($EC_{50}\;=\;0.75$, 0.42 and $0.81\;{\mu}g/ml$, respectively), while iristectorigenin A exhibited weak estrogenic activity ($EC_{50}{\geq}4\;{\mu}g/ml$).

Antiinflammatory and Antiangiogenic Activities of Flavonoids Isolated from Belamcandae Rhizoma

  • The present study was carried out to clarify whether isoflavonoids isolated from Belamcandae Rhizoma (Iridaceae) inhibit inflammation and angiogenesis by the experimental methods in vitro and in vivo. Among the isolated isoflavonoids, such as irigenin, irisflorentine, and iristectorene B inhibited nitric oxide (NO) production, as measured by nitrite formation at 3-30 ${\mu}M$. Also these compounds reduced cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) enzyme expression in a concentration dependent manner, when measured by western blotting, at 3-30 ${\mu}M$. Irigenin, irisflorentine and iristectoren B decreased angiogenesis of chick embryos in the chorioallantoic membrane assay. These compounds also reduced the proliferation of calf pulmonary arterial endothelial (CPAE) cells and found to possess relatively weak gelatinase/collagenase inhibitory activity in vitro. These compounds, when administered subcataneously at the dose of 30mg/kg for 20 days to mice implanted with murine Lewis lung carcinoma (LLC), caused a significant inhibition of tumor volume. Therefore, antiangiogenic activities of isoflavonoids from Belamcandae Rhizoma might be due to antiproliferative activities under inhibition the induction of COX-2 and iNOS enzyme.

Decursin from the Rhizome of Belamcanda chinensis

  • Six components were isolated from the $CH_2Cl_2$ fraction of Belamcanda chinensis rhizome by open column chromatography. Their structures were elucidated as ${\beta}-sitosterol$ (1), apocynin (2), decursin (3), iristectorigenin A (4), irigenin (5) and tectorigenin (6) by spectral analysis. Among these compounds, decursin (3) was isolated for the first time from a plant of the family Iridaceae.

Cytoprotective Activity of Belamcanda chinensis Rhizome Against Glutamate-Induced Oxidative Injury in HT22 Cells

  • Four isoflavones including tectorigenin (1), irisflorentin (2), irigenin (3), and tectoridin (4) have been isolated from the 70% EtOH extract of Belamcanda chinensis rhizome. The evaluation for protective effect of compounds 1-4 against glutamate-induced cytotoxicity in hippocampal HT22 cell line was conducted. Compound 1 showed significant protective effect with an EC$_{50}$ value of 67.25 ${\pm}$ 1.2 ${\mu}$M, whereas compounds 2-4 were inactive. These results suggest that compound 1 may possess the neuroprotective activity against oxidative cellular injures.

Isoflavonoids from the Rhizomes of Belamcanda chinensis and Their Effects on Aldose Reductase and Sorbitol Accumulation in Streptozotocin Induced Diabetic Rat Tissues

  • Aldose reductase, the key enzyme of the polyol pathway, is known to play important roles in the diabetic complication. The inhibitors of aldose reductase, therefore, would be potential agents for the prevention of diabetic complications. To evaluate active principles for the inhibition of aldose reductase from the rhizomes of Belamcanda chinensis, twelve phenolic compounds were isolated and tested for their effects on rat lens aldose reductase. As a result, isoflavones such as tectorigenin, irigenin and their glucosides were found to show a strong aldose reductase inhibition. Tectoridin and tectorigenin, exhibited the highest aldose reductase inhibitory potency, their $IC_{50}$ values, being $1.08{\times}10^{-6}{\;}M{\;}and{\;}1.12{\times}10^{-6}{\;}M$, respectively, for DL-glyceraldehyde as a substrate. Both compounds, when administered orally at 100 mg/kg for 10 consecutive days to streptozotocin-induced diabetic rats, caused a significant inhibition of sorbitol accumulation in the tissues such as lens, sciatic nerves and red blood cells. Tectorigenin showed a stronger inhibitory activity than tectoridin. From these results, it is suggested that tectorigenin is attributed to be a promising compound for the prevention and/or treatment of diabetic complications.

Antioxidant, Antimicrobial and Cytoprotective Effects of the Extract and Its Fraction Obtained from Rhizomes of Belamcanda chinensis (L.) DC (범부채 뿌리 추출물 및 분획물의 항산화, 항균 및 세포 보호 효과)

  • In this study, we investigated antioxidant, antimicrobial and cytoprotective effects of 50% ethanol extract and ethyl acetate fraction from rhizomes of Belamcanda chinensis (L.) DC. 1,1-Diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activities ($FSC_{50}$) of the 50% ethanol extract and ethyl acetate fraction were 621.5 and $253.0{\mu}g/mL$, respectively. Total antioxidant capacities ($OSC_{50}$) of the extract and fraction were 13.6 and $3.0{\mu}g/mL$, respectively. Minimum inhibitory concentrations (MIC) of the ethyl acetate fraction for Staphylococcus aureus and Candida albicans were 156, $1,250{\mu}g/mL$, respectively, indicating similar or higher levels of those of using methyl paraben. Cytoprotective effects of the 50% ethanol extract against $^1O_2$-induced cellular damage (${\tau}_{50}$) showed in a dose dependent manner at 4 to $64{\mu}g/mL$. ${\tau}_{50}$ of the 50% ethanol extract, ethyl acetate fraction and (+)-${\alpha}$-tocopherol at $16{\mu}g/mL$ were 36.4, 45.0 and 45.8 min respectively, and the ethyl acetate fraction showed cytoprotective effects similar to (+)-${\alpha}$-tocopherol. In ultraviolet B radiation-induced HaCaT cell damage, the ethyl acetate fraction decreased intracellular reactive oxygen species (ROS) up to 45.9% at $8{\mu}g/mL$. Also in $H_2O_2$-induced HaCaT cell damage, the ethyl acetate fraction significantly increased the cell viability at $0.5{\sim}8.0{\mu}g/mL$. As a result of chemical analyses of the ethyl acetate fraction, the presence of flavonoids and polyphenol such as irisflorentin, irigenin, tectorigenin, resveratrol, iridin and tectoridin were identified. In conclusion, the extract/fraction from rhizomes of B. chinensis can be applied as a natural antioxidant and antimicrobial material to cosmetics.
