Purpose: We previously found that the histone methyltransferase suppressor of variegation, enhancer of zeste, trithorax and myeloid-nervy-deformed epidermal autoregulatory factor-1 domain-containing protein 3 (SMYD3) is a potential independent predictive factor or prognostic factor for overall survival in gastric cancer patients, but its roles seem to differ from those in other cancers. Therefore, in this study, the detailed functions of SMYD3 in cell proliferation and migration in gastric cancer were examined. Materials and Methods: SMYD3 was overexpressed or suppressed by transfection with an expression plasmid or siRNA, and a wound healing migration assay and Transwell assay were performed to detect the migration and invasion ability of gastric cancer cells. Additionally, an MTT assay and clonogenic assay were performed to evaluate cell proliferation, and a cell cycle analysis was performed by propidium iodide staining. Furthermore, the expression of genes implicated in the ataxia telangiectasia mutated (ATM) pathway and proteins involved in cell cycle regulation were detected by polymerase chain reaction and western blot analyses. Results: Compared with control cells, gastric cancer cells transfected with si-SMYD3 showed lower migration and invasion abilities (P<0.05), and the absence of SMYD3 halted cells in G2/M phase and activated the ATM pathway. Furthermore, the opposite patterns were observed when SMYD3 was elevated in normal gastric cells. Conclusions: To the best of our knowledge, this study provides the first evidence that the absence of SMYD3 could inhibit the migration, invasion, and proliferation of gastric cancer cells and halt cells in G2/M phase via the ATM-CHK2/p53-Cdc25C pathway. These findings indicated that SMYD3 plays crucial roles in the proliferation, migration, and invasion of gastric cancer cells and may be a useful therapeutic target in human gastric carcinomas.
초음파는 세포사멸을 포함하여 의학 및 생물학분야에 널리 응용되고 있으나 그 정확한 기작에 대해선 논쟁의 여지가 있다. 본 연구에서는 40 kH 초음파 조사시스템을 단세포 효모에 적합하게 개발하고 세포사멸 유도시 40 kH 초음파의 생물학적 현상을 살펴보았다. 아이오딘화 칼륨 선량 측정법을 이용하여 1.5 ml 실험튜브에 40 kH 초음파 조사 시스템의 최적 조건을 맞추어 세포사멸을 시간 의존적 방식으로 연구하였고 초음파 조사과정동안 온열효과와는 별개로 세포 사멸이 관찰되었다. 40 kH 초음파와 과산화수소의 동시 처리는 세포사멸에 상조적인 효과가 관찰되어 활성산소가 40 kH 초음파사멸에 관련이 있었다. 그러나 활성산소 저해제, NAC(N-acetyl-Lcysteine)는 초음파에 의한 세포사멸에 약한 영향만을 미쳤고 다른 세포사멸, 괴사억제제[글리실리진(glycyrrhizin) 또는 zVAD-fmk] 역시도 세포사멸을 완전히 억제하진 못하였다. 본 연구를 통하여 40 kH 초음파에 의한 세포사멸에는 온열효과나 활성산소만으로 사멸이 유도되지는 않는 것으로 보인다.
An analytical method the determination of benzo(a)pyrene (BaP) and its hydroxylated metabolites, 1-hydroxybenzo(a)pyrene (1-OHBaP), 3-hydroxybenzo(a)pyrene (3-OHBaP), benzo(a)pyrene-4,5-dihydrodiol (4,5-diolBaP) and benzo(a)pyrene-7,8-dihydrodiol (7,8-diolBaP), in rat urine and plasma has been developed by HPLC/FLD and GC/MS. The derivatization with alkyl iodide was employed to improve the resolution and the detection of two mono hydroxylated metabolites, 1-OHBaP and 3-OHBaP, in LC and GC. BaP and its four metabolites in spiked urine were successfully separated by gradient elution on reverse phase ODS $C_{18}$ column (4.6 mm I.D., 100 mm length, particle size 5 ㎛) using a binary mixture of MeOH/H₂O (85/15, v/v) as mobile phase after ethylation at 90 ℃ for 10 min. The extraction recoveries of BaP and its metabolites in spiked samples with liquid-liquid extraction, which was better than solid phase extraction, were in the range of 90.3- 101.6% in n-hexane for urine and 95.7-106.3% in acetone for plasma, respectively. The calibration curves has shown good linearity with the correlation coefficients (R²) varying from 0.992 to 1.000 for urine and from 0.996 to 1.000 for plasma, respectively. The detection limits of all analytes were obtained in the range of 0.01-0.1 ng/mL for urine and 0.1-0.4 ng/mL for plasma, respectively. The metabolites of BaP were excreted as mono hydroxy and dihydrodiol forms after intraperitoneal injection of 20 mg/kg of BaP to rats. The total amounts of BaP and four metabolites excreted in dosed rat urine were 3.79 ng over the 0-96 hr period from adminstration and the excretional recovery was less than 0.065% of the injection amounts of BaP. The proposed method was successfully applied to the determination of BaP and its hydroxylated metabolites in rat urine and plasma for the pharmacokinetic studies.
본 연구에서는 체외수정, 난자내 정자 직접주입, 난자내 정자 두부 미두 주입 후의 핵과 미세소관의 변화를 관찰하였다. 핵과 미세소관의 움직임은 형광염색을 실시한 후 공초점주사현미경을 이용하여 관찰하였다. 체외수정에서 관찰된 바와 동일하게 정자를 난자에 직접주입 한 직후 정자 중편부에서 성상체가 형성되었고, 이 성상체에 의해 자성 웅성 전핵이 융합되는 것으로 관찰되었다. 그러나 난자내 정자를 직접주입하였을 경우 웅성전핵으로 발달하는 비율이 낮았다. 이는 주입된 정자가 원형질막과 perinuclear theca에 싸인 체 난자내로 들어가 난자내의 sperm nucleus decondensing factor와 정자 핵과의 반응이 억제되기 때문으로 생각된다. 정자 두부 만을 주입하였을 경우 성상체가 형성되지 않았지만 자성 웅성 전핵 사이 또는 그 주위에서 두터운 미세소관층이 관찰이 되었다. 따라서 소에 있어서는 정자의 중편부에 위치하여 microtubule organizing center (MTOC)의 역할을 하는 중심립 또는 중심체 없이도 모계에서 유래된 미세소관이 형성되어 이것이 전핵의 융합과 세포분열에 관여하는 것으로 생각된다. 정자의 미부 만을 주입하였을 경우 성상체가 형성이 되지 않았으며, 자성핵 사이에 형성된 미세소관과 떨어져서 관찰되었다. 따라서 주입된 정자의 꼬리는 미세소관형성과 관련이 없는 것으로 생각된다. 이러한 결과는 소에 있어서, 수정 시 정자로부터 유래되는 중심립 또는 중심체가 없이도 미세소관을 형성하여 미세소관에 의해 이후의 배발달이 정상적으로 일어남을 보여주고 있다.

산화제 첨가없이 고온동위원소 교환법으로 오르토 요오도히퓨린산(OIH)을 방사성요오드로 표지하면 효과적이라고 하나 그 교환메카니즘은 불확실하였다. 본 연구에서는 OIH와 $^{125}$/ $I^{-}$ 및 OIH와 $^{125}$/ $I_2$사이의 동위원소 교환반응을 두 다른 온도에서 진행시켜 교환속도 상수와 활성화파라미터들을 방사종이 크로마토그래피법으로 측정함으로써 반응속도론적으로 고찰하였다. OIH표지반응 때의 부산물로 알려진 오르토요오도벤조산(OIB)에 대해서도 같은 방범으로 진행시켰던바 교환속도 상수가 큰 순서는 OIB...$^{125}$/ $I^{-}$>OIB...$^{125}$/ $I_2$>OIH..$^{125}$/ $I^{-}$>OIH...$^{125}$/ $I_2$이었고 OIH의 활성화 파라미터가 일반적으로 OIB의 그것들보다 컸다. ($\Delta$H$\neq$$_{OIH}$>$\Delta$H$\neq$$_{OIB}$, $\Delta$S$\neq$$_{OIH}$>$\Delta$S$\neq$$_{OIB}$). 조건에 따라서는 친전자적 교환메카니즘도 가능하겠으나 위의 결과는 고온 교환반응에서 친핵적 교환메카니즘이 압도적임을 말해주었다. 그와같은 원인은 OIH의 CONHC $H_2$COOH의 오르토치환기 효과에 의한 수소결합형 전이상태 생성이 용이하기 때문이라고 보았다. 따라서 고온 교환법은 환원제가 함유되지 않은 $^{131}$ I가 생산되지 않는 작은 연구기관에서의 Hippuran표지 화합물 제조에 효과적임을 알수 있었다. 알수 있었다.
There seems to be some controversy about the effect of total ginseng saponin (TGS) on the secretion of catecholamines (CA) from the adrenal gland. Therefore, the present study aimed to determine whether TGS can affect the CA release in the perfused model of the adrenal medulla isolated from spontaneously hypertensive rats (SHRs). TGS (15-150 ${\mu}g/mL$), perfused into an adrenal vein for 90 min, inhibited the CA secretory responses evoked by acetylcholine (ACh, 5.32 mM) and high $K^+$ (56 mM, a direct membrane depolarizer) in a dose- and time-dependent fashion. TGS (50 ${\mu}g/mL$) also time-dependently inhibited the CA secretion evoked by 1.1-dimethyl-4 -phenyl piperazinium iodide (DMPP; 100 ${\mu}M$, a selective neuronal nicotinic receptor agonist) and McN-A-343 (100 ${\mu}M$, a selective muscarinic M1 receptor agonist). TGS itself did not affect basal CA secretion (data not shown). Also, in the presence of TGS (50 ${\mu}g/mL$), the secretory responses of CA evoked by veratridine (a selective $Na^+$ channel activator (50 ${\mu}M$), Bay-K-8644 (an L-type dihydropyridine $Ca^{2+}$ channel activator, 10 ${\mu}M$), and cyclopiazonic acid (a cytoplasmic $Ca^{2+}$-ATPase inhibitor, 10 ${\mu}M$) were significantly reduced, respectively. Interestingly, in the simultaneous presence of TGS (50 ${\mu}g/mL$) and N${\omega}$-nitro-L-arginine methyl ester hydrochloride [an inhibitor of nitric oxide (NO) synthase, 30 ${\mu}M$], the inhibitory responses of TGS on the CA secretion evoked by ACh, high $K^+$, DMPP, McN-A-343, Bay-K-8644, cyclopiazonic acid, and veratridine were considerably recovered to the extent of the corresponding control secretion compared with the inhibitory effect of TGS-treatment alone. Practically, the level of NO released from adrenal medulla after the treatment of TGS (150 ${\mu}g/mL$) was greatly elevated compared to the corresponding basal released level. Taken together, these results demonstrate that TGS inhibits the CA secretory responses evoked by stimulation of cholinergic (both muscarinic and nicotinic) receptors as well as by direct membrane-depolarization from the isolated perfused adrenal medulla of the SHRs. It seems that this inhibitory effect of TGS is mediated by inhibiting both the influx of $Ca^{2+}$ and Na+ into the adrenomedullary chromaffin cells and also by suppressing the release of $Ca^{2+}$ from the cytoplasmic calcium store, at least partly through the increased NO production due to the activation of nitric oxide synthase, which is relevant to neuronal nicotinic receptor blockade, without the enhancement effect on the CA release. Based on these effects, it is also thought that there are some species differences in the adrenomedullary CA secretion between the rabbit and SHR.
Background: Oxidative stress-induced cardiomyocytes apoptosis is a key pathological process in ischemic heart disease. Glutathione reductase (GR) reduces glutathione disulfide to glutathione (GSH) to alleviate oxidative stress. Ginsenoside Rb1 (GRb1) prevents the apoptosis of cardiomyocytes; however, the role of GR in this process is unclear. Therefore, the effects of GRb1 on GR were investigated in this study. Methods: The antiapoptotic effects of GRb1 were evaluated in H9C2 cells by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide, annexin V/propidium iodide staining, and Western blotting. The antioxidative effects were measured by a reactive oxygen species assay, and GSH levels and GR activity were examined in the presence and absence of the GR inhibitor 1,3-bis-(2-chloroethyl)-1-nitrosourea. Molecular docking and molecular dynamics simulations were used to investigate the binding of GRb1 to GR. The direct influence of GRb1 on GR was confirmed by recombinant human GR protein. Results: GRb1 pretreatment caused dose-dependent inhibition of tert-butyl hydroperoxide-induced cell apoptosis, at a level comparable to that of the positive control N-acetyl-L-cysteine. The binding energy between GRb1 and GR was positive (-6.426 kcal/mol), and the binding was stable. GRb1 significantl reduced reactive oxygen species production and increased GSH level and GR activity without altering GR protein expression in H9C2 cells. Moreover, GRb1 enhanced the recombinant human GR protein activity in vitro, with a half-maximal effective concentration of ≈2.317 μM. Conversely, 1,3-bis-(2-chloroethyl)-1-nitrosourea co-treatment significantly abolished the GRb1's apoptotic and antioxidative effects of GRb1 in H9C2 cells. Conclusion: GRb1 is a potential natural GR agonist that protects against oxidative stress-induced apoptosis of H9C2 cells.
식물에서 추출한 파이토케미컬은 암세포의 여러 신호전달 기작에 관여함으로써 apoptosis를 유도한다. 본 연구에서는 파이토케미컬의 한 종류인 레스베라트롤을 MCF-7 세포에 처리함으로써 암세포의 증식 억제와 apoptosis 유도 효과를 알아보았고, 이러한 효과가 암세포의 성장과 증식에 관여하는 단백질인 mTOR와 COX-2의 발현 양상에 어떠한 영향을 미치는지 알아보고자 하였다. 그 결과 MCF-7 세포에 레스베라트롤을 처리했을 때 농도가 증가함에 따라 암세포의 생존률이 감소하였고, Hoechst 33342를 이용한 chromatin 염색과 Annexin V-propodium iodide staning을 통하여 암세포의 세포증식 효과가 apoptosis에 의해 유도된 것임을 알 수 있었다. MCF-7 세포에 레스베라트롤을 처리했을 때 mTOR 및 COX-2의 발현 양상을 확인하기 위해 Western blotting을 실시한 결과, 레스베라트롤의 농도가 높아짐에 따라 mTOR 및 COX-2의 발현이 감소함을 확인 하였다. 이와 같은 결과는 MCF-7 유방암 세포에서 레스베라트롤에 의한 암세포의 증식 억제 및 apoptosis 유도가 mTOR 신호경로 저해를 통한 COX-2의 발현을 감소시킴으로써 나타나는 것으로 보인다.
제주조릿대(Sasa quelpaertensis Nakai)는 한라산에 넓게 분포되어 자생하는 식물로 최근 연구에서 항염증, 항당뇨, 항산화, 항암 효능을 가지는 것으로 알려져 있으나 대장암에서의 항암 효능 및 그에 따른 mechanism에 대해서는 명확히 밝혀지지 않았다. 본 연구에서는 인간 대장암 HT-29 세포를 대상으로 제주조릿대에 의한 항암작용과 기전에 대해 조사하였다. 제주조릿대에 의한 HT-29 세포의 증식 억제가 apoptosis 유도와 연관성이 있음을 DNA fragmentation와 flow cytometry 분석에 의한 sub-G1기의 세포빈도의 증가로 확인하였다. 제주조릿대에 의한 apoptosis 유발은 HT-29 세포의 S arrest 현상을 동반하였을 뿐만 아니라 발생한 산화질소의 증가와 anti-apoptotic factor인 IAP family (survivin, XIAP, cIAP-1, cIAP-2) 발현이 감소함으로써 촉진되었음을 확인할 수 있었다. 이러한 결과들은 제주조릿대가 대장암에 대한 치료제로서의 사용 가능성을 확인할 수 있었지만 이를 입증하기 위해서는 더 자세한 항암기전에 관한 연구가 진행되어야 한다고 사료된다.
