  • Ion-pair reverse phase HPLC techniques were used to compare the contents of IAA and IAAsp in the embryogenic and non-embryogenic calli derived from ginseng (Panax ginseng C.A. Meyer) root tissues. The contents of IAA and IAAsp of the embryogenic callus were much higher (7 to 18 X respectively) than those of non-embryogenic callus. There is a distinct fluorescent peak of an unknown component in the HPLC profile of the extract for indolic compounds from non-embryo-genic callus. This distinct difference may be employed as a promising parameter to screen the culture pieces for obtaining the calli with high potential for embryoid formation.

  • Determination of Aconitum alkaloids in processed Buza (Cho-0, Salted Buza, Moist-heating Buza, Limed Buza), which had been prepared from the raw tubers of Aconitum chiisanenseb(Ranunculaceae), was established using visible spectrophotometry and high-performance liquid chromatography (HPLC) method especially for Aconitine analysis. Aconitum alkloids were reacted with tetra- thiocy-anatocobalt[II] complex ion to form a stable ion pair. The reaction product was insoluble in water but freely soluble in several organic solvents. 1.2-Dichloroethane was the best extracting solvent among the examined solvents. Spectrophotometry of Aconitum alkaloids at nax. 625 was carried out. The HPLC method for aconitine was carried out using Radial PAK-CN column with gradient solvent system by solvent mixture of acetonitrile and phosphate buffer (pH 3.0) at 4$0^{\circ}C$ and 254 nm. Linear relationship was found between absorbance response and concentration of aconitine in range of 0.45 mM~0.9 mM ($r^2$=0.9949) by spectrophotometry and 0.3 mM~1.2mM($r^2$=0.9983) by HPLC method. These methods have been found to be suitable and reproducible for routine analysis of Aconitum alkaloids and its pharmaceutical preparations.

  • A new method for the analysis of metocurine iodide in biological fluids was developed. Metocurine iodide was quantitatively extracted with rose bengal from aqueous layer into dichloromethane layer and the amount of metocurine iodide was calculated from the amount of rose bengal which was determined by HPLC with fluorescence detector. It was possible to analyze metocurine iodide without the effect of co-prescribed drugs in the concentration range of $0.09{\sim}9.10\;{\mu}g/ml$. The detection limits of metocurine iodide in urine and blood were 0.8 and 1.2 ng at S/N=3, each respectively.

  • High-performance liquid chromatographic method with UV detecction was developed for the determination of theophylline and its metabolites in human urine using ${beta}$-hydroxyethyl theophylline$({beta} -HET)$ as an internal standard. For extraction of urine sample, quality control sample and xanthine-free blank urine were mixed with decylamine (ion-paring reagent) and ${beta}$-HET. After saturation with ammonium sulfate, the mixture was then extracted with organic solvent at pH values of 4.0-4.5. All separations were performed with ion-pair chromatography using decylamine as an ion-pairing reagent and 3mM sodium acetate buffered mobile phase (pH 4.0) containing 1% (v/v) acetonitrile and 0.75 mM decylamine. The detection limits of theophylline, 1, 3-DMU, 1-MU, 3-MX and 1-MX in human urine were 0.17, 0.17, 0.39, 0.19 and 0.19 ${\mu}g$/ml, based on a signal-to-noise ratios of 3.0. The mean intraday coefficients of variation (C.V.s) of each compound on nine replicates were lower than 2.0%, while mean interday C.V.s on three days were lower than 1.6%. All separations were finished within 40miutes.

  • Micellar electrokinetic capillary chromatography(MECC) and HPLC with ion-pairing mechanism were applied for the separation of the well known environmental wastes from dye industry. These compounds include H-acid, J-acid, ${\gamma}$-acid, orthanilic acid, sulfanilic acid and 2-naphthylamine-1,5-disulfonic acid, and are known to be the diazo components of the azo dye. MECC method was also applied to separate few acid dyes including Acid Orange 7, Acid Orange 5 and Acid Blue 92 and direct dye such as Direct Red 80. Informations about the diazo components of any azo dye could be obtained by comparison of electropherogram of the reduction solution of a given dye with those obtained from standard materials such as H-acid, J-acid, ${\gamma}$-acid, orthanilic acid, sulfanilic acid and 2-naphthylamine-1,5-disulfonic acid. It has been concluded that MECC and HPLC with ion-pairing mechanism could be successfully applied for the analysis of unknown dyes and their diazo components.

Mass Spectrometric Analysis for Discrimination of Diastereoisomers

  • A liquid chromatography mass spectrometry (LC-MS) system was used to identify and distinguish oligostilbene diastereoisomers. A polyphenolic extract from Neobalanocarpus heimii known to be rich in oligostilbenes of various degrees of condensation was used as test material. Fourteen oligostilbenes were isolated from this extract on a fully automated semi-preparative HPLC system. Out of these, two pairs of dimers, one pair of trimers, two pairs of tetramers and a group of four tetramers with similar skeleton were identified as diastereoisomers. Their structures and configurations were established by spectroscopic methods. All isolated compounds were subjected to an LC-MS/MS to study their fragmentation patterns. The experiments were performed on a liquid chromatography-mass spectrometry (LC-MS) with electrospray-ionization (ESI) interface in positive mode. MS/MS spectra of each pure compound were recorded by direct infusion in identical conditions and their product ion spectra were analysed. Some subtle yet significant differences were observed between the spectra of oligostilbenes from the various diastereoisomeric series.

  • When cationic basic compounds are chromatographed using hydro-organic mobile phase, the presence of anionic free silanols in the silica-based stationary phases results in broad and asymmetrical peaks. The addition of an ionic reagent to the mobile phase prevents analytes from accessing free silanols, improving peak shape. In this study, the chromatographic behavior of salbutamol sulfate as a basic compound was investigated under various conditions, including the use of different columns, mobile phases, and ion-pair reagents such as triethanolamine (TEA) and sodium heptane sulfonate (SHS). The retention and peak shape of chromatograms were both evaluated. The results show that pre-conditioning the column with TEA and including it in the mobile phase can prevent cationic analytes from accessing anionic silanols, resulting in improved peak shape. Furthermore, buffering the mobile phase is an important factor in keeping the pH constant throughout the process. The chosen method was validated in part. This study could be helpful for researchers and analyst to solve such problems with cationic basic components.

Studies of separation and quantitation for selenium species in food (식품중의 셀레늄 화학종의 분리 및 정량연구)

  • The purpose of this research is to separate and quantitate selenium species in some food samples with HPLC-ICP-MS. Cation exchange chromatography showed efficient separation only for inorganic Se species while reversed phase ion pair chromatography showed good separation for both inorganic and organic Se species. $C_8$ column ($Symmetryshield^{TM}\;RP_8$, 3.5 ${\mu}m$, $4.6{\times}150$ mm) was used with optimum condition of 5% methanol mobile phase, 0.05% of nonafluorovaleric acid ion pairing reagent. Five standard Se species of Se(IV), Se(VI), SeCys(selenocystein), SeMet(selenomethionine) and Se-M-C(seleno methyl cystein) were separated successfully under the optimum condition (mobile phase; 5% methanol, ion-pairing reagent; 0.05% nonafluorovaleric acid, flow rate; 0.9 mL $min^{-1}$). To extract Se species, microwave assisted and enzyme-assisted extraction methods were studied. In enzyme-assisted extraction method, protease I for garlic, protease I plus trypsin for pork and mackerel, and protease XIV for tuna showed the best extraction efficiency. With the optimum condition for each sample, it was found that mostly inorganic Se, SeCys and SeMet are present in the sample studied ranging from few ${\mu}g$ $g^{-1}$ to few tens of ${\mu}g$ $g^{-1}$.

The results of the separation were compared with the result obtained by the HPLC using ion-pairing mechnism. MECC method was also applied to separate a few direct dyes including Direct Blue 2, Direct Blue 6 and Direct Blue 15, and reactive dye such as Reactive Orange 4. Informations about the diazo components of any azo dye could be obtained by comparison of electropherogram of the reduction solution of given dye with those obtained from standard materials such as H-acid, J-acid, ${\gamma}$-acid, orthanilic acid, sulfanilic acid and 2-naphthylamine-1,5-disulfonic acid which are used as diazo components of the typical azo dyes. It has been concluded that MECC and HPLC with ion-pairing mechanism could be successfully applied for the analysis of unknown dyes and their diazo components.

Simultaneous Determination of Water-Soluble Vitamins (Vitamin $B_1$, $B_2$, $B_3$, $B_6$ and C) in Dietary Supplements by High-Performance Liquid Chromatography (영양보충용 식품 중 수용성비타민(Vitamin $B_1$, $B_2$, $B_3$, $B_6$ and C)의 HPLC를 이용한 동시분석법)

  • 시중에 유통 중인 영양보충용 제품의 수용성 비타민 $B_1$(thiamin), $B_2$(riboflavin), $B_3$(nicotinic acid and nicotine amide), $B_6$(pyridoxine), C(ascorbic acid)의 신속한 동시분석 방법을 확립하기 위하여 본 연구를 실시하였다. 영양보충용 제품은 정제, 연질캅셀, 분말, 액상의 4가지 제형에 대해 27종의 제품을 구입한 후, Ion-pair 분리기법을 사용하여 HPLC-UVD를 이용한 동시분석 방법을 검토하였다. 비타민 $B_1$, $B_2$, $B_3$, $B_6$, C의 HPLC에 의한 동시분석 조건을 검토한 결과, 이동상은 0.02% triethylamine, 17.5% 메탄올, $5{\mu}M$ sodium hexanesulfonic acid가 함유된 pH 3.5(acetic acid로 조절)의 수용액을 사용하였고, 용출시간은 다른 피크의 영향을 받지 않도록 30분으로 하였다. 수용성 비타민의 회수율은 96% 이상이었다. 본 연구에 의해 확립된 수용성 비타민의 동시분석 조건은 검량선의 직선성, 정밀성, 정확성, 기기적합성 등이 USP 및 ICH 기준에 적합하여 HPLC의 동시분석 방법으로 합당하였다. 수용성 비타민의 추출 용매는 제형에 따라 약간의 차이를 보이긴 했으나, 물이나 산성조건을 갖춘 HPLC의 이동상이 에탄올이나 메탄올보다 높은 추출 효율을 보였다. 초음파 추출기에 의한 추출 시간은 20분이 가장 적당하였다. 본 연구의 결과는 수용성 비타민의 신속한 추출 및 분석에 매우 효율적으로 이용될 것으로 기대된다.
