HPLC-DAD-MS/MS를 이용한 금은화 생리활성 물질의 동시분석 (Simultaneous Analysis of Bioactive Metabolites from Lonicera japonica Flower Buds by HPLC-DAD-MS/MS)

  • A high-performance liquid chromatography (HPLC) with diode array detector (DAD) and electrospray ionization mass spectrometry (ESI-MS) was established for the simultaneous determination of chlorogenic acid (1), sweroside (2), luteolin-7-O-glucoside (3), (E)-aldosecologanin (4) and 3,5-dicaffeoylquinic acid (5) from Lonicera joponica flower buds. The optimal chromatographic conditions were obtained on an ODS column (5 ${\mu}m$, 4.6${\times}$150 mm) with the column temperature $25^{\circ}C$. The mobile phase was composed of (A) water with 0.1% formic acid and (B) acetonitrile with 0.1% formic acid using a gradient elution, the flow rate was 0.3 ml/min. Detection wavelength was set at 250 nm. All calibration curves showed good linear regression ($r^2$>0.994) within test ranges. The developed method provided satisfactory precision and accuracy with overall intra-day and inter-day variations of 0.05${\sim}$1.95% and 0.15${\sim}$2.26%, respectively, and the overall recoveries of 97.71${\sim}$103.65% for the five compounds analyzed. The verified method was successfully applied to quantitative determination of the three types (phenolic compounds, iridoids and flavonoids) of bioactive compounds in 21 commercial L. japonica flower buds samples from different markets in Korea and China. The analytical results demonstrated that the contents of the five analytes vary significantly with sources.

  • The aerial parts of Verbascum sinaiticum Benth. (Scrophulariaceae) yielded two iridoids that were idenified as ajugol 3 and aucubin 4. Also, investigation of the flavonoid constituents revealed the isolation and identification of luteolin 1 and chrysoeriol-7-glucoside 2. All the isolated compounds were identifid by spectroscopicmethods (UV, MS, $^{1}H-NMR$ and $^{13}C-NMR$) and in comparison with the literature data. Both alcohol and methylene chloride/methanol extracts (1 : 1) exhibited a hepatoprotective effect by $CCl_{4}challenge$ test.

  • The genus Patrinia, belonging to the Valerianaceae family, includes ca. 20 species of herbaceous plants with yellow or white flowers, distributed in Korea, China, Siberia, and Japan. Among them, P. scabiosaefolia (yellow Patrinia), P. saniculaefolia, P. villosa (white Patrinia), and P. rupestris are found in Korea. Several members of this genus have long been used in folk medicine for the treatment of inflammation, wound healing, ascetics, and abdominal pain after childbirth. Thus far, ca. 217 constituents, namely flavonoids, iridoids, triterpenes, saponins, and others have been identified in this genus. Crude extract and isolated compounds have been found to exhibit anticancer, anti-inflammatory, antioxidant, antifungal, antibacterial, cytotoxic activities, lending support to the rationale behind several of its traditional uses. The present review compiles information concerning the phytochemistry and biological activities of Patrinia, with particular emphasis on P. villosa, as studied by our research group.

  • Acute bronchitis and chronic obstructive pulmonary diseases (COPD) are essentially lung inflammatory disorders. Various plant extracts and their constituents showed therapeutic effects on several animal models of lung inflammation. These include coumarins, flavonoids, phenolics, iridoids, monoterpenes, diterpenes and triterpenoids. Some of them exerted inhibitory action mainly by inhibiting the mitogen-activated protein kinase pathway and nuclear transcription $factor-{\kappa}B$ activation. Especially, many flavonoid derivatives distinctly showed effectiveness on lung inflammation. In this review, the experimental data for plant extracts and their constituents showing therapeutic effectiveness on animal models of lung inflammation are summarized.

  • Previously, we reported that the n-butanol fraction of Lonicera japonica showed anti-flammatory activity in mice and rats. Several constituents such a loniceroside A and B, flavonoids and iridoids were isolated from this fraction. In this investigation, the anti-inflammatory activity of the major constituents of L. japonica was studied. Loniceroside A, lonicerin and loganin showed anti-inflammatory activity comparable to aspirin.

Three New Iridoid Glucosides from the Roots of Patrinia scabra

  • To probe the chemical constituents of Patrinia scabra, we undertook the phytochemical investigation on its roots, which led to the isolation and elucidation of three new iridoid glucosides, scabroside A-C (1-3), along with three known iridoids, jatamanin J (4), isopatriscabroside I (5) and loganic acid (6) from the aqueous fraction of the ethanolic extract of the roots. The structures and relative configurations of the three new compounds were elucidated by spectroscopic methods including IR, UV, MS, 1D and 2D NMR experiments. Compound 3 was an unusual iridoid with an oxygen bridge connecting C-3 and C-8.

지황 (地黃)의 성분연구 (Phytochemical Studies on Rehmanniae Radix)

  • Twenty-four compounds were isolated from the 70% ethanolic extract of Rehmanniae Radix (Scrophulariaceae) cultivated in Ubo-myeon, Gunwi-gun, Gyeongbuk province, Korea and their structures were identified as four iridoids [6-O-(4''-O-${\alpha}$-L-rhamnopyranosyl) vanilloyl ajugol (17), ajugol (18), aucubin (19), and catalpol (20)], three phenethyl alcohol glycosides [decaffeoyl acteoside (15), isoacteoside (16), and acteoside (21)], five sugar derivatives [ethyl ${\beta}$-D-fructofuranoside (7), eleutheroside C (14), mannitol (22), raffinose (23), and stachyose (24)], two terpenoids [remophilanetriol (4) and glutinolic acid (11)], a lignan, paulownin (2), and eight others [${\beta}$-sitosterol (1), daucosterol (6), monopalmitin (3), pinellic acid (9), uracil (5), adenosine (12), jio-cerebroside (10), aeginetic acid 5-O-${\beta}$-D-quinovoside (8), aeginetoyl ajugol 5''-O-${\beta}$-D-quinovoside (13)]. The chemical structures of these compounds were identified on the basis of spectroscopic methods and comparison with literature values. Among these compounds, paulownin (2), monopalmitin (3), uracil (5), daucosterol (6), ethyl ${\beta}$-D-fructo-furanoside (7), and eleutheroside C (14) were isolated from this plant for the first time.

포제에 따른 두충의 지표성분 함량분석 (Quantitative Determination of the Six Marker Compounds in Eucommiae Cortex by Processing Method)

  • In this study, we carried out quantification analysis of the six marker components, geniposidic acid, chlorogenic acid, geniposide, pinoresinol diglucoside, liriodendrin, and genipin in the 70% ethanol extracts of non-processed Eucommiae Cortex and processed Eucommiae Cortex using a high-performance liquid chromatography coupled with photodiode array detector. The six components were separated on Gemini C18 column (5 μm, 4.6×250 mm) by the gradient elution with 1.0% (v/v) acetic acid in water and 1.0% (v/v) acetic acid in acetonitrile as mobile phase. The flow rate was 1.0 mL/min and the injection volume was 10 mL. The amount of geniposidic acid, chlorogenic acid, geniposide, pinoresinol diglucoside, liriodendrin, and genipin in non-processed Eucommiae Cortex were 1.31, 0.31, 0.66, 0.46, 0.46, and 0.03%, respectively, while the amount of the six compounds in non-processed Eucommiae Cortex were 0.04-0.78, 0.01-0.14%, 0.05-0.63%, 0.01-0.37%, 0.15-0.42%, and not detected, respectively. After processing treatment, the contents of three iridoids, two lingnan, and one phenylpropanoid decreased in Eucommiae Cortex.

숙지황(Rehmannia glutinosa)의 약리작용과 독성 (Pharmacological Action and Toxicity of Rehmannia glutinosa)

  • Objectives : Rehmannia glutinosa has been used extensively in Korean traditional medicine. Although thorough clinical trials are lacking, Various pharmacological actions for Rehmannia glutinosa has been identified newly using animal models. In addition, it was reported that reactive intermediates, potentially causing toxic effects, was isolated from one of components in Rehmannia glutinosa. In this article, it is purposed for explanation and introduction of new studies for Rehmannia glutinosa in terms of pharmacological action and toxicology. Methods : New studies for Rehmannia glutinosa were reviewed and summarized in terms of pharmacological action and toxicity. Results and Conclusions : Rhmannia glutinosa and its components including iridoids, saccharides, as well as amino acid, showed a variety of pharmacological actions on the blood system, immune system, endocrine system, cardiovascular system and the nervous system. In addition, it was identified that aucubin, one of major components of Rhmannia glutinosa was biotransformed to reactive intermediates by ${\beta}$-glycosidase and acid-hydrolysis, resulting in forming aucubigenin- albumin adduct. Even if a lot of new pharmacological actions has been identified, it should be considered for Rhmannia glutinosa to contain the material producing reactive intermediates which may induce the side effects.

Simultaneous Analysis of Bioactive Metabolites from Rehmannia glutinosa by HPLC-DAD-MS/MS

The optimal chromatographic conditions were obtained on an ODS column (5 mm, $4.6{\times}250\;mm$) with the column temperature at $25^{\circ}C$. The mobile phase was composed of water and acetonitrile using a gradient elution with the flow rate 0.3 mL/min. Detection wavelength was set at 205 nm. All calibration curves showed good linear regression ($r^2$ > 0.997) within test ranges. Limits of detection (LOD) and quantitation (LOQ) values were lower than 0.123 and $0.373\;{\mu}g/mL$, respectively. The developed method provided satisfactory precision and accuracy with overall intra-day and inter-day variations of 0.09 - 0.76% and 0.16 - 1.41%, respectively, and the overall recoveries of 99.03 - 102.67% for all of the compounds analyzed. In addition, effectiveness of diverse extraction methods was compared to each other for the development of standard analytic method. The verified method was successfully applied to the quantitative determination of five representative metabolites in twenty-one commercial Rehmannia glutinosa samples from different markets in Korea and China. The analytical results showed that the contents of the five analytes vary significantly with sources.
